Cryopreservation of Queen Honeybee (Apis mellifera carnica) born Worker Eggs by Vitrification

Many species of insect egg can be targeted individually or （and） collectively for cryopreservation by vitrification. However, there has been no report on cryopreservation of honeybee eggs by vitrification. In an attempt to define a preliminary procedure of cryopreservation of honeybee eggs by vitrification, queen honeybee born worker eggs （worker eggs） were stored through vitrification in liquid nitrogen up to 1 h, and then post-vitrification survival of the vitrified worker eggs in vitro and their hatching rates during maturation in vivo were observed using microscopic and close visual inspections. The procedure of cryopreservation by vitrification included dechorionation with sodium hypochlorite and permeabilization with isopropyl alcohol; equilibration by addition of loading solution （i.e., 25% vitrification storage solution） and dehydration by gradual replacement of loading solution with vitrification storage solution; cooling in liquid nitrogen vapor right before droplet vitrification in liquid nitrogen; and recovery in liquid nitrogen vapor right after storage in liquid nitrogen, thawing at temperature of thawing medium （5% sucrose in TC 100-insect medium） and rehydration by gradual replacement of vitrification storage solution with rehydration solution （5% fetal bovine serum in TC 100-insect medium）. It was found that among the worker eggs experiencing cyropreservation by vitrification, 1.25% of them were successfully passed through the four life stages, viz., egg, larva, pupa, and adult. In summary, it can be inferred that although a majority of worker eggs were dead after cyropreservation by vitrification, a few of them were developed into larvae, pupae, and finally emerged as adults.

Assessing the concentration and potential health risk of heavy metals in China's main deciduous fruits

To assess levels of contamination and human health risk, we analyzed the concentrations of the heavy metals lead （Pb）, cadmium （Cd）, chromium （Cr）, and nickel （Ni） in China’s main deciduous fruits-apple, pear, peach, grape, and jujube. The concentration order of the heavy metals was Ni〉Cr〉Pb〉Cd. In 97.5% of the samples, heavy metal concentrations were within the maximum permissible limits. Among the fruits studied, the heavy metal concentrations in jujube and peach proved to be the highest, and those in grape proved to be the lowest. Only 2.2% of the samples were poluted by Ni, only 0.4% of the samples were poluted by Pb, and no samples were poluted by Cd or Cr. Compared with the other fruits, the combined heavy metal polution was signiifcantly higher （P〈0.05） in peach and signiifcantly lower （P〈0.05） in grape. For the combined heavy metal polution, 96.9% of the samples were at safe level, 2.32% at warning level, 0.65% at light level, and 0.13% at moderate level. In the fruits studied, the contribution of heavy metals to the daily intake rates （DIR） folowed the order of Ni〉Cr〉Pb〉Cd. The highest DIR came from apple, while the lowest DIR came from grape. For each of the heavy metals, the total DIR from ifve studied fruits corresponded to no more than 1.1% of the tolerable daily intake, indicating that no signiifcant adverse health effects are expected from the heavy metals and the fruits studied. The target hazard quotients and the total target hazard quotients demonstrated that none of the analyzed heavy metals may pose risk to consumers through the fruits studied. The highest risk was posed by apple, folowed in decreasing order by peach and pear, jujube, and grape. We suggest that the main deciduous fruits （apple, pear, peach, grape, and jujube） of China’s main producing areas are safe to eat.

Coexistence of multiple coronaviruses in several bat colonies in an abandoned mineshaft

Since the 2002–2003 severe acute respiratory syndrome(SARS) outbreak prompted a search for the natural reservoir of the SARS coronavirus, numerous alpha- and betacoronaviruses have been discovered in bats around the world. Bats are likely the natural reservoir of alpha- and betacoronaviruses, and due to the rich diversity and global distribution of bats, the number of bat coronaviruses will likely increase. We conducted a surveillance of coronaviruses in bats in an abandoned mineshaft in Mojiang County, yunnan Province, China, from 2012–2013. Six bat species were frequently detected in the cave: Rhinolophus sinicus, Rhinolophus affinis, Hipposideros pomona, Miniopterus schreibersii, Miniopterus fuliginosus, and Miniopterus fuscus. By sequencing PCR products of the coronavirus RNA-dependent RNA polymerase gene(Rd Rp), we found a high frequency of infection by a diverse group of coronaviruses in different bat species in the mineshaft. Sequenced partial Rd Rp fragments had 80%–99% nucleic acid sequence identity with well-characterized Alphacoronavirus species, including Bt CoV HKU2, Bt CoV HKU8, and Bt CoV1,and unassigned species Bt CoV HKU7 and Bt CoV HKU10. Additionally, the surveillance identified two unclassified betacoronaviruses, one new strain of SARS-like coronavirus, and one potentially new betacoronavirus species. Furthermore, coronavirus co-infection was detected in all six bat species, a phenomenon that fosters recombination and promotes the emergence of novel virus strains. Our findings highlight the importance of bats as natural reservoirs of coronaviruses and the potentially zoonotic source of viral pathogens.

Contribution of ear photosynthesis to grain yield under rainfed and irrigation conditions for winter wheat cultivars released in the past 30 years in North China Plain

To understand the contribution of ear photosynthesis to grain yield and its response to water supply in the improvement of winter wheat, 15 cultivars released from 1980 to 2012 in North China Plain（NCP） were planted under rainfed and irrigated conditions from 2011 to 2013, and the ear photosynthesis was tested by ear shading. During the past 30 years, grain yield significantly increased, the flag leaf area slightly increased under irrigated condition but decreased significantly under rainfed condition, the ratio of grain weight：leaf area significantly increased, and the contribution of ear photosynthesis to grain yield changed from 33.6 to 64.5% and from 32.2 to 57.2% under rainfed and irrigated conditions, respectively. Grain yield, yield components, and ratio of grain weight：leaf area were positively related with contribution of ear photosynthesis. The increase in grain yield in winter wheat was related with improvement in ear photosynthesis contribution in NCP, especially under rainfed condition.

Hepatitis E virus(HEV)is the prototype of the family Hepeviridae and the causative agent of common acute viral *** diverse HEV-related viruses have been detected in a variety of mammals and some of them may have zoonotic *** this study,we tested 278 specimens collected from seven wild small mammal species in yunnan province,China,for the presence and prevalence of orthohepevirus by broad-spectrum reverse transcription(RT)-***-related sequences were detected in two rodent species,including Chevrier’s field mouse(Apodemus chevrieri,family Muridae)and Père David’s vole(Eothenomys melanogaster,family Cricetidae),with the infection rates of 29.20%(59/202)and 7.27%(4/55),*** four representative full-length genomes were generated:two each from Chevrier’s field mouse(named Rd HEVAc14 and Rd HEVAc86)and Père David’s vole(Rd HEVEm40 and Rd HEVEm67).Phylogenetic analyses and pairwise distance comparisons of whole genome sequences and amino acid sequences of the gene coding regions showed that orthohepeviruses identified in Chinese Chevrier’s field mouse and Père David’s vole belonged to the species Orthohepevirus C but were highly divergent from the two assigned genotypes:HEV-C1 derived from rat and shrew,and HEV-C2 derived from ferret and possibly *** real-time RT-PCR demonstrated that these newly discovered orthohepeviruses had hepatic *** summary,our work discovered two putative novel genotypes orthohepeviruses preliminarily named HEVC3 and HEV-C4 within the species Orthohepevirus C,which expands our understanding of orthohepevirus infection in the order Rodentia and gives new insights into the origin,evolution,and host range of orthohepevirus.

Risk assessment and ranking of pesticide residues in Chinese pears

The presence of pesticide residues in pears is a serious health concern. This study presents the results from a 2-year investigation （2013-2014） that used gas chromatography, GS/MS and UPLC/MS-MS to measure the levels of 104 pesti- cides in 310 pear samples. In 93.2% of the samples, 43 pesticides were detected, of which the maximum residue levels （MRLs） were exceeded in 2.6% of the samples. Multiple residues （two to eight compounds） were present in 69.7% of the samples; one sample contained nine pesticides and one sample contained 10. Only 6.8% of the samples did not contain residues. To assess the health risks, the pesticide residue data have been combined with daily pear consumption data for children and adult populations. A deterministic model was used to assess the chronic and acute exposures based on the Joint Meeting on Pesticide Residues （JMPR） method. A potential acute risk was demonstrated for children in the case of bifenthrin, which was found to be present at 105.36% of the acute reference dose （ARfD） value. The long- term exposure of the Chinese consumer to pesticide residues through the consumption of raw pears was far below the acceptable daily intake （ADI） criterion. Additionally, the matrix ranking scheme was used to classify risk subgroups of pesticides and pear samples. In general, 95.5% of samples were deemed to be safe and nine pesticides were classified as being of a relatively high risk. The findings indicated that the occurrence of pesticide residues in pears should not be considered a serious public health problem. Nevertheless, a more detailed study is required for vulnerable consumer groups, especially children. Continuous monitoring of pesticides in pears and tighter regulation of pesticide residue standards are recommended.

Achieving a High-Strength CoCrFeNiCu High-Entropy Alloy with an Ultrafine-Grained Structure via Friction Stir Processing

High-entropy alloys(HEAs) are a new class of materials with a potential engineering application,but how to obtain ultrafine or nano-sized crystal structures of HEAs has been a ***,we first presented an equiatomic CoCrFeNiCu HEA with excellent mechanical properties obtained via friction stir processing(FSP).After FSP,the Cu element segregation in the cast CoCrFeNiCu HEA was almost eliminated,and the cast coarse two-phase structure(several micrometers) was changed into an ultrafine-grained single-phase structure(150 nm) with a large fraction of high-angle grain boundaries and nanoscale deformation *** unique microstructure was mainly attributed to the severe plastic deformation during FSP,and the sluggish diffusion effect in dynamics and the lattice distortion effect in crystallography for ***,FSP largely improved the hardness and yield strength of the CoCrFeNiCu HEA with a value of 380 HV and more than 1150 MPa,respectively,which were> 1.5 times higher than those of the base *** great strengthening after FSP was mainly attributed to the significant grain refinement with large lattice distortion and *** study provides a new method to largely refine the microstructure and improve the strength of cast CoCrFeNiCu HEAs.
