OBJECTIVE The alcoholism-related social problems have burdened the public health heavily.A better therapy for alcohol dependence as a chronic brain disease is highly required and interests the scientists worldwide. Our group has focused on screening the right drug with low toxicity and a sound curative effect from traditional Chinese medicine. METHODS Alcohol-preferring Fawn-Hooded(FH/Wjd) rat was used as an animal model of alcoholism to evaluate the effects of brucine N-oxide(BNO),an alkoloid naturally existing in the seeds of Strychnos nux-vomica L,on the alcohol-drinking behaviors.Furthermore,its adverse action and toxicity were investigated. RESULTS Treatment with BNO at the doses of 30,50 and 70 mg · kg^(-1)reduced the voluntary alcohol consumption and preference dosedependently and selectively without altering their water intake,total fluid intake,food consumption,body weight as well as sucrose preference. Remarkably,70 mg·kg^(-1)of BNO did suppress the deprivationinduced elevation of alcohol ingestion. Moreover,BNO used at the same doses as above had no influence on locomotion in an open field test and could not result in the place preference effect. CONCLUSION Taken together,BNO is of some significant pharmacological profiles to inhibit symptoms of alcohol dependence with high safety,and thence may be a potential pharmacotherapy.

OBJECTIVE Alcoholism is one of the most damaging psychiatric disorders and causes serious social and health problems in the world. However,there are no ideal treatments for this disease in clinic.Phosphodiesterases(PDEs) are a superfamily of enzymes consisting of 11 PDE families that hydrolyze cyclicAMP(cA MP) and/or cyclicGMP(cGMP). Among them,PDE4 is critical in the control of intracellular cAMP levels and has been shown to play an important role in the regulation of ethanol consumption.However,the functional role of PDE4 in mediating alcoholism remains unclear. METHODS Ethanol drinking and preference were examined using the two-bottle choice and/or drinking-in-dark(DID) test in high alcohol preferring(HAP) animals,including C57,HAP,and PDE4-subtype knockout mice,and Fawn-Hooded(FH/Wjd) rats,treated with or without the PDE4 inhibitor rolipram or roflumilast. Ethanol withdrawal-induced anxiety-and depressive-like behaviors were examined using the elevated plusmaze,holeboard,forced-swim,and tail-suspension tests in C57 mice or FH rats in the presence of PDE4 inhibition. Levels of cAMP,CREB were determined in brain regions. RESULTS Treatment with rolipram or roflumilast decreased ethanol intake and preference in two-bottle choice and DID tests in C57 and HAP mice as well as FH rats. Mice deficient in PDE4 B,but not PDE4 D,displayed similar effects to general PDE4 inhibition. In addition,rolipram reversed ethanol withdrawal-induced anxietyand depressive-like behaviors 1 d and 14 d,respectively,following withdrawal from ethanol drinking in the two-bottle choice in C57 mice or FH rats. Locomotor activity was not changed in either mice or rats treated with the PDE4 inhibitors. Levels of cAMP,p CREB in the brain were increased by rolipram.CONCLUSION The results provide solid evidence for the important role of PDE4 in ethanol consumptionand ethanol withdrawal-induced symptoms. Inhibitors of PDE4,in particular the PDE4 B isoform,can be a novel class of treatment for alcoholism.

Interface interaction between SiO_(2) and magnetite under high temperature:particle migration and inhibition mechanism

Silicon is one of the main gangue components in iron ore,usually in the form of quartz and olivine.Numerous studies have shown that SiO_(2) has a two-sided effect on the consolidation of pellets during high-temperature oxidation roasting of magnetite.However,it is very difficult to capture the structural evolution and migration mechanisms during high-tem-perature roasting process by existing experimental methods.Therefore,the influence of SiO_(2) on the consolidation behavior of magnetite was studied through a series of roasting experiments and molecular dynamic simulation.The results show that the consolidation index and particle growth index decrease with the increase in SiO_(2) content in the particles.At 1573 K,the liquid phase promotes the recrystallization growth of hematite at high temperature.Molecular dynamic study shows that it is difficult for quartz SiO_(2) to form sintering neck with Fe_(2)O_(3).When the calcination temperature is higher than 1400 K,the Fe_(2)O_(3)-Fe_(2)SiO_(4) system produces a considerable sintering neck structure after relaxation.The atomic migration ability of Fe_(2)SiO_(4) is much higher than that of Fe_(2)O_(3).The higher atomic migration ability of Fe_(2)SiO_(4) is the main reason for the formation of the sintering neck.

OBJECTIVE To investigate the effects of heat shock protein 70(Hsp70)in the development as well as expression of morphine induced conditioned place preference(CPP)in rats using Hsp70 inhibitors.METHODS The unbi⁃ased procedures of CPP lasted for 7 d and included three phases:preconditioning(D1-D3),conditioning(D4-D6,6 sessions)and test(D7).Here,morphine 5 mg·kg-1 injected in a subcuta⁃neous(sc)manner can induce significant place preference.Inhibitors of Hsp70 were injected into the right lateral ventricle during either the condi⁃tioning phase or the test phase separately.Fur⁃thermore,the expression of Hsp70 in certain areas of the mesocorticolimbic system was also studied following the intervention of N-formyl-3,4-methylenedioxybenzylidine-γ-butyrolactam(KNK437),a transcriptional inhibitor of Hsp70.RESULTS Pifithrin-μ(PES),a selective functional inhibitor acting on the substrate binding domain(SBD)of Hsp70,dose-dependently suppressed both the acquisition and expression of morphine-induced CPP.Similar function was observed after the intracerebroventricular injection(icv)of KNK437.The other functional inhibitor methy⁃lene blue,targeting the nucleotide-binding area,showed a significant tendency of inhibitory phar⁃macological effect on the expressional and devel⁃opment phases of morphine-induced CPP.Following the interventions of KNK437,we found that the level of Hsp70 was significantly decreased in the NAcs both in the acquisition and expres⁃sion of morphine induced CPP.CONCLUSION Hsp70 in NAcs plays a critical role in mediating the psychological dependence induced by morphine.

Uterine fibroids are benign tumors that originate from smooth muscle cells of the uterus.It is the most common gynecological disorder,affecting up to 80%of women of reproductive age.Uterine fibroids can cause various symptoms such as abnormal uterine bleeding,pelvic pain,infertility,and pregnancy complications.The treatment options for uterine fibroids include medical therapy,surgical intervention,and minimally invasive techniques.AIM To compare ovarian function of women with uterine fibroids who did or did not undergo uterine artery embolization(UAE).METHODS This prospective cohort study enrolled 87 women with symptomatic uterine fibroids who underwent UAE,and 87 women with the same symptoms who did not undergo UAE but received conservative management or other treatments.The two groups were matched for age,body mass index,parity,and baseline characteristics of uterine fibroids.The primary outcome was ovarian function that was evaluated by serum levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2),and anti-Müllerian hormone(AMH),as well as ovarian reserve tests,such as antral follicle count(AFC)and ovarian volume(OV).The secondary outcome was fertility that was evaluated based on the menstrual cycle,ovulation,conception,pregnancy,and delivery.The participants were followed-up for 36 months and assessed at 1,3,6,12,24,and 36 months after treatment.RESULTS The study found that the most common minor complication of UAE was postembolization syndrome in 73.6% of women,resolving within a week.No significant differences were observed between the UAE group and the control group in serum levels of reproductive hormones(FSH,LH,E2,AMH)and ovarian reserve indicators(AFC,OV)at any point up to 36 months post-treatment.Additionally,there were no significant differences in conception,pregnancy,or delivery rates,with the average time to conception and gestational age at delivery being similar between the two groups.Birth weights were also comparable.Finally,there was no significant correlation between

At high temperature,the chemical reaction mechanism at the interface of magnetite and the influence mechanism of gangue element Al on the oxidation performance of magnetite are not clear.In addition,due to the limitation of existing experimental equipment,it is difficult to clarify the interface reaction mechanism in the oxidation process and the influence mechanism of Al on the surface oxidation reaction of Fe_(3)O_(4) at an atomic scale at high temperature.The surface oxidation reaction mechanism of magnetite and the influence mechanism of Al on the oxidation of magnetite were studied by experiments and AIMD(ab initio molecular dynamics).The experimental results show that the existence of Al_(2)O_(3) will reduce the initial oxidation temperature and comprehensive oxidation performance of magnetite.The AIMD results show that aluminum can accelerate the interface reaction rate of Fe_(3)O_(4) in the high-temperature oxidation atmosphere,but the strong Al-O binding ability is not conducive to the transfer of O atoms in the oxidation process.

Rationale:Comorbidity of depression and anxiety is one of the most common mental disorders in practice.Few experimental procedures,however,have focused on identifying mixed depression-and anxiety-like behaviors in animal models.Objectives:To develop a novel testing paradigm,the so-called liang’s contextual-stress Box(briefly,liang’s Box),for evaluating depression-and/or anxiety-like behaviors in mice.Methods:liang’s Box consisted of a central area(CA)with three peripheral arms(PA).Mice were placed individually in the end of a peripheral arm facing to the central area and were allowed to travel around liang’s Box.During a test period,behavioral parameters were recorded and collected by a video-tracking system named SMART 3.0(Panlab,Spain),including①Latency to CA,②Time spent in CA(CA-time),③Distance traveled in CA(CA-distance),④Distance traveled in PA(PA-distance),⑤Transition number between CA and PA,⑥Global activity,⑦CA-time/cm2,⑧PA-time/cm2,⑨CAdistance/cm2,and⑩PA-distance/cm2.In order to assess the reliability and validity of liang’s Box and get baseline information,we undertook a series of experiment,including verifying consistency and stability in the various strains of mice(KM,ICR,and C57 BL/6 J mice),imposing high illumination,restraint and tail suspension stress,pre-treating with an anxiolytic(diazepam,buspirone)or antidepressant(desipramine,mianserin).Results:①There were not remarkable differences in all the parameters among KM,ICR,and C57 BL/6 J mice;②High illumination,restraint and tail suspension significantly increased the latency to CA and decreased the CA-time,CA-distance and transition number.However,these three types of stresses evoked different effect on global activity.High illumination increased activity,but restraint stress had no significant influence.In contrast with high illumination,tail suspension stress markedly reduced activity in mice;③The anxiolytics diazepam(0.25 mg·kg-1 and 0.5 mg·kg-1,i.p.)and buspirone(0.5 mg·kg-1 and 1.0 mg·kg-1,i.p.)significantly decreased latency to CA and transi

BACKGROUND Previous studies have shown that the Shi-pi-xiao-ji(SPXJ)herbal decoction formula is effective in suppressing hepatocellular carcinoma(HCC),but the underlying mechanisms are not known.Therefore,this study investigated whether the antitumor effects of the SPXJ formula in treating HCC were mediated by acetyl-coA acetyltransferase 1(ACAT1)-regulated cellular stiffness.Through a series of experiments,we concluded that SPXJ inhibits the progression of HCC by upregulating the expression level of ACAT1,lowering the level of cholesterol in the cell membrane,and altering the cellular stiffness,which provides a new idea for the research of traditional Chinese medicine against HCC.AIM To investigate the anti-tumor effects of the SPXJ formula on the malignant progression of HCC.METHODS HCC cells were cultured in vitro with SPXJ-containing serum prepared by injecting SPXJ formula into wild-type mice.The apoptotic rate and proliferative,invasive,and migratory abilities of control and SPXJ-treated HCC cells were compared.Atomic force microscopy was used to determine the cell surface morphology and the Young’s modulus values of the control and SPXJ-treated HCC cells.Plasma membrane cholesterol levels in HCC cells were detected using the Amplex Red cholesterol detection kit.ACAT1 protein levels were estimated using western blotting.RESULTS Compared with the vehicle group,SPXJ serum considerably reduced proliferation of HCC cells,increased stiffness and apoptosis of HCC cells,inhibited migration and invasion of HCC cells,decreased plasma membrane cholesterol levels,and upregulated ACAT1 protein levels.However,treatment of HCC cells with the water-soluble cholesterol promoted proliferation,migration,and invasion of HCC cells as well as decreased cell stiffness and plasma membrane cholesterol levels,but did not alter the apoptotic rate and ACAT1 protein expression levels compared with the vehicle control.CONCLUSION SPXJ formula inhibited proliferation,invasion,and migration of HCC cells by decreasing plasma membrane chole

AIM:To investigate the types of bacteria in patients with eye infections in Suzhou and their drug resistance to commonly used antibacterial drugs.METHODS:The clinical data of 155 patients were retrospectively collected in this study,and the pathogenic bacteria species and drug resistance of each pathogenic bacteria were analyzed.RESULTS:Among the 155 patients(age from 12 to 87 years old,with an average age of 57,99 males and 56 females)with eye infections(160 eyes:74 in the left eye,76 in the right eye and 5 in both eyes,all of which were exogenous),71(45.81%)strains were gram-positive bacteria,23(14.84%)strains were gram-negative bacteria and 61(39.35%)strains were fungi.Gram-positive bacteria were highly resistant to penicillin and erythromycin(78.87%and 46.48%respectively),but least resistant to vancomycin at 0.Gram-negative bacteria were highly resistant to cefoxitin and compound sulfamethoxazole(100%and 95.65%respectively),but least resistant to meropenem at 0.Comparison of the resistance of gram-positive and gram-negative bacteria to some drugs revealed statistically significant differences(P<0.05)in the resistance of both to cefoxitin,cotrimoxazole,levofloxacin,cefuroxime,ceftriaxone and ceftazidime,and both had higher rates of resistance to gram-negative bacteria than to gram-positive bacteria.The distribution of bacterial infection strains showed that Staphylococcus epidermidis was the most common strain in the conjunctiva,cornea,aqueous humor or vitreous body and other eye parts.Besides,Fusarium and Pseudomonas aeruginosa were also among the most common strains of conjunctival and corneal infections.CONCLUSION:Gram-positive bacteria are the dominant bacteria in eye infections,followed by gram-negative bacteria and fungi.Considering the resistance of gramnegative bacteria to multiple drugs,monitoring of bacteria should be strengthened in eye bacterial infections for effective prevention and control to reduce complications caused by eye infections.
