Transcriptome profile and its partly verification of human hepatocellular carcinoma cells exposed to Yuzhizi(Fructus Akebiae Quinatae) seed extract

OBJECTIVE: To investigate the efficacy of Yuzhizi(Fructus Akebiae Quinatae) seed extract(FAQSE) on inhibiting the proliferation of hepatocellular carcinoma(HCC) cells in vitro and to explore the anti-HCC action mechanism of FAQSE. METHODS: Human HCC Hep G2 and Huh7 cells were used to investigate the anti-HCC effect of FAQSE in vitro. 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) method was used to measure cell viability. Affymetrix microarray was adopted to detect the expression of transcriptome. The differentially expressed genes(DEGs) of each cell line were identified. For coDEGs of both cell lines, the gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway were enriched using the Database for Annotation, Visualization and Integrated Discovery(DAVID), and the network analysis of protein-protein interaction(PPI) was mapped using the Retrieval of Interacting Genes/Proteins(STRING) and Cytoscape software. Some important genes in the PPI network of coDEGs were selected to verify by quantitative real-time reverse transcription-polymerase chain reaction, Western blot and enzyme-linked immunosorbent assay. RESULTS: FAQSE decreased the viability of Hep G2 and Huh7 cells. There were 211 co-upregulated and 86 codownregualted genes in both cell lines after FAQSE treatment. The enriched GO terms of co-upregulated DEGs were primarily involved cell-cell adhesion, viral process, transcription initiation from RNA polymerase II promoter, positive regulation of transcription from RNA polymerase II promoter and actin cytoskeleton organization. The GO terms of co-downregulated DEGs were mainly enriched in the processes of SRP-dependent cotranslational protein targeting to membrane, viral transcription, nuclear-transcribed m RNA catabolic process, nonsense-mediated decay, translational initiation and r RNA processing. Grey-based approach for estimating software reliability under nonhomogeneous Poisson process

Due to the randomness and time dependence of the factors affecting software reliability, most software reliability models are treated as stochastic processes, and the non-homogeneous Poisson process(NHPP) is the most used ***, the failure behavior of software does not follow the NHPP in a statistically rigorous manner, and the pure random method might be not enough to describe the software failure behavior. To solve these problems, this paper proposes a new integrated approach that combines stochastic process and grey system theory to describe the failure behavior of software. A grey NHPP software reliability model is put forward in a discrete form, and a grey-based approach for estimating software reliability under the NHPP is proposed as a nonlinear multi-objective programming problem. Finally, four grey NHPP software reliability models are applied to four real datasets, the dynamic R-square and predictive relative error are calculated. Comparing with the original single NHPP software reliability model, it is found that the modeling using the integrated approach has a higher prediction accuracy of software reliability. Therefore, there is the characteristics of grey uncertain information in the NHPP software reliability models, and exploiting the latent grey uncertain information might lead to more accurate software reliability estimation.

Refining mechanism of yttrium on primary α-Al in semi-solid Al alloy

Semi-solid Al7Si0.3Mg alloy slurry, which is grain-refined by yttrium, is manufactured by low temperature pouring. The effects of grain-refined by yttrium on the morphology and the grain size of the primary α-Al in semi-solid Al7Si0.3Mg alloy are researched. The results indicate that semi-solid Al7Si0.3Mg alloy with particle-like and rosette-like primary α-Al can be prepared by low temperature pouring from liquid Al7Si0.3Mg alloy grain-refined by yttrium. The grain size and particle morphology of primary α-Al in Al7Si0.3Mg alloy are markedly improved by 0.5 wt pet Y. The refining mechanism of Y on the morphology and the grain size of the primary α-Al in semi-solid Al7Si0.3Mg alloy are delved.

Research on fractal characteristics of primary phase morphology in semi-solid A356 alloy

Semi-solid slurry of A356 alloy was prepared by low superheat pouring and slightly electromagnetic stirring, and the fractal characteristics of morphology in semi-solid primary phase was researched. The fractal dimensions of primary phase morphology in the semi-solid A356 alloy were calculated by the program written to calculate the fractal dimensions of box-counting in the image of solid phase morphology in semi-solid metal slurry. The results indicated that the morphology of primary phase in semi-solid A356 prepared by low superheat pouring and slightly electromagnetic stirring is characterized by fractal dimension, and the primary phase morphology obtained by the different processing parameters has different fractal dimensions. The morphology of primary phase at the different position of ingot has different fractal dimensions, which reflected the effect of solidified conditions at different positions in the same ingot on the morphology of semi-solid primary phase. Solidification of semi-solid alloy is a course of change in fractal dimension.

In this paper,an optimal switch-time control problem is solved for a class of impulsive switched autonomous *** considered systems jump at the switching times,and the sequence of active subsystems is *** control variables consist of the impulse times and a set of scalars which determine the jump ***,the subsystems do not require a refractory period,which can bring more *** the calculus of variation,the partial derivatives of the cost with respect to the control variables are derived,based on which the optimality conditions are ***,the obtained formulas can be used in some gradient descent algorithms to locate the optimal control ***,the viability of the proposed method is illustrated through two numerical examples.

CAVITATION INCEPTION OF GATE SLOTS

Starting with field investigation this paper presented a comprehensive review of the hydraulic design and performance of gate slots in 85 existing projects covering 158 gates of various dimension and operating head. Systematic model tests were conducted in a water tunnel to optimize the configuration of gate slots. Main parameters affecting cavitation inception were studied, such as the ratio of width to depth, relative offset, the slope of side wall, as well as the radius of curvature of slot's corner. The scale effects of cavitation modelling were preliminarily verified. The conformity between model studies and prototype inspections is quite agreeable. Conclusion and suggestion were made to meet the demand of hydraulic design of gate slots.

Experimental and numerical investigations on fatigue behavior of aluminum alloy 7050-T7451 single lap four-bolted joints

The fatigue behavior of aluminum alloy 7050-T7451 single lap four-bolted joints was studied by high- frequency fatigue test and finite element （FE） methods. The fatigue test results showed that a better enhancement of fatigue life was achieved for the joints with highlocked bolts by employing the combinations of cold expansion, interference fit, and clamping force. The fractography revealed that fatigue cracks propagated tortuously; more fatigue micro-cliffs, tearing ridges, lamellar structure were observed, and fatigue striation spacing was simultaneously reduced. The evaluation of residual stress conducted by FE methods confirmed the experimental results and locations of fatigue crack initiation. The extension of fatigue lives can be attributed to the evolution of fatigue damage and effect of beneficial compressive residual stresses around the hole, resulting in the delay of crack initiation, crack deflection, and plasticityinduced crack closure.2017 Published by Elsevier Ltd on behalf of The editorial office of Journal of Materials Science ＆ Technology.

Semi-solid A356 alloy slurry for rheocasting prepared by a new process

To obtain the semi-solid slurry with uniform and fine structure morphology that satisfies the requirement of rheocasting process, a new process for preparing semi-solid AI alloy slurry was developed, in which local chilling was combined with low superheat pouring and slight electromagnetic stirring （LSPSEMS）. The morphology and the size of primary a-AI in the A356 alloy slurry prepared with the new process, i.e., LSPSEMS with local chilling, were investigated using MIAPS image analyzing software, and the grain refinement mechanism was discussed. The results indicate that the semi-solid primary phase consists of particle-like or globular-like a-AI, and the morphology and grain size of primary a-AI in the slurry can be markedly improved by the new process. The fine primary a-AI distributes uniformly in the slurry, which satisfies the requirement of rheocasting. Compared with the alloy prepared by LSPSEMS, the average equal-area-circle grain diameter of primary a-AI in semi-solid A356 alloy ingot prepared by the new process is decreased from 85.6μm to 68.8 μm at the central area, 112.6 μm to 77.6 μm at the transition area and is 84.7 μm in the edge area, respectively. The corresponding shape factor of primary a-AI is increased from 0.78 to 0.83, 0.54 to 0.77 and 0.28 to 0.59, respectively. In addition, the pouring temperature could be suitably raised from 620-630℃ of the traditional process to 650℃ using this technique, which is convenient for practical operation. The mechanism of grain refinement, in the new process, is that the local chilling quickens up the temperature decrease in the center of the melt. The nuclei could not grow up in a short time so the finer grains are formed in the melt.

Biochar has been widely applied for the remediation of petroleum-contaminated ***,the effect of biochar on the transport of petroleum degradation bacteria has not been studied.A typical Gram-positive petroleum degradation bacteria-Corynebacterium variabile HRJ4 was used to study the effect of different biochars on bacterial transport and *** indicated that the addition of biochar in sand was effective for reducing the transport of bacteria and poplar sawdust biochar(PSBC)had a stronger hinder effect than corn straw biochar(CSBC).The hindrance was more evident with pyrolysis temperature of biochar raised from 300℃ to 600℃,which was attributed to the increase of specific surface area(309 times).The hindrance effect also enhanced with higher application rate of ***,the reduction of HRJ4 transport was more obvious in higher(25 mmol/L)concentration of Na Cl solution owing to electrostatic attraction *** adsorption of biochar to HRJ4 was defined to contribute to the hindrance of HRJ4 transport *** the influence of feedstocks and pyrolysis temperature on HRJ4 transport,it suggested that specific surface area had the greatest effect on HRJ4 transport,and pore-filling,electrostatic force also contributed to HRJ4 retained in quartz sand *** last,phenol transportation experiment indicated that the restriction of biochar on HRJ4 enhanced the phenol removal rate in the *** study provides a theoretical basis for the interaction of biochar and bacteria,which is vital for the remediation of oil-contaminated soil and groundwater in the field.

The development of MOFs-based nanomaterials in heterogeneous organocatalysis

Metal-organic framework(MOF) is a class of inorganic-organic hybrid material assembled periodically with metal ions and organic ligands. MOFs have always been the focuses in a variety of frontier fields owing to the advantageous properties, such as large BET surface areas, tunable porosity and easyfunctionalized surface structure. Among the various application areas, catalysis is one of the earliest application fields of MOFs-based materials and is one of the fastest-growing topics. In this review, the main roles of MOFs in heterogeneous organocatalysis have been systematically summarized, including used as support materials(or hosts), independent catalysts, and sacrificial templates. Moreover, the application prospects of MOFs in photocatalysis and electrocatalysis frontiers were also ***, the key issues that should be conquered in future were briefly sketched in the final parts of each item. We hope our perspectives could be beneficial for the readers to better understand these topics and issues, and could also provide a direction for the future exploration of some novel types of MOFs-based nanocatalysts with stable structures and functions for heterogeneous catalysis.
