Protective role of adiponectin in a rat model of intestinal ischemia reperfusion injury

AIM: To determine the potential protective role of adiponectin in intestinal ischemia reperfusion(I/R) ***: A rat model of intestinal I/R injury was established. The serum level of adiponectin in rats with intestinal I/R injury was determined by enzymelinked immunosorbent assay(ELISA). The serum levels of interleukin(IL)-1β, IL-6, and tumor necrosis factor(TNF)-α were also measured by ELISA. Apoptosis of intestinal cells was detected using the terminal deoxynucleotidyl transferase d UTP nick end labeling assay. The production of malondialdehyde(MDA) and superoxide dismutase(SOD) and villous injury scores were also ***: Adiponectin was downregulated in the serum of rats with intestinal I/R injury compared with sham rats. No significant changes in the expression of adiponectin receptor 1 and adiponectin receptor 2 were found between sham and I/R rats. Pre-treatment with recombinant adiponectin attenuated intestinal I/R injury. The production of pro-inflammatory cytokines,including IL-6, IL-1β, and TNF-α, in rats with intestinal I/R injury was reduced by adiponectin pre-treatment. The production of MDA was inhibited, and the release of SOD was restored by adiponectin pre-treatment in rats with intestinal I/R injury. Adiponectin pre-treatment also inhibited cell apoptosis in these rats. Treatment with the AMP-activated protein kinase(AMPK) signaling pathway inhibitor, compound C, or the heme oxygenase 1(HO-1) inhibitor, Snpp, attenuated the protective effects of adiponectin against intestinal I/R injury. CONCLUSION: Adiponectin exhibits protective effects against intestinal I/R injury, which may involve the AMPK/HO-1 pathway.

Groundwater leakage in shield tunnels poses a threat to the safety and durability of tunnel structures. Disturbance of adjacent constructions during the operation of shield tunnels frequently occurs in China, leading to deformation of tunnel lining and leakage in joints. Understanding the impact of adjacent constructions on the waterproofing performance of the lining is critical for the protection of shield tunnels. In this study, the weakening behavior of waterproof performance was investigated in the joints of shield tunnels under transverse deformation induced by adjacent construction. First, the relationship between the joint opening and transverse deformation under three typical adjacent constructions (upper loading, upper excavation, and side excavation) was investigated via elaborate numerical simulations. Subsequently, the evolution of the waterproof performance of a common gasket with a joint opening was examined by establishing a coupled Eulerian-Lagrangian model of joint seepage, and a formula describing the relationship between waterproof performance and joint opening was proposed. Finally, the weakening law of waterproofing performance was investigated based on the results of the aforementioned studies. It was determined that the joints with the greatest decline in waterproof performance were located at the tunnel shoulder in the upper loading case, tunnel crown in the upper excavation case, and tunnel shoulder in the side excavation case. When the waterproof performance of these joints decreased to 50% and 30%, the transverse deformations were 60 and 90 mm under upper loading, 90 and 140 mm under upper excavation, and 45 and 70 mm under side excavation, respectively. The results provide a straightforward reference for setting a controlled deformation standard considering the waterproof performance.

The Qinghai-Tibet Plateau(QTP)possesses the largest areas of permafrost in the midand low latitude regions on the earth and many large lakes in the permafrost *** on a comprehensive investigation around certain typical lakes,this study found that although the presence of lakes formed different ranges of unfrozen zones in permafrost,the heating effect of lake water on surrounding permafrost is limited to a small *** temperature of permafrost around the lake is closely related to the distance to the lake and the ice content of the *** lakes are ice-rich permafrost zones and permafrost temperature in this area is significantly lower than that far away from the lake,which indicates that the existence of lakes in the QTP has special effect on the permafrost *** on the monitoring results,this study presents the typical distribution pattern of the permafrost around large lakes and discusses the reasons for the distribution *** to the huge area of lakes and the significant impact of lakes on permafrost distribution,it is suggested to re-estimate the total permafrost area and underground ice storage in the QTP.

Evaluation of Four Candidate VNTR Loci for Genotyping 225 Chinese Clinical Mycobacterium Tuberculosis Complex Strains

Objective To evaluate four candidate variable number tandem repeat （VNTR） loci for genotyping Mycobacterium tuberculosis complex strains. Methods Genomic sequences for two M. tuberculosis strains （CCDC5079 and CCDC5180） were generated, and using published sequence data, four candidate VNTR loci were identified. The VNTRs were used to genotype 225 Chinese clinical M. tuberculosis complex strains. The discriminatory power of the VNTRs was evaluated using BioNumerics 5.0 software. Results The Hunter-Gaston Index （HGI） for BJ1, BJ2, BJ3, and BJ4 loci was 0.634, 0.917, 0.697, and 0.910, respectively. Combining all four loci gave an HGI value of 0.995, thus confirming that the genotyping had good discriminatory power. The HGI values for BJ1, BJ2, BJ3, and BJ4, obtained from Beijing family strain genotyping, were 0.447, 0.878, 0.315, and 0.850, respectively. Combining all four loci produced an HGI value of 0.988 for genotyping the Beijing family strains. We observed unique patterns for M. boris and M. africanum strains from the four loci. Conclusion We have shown that the four VNTR loci can be successfully used for genotyping M. tuberculosis complex strains. Notably, these new loci may provide additional information about Chinese M. tuberculosis isolates than that currently afforded by established VNTR loci typing.

AIM:To investigate the effects of rosiglitazone(RGZ) on expression of interleukin-18(IL-18) and caspase-1 in liver of non-alcoholic fatty liver disease(NAFLD) ***:Twenty-eight Sprague-Dawley(SD) rats were randomly divided into control,NAFLD,and RGZ treated NAFLD groups.A NAFLD rat model of NAFLD was established by feeding the animals with a high-fat diet for 12 *** NAFLD animals were treated with RGZ or vehicle for the last 4 wk(week 9-12) and then sacrificed to obtain liver *** changes were analyzed with HE,oil red O and Masson's trichrome *** of IL-18 and caspase-1 were detected using immunohistochemical staining and semi-quantitative reverse-transcription polymerase chain reaction(RT-PCR) ***:The expression levels of both IL-18 and caspase-1 were higher in the liver of NAFLD group than in the control ***,inflammation and fibrosis,found in the liver of NAFLD rats,were significantly improved 4 wk after RGZ *** elevated hepatic IL-18 and caspase-1 expressions in NAFLD group were also significantly attenuated after RGZ ***:RGZ treatment can ameliorate increased hepatic IL-18 production and histological changes in liver of NAFLD *** beneficial effects of RGZ on NAFLD may be partly due to its inhibitory effect on hepatic IL-18 production.

The structural evolution of Cu_（45）Zr_（45）Ag_（10） metallic glass was investigated by in situ transmission electron microscopy heating experiments. The relationship between phase separation and crystallization was elucidated. Nucleation and growth-controlled nanoscale phase separation at early stage were seen to impede nanocrystallization, while a coarser phase separation via aggregation of Ag-rich nanospheres was found to promote the precipitation of Cu-rich *** of composition and dynamics heterogeneities was supposed to play a key role during phase separation preceding crystallization.

Alcoholic liver disease(ALD) has become one of the leading causes of death in the world. Berbamine(BM), a natural product mainly derived from Berberis vulgaris L, possesses multiple bioactivities as a traditional medicine. However, the protective effect of BM on ALD remains unknown. In this study, we investigated the effect of BM on ethanol-induced hepatic injury in mice and its underlying mechanism. It was shown that BM at 0.3125-40 μmol·L^-1 had no effect on macrophages and hepatocytes proliferation. BM at 5-20 μmol·L^-1 significantly inhibited lipopolysaccharide(LPS) or acetate-induced IL-1β and IL-6 m RNA expression in RAW264.7 cells. Moreover, BM treatment significantly inhibited LPS-induced p65 and STAT3 phosphorylation in RAW264.7 cells. Hepatic histopathology analysis showed that inflammatory cells infiltration and lipid accumulation were suppressed by 25 and 50 mg·kg^-1 BM administration in ethanol-induced hepatic injury mouse model. Meanwhile, BM treatment significantly inhibited serum ALT and AST levels in ethanol-fed mice. Oil red O staining results showed that BM administration ameliorated hepatic lipid accumulation in ethanolfed mice. Preventions of ethanol-induced hepatic injury by BM were reflected by markedly decreased serum and hepatic triglyceride(TG) and total cholesterol(TC) contents. Real-time PCR results showed that BM treatment significantly inhibited pro-inflammatory cytokines m RNA expression in ethanol-fed mouse liver. Remarkably, the mechanism of action of BM was related to the reduction of ethanol-induced NF-κB and STAT3 phosphorylation levels in liver. In addition, BM treatment significantly inhibited ERK phosphorylation but not JNK and p38 of MAPK pathway. Taken together, our results demonstrate a beneficial effect of BM on ethanol-induced liver injury via a mechanism associated with inactivation of NF-κB, STAT3 and ERK pathway, which gives insight into the further evaluation of the therapeutic potential of BM for ALD.
