To the Editor:Recurrent pregnancy loss(RPL)is defined as the failure of two or more clinically recognized pregnancies.[1]Both parental and embryonic/fetal factors are associated with RPL.Parental factors include balanced chromosome rearrangements,maternal antiphospholipid syndrome,uterine anomalies,and hormonal or metabolic disorders.[2]Of the examined products of conception(POC),approximately 60%of early pregnancy losses result from sporadic chromosomal abnormalities in embryos,specifically numeric chromosome errors.However,up to 50%of RPL cases remain unexplainable by known causes.Here,we report a couple who have experienced four consecutive clinical pregnancy losses within 10 weeks of gestation,and describe a novel(elongation factor Tu GTP-binding domain containing 2[EFTUD2],Online Mendelian Inheritance in Man[MIM]#603892)nonsense mutation(c.1012G>T,p.E338∗)found in embryonic tissues from each of the last three miscarriages by whole exome sequencing(WES)analysis.In addition,we generated a zebrafish line with a mutation homologous to the human EFTUD2 disruption to investigate the relationship between this mutation and RPL.We hypothesized that the mutation was associated with embryonic lethality,which may induce RPL in patients.The study was approved by the Medical Ethics Committee of the West China Second University Hospital,Sichuan University,China(No.2016[029]).Written informed consent was obtained from the couple before genetic testing.

Microneedles are considered to be an effective,convenient,non-invasive,biosafety and compliant medical technology for vaccinations,biomarker testing,medical aesthetics and other related fields.Nonetheless,further clinical and commercial translation of regular microneedles is hampered by challenges in manufacturability,cost variability,insufficient comfort,contamination and so on.Recent innovations in functional biomaterials and chemical engineering technologies have been applied to develop extensible and swellable hydrogel-forming microneedles,achieving precise and controlled drug delivery and localized sampling from the target tissues.In this review,we systematically summarize the latest development of the extensible and swellable hydrogel-forming microneedles,including deep point-of-care testing,drug deployment,wound healing and mucoadhesion improvement.In addition,further analysis of the challenges and prospects for clinical application of current strategies is well presented.It is believed that the combined efforts of engineering,material,pharmaceutical and clinical research will contribute to the future success of this clinical and commercial translation.

A turbot(Scophthalmus maximus)cell line named SMSP was obtained from the spleen.The origin of the cells was identified by morphology,chromosome number and COI gene.The optimal basic medium,serum concentration and growth temperature of the cells were detected.SMSP cell line is mainly composed of fibroblast-like cells.Most of the SMSP cells contained 44 chromosomes,and the sequence of COI gene confirmed that the cells were originated from turbot.The optimal culture conditions were 24℃,DMEM+10%FBS.The cell line had high transfection efficiency for siRNA and plasmid.After stimulation with lipopolysaccharide(LPS)or poly(I:C),the expressions of immune-related genes such as TNF-β,IL-12s,IL-10 and IL-1βwere up-regulated significantly in the early stage(P<0.05).This study will provide a model for exploring immune mechanism of turbot against pathogen in vitro.

The drug resistance of chemotherapy is a major challenge to overcome for antineoplastic agents and the reverse of drug resistant is essential for cancer therapy. Herein, we developed a drug delivery system which can simultaneously detect/reverse the drug resistance and perform synergetic treatment of cancer. In this work, we integrated cyanine5(Cy5) modified mi RNA(let-7 i)(Cy5-mi RNA) and platinum onto nano-graphene oxide(NGO)(30-50 nm) platform to achieve simultaneously detection/reversion of drug resistance and synergetic treatment of cisplatin resistant SKOV3 cells(SKOV3 DDP cells). The Cy5-mi RNA adsorbed on NGO could selectively bind the drug resistance related m RNA follow by suppress the expression of drug resistance m RNA, and the binding simultaneously induced the release of Cy5-mi RNA from the NGO, thus the fluorescence signal of Cy5 recovered and could be used for drug resistance monitoring.Moreover, the mi RNA suppressed the Cyclin D1 protein expressions thus reversed the drug resistance. The loaded platinum(Ⅳ)(Pt(Ⅳ)) was converted to the therapeutic platinum(Ⅱ)(Pt(Ⅱ)) in both tumor acidic and reductive environment responsive behavior. NGO furtherly performed photothermal therapy under near infrared(NIR) laser irradiation and enhanced the therapeutic effect. All in all, this nanoplatform realized detection/reversion of the drug resistance as well as synergetic chemical-photothermal treatment of ovarian cancer cells, which holds great promise in the treatment of drug resistant cancer cells.

Online monitoring of the curing temperature field is essential to improving the quality and efficiency of the manufacturing process of composite parts.Traditional embedded sensor-based technologies have difficulty monitoring the full temperature field or have to introduce heterogeneous items that could have an undesired impact on the part.In this paper,a non-contact,full-field monitoring method based on deep learning that predicts the internal temperature field of composite parts in real time using surface temperature measurements of auxiliary materials is proposed.Using the proposed method,an average temperature monitoring accuracy of 97%is achieved in various heating patterns.In addition,this method also demonstrates satisfying feasibility when a stronger thermal barrier covers the part.This method was experimentally validated during the self-resistance electric heating process,in which the monitoring accuracy reached 93.1%.This method can potentially be applied to automated manufacturing and process control in the composites industry.

Text similarity measurements are the basis for measuring the degree of matching between two or more texts. Traditional large-scale similarity detection methods based on a digital fingerprint have the advantage of high detection speed, which are only suitable for accurate detection. We propose a method of Chinese text similarity measurement based on feature phrase semantics.Natural language processing(NLP) technology is used to pre-process text and extract the keywords by the Term frequency-Inverse document frequency(TF-IDF) model and further screen out the feature words. We get the exact meaning of a word and semantic similarities between words and a How Net semantic dictionary. We substitute concepts to get the feature phrases and generate a semantic fingerprint and calculate similarity. The experimental results indicate that the method proposed is superior in similarity detection in terms of its accuracy rate,recall rate, and F-value to the traditional and digital fingerprinting method.

Dysregulation of microRNAs(miRNAs)in adipocytes plays a critical role in the pathogenesis of obesity.However,the signaling mechanisms regulating miRNAs production in adipose tissue remain largely unclear.Here,we show that adipose tissue-specific knockout of Ras homolog enriched in brain(Rheb),a direct upstream activator of mTOR,increases miR-182-5p level in mouse subcutaneous white adipose tissues.Interestingly,the inhibition of mTOR signaling by rapamycin has no effect on miR-182-5p level in primary subcutaneous white adipocytes,suggesting the presence of a mTOR-independent mechanism regulating Rheb-mediated miR-182-5p expression.Consistent with this view,Rheb-ablation activates the cAMP/PPARγsignaling pathway.In addition,treatment of white adipocytes with pioglitazone,a PPARγagonist,dramatically upregulates miR-182-5p levels.Our study reveals a unique mechanism by which Rheb regulates miR-182-5p in adipocytes.Given that increasing miR-182-5p in adipose tissue promotes beige fat development,our study also suggests a unique mechanism by which Rheb promotes thermogenesis and energy expenditure.

Perovskite-based tandem solar cells have attracted increasing interest because of its great potential to surpass the Shockley-Queisser limit set for single-junction solar cells.In the tandem architectures,the wide-bandgap(WBG)perovskites act as the front absorber to offer higher open-circuit voltage(VOC)for reduced thermalization losses.Taking advantage of tunable bandgap of the perovskite materials,the WBG perovskites can be easily obtained by substituting halide iodine with bromine,and substituting organic ions FA and MA with Cs.To date,the most concerned issues for the WBG perovskite solar cells(PSCs)are huge VOC deficit and severe photo-induced phase separation.Reducing VOC loss and improving photostability of the WBG PSCs are crucial for further efficiency breakthrough.Recently,scientists have made great efforts to overcome these key issues with tremendous progresses.In this review,we first summarize the recent progress of WBG perovskites from the aspects of compositions,additives,charge transport layers,interfaces and preparation methods.The key factors affecting efficiency and stability are then carefully discussed,which would provide decent guidance to develop highly efficient and stable WBG PSCs for tandem application.

The printing quality of components manufactured using laser powder bed fusion(LPBF)generally depends on the presence of various defects such as massive porosity.Thus,the efficient elimination of pores is an important factor in the production of a sound LPBF product.In this study,the efficacy of two in situ laser remelting approaches to eliminating pores during the LPBF of a titanium alloy Ti–6.5 Al–3.5 Mo–l.5 Zr–0.3 Si(TC11)was assessed both experimentally and computationally.These two remelting methods are surface remelting and layer-by-layer printing and remelting.A multi-track multi-layer phenomenological model was established to analyze the variation of pores with the temperature and velocity fields.The results showed that surface remelting with a high laser power,such as 180 W,can eff ectively eliminate pores within three deposited layers.However,such remelting could not reach defects in deeper regions.Layer-by-layer remelting with a 180 W laser could eff ectively eliminate the pores formed in the previous layer in real time.The results obtained in this study can provide useful guidance for the in situ control of printing defects supported by real-time monitoring,feedback,and operating systems of an intelligent LPBF equipment.
