Apple stem grooving virus is associated with leaf yellow mottle mosaic disease on Citrus grandis cv. Huangjinmiyou in China

Although it is usually latent on citrus, apple, and pear, apple stem grooving virus(ASGV) poses a great risk to many sensitive cultivars. Since severe leaf yellow mottle mosaic(LYMM) symptoms have been observed on Huangjinmiyou(HJY) pummelos(Citrus grandis cv. Huangjinmiyou), a commercial variety that is widely cultivated in South China, high throughput sequencing(HTS) was used to find potential pathogens and only three divergent ASGV variants were identified. The three ASGV variants shared 81.03–82.34% genome-wide pairwise identities with each other, and were separately closest to other ASGV variants from different hosts and/or geographical regions, as indicated by viral phylogenies. However, these new variants may have developed from viral interstrain interactions, based on the results of recombination analysis. A large-scale survey using reverse transcription-PCR(RT-PCR) protocols designed for the three ASGV variants revealed a high incidence(92.7–100%) of ASGV in symptomatic HJY trees from 11 major citrusproducing regions in China. None of ASGV were detected in asymptomatic trees. Temperature treatments applied to the symptomatic HJY plants showed that ASGV is sensitive to high temperatures(30–35°C), at which not only the plants recovered, but also the viruses were not detected by RT-PCR, while at low temperatures(20–24°C), both the symptoms and viruses remained detectable. These data show that ASGV is associated with the LYMM disease prevalent on HJY in China, and this is the significant basis especially of taking appropriate measures timely to manage the disease.

Complete genome sequence analysis of two Citrus tatter leaf virus(CTLV) isolates from China

In order to understand molecular characterization of Citrus tatter leaf virus（CTLV） isolated from China,full-length cDNAs of CTLV-MTH and CTLV-XHC from Citrus reticulata and Citrus sinensis were cloned and sequenced based on whole-genome amplification by RT-PCR.The complete nucleotide sequences of CTLV-MTH and CTLV-XHC were determined to be 6497 nucleotides in length and shared 79.9-91.0%and 78.8-98.0%nucleotide sequence identity,respectively,with other Apple stem grooving virus（ASGV） or CTLV strains available in GenBank.Unexpectedly,CTLV-MTH showed the highest nucleotide sequence identity（91%） with an apple isolate of ASGV,followed by 86.5%with ASGV-HH and 85.7%with ASGV-CHN.Furthermore,CTLV-MTH and three ASGV strains were grouped to a separate cluster in the phylogenetic tree,suggesting it has a closer relationship to ASGV than to CTLV.Therefore,it can be concluded roughly that CTLV may be not a distinct strains of ASGV.We proposed that Citrus tatter leaf virus should be renamed Apple stem grooving virus.

前数字时代的传统侦查监督范式具有明显的被动性与事后性,导致检察机关侦查监督工作流于形式而质效不高。随着数字时代的到来,检察机关以数字正义为价值追求、以调查核实权为依托、以能动检察为理念支撑,借助数字技术来构建大数据侦查监督这一全新范式,实现了以技术集中促进侦查监督实质化、以类案监督促进社会治理、以主动全程侦查监督促进权利保障。值得注意的是,大数据侦查监督范式虽有万般利好,却仍存在立案监督权不完整、司法公正面临挑战及降低侦查效率等问题,未来需要进一步通过充分延伸立案监督范围、有限度地进行数字公开和适度监督等措施予以完善,以此平衡全新范式与原有司法秩序之间的冲突。

Decreasing detection frequency of MITE(MCLas-A)in the population of‘Candidatus Liberibacter asiaticus'recently collected in southern China

An active miniature inverted-repeat transposable element(MITE),MCL as-A,was previously identified from'Candidatus Liberibacter asiaticus'known to be associated with citrus Huanglongbing(HL B,yellow shoot disease).To explore the recent transposition status of MCLas-A,389'Ca.L.asiaticus'strains collected from nine regions in China were amplifed using a specific primer set and three representative'Ca.L.asiaticus'strains were analyzed by next-generation sequencing(NGS)approach.PCR and genomic analysis showed that the entire MCLas A was only present in 1.80%(7/389)and the jumping-out type of the MITE was predominant(8123%)in samples tested,suggesting high frequency transposition occurred in'Ca.L.asiaticus'strains recently collected from China.Biological roles of transposition of the active MITE remain to be determined.

AIM:To directly quantify peroxynitrite(ONOO-)using a highly sensitive fluorescence resonance energy transfer probe RN-NA,investigate the association between ONOOand primary open angle glaucoma(POAG),and clarify whether RN-NA could be used as a potential tool for POAG diagnosis.METHODS:Plasma and aqueous humor(AH)samples were collected from POAG patients(n=100,age:59.70±6.87y)and age-related cataract(ARC)patients(n=100,age:61.15±4.60y)admitted to our hospital.Next,RN-NA was used to detect ONOO-in plasma and AH samples,and the relationship between ONOO-level and POAG was analyzed using binary logistic regression.Besides,Pearson correlation analysis was applied to characterize the correlation of the levels of ONOO-with the patients’age,intraocular pressure(IOP),and mean deviation of visual field testing.The ONOO-scavenger MnTMPyP was employed to treat the 3-morpholinosyndnomine(SIN-1)-induced ocular hypertension in mice(n=7,6-8wk).Finally,the IOP and ONOO-in both eyes were measured 30min after the last drug treatment.RESULTS:ONOO-levels of AH and plasma were significantly higher in the POAG group than in the ARC group(Pker of POAG,especially advanced POAG.Besides,anti-nitration compounds may be novel ocular hypotensive agents based on the animal study.

In this editorial,we comment on an article by Ruan et al published in a recent issue of the World Journal of Clinical Case.Pulmonary meningothelial proliferative lesions,including primary pulmonary meningiomas,minute pulmonary meningothelial-like nodules,and metastatic pulmonary meningiomas are rare pulmonary lesions.These lesions are difficult to differentiate from lung cancers based on clinical and imaging manifestations.Herein,we briefly introduce the clinical,imaging,and pathological characteristics of these lesions and discuss their pathogenesis to strengthen the current understanding of pulmonary meningothelial proliferative lesions in clinical diagnosis and therapy.

Sensing properties and mechanisms of LaF3–Co3O4nanorods for low-concentration methanol detection

Methanol is one of the characteristic gases that distinguish between healthy individuals and lung cancer（LC） lesions in exhaled human breath.Its concentration in exhaled breath is generally below one part per million(1×10-6).The second-phase composite is widely regarded as one of the methods to improve the gas-sensing performance of metal oxide semiconductor （MOS） materials.In this study,LaF3DCo3O4was synthesized by a simple hydrothermal method to enhance its low-concentration methanol gas-sensing performance.5 at%LaF3DCo3O4nanorods exhibited excellent methanol detection performance,including a wider linear detection concentration range (0.2×10-6D5×10-6),a response value exceeding 4.0for 1×10-6methanol at 275°C and 75%relative humidity（RH）,long-term stability （maximum deviation within 15%over 2 weeks）,and excellent selectivity.The mechanism of performance enhancement was studied using various techniques,and density functional theory.The specia spinel structure of Co3O4,the high ionic migration of F-in LaF3,the larger specific surface area of 5 at%LaF3DCo3O4nanorods,and the generated crystal defects all explain the excellent methanol gas-sensing property.This work provides a novel route to prepare MOS composite materials for low-concentration methanol gas detection.

Bayesian Filtering for High-Dimensional State-Space Models With State Partition and Error Compensation

In the era of exponential growth of data availability,the architecture of systems has a trend toward high dimensionality,and directly exploiting holistic information for state inference is not always computationally affordable.This paper proposes a novel Bayesian filtering algorithm that considers algorithmic computational cost and estimation accuracy for high-dimensional linear systems.The high-dimensional state vector is divided into several blocks to save computation resources by avoiding the calculation of error covariance with immense dimensions.After that,two sequential states are estimated simultaneously by introducing an auxiliary variable in the new probability space,mitigating the performance degradation caused by state segmentation.Moreover,the computational cost and error covariance of the proposed algorithm are analyzed analytically to show its distinct features compared with several existing methods.Simulation results illustrate that the proposed Bayesian filtering can maintain a higher estimation accuracy with reasonable computational cost when applied to high-dimensional linear systems.

NCAD v1.0: a database for non-coding variant annotation and interpretation

The application of whole genome sequencing is expanding in clinical diagnostics across various genetic disorders, and the significance of non-coding variants in penetrant diseases is increasingly being demonstrated. Therefore, it is urgent to improve the diagnostic yield by exploring the pathogenic mechanisms of variants in non-coding regions. However, the interpretation of non-coding variants remains a significant challenge, due to the complex functional regulatory mechanisms of non-coding regions and the current limitations of available databases and tools. Hence, we develop the non-coding variant annotation database (NCAD, http://***/), encompassing comprehensive insights into 665,679,194 variants, regulatory elements, and element interaction details. Integrating data from 96 sources, spanning both GRCh37 and GRCh38 versions, NCAD v1.0 provides vital information to support the genetic diagnosis of non-coding variants, including allele frequencies of 12 diverse populations, with a particular focus on the population frequency information for 230,235,698 variants in 20,964 Chinese individuals. Moreover, it offers prediction scores for variant functionality, five categories of regulatory elements, and four types of non-coding RNAs. With its rich data and comprehensive coverage, NCAD serves as a valuable platform, empowering researchers and clinicians with profound insights into non-coding regulatory mechanisms while facilitating the interpretation of non-coding variants.

Multi-Perspective Data Fusion Framework Based on Hierarchical BERT: Provide Visual Predictions of Business Processes

Predictive Business Process Monitoring(PBPM)is a significant research area in Business Process Management(BPM)aimed at accurately forecasting future behavioral events.At present,deep learning methods are widely cited in PBPM research,but no method has been effective in fusing data information into the control flow for multi-perspective process prediction.Therefore,this paper proposes a process prediction method based on the hierarchical BERT and multi-perspective data fusion.Firstly,the first layer BERT network learns the correlations between different category attribute data.Then,the attribute data is integrated into a weighted event-level feature vector and input into the second layer BERT network to learn the impact and priority relationship of each event on future predicted events.Next,the multi-head attention mechanism within the framework is visualized for analysis,helping to understand the decision-making logic of the framework and providing visual predictions.Finally,experimental results show that the predictive accuracy of the framework surpasses the current state-of-the-art research methods and significantly enhances the predictive performance of BPM.
