This paper studies the linear EV model when replicate observations are made only on independent variables. We construct the estimates of regression coefficients and prove the consistency and asymptotic normality under some proper conditions. Results obtained reveal the difference between the case where the independent and dependent variables are observed repeatedly and simultaneously and the case studied in this article.

原发性肺癌与结直肠癌的孟德尔随机化研究

目的:随着低剂量胸部CT技术的研发和普及,早期肺癌(lung cancer,LC)患者的诊断率和生存率显著提升,首位原发性肺癌(primary lung cancer,PLC)幸存者中第二原发癌(second primary cancer,SPC)为结直肠癌(colorectal cancer,CRC)的发生已成为影响早期LC预后的重要因素,本研究试图从遗传学上探究PLC和CRC之间的潜在关联,为原发性早期LC术后SPC-CRC预防策略的制定奠定基础。方法:本研究基于两样本的孟德尔随机化设计,以PLC及CRC的全基因组关联研究为基础,系统性地筛选遗传工具变量,应用逆方差加权法和遗传风险评分法评估两种癌症之间的发病关联,并使用敏感性分析进一步验证结果的稳健性。结果:共筛选出4.个遗传工具变量,其中肺癌总体2.个,肺腺癌(lung adenocarcinoma,LUAD)12.,肺鳞癌(lung squamous cell carcinoma,LUSC)12.。逆方差加权法及遗传风险评分法发现,PLC与CRC在基因水平上未见显著关联。亚组分析同样显示LUAD、LUSC与结肠癌及直肠癌的发生均不相关。敏感性分析与主分析结果一致,证实了研究的稳健性。结论:本研究证实PLC和CRC在遗传学上不存在关联。
