BACKGROUND Transcatheter device closure of atrial septal defect(ASD) guided by fluoroscopy and/or transesophageal echocardiography is a mature technology. Little study has focused on whether the technology can be guided totally by transthoracic echocardiography(TTE),even in pregnant women with *** To evaluate the safety and efficacy of totally TTE guided transcatheter device closure of ASD in pregnant *** Six pregnant women(gestational age 20-26 wk) with ASD underwent transcatheter device closure totally guided by TTE at our cardiac center from January 2015 to August 2017. A routine transcatheter procedure without fluoroscopy or intubation and a domestic occluder were used in this *** All patients had successful closure with good clinical results,and the overall immediate complete closure rate was 100%. The size of the occluder deployed ranged from 20 to 32 mm(26.7 ± 4.3 mm),the procedure time ranged from 30 to50 min(41.7 ± 7.5 min),and the length of hospital stay was 2-3 d(mean 2.2 ± 0.4 d). There were no serious cardiovascular related complications,and transient arrhythmias occurred in one patient during the procedure. During the follow-up period(3 mo to 2 years),no occluder dislodgement,residual fistulas,or thromboses occurred. 黑龙江省大豆生长季冷害发生频次高、影响重,构建黑龙江省大豆延迟型冷害指标,对大豆防灾减灾、保障大豆安全生产具有重要意义。利用1961-2020年黑龙江省73县(市、区)大豆产量资料及气象站逐日温度资料,以5-9月各月月平均温度之和的距平值为致灾因子,利用致灾因子和减产率的线性关系确定冷害阈值,构建黑龙江省大豆延迟型冷害指标并验证。结果表明:研究区大豆在5个种植区轻度冷害阈值分别为-1.8℃、-1.9℃、-1.9℃、-2.0℃、-2.0℃,中度冷害阈值分别为-2.0℃、-2.2℃、-2.3℃、-2.4℃、-2.6℃,重度冷害阈值分别为-2.2℃、-2.5℃、-2.7℃、-2.9℃、-3.1℃。经检验,大豆延迟型冷害指标对冷害发生趋势判识平均准确率83%,对冷害发生等级判识平均准确率62%,典型冷害年判识冷害平均准确率93%。综上所述,大豆延迟型冷害指标界定及评价黑龙江省大豆生长季延迟型冷害准确率较高,指标获取容易且实用性强。

Taxol is a 'blockbuster' antitumor drug produced by Taxus species with extremely low amount, while its analogue 7-β-xylosyl-10-deacetyltaxol is generally much higher in the plants. Both the fungal enzymes LXYL-P1à1 and LXYL-P1à2 can convert 7-β-xylosyl-10-deacetyltaxol into10-deacetyltaxol for Taxol semi-synthesis. Of them, LXYL-P1à2 is twice more active than LXYLP1à1, but there are only 11 significantly different amino acids in terms of the polarity and acidic-basic properties between them. In this study, single and multiple site-directed mutations at the 11 sites from LXYL-P1à1 to LXYL-P1à2 were performed to define the amino acids with upward bias in activities and to acquire variants with improved catalytic properties. Among all the 17 mutants, E12(A72 T/V91 S) was the most active and even displayed 2.8-and 3-fold higher than LXYL-P1à2 on β-xylosidase andβ-glucosidase activities. The possible mechanism for such improvement was proposed by homology modeling and molecular docking between E12 and 7-β-xylosyl-10-deacetyltaxol. The recombinant yeast GS115-P1 E12-7 was constructed by introducing variant E12, the molecular chaperone gene pdi and the bacterial hemoglobin gene vhb. This engineered yeast rendered 4 times higher biomass enzyme activity than GS115-3.5 K-P1à2 that had been used for demo-scale fermentation. Thus, GS115-P1 E12-7 becomes a promising candidate to replace GS115-3.5 K-P1à2 for industrial purpose.

Temperature tolerance restricts the distribution of a species. However, the molecular and cellular mechanisms that set the thermal tolerance limits of an organism are poorly understood. Here, we report on the function of dual-specificity phosphatase 1(DUSP1) in thermal tolerance regulation. Notably, we found that dusp1-/- zebrafish grew normally but survived within a narrowed temperature range. The higher susceptibility of these mutant fish to both cold and heat challenges was attributed to accelerated cell death caused by aggravated mitochondrial dysfunction and over-production of reactive oxygen species in the gills. The DUSP1-MAPK-DRP1 axis was identified as a key pathway regulating these processes in both fish and human cells. These observations suggest that DUSP1 may play a role in maintaining mitochondrial integrity and redox homeostasis. We therefore propose that maintenance of cellular redox homeostasis may be a key mechanism for coping with cellular thermal stress and that the interplay between signaling pathways regulating redox homeostasis in the most thermosensitive tissue(i.e., gills) may play an important role in setting the thermal tolerance limit of zebrafish.

为明确华北大黑鳃金龟Holotrichia oblita周期蛋白基因(Period,Per)的表达模式,本研究采用RACE技术从2龄幼虫中克隆到生物钟基因HoPer,并进行生物信息学分析;再采用RT-qPCR技术检测HoPer在不同发育阶段、雌雄成虫不同组织以及不同光周期下的表达水平。结果表明:HoPer编码1 172个氨基酸,蛋白分子量129.67 kDa,等电点为5.62。HoPer推导出的氨基酸序列中具有2个PAS和1个Period C结构域,是昆虫PER蛋白的典型结构特征。表达模式分析得出:HoPer在各个发育阶段均有表达,但在卵中的表达量显著高于其他发育时期;HoPer在雌雄成虫的头和触角中表达水平最高,其次为翅和足,在胸和腹中表达水平最低;雌雄成虫中HoPer在试验设定的5个光周期中未发现明显的表达差异。本研究为进一步阐明华北大黑鳃金龟HoPer基因功能以及其在生物钟网络中的调控作用提供了一定的参考。

For the purpose of developing flexible organic photovoltaic devices, we have fabricated two flexible devices using 5-formyl- 2,2′：5′,2″：5″,2′″-quaterthiophene （4T-CHO）, 5-formyl-2,2′：5′, 2″：5″,2′″：5′″,2″″-quinquethiophene （5T-CHO） and 3,4,9,10-perylenetertracarboxylic dianhydride （PTCDA）. The PET-ITO/4T-CHO/PTCDA/A1 device has an open circuit voltage （Voc） of 1.56 V, photoelectric conversion efficiency of 0.77%. The PET-ITO/5T-CHO/PTCDA/A1 device has a Voc of 1.70 V, photoelectric conversion efficiency of 0.84%. The two flexible devices have high Voc （1.56 and 1.70 V）. It is possible that intermolecular hydrogen bonding between -CHO group of nT-CHO and carboxylic dianhydride of PTCDA contributes to enhancing the efficiency by promoting interfacial electron transfer and eliminating the subconducting band trap sites.
