Prevalence of and risk factors for diabetic macular edema in a northeastern Chinese population

AIM:To estimate the prevalence of diabetic macular edema(DME) and clinically significant macular edema(CSME),and to assess their risk factors in a population with type 2 diabetic mellitus(T2DM) located in northeast China.METHODS:Patients were included from the Fushun Diabetic Retinopathy Cohort Study(FS-DIRECT),a community-based study conducted in northeast China.The presence of DME and CSME was determined by the Early Treatment Diabetic Retinopathy Study(ETDRS) retinopathy scale of fundus photographs.The age-standardized prevalence of DME and CSME was estimated.The association between DME/CSME and risk factors was analyzed in a multivariate Logistical analysis.RESULTS:A total of 292(15.4%) and 166(8.8%) patients were diagnosed as DME and CSME,yielding the age and sex standardized prevalence of 13.5%(95%CI:11.9%-15.0%),and 7.1%(95%CI:5.9%-8.3%),respectively.Female patients had a higher prevalence of DME compared to their male counterparts(15.7% vs 10.4%,P=0.03).Multivariable Logistic regression analysis showed that younger age,insulin use,proteinuria,longer duration of diabetes,and higher glycosylated hemoglobin A1c,were associated with the prevalence of DME and CSME.Patients with higher fasting plasma glucose,systolic blood pressure,and blood urea nitrogen were also found to be associated with DME.CONCLUSION:Early fundus screening in diabetic patients is invaluable and given the relatively high prevalence of DME and CSME in this study cohort,those with a high risk of sight threatening maculopathy would invariably benefit from earlier detection.

An ideal animal model of azoospermia would be a powerful tool for the evaluation of spermatogonial stem cell(SSC)transplantation.Busulfan has been commonly used to develop such a model,but 30%–87%of mice die when administered an intraperitoneal injection of 40 mg kg^?1.In the present study,hematoxylin and eosin staining,Western blot,immunofluorescence,and quantitative real-time polymerase chain reaction were used to test the effects of busulfan exposure in a mouse model that received two intraperitoneal injections of busulfan at a 3-h interval at different doses(20,30,and 40 mg kg^?1)on day 36 or a dose of 40 mg kg^?1 at different time points(0,9,18,27,36,and 63 days).The survival rate of the mice was 100%.When the mice were treated with 40 mg kg^?1 busulfan,dramatic SSC depletion occurred 18 days later and all of the germ cells were cleared by day 36.In addition,the gene expressions of glial cell line-derived neurotrophic factor(GDNF),fibroblast growth factor 2(FGF2),chemokine(C-X-C Motif)ligand 12(CXCL12),and colony-stimulating factor 1(CSF1)were moderately increased by day 36.A 63-day,long-term observation showed the rare restoration of endogenous germ cells in the testes,suggesting that the potential period for SSC transplantation was between day 36 and day 63.Our results demonstrate that the administration of two intraperitoneal injections of busulfan(40 mg kg^?1 in total)at a 3-h interval to mice provided a nonlethal and efficient method for recipient preparation in SSC transplantation and could improve treatments for infertility and the understanding of chemotherapy-induced gonadotoxicity.

目的:本研究旨在评估吻合器经肛直肠切除术(STARR)治疗梗阻性排便综合征(ODS)的功能学结果,并探讨肠道Cajal细胞(ICC)数量与STARR手术疗效的关系。方法:将50例行STARR手术的ODS患者的全层直肠标本,采用免疫组化方法检测直肠样本中的ICC细胞数量,并取20例来自直肠癌癌旁组织的直肠样本作为对照。收集排粪造影和直肠测压的功能学参数。结果:无论是在黏膜下层表面(SM)、肌纤维(IM)还是环形肌与纵形肌的肌间隙(MY)中,与对照标本相比,ODS直肠标本中的ICC细胞数量均显著减少(均P<0.05)。术前克利夫兰便秘评分(CCS)为24.2˘64.1,术后1、2、3、4和5年CCS评分均显著下降(均P<0.05)。术后3年,58.3%(28/48)的患者疗效满意(CCA≤10)。单因素分析显示,术前需手辅助排便(P=0.017)、ICC-MY细胞数量减少(P=0.067)、直肠感觉阈值增加(P=0.073)的ODS患者,术后功能学结果不良。多因素分析证实,ICC-MY细胞数量减少是术后不良功能学结果的独立预测因素(OR=0.097,95%CI:0.012-0.766)。结论:STARR手术治疗ODS总体疗效满意,但随时间推移,肛门功能会有所恶化。直肠标本中ICC细胞数量减少的患者术后肛门功能不良,提示术前对直肠进行全层组织学评估或可预测STARR手术的功能学结局.

Adzuki bean(Vigna angularis(Willd.)Ohwi&Ohashi)is an annual cultivated leguminous crop commonly grown in Asia and consumed worldwide.However,there has been limited research regarding adzuki bean genetics,which has prevented the efficient application of genes during breeding.In the present study,we constructed a high-density genetic map based on whole genome re-sequencing technology and validated its utility by mining QTLs related to seed size.Moreover,we analyzed the sequences flanking insertions/deletions(In Dels)to develop a set of PCR-based markers useful for characterizing adzuki bean genetics.A total of 2904 markers were mapped to 11 linkage groups(LGs).The total length of the map was 1365.0 cM,with an average distance between markers of 0.47 cM.Among the LGs,the number of markers ranged from 208(LG7)to 397(LG1)and the total distance ranged from 97.4 cM(LG9)to 155.6 cM(LG1).Twelve QTLs related to seed size were identified using the constructed map.The two major QTLs in LG2 and LG9 explained 22.1 and 18.8%of the total phenotypic variation,respectively.Ten minor QTLs in LG4,LG5 and LG6 explained 3.0–10.4%of the total phenotypic variation.A total of 9718 primer pairs were designed based on the sequences flanking In Dels.Among the 200 selected primer pairs,75 revealed polymorphisms in 24 adzuki bean germplasms.The genetic map constructed in this study will be useful for screening genes related to other traits.Furthermore,the QTL analysis of seed size and the novel markers described herein may be relevant for future molecular investigations of adzuki bean and will be useful for exploiting the mechanisms underlying legume seed development.

Phenotype and mechanism analysis of plant dwarfing in pear regulated by abscisic acid

Close planting of dwarf varieties is currently the main cultivation direction for pear trees,and the screening of excellent dwarf varieties is an important goal for breeders.In this study,the dwarfing pear variety‘601D’and its vigorous mutant‘601T’were used to show their biological characteristics and further explore the dwarfing mechanism in‘601D’.The biological characteristics showed that‘601D’had a shorter internode length,a shorter and more compact tree body,thicker and broader leaves,lower stomata density,larger stomata size(dimension),and higher photosynthetic capacity.The biological characteristics of‘601T’showed notable contrasts.The results of endogenous hormone tests indicated that the contents of abscisic acid(ABA),ABA-glucosyl ester,and GA_(4) were higher in‘601D’,but the trans-zeatin content was lower.By transcriptomic analysis,significant differences were found in the biosynthetic and metabolic pathways of ABA.Related transcription factors such as bHLH,WRKY,and homeobox also participated in the regulation of plant dwarfing.We therefore examined three hormones with obvious differences with‘601T’,and found that only ABA could induce‘601T’to return to a dwarfing plant phenotype.Therefore,we conclude that the dwarfing of‘601D’is caused by an excessive accumulation of ABA.This study provides a new theoretical basis for breeding dwarf varieties.

Methamphetamine is an amphetamine-type psychostimulant that can damage dopaminergic neurons and cause characteristic pathological changes similar to neurodegenerative diseases such as Parkinson's disease. However, its specific mechanism of action is still unclear. In the present study, we established a Parkinson's disease pathology model by exposing SH-SY5 Y cells and C57 BL/6 J mice to methamphetamine. In vitro experiments were performed with 0, 0.5, 1.0, 1.5, 2.0 or 2.5 mM methamphetamine for 24 hours or 2.0 mM methamphetamine for 0-, 2-, 4-, 8-, 16-, and 24-hour culture of SH-SY5 Y cells. Additional experimental groups of SH-SY5 Y cells were administered a nitric oxide inhibitor, 0.1 mM N-nitro-L-arginine, 1 hour before exposure to 2.0 mM methamphetamine for 24 hours. In vivo experiments: C57 BL/6 J mice were intraperitoneally injected with N-nitro-L-arginine(8 mg/kg), eight times, at intervals of 12 hours. Methamphetamine 15 mg/kg was intraperitoneally injected eight times, at intervals of 12 hours, but 0.5-hour after each N-nitro-L-arginine injection in the combined group. Western blot assay was used to determine the expression of nitric oxide synthase, α-synuclein(α-Syn), 5 G4, nitrated α-synuclein at the residue Tyr39(nT39 α-Syn), cleaved caspase-3, and cleaved poly ADP-ribose polymerase(PARP) in cells and mouse brain tissue. Immunofluorescence staining was conducted to measure the positive reaction of NeuN, nT39 α-Syn and 5 G4. Enzyme linked immunosorbent assay was performed to determine the dopamine levels in the mouse brain. After methamphetamine exposure, α-Syn expression increased; the aggregation of α-Syn 5 G4 increased; nT39 α-Syn, nitric oxide synthase, cleaved caspase-3, and cleaved PARP expression increased in the cultures of SH-SY5 Y cells and in the brains of C57 BL/6 J mice; and dopamine levels were reduced in the mouse brain. These changes were markedly reduced when N-nitro-L-arginine was administered with methamphetamine in both SH-SY5 Y cells and C57 BL/6 J mice. These results suggest that

RNA can interact with RNA-binding proteins(RBPs),mRNA,or other non-coding RNAs(ncRNAs)to form complex regulatory networks.High-throughput CLIP-seq,degradome-seq,and RNA-RNA interactome sequencing methods represent powerful approaches to identify biologically relevant ncRNA-target and protein-ncRNA interactions.However,assigning ncRNAs to their regulatory target genes or interacting RNA-binding proteins(RBPs)remains technically challenging.Chemical modifications to mRNA also play important roles in regulating gene expression.Investigation of the functional roles of these modifications relies highly on the detection methods used.RNA structure is also critical at nearly every step of the RNA life cycle.In this review,we summarize recent advances and limitations in CLIP technologies and discuss the computational challenges of and bioinformatics tools used for decoding the functions and regulatory networks of ncRNAs.We also summarize methods used to detect RNA modifications and to probe RNA structure.

Bottom simulating reflector(BSR)has been recognized as one of the indicators of gas hydrates.However,BSR and hydrate are not one-to-one correspondence.In the Xisha area of South China Sea(SCS),carbonate rocks wildly develop,which continuously distribute parallel to the seafloor with high amplitude on seismic sections,exhibiting reflections similar to BSRs in the Shenhu area nearby.This phenomenon causes some interference to hydrates identification.In this paper,the authors discussed the typical geophysical differences between carbonate rocks and hydrates,indicating that the main difference exists in relationship between porosity and velocity,causing different amplitude versus offset(AVO)characters.Then the authors proposed a new model assuming that the carbonates form the matrix and the hydrate fill the pore as a part of the matrix.The key modeling parameters have been optimized constrained by Pvelocities and S-velocities simultaneously,and the model works well both for carbonate rock and gas hydrate bearing sediments.For quantitative identification,the authors calculated the velocities when carbonates and hydrates form the matrix together in different proportions.Then they proposed a carbonate and hydrate identification template(CHIT),in which the possible hydrate saturation(PHS)and possible carbonate content(PCC)can be both scaled out for a group of sample composed by P-velocity and S-velocity.If PHS is far larger than PCC,it is more likely to be a hydrate sample because carbonates and hydrates do not coexist normally.The real data application shows that the template can effectively distinguish between hydrates and carbonate rocks,consequently reducing the risk of hydrate exploration.
