Overexpression of a modified AM79 aroA gene in transgenic maize confers high tolerance to glyphosate

It has previously been shown that a bacterial 5-enolpyruvylshikimate-3-phosphate synthase（EPSPS） encoding gene AM79 aroA can be a candidate gene to develop glyphosate-tolerant transgenic crops（Cao et al. 2012）. In this study, AM79 aroA was redesigned using the plant biased codons and eliminating the motifs which would lead to the instability of mRNA, to create a synthetic gene that would be expressed highly in plant cells. The redesigned and artificially synthesized gene, named as mAM79, was cloned into plant expression vector pM3301 Ubi Sp AM79, where mAM79 is fused with signal peptide sequence of pea rib-1,5-bisphospate carboxylase（rbcS） small subunit and controlled by ubiquitin promoter. The plasmid was transformed into maize（Zea mays） immature embryos using Agrobacterium-mediated transformation method. Total 74 regenerated plants were obtained and PCR analysis showed that these transgenic plants had the integration of mAM79. Southern blot analysis was performed on the genomic DNA from four transgenic lines, and the result showed that one or two copies of mAM79 were integrated into maize genome. RT-PCR analysis result indicated that mAM79 was highly transcribed in transgenic maize plants. When sprayed with glyphosate, transgenic maize line AM85 and AM72 could tolerate 4-fold of commercial usage of glyphosate; however, all the non-transgenic maize plants were killed by glyphosate. The results in this study confirmed that mAM79 could be used to develop glyphosate-tolerant maize, and the obtained transgenic maize lines could be used for the breeding of glyphosate-tolerant maize.

The core-mantle differentiation process plays a pivotal role in redistributing material on a massive scale,shaping the long-term evolution of rocky *** this process is crucial for gaining insights into the accretion and evolution of planets like ***,the details of Mars’s core-mantle differentiation remain poorly understood due to limited compositional data for its core and *** this study,we aim to constrain the Martian core-mantle differentiation by examining FeO partitioning between core and mantle materials,incorporating improved Martian compositional data from the InSight *** ab initio thermodynamic techniques,we calculated the FeO partition coefficient between liquid iron and silicate *** results align with previous studies while also clarifying the factors affecting partitioning *** on these findings and estimates of oxygen concentration in the core,we infer that Mars’s core and mantle likely differentiated at temperatures above 2440 K and pressures ranging from 14 to 22 *** these estimates are higher than previously reported,they are consistent with observed abundances of moderately siderophile elements and Mars’s accretion models.

The phenomenon of induced transparency,characterized by a high quality(Q)factor,holds significant importance in the fields of high-precision sensing,energy storage and wave *** transparency systems are often constrained by the limited experimentally accessible parameter space and the lack of independent control over intrinsic parameters,thereby hindering advanced research on high-Q *** we propose a virtualized coupled-oscillator system that enables reconfigurable induced transparency in the reflection *** virtually incorporating a pair of coupled oscillators into an individual acoustic meta-atom,we achieve the manipulation of induced transparency in a decoupled,wide-ranging,and elaborate *** the diverse evolutions across the intrinsic parameter space,we show experimentally a counter-intuitive increase in the Q-factor of the transparency window,which is attributed to the enhanced dissipation of the bright ***,a constant group delay at the center frequency is observed accompanying this *** work demonstrates an intriguing platform as well as a novel strategy to engineer high-Q induced transparency phenomenon,and paves the way towards applications in programmable acoustic sensors,filters and slow-wave devices.

Composition and Content of High-Molecular-Weight Glutenin Subunits and Their Effects on Wheat Quality

Sedimentation values, flour glutenin macropolymer (GMP) contents, composition and contents of high-molecular-weight (HMW) glutenin subunits (GS) of 233 flour samples were determined. Our data indicated that subunit 1 occurred more frequently at Glu-A1 , subunit pair 7 + 8 at Glu-B1 and 2 + 12 at Glu-D1. The significant relationships between Glu-1 quality score and total HMW glutenin content, sedimentation value and GMP content suggested that the composition of HMW-GS affects wheat quality strongly. Moreover, the total content of HMW-GS was correlated with certain quality parameters more significantly. Relationship between subunit 5 + 10 content and breadmaking quality was better than others, but 2 + 12, 7 + 8, 7 + 9 and 4 + 12 also correlated with certain quality parameters significantly. The contents of total HMW-glutenin, x-type subunits and y-type subunits related with sedimentation value, flour GMP content, and Glu-1 quality score more strongly than that of individual subunit or subunit pair. The flour GMP content, with excellent correlation to sedimentation value, total contents of HMW glutenin, x- and y-type subunits and many other quality parameters, could be an ideal indicator of breadmaking quality at earlier generations for breeding purpose for its simple procedure and small scale.
