High-Resolution Data on Human Behavior for Effective COVID-19 Policy-Making—Wuhan City,Hubei Province,China,January 1–February 29,2020

Introduction:High-resolution data is essential for understanding the complexity of the relationship between the spread of coronavirus disease 2019(COVID-19),resident behavior,and interventions,which could be used to inform policy responses for future prevention and control.Methods:We obtained high-resolution human mobility data and epidemiological data at the community level.We propose a metapopulation Susceptible-Exposed-Presymptomatic-Infectious-Removal(SEPIR)compartment model to utilize the available data and explore the internal driving forces of COVID-19 transmission dynamics in the city of Wuhan.Additionally,we will assess the effectiveness of the interventions implemented in the smallest administrative units(subdistricts)during the lockdown.Results:In the Wuhan epidemic of March 2020,intra-subdistrict transmission caused 7.6 times more infections than inter-subdistrict transmission.After the city was closed,this ratio increased to 199 times.The main transmission path was dominated by population activity during peak evening hours.Discussion:Restricting the movement of people within cities is an essential measure for controlling the spread of COVID-19.However,it is difficult to contain intra-street transmission solely through citywide mobility restriction policies.This can only be accomplished by quarantining communities or buildings with confirmed cases,and conducting mass nucleic acid testing and enforcing strict isolation protocols for close contacts.

Single-cell joint analysis of methylome and transcriptome reveals how the methylation regulates the transcriptional activity.However,traditional bench-top protocols for single-cell DNA methylation and RNA transcription co-detection are laborintensive,cost-ineffective and contaminant-prone.Herein,we establish the DMF-sc MT-seq,a highly-efficient and cost-effective method to simultaneously analyze single-cell DNA methylation and transcriptional activity based on digital microfluidics.DMFsc MT-seq automates the workflow of single-cell isolation,cellular hypotonic lysis,nucleic acid separation and methylome/transcriptome library construction in a contactless and addressable way.The system ensures high accuracy(R>0.85),high gene detection ability(14,697 genes per cell at 4 million sequencing depth),and high CpG coverage(677,198 CpG sites per cell at 1million sequencing depth).By using DMF-sc MT-seq,the relationship of DNA methylation and RNA transcription under different genomic contexts is resolved.We further apply DMF-sc MT-seq to study the dynamics of transcription regulation with methylation-inhibiting anti-tumor Decitabine,and identify the methylated promoter/gene body driven genes in response to Decitabine treatment.DMF-sc MT-seq facilitates the construction of the correlation of DNA methylation and transcriptional activity at the single-cell level in a flexible,sensitive and accurate way,which is anticipated to be a powerful tool in studying single-cell biological systems.

Investigation on bonding interfaces of an SA508 steel billet manufactured by additive forging

An experimental steel billet of SA508 reactor pressure vessel material was manufactured by the additive forging method,and microstructure and mechanical properties of the hot-compression bonding interface were systematically investigated.The result indicated that oxidation levels of bonding interfaces were well controlled using vacuum electron beam welding.It was easy to discriminate interfaces from base materials during the optical microstructure observation,since interfaces were characterized by grain or phase boundaries in a straight line.Test results of uniaxial tensile experiments(at 20 and 350°C)and Charpy V-notched impact tests(at 0 and 20°C)showed that fracture behaviour of all those samples appeared at the base material,and bonding interfaces showed advantage of strength and toughness at the forge bonding state.

Reperfusion after hypoxia-ischemia exacerbates brain injury with compensatory activation of the antiferroptosis system:based on a novel rat model

Hypoxic-ischemic encephalopathy,which predisposes to neonatal death and neurological sequelae,has a high morbidity,but there is still a lack of effective prevention and treatment in clinical practice.To better understand the pathophysiological mechanism underlying hypoxic-ischemic encephalopathy,in this study we compared hypoxic-ischemic reperfusion brain injury and simple hypoxic-ischemic brain injury in neonatal rats.First,based on the conventional RiceVannucci model of hypoxic-ischemic encephalopathy,we established a rat model of hypoxic-ischemic reperfusion brain injury by creating a common carotid artery muscle bridge.Then we performed tandem mass tag-based proteomic analysis to identify differentially expressed proteins between the hypoxic-ischemic reperfusion brain injury model and the conventional Rice-Vannucci model and found that the majority were mitochondrial proteins.We also performed transmission electron microscopy and found typical characteristics of ferroptosis,including mitochondrial shrinkage,ruptured mitochondrial membranes,and reduced or absent mitochondrial cristae.Further,both rat models showed high levels of glial fibrillary acidic protein and low levels of myelin basic protein,which are biological indicators of hypoxic-ischemic brain injury and indicate similar degrees of damage.Finally,we found that ferroptosis-related Ferritin(Fth1)and glutathione peroxidase 4 were expressed at higher levels in the brain tissue of rats with hypoxic-ischemic reperfusion brain injury than in rats with simple hypoxic-ischemic brain injury.Based on these results,it appears that the rat model of hypoxic-ischemic reperfusion brain injury is more closely related to the pathophysiology of clinical reperfusion.Reperfusion not only aggravates hypoxic-ischemic brain injury but also activates the anti-ferroptosis system.

Epilepsy can be defined as a dysfunction of the brain network,and each type of epilepsy involves different brain-network changes that are implicated diffe rently in the control and propagation of interictal or ictal discharges.Gaining more detailed information on brain network alterations can help us to further understand the mechanisms of epilepsy and pave the way for brain network-based precise therapeutic approaches in clinical practice.An increasing number of advanced neuroimaging techniques and electrophysiological techniques such as diffusion tensor imaging-based fiber tra ctography,diffusion kurtosis imaging-based fiber tractography,fiber ball imagingbased tra ctography,electroencephalography,functional magnetic resonance imaging,magnetoencephalography,positron emission tomography,molecular imaging,and functional ultrasound imaging have been extensively used to delineate epileptic networks.In this review,we summarize the relevant neuroimaging and neuroelectrophysiological techniques for assessing structural and functional brain networks in patients with epilepsy,and extensively analyze the imaging mechanisms,advantages,limitations,and clinical application ranges of each technique.A greater focus on emerging advanced technologies,new data analysis software,a combination of multiple techniques,and the construction of personalized virtual epilepsy models can provide a theoretical basis to better understand the brain network mechanisms of epilepsy and make surgical decisions.

All-trans Retinoic Acid-incorporated Glycol Chitosan Nanoparticles Regulate Macrophage Polarization in Pg-LPS-Induced Inflammation

Objective The occurrence and development of inflammation are closely correlated to the polarization of macrophages.All-trans retinoic acid(ATRA)has been proven to promote the polarization of macrophages from M1 to M2,but this lacks an effective carrier to participate in the biological response.The present study aims to determine whether retinoic acid-incorporated glycol chitosan(RA-GC)nanoparticles can regulate macrophage polarization in Porphyromonas gingivalis-lipopolysaccharide(Pg-LPS)-induced inflammation.Methods Mouse 264.7 cell lines were treated with 1µg/mL Pg-LPS to induce inflammation.After the effects of ATRA and RA-GC on the activity of macrophages were detected by CCK-8 assay,cells induced with Pg-LPS were assigned to the blank control group(GC)nanoparticles without ATRA,and experimental groups(GC nanoparticles loaded with different concentrations of ATRA:1,10 and 100µg/mL).The effects of RA-GC on inflammatory cytokines tumor necrosis factor-α,interleukin(IL)-10 and IL-12 in macrophages were detected by enzyme-linked immunosorbent assay(ELISA).Subsequently,the effects of GC nanoparticles loaded with/without ATRA on macrophage polarization in an inflammatory environment were detected by RT-PCR and Western blotting.Results The results revealed that RA-GC had no significant effect on macrophage activity.However,RA-GC could effectively inhibit the Pg-LPS-induced inflammatory factor expression in macrophages.Meanwhile,the experimental results confirmed that RA-GC could downregulate the expression of inducible nitric oxide synthase(iNOS)(a marker of M1 macrophages)and upregulate the expression of mannose receptor and Arginase-1(a marker of M2 macrophages)in a dose-dependent manner.Conclusion The present study confirms that RA-GC can promote the M2 polarization of macrophages in an inflammatory environment,and proposes this as a promising target for the clinical treatment of Pg-LPS-related diseases.

In this study,the first methyl sulfonate deepultraviolet(DUV)nonlinear optical(NLO)crystal,Ba(SO_(3)CH_(3))_(2),was successfully synthesized with the polar non-π-conjugated tetrahedron,SO_(3)CH_(3)^(−),as a novel DUV NLO building unit.Results showed that Ba(SO_(3)CH_(3))_(2)not only had a very short absorption edge of 159 nm,which is the shortest among reported phase-matchable sulfates or sulfonates,but also exhibited excellent optical properties.(Powder X-ray diffractionwas ca.1.5×KH_(2)PO_(4)and birefringence was 0.04 at 589.3 nm.)It also exhibited high thermal stability and remarkable stability against air and moisture.Additionally,bulk Ba(SO_(3)CH_(3))_(2)crystal could be obtained conveniently from a simple solution evaporation process.Therefore,Ba(SO_(3)CH_(3))_(2)should have great potential as a DUV NLO crystal in the near future.

Background Bronchiolitis is a common acute lower respiratory tract infection(ALRTI)and the most frequent cause of hospitalization of infants and young children with ALRTI.Respiratory syncytial virus is the main pathogen that leads to severe bronchiolitis.The disease burden is relatively high.To date,few descriptions of the clinical epidemiology and disease burden of children hospitalized for bronchiolitis are available.This study reports the general clinical epidemiological characteristics and disease burden of bronchiolitis in hospitalized children in China.Methods This study included the face sheet of discharge medical records collected from 27 tertiary children’s hospitals from January 2016 to December 2020 that were aggregated into the FUTang Update medical REcords(FUTURE)database.The sociodemographic variables,length of stay(LOS)and disease burden of children with bronchiolitis were analyzed and compared using appropriate statistical tests.Results In total,42,928 children aged 0–3 years were hospitalized due to bronchiolitis from January 2016 to December 2020,accounting for 1.5%of the total number of hospitalized children of the same age in the database during the period and 5.31%of the hospitalizations for ALRTI.The male to female ratio was 2.01:1.Meanwhile,more boys than girls were observed in different regions,age groups,years,and residences.The 1–2 year age group had the greatest number of hospitalizations for bronchiolitis,while the 29 days–6 months group had the largest proportion of the total inpatients and inpatients with ALRTI in the same age group.In terms of region,the hospitalization rate of bronchiolitis was the highest in East China.Overall,the number of hospitalizations from 2017 to 2020 showed a decreasing trend from that in 2016.Seasonally,the peak hospitalizations for bronchiolitis occurred in winter.Hospitalization rates in North China in autumn and winter were higher than those in South China,while hospitalization rates in South China were higher in spring and summer.Approximately,half of

Perfusion and plaque evaluation to predict recurrent stroke in symptomatic middle cerebral artery stenosis

Background and purpose We investigated the baseline demographics of patients with severe unilateral atherosclerotic stenosis of the middle cerebral artery(MCA)using multimodal MRI and evaluated the haemodynamic impairments and plaque characteristics of patients who had a recurrent stroke.Materials and methods We retrospectively recruited consecutive patients with severe unilateral atherosclerotic MCA stenosis who underwent arterial spin labelling(ASL)with postlabelling delay(PLD)of 1.5 and 2.5 s,and vessel wall MRI.For each PLD,cerebral blood flow(CBF)maps were generated.Hypoperfusion volume ratio(HVR)from 2 PLD CBF was calculated.An HVR value≥50%was considered as severe HVR.Plaque areas,plaque burden,plaque length and remodelling index were measured.Plaque enhancement at maximal lumen narrowing site were graded.Baseline clinical and imaging characteristics were compared between patients with(event+)and without(event?)1 year ischaemic events.Results Forty-three patients(47.23±12.15 years;28 men)were enrolled in this study.Seven patients had an HVR≥50%.During the 1-year follow-up,7 patients had experienced a recurrent stroke.HVR were significantly higher in the event+than event?(53.17%±29.82%vs 16.9%±15.57%,p=0.0002),whereas no significant difference was detected in plaque areas,plaque burden,remodelling index,plaque length and plaque enhancement grade.The multivariable analysis revealed that a severe HVR was significantly associated with a recurrent stroke(Odds ratio=12.93,95%confidence interval 1.57 to 106.24,p=0.017)after adjusted by hypertension and smoking.Conclusion HVR obtained from two PLD ASL may be a useful imaging predictor of recurrent stroke.
