Dear editor,A scheme for treating COVID-19 was published in NSR in April [1] based on blocking the translation of very large viral proteins. Since then, additional supporting data have greatly strengthened the proposal. The drug, HHT (homoharringtonine, or omacetaxine), is readily available and inexpensive as it has been approved since 2012 for treating leukemia.

The long-lived isotope ^(53)Mn cosmogenically produced in situ is a very useful indicator for geochronological dating.A new ^(53)Mn accelerator−mass-spectrometry(AMS)measurement method is developed based on the HI-13 Tandem Accelerator andΔE−Q3D detection system at China Institute of Atomic *** performance of ^(53)Cr isobar separation and suppression is tested by a series of ^(53)Mn standards and commercial MnO *** results show that theΔE−Q3D detection system brought about an overall suppression factor of more than 107 for the interfering isobar ^(53)Cr.

Toll-like receptor 4/nuclear factor-kappa B signaling detected in brain after early subarachnoid hemorrhage

Background Inflammation and immunity play a vital role in the pathogenesis of early brain injury after subarachnoid hemorrhage （SAH）. Nuclear factor-kappa B （NF-κB） regulates many genes essential for inflammation and immunity and is activated by toll-like receptor （TLR）. This study aimed to detect the expression of the toll-like receptor 4/nuclear factor-kappa B （TLR4/NF-κB） signaling in the rat brain after early SAH. Methods The rats were decapitated and their brains were removed at 0, 2, 4, 6, 12, 24 and 48 hours after a single injection of blood into the prechiasmatic cistern, mRNA expression of TLR4 was measured by Taqman real-time RT-PCR, and protein expression by immunohistochemistry and Western blotting. NF-κB activity and concentrations of tumor necrosis factor-alpha （TNF-α）, interleukin-lbeta （IL-1β） and interleukin-6 （IL-6） were measured by enzyme-linked immunosorbent assay （ELISA）. Results TaqMan real-time RT-PCR and Western blotting identified a biphasic change in TLR4 expression in both mRNA and protein： an initial peak （2-6 hours） and a sustained elevation （12-48 hours）. Immunohistochemical staining showed the inducible expression of TLR4-like immunoreactions predominantly in glial cells and vascular endothelium. A similar biphasic change in the activation of NF-κB subunit p65 as well as the production of NF-κB-regulated proinflammatory cytokines （TNF-α, IL-1β and IL-6） were detected by ELISA. Conclusions These data suggest that experimental SAH induces significant up-regulation of TLR4 expression and the NF-κB signaling in early brain injury. Activation of the TLR4/NF-κB signaling may regulate the inflammatory responses after SAH.

BACKGROUND Chronic kidney disease(CKD)is highly prevalent in patients with atrial fibrillation(AF).However,the asso-ciation between CKD and clinical consequences in AF patients is still under *** We included 19,079 nonvalvular AF patients with available estimated glomerular filtration rate(eGFR)values in the Chinese Atrial Fibrillation Registry from 2011 to *** were classified into no CKD(eGFR≥90 mL/min per 1.73 m2),mild CKD(60≤eGFRding,and cardiovascular mortality were estimated with Fine-Gray regression analysis according to CKD *** regression was performed to assess the risk of all-cause mortal-ity associated with *** Over a mean follow-up of 4.1±1.9 years,there were 985 thromboembolic events,414 major bleeding events,956 car-diovascular deaths,and 1,786 all-cause *** multivariate adjustment,CKD was not an independent risk factor of throm-boembolic *** compared to patients with no CKD,those with mild CKD,moderate CKD,and severe CKD had a 45%,47%,and 133%higher risk of major bleeding,*** was a graded increased risk of cardiovascular mortality associated with CKD status compared with no CKD group:adjusted hazard ratio[HR]was 1.34(95%CI:1.07−1.68,P=0.011)for mild CKD group,2.17(95%CI:1.67−2.81,Pher risks of major bleeding and mortality,but didn’t seem to be an independent predictor of thromboembolism in AF patients.

Frequent ventricular premature beats increase blood pressure variability in rats

AIM: The present study was designed to test a hypothesis that nonfatal ventricular arrhythmia such as ventricularpremature beats (VPB) is a contributing factor in the elevation of blood pressure variability (BPV). METHODS:Blood pressure (BP) and electrocardiogram were continuously recorded. The relation between VPB and BPV wasobserved under conscious state in chronic myocardial infarction (MI) rats one month after ligation of the leftcoronary artery, and further verified under anesthetized state in rat model of ventricular arrhythmia produced byacute intravenous infusion of aconitine. RESULTS: MI rats exhibited a big difference in the count and pattern ofVPB, and were divided into no VPB, occasional VPB, and frequent VPB groups. Among the three groups, therewere no differences in BP, heart period (HP), and MI size. However, BPV was markedly higher in frequent notoccasional VPB rats, and HP variability (HPV) was larger in both frequent and occasional VPB rats, when comparedwith no VPB rats. In the whole population of MI rats, BPV was positively correlated with VPB and HPV, not withBP, HP and MI size. Infusion of aconitine had no effect on BP, HP, BPV, and HPV during the period without *** VPB after several minutes of aconitine infusion induced significant increase in BPV and HPV with nochange in BP and HP. BPV was also positively correlated with VPB and HPV, not with BP and HP. hemodynamicsin aconitine-evoked ventricular tachycardia was characterized as lower BP, higher BPV, and higher ***: High BPV can be caused by frequent not occasional VPB in rats.

Since severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)-specific T cells have been found to play essential roles in host immune protection and pathology in patients with coronavirus disease 2019(COVID-19),this study focused on the functional validation of T cell epitopes and the development of vaccines that induce specific T cell responses.A total of 120 CD8^(+)T cell epitopes from the E,M,N,S,and RdRp proteins were functionally *** these,110,15,6,14,and 12 epitopes were highly homologous with SARS-CoV,OC43,NL63,HKU1,and 229E,respectively;in addition,four epitopes from the S protein displayed one amino acid that was distinct from the current SARS-CoV-2 ***,31 epitopes restricted by the HLA-A2 molecule were used to generate peptide cocktail vaccines in combination with Poly(I:C),R848 or poly(lactic-co-glycolic acid)nanoparticles,and these vaccines elicited robust and specific CD8^(+)T cell responses in HLA-A2/DR1 transgenic mice as well as wild-type *** contrast to previous research,this study established a modified DC-peptide-PBL cell coculture system using healthy donor PBMCs to validate the in silico predicted epitopes,provided an epitope library restricted by nine of the most prevalent HLA-A allotypes covering broad Asian populations,and identified the HLA-A restrictions of these validated epitopes using competitive peptide binding experiments with *** cell lines expressing the indicated HLA-A allotype,which initially confirmed the in vivo feasibility of 9-or 10-mer peptide cocktail vaccines against *** data will facilitate the design and development of vaccines that induce antiviral CD8^(+)T cell responses in COVID-19 patients.

用邻氨基噻吩腈与乙酰乙酸甲酯为原料,利用串联Aza-Wittig法合成了9个未见文献报道的噻吩并吡啶并嘧啶类化合物,通过IR,1H NMR,EI-MS,元素分析等方法对所合成的化合物进行了结构表征,并初步测定了所合成化合物的杀菌活性.生测结果表明:此类化合物对棉花枯萎(Fusarium oxysporium)、水稻纹枯(Rhizoctonia solani)、小麦赤霉(Gibberella zeae)、苹果轮纹菌(Dothiorella gregaria)、棉花炭疽菌(Colletotrichum gossypii)等5种菌均有一定的抑制活性.其中对苹果轮纹菌抑制活性最好,在浓度为5.0×10-5g/mL时抑制率为31.80%～82.84%.

＜正＞The synthesis of derivatives of pyridopyrimidine has been the focus of great interest recently. This is due, in part, to the broad spectrum of biological properties of these compounds. Some of these activities include antibacterial, antifungal,anticancer, insecticidal, antifloate, antivirus properties. The range of biological activities and characteristic chemical structures have made synthetic studies of pyridopyrimidine very attractive. here we report a new and facile regioselective annulation process to the synthesis of tetrahydrobenzo [4’,5’]thieno-[3’,2’:5,6]pyrido[4,3-d]pyrimidin-4（3H）-one derivatives by tandem aza-Wittig and cyclization reaction （Scheme 1）. All the products were confirmed by 1H NMR, IR spectra and elemental analysis. Their biological activities are being tested.

As one of the three payloads for the Advanced Space-based Solar Observatory(ASO-S)mission,the Lyman-alpha(Lyα)Solar Telescope(LST)is composed of three instruments:a Solar Corona Imager(SCI),a LyαSolar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST).When working in-orbit,LST will simultaneously perform high-resolution imaging observations of all regions from the solar disk to the inner corona up to 2.5 R⊙(R⊙stands for the mean solar radius)with a spatial resolution of 4.8′′and 1.2′′for coronal and disk observations,respectively,and a temporal resolution of 30–120 s and 1–120 s for coronal and disk observations,*** maximum exposure time can be up to20 s due to precise pointing and image stabilization *** the three telescopes of LST,SCI is a dual-waveband coronagraph simultaneously and independently observing the inner corona in the HI Lyα(121.6±10 nm)line and white light(WL)(700±40 nm)wavebands by using a narrowband Lyαbeam splitter and has a field of view(FOV)from 1.1 to 2.5 R⊙.The stray-light suppression level can attainhe mean brightness of the solar disk)at 1.1 R⊙and≤5×10^-8 B⊙at 2.5 R⊙.SDI and WST are solar disk imagers working in the Lyαline and 360.0 nm wavebands,respectively,which adopt an off-axis two-mirror reflective structure with an FOV up to 1.2 R⊙,covering the inner coronal edge area and relating to coronal *** present the up-to-date design for the LST payload.
