Protective effects of taurine against muscle damage induced by diquat in 35 days weaned piglets

Background: Oxidative stress is a key factor that influences piglets' health. Taurine plays an imperative role in keeping the biological system from damage. This study was conducted to investigate the protective effect of taurine against muscle injury due to the secondary effect of diquat ***: Our study found that taurine effectively and dose-dependently alleviated the diquat toxicity induced rise of feed/gain, with a concurrent improvement of carcass lean percentage. The plasma content of taurine was considerably increased in a dose-dependent manner. Consequently, dietary taurine efficiently improved the activity of plasma antioxidant enzymes. Furthermore, taurine attenuated muscle damage by restoring mitochondrial micromorphology, suppressing protein degradation and reducing the percentage of apoptotic cells in the skeletal muscle. Taurine supplementation also suppressed the genes expression levels of the antioxidant-, mitochondrial biogenesis-, and muscle atrophy-related genes in the skeletal muscle of piglets with oxidative ***: These results showed that the dose of 0.60% taurine supplementation in the diet could attenuate skeletal muscle injury induced by diquat toxicity. It is suggested that taurine could be a potential nutritional intervention strategy to improve growth performance.

Engineered probiotics can serve as therapeutics based on their ability of produce recombinant immune-stimulating *** this study,we built the recombinant Bacillus subtilis WB800 expressing antimicrobial peptide KR32(WB800-KR32)using genetic engineering methods and investigated its protective effects of nuclear factor-E2-related factor 2(Nrf2)-Kelch-like ECH-associated protein 1(Keap1)pathway activation in intestinal oxidative disturbance induced by enterotoxigenic Escherichia coli(ETEC)K88 in weaned ***-eight weaned piglets were randomly distributed into four treatment groups with seven replicates fed with a basal *** feed of the control group(CON)was infused with normal sterilized saline;meanwhile,the ETEC,ETEC+WB800,and ETEC+WB800-KR32 groups were orally administered normal sterilized saline,5×10^(10)CFU(CFU:colony forming units)WB800,and 5×10^(10)CFU WB800-KR32,respectively,on Days 1-14 and all infused with ETEC K881×10^(10)CFU on Days *** results showed that pretreatment with WB800-KR32 attenuated ETEC-induced intestinal disturbance,improved the mucosal activity of antioxidant enzyme(catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GPx))and decreased the content of malondialdehyde(MDA).More importantly,WB800-KR32 downregulated genes involved in antioxidant defense(GPx and SOD1).Interestingly,WB800-KR32 upregulated the protein expression of Nrf2 and downregulated the protein expression of Keap1 in the ***800-KR32 markedly changed the richness estimators(Ace and Chao)of gut microbiota and increased the abundance of Eubacterium_rectale_ATCC_33656 in the *** results suggested that WB800-KR32 may alleviate ETEC-induced intestinal oxidative injury through the Nrf2-Keap1 pathway,providing a new perspective for WB800-KR32 as potential therapeutics to regulate intestinal oxidative disturbance in ETEC K88 infection.

本研究旨在建立一种环介导等温扩增技术(loop-mediated isothermal amplification,LAMP)检测食品中的肺炎克雷伯菌。选取肺炎克雷伯菌特有的毒力基因区域,设计2对引物(1对内引物、1对外引物),进行体外恒温扩增反应。对反应体系内的甜菜碱、核酸原料(dNTPs)、内外引物比例、Mg^(2+)浓度、反应时间和温度进行优化和选择,并进行特异性和灵敏性试验检测。应用建立的LAMP方法对灭菌鸡肉人工污染加标样品进行检测。结果表明,通过优化试验,筛选到最优反应体系和反应条件;应用优化后的LAMP反应体系对包括肺炎克雷伯菌在内的18种常见微生物进行特异性检测试验,特异性良好。肺炎克雷伯菌LAMP反应体系对基因组的最低检测限是0.875pg/μL,比传统PCR方法灵敏性高100倍。对人工鸡肉阳性样品进行检测,检测限为30CFU/mL。本试验成功建立了一种能检测食物中致病肺炎克雷伯菌的LAMP检测方法,该方法利用2对引物针对肺炎克雷伯菌目的基因6个区域进行体外恒温核酸阶梯性扩增,灵敏性更高,反应时间更短,检测成本更低。

Natural frequency for exponential shaped thermoacoustic resonator

The two-dimensional nonlinear acoustic field of eleven exponential shaped res- onators was simulated with a computational fluid dynamics software Fluent. The influence of driving frequency and driving intensity on pressure in resonator as well as its natural frequency was investigated. The relationship between natural frequency and theoretical calculation res- onance frequency was also explored. It is found that beating phenomena can be observed in the resonator when the driving frequency deviates from the natural frequency. Moreover, the natural frequency of resonator increases with the increasing of driving intensity, which shows a hard spring effect. However, the driving intensity plays little effect on natural frequency and the natural frequencies are smaller than the theoretical calculation values in any driving intensity. Meanwhile, a formula between the natural frequency and its first-order resonance frequency from theoretical calculation was obtained by linear fitting for all these exponential shaped resonators under consideration. It is also found that the highest pressure amplitude and highest pressure ratio can be obtained from the exponential shaped resonator of m = 2.8 under the same driving intensity. Moreover, the relation between natural frequency and the theoretical resonance frequency for m = 2.8-tube is slightly different from other tubes in consideration.

Coral fluorescence phenotypes have been suggested as an adaptation to a broad range of environmental conditions,yet the mechanisms linking thermal bleaching tolerance in reef-building coral populations,associated with fluorescence phenotypes due to GFP-like proteins,remains *** this study,the relationship between the thermal sensitivity and phenotypic plasticity of corals was investigated using two phenotypes of Galaxea fascicularis,green and *** results reveal that brown *** was more susceptible to bleaching than green *** when exposed to a higher growth temperature of 32℃.Both phenotypes of *** were associated with the thermotolerant Symbiodiniaceae symbiont,Durusdinium ***,the brown *** showed a significant decrease in Symbiodiniaceae cell density and a significant increase in pathogenic bacteria abundance when the growth temperature was raised from 29 to 32℃.The physiological traits and transcriptomic profiles of Symbiodiniaceae were not notably affected,but there were differences in the transcriptional levels of certain genes between the two phenotype hosts of *** heat stress of 32℃,the gene encoding green fluorescent protein(GFP)-like and chromosome-associated proteins,as well as genes related to oxidative phosphorylation,cell growth and death showed lower transcriptional levels in the brown *** compared to the green ***,the results demonstrate that the green form of *** is better able to tolerate ocean warming and defend against pathogenic bacteria,likely due to higher gene transcription levels and defense ability.

Antibacterial substance from mucus of a scleractinian coral,Symphyllia gigantea

Coral mucus covers the surface of coral and contains antibacterial substances as a first line of defense. Coral mucus not only enables the coral itself to resist disease, but also provides antibacterial agents for people. We collected mucus from a scleractinian coral （Symphyllia gigantea） at Sta. Sanya （China）, then extracted the antibacterial substances using 10% glacial acetic acid with the help of antiprotease inhibitors, and tested the antibacterial activity by a terrestrial bacterium （Staphylococcus aurevs） and a marine bacterium （Vibrio anguillarum）. The result showed that, there were antibacterial agents in the mucus, and their antibacterial activities were lost by treatment of the sample at 90 ℃ water for 10 min.

Effects of dietary protein restriction on muscle fiber characteristics and m TORC1 pathway in the skeletal muscle of growing-finishing pigs

Background： To investigate the effects of dietary crude protein（CP） restriction on muscle fiber characteristics and key regulators related to protein deposition in skeletal muscle, a total of 18 growing-finishing pigs（62.30 ± 0.88 kg）were allotted to 3 groups and fed with the recommended adequate protein（AP, 16 % CP） diet, moderately restricted protein（MP, 13 % CP） diet and low protein（LP, 10 % CP） diet, respectively. The skeletal muscle of different locations in pigs, including longissimus dorsi muscle（LDM）, psoas major muscle（PMM） and biceps femoris muscle（BFM） were collected and ***： Results showed that growing-finishing pigs fed the MP or AP diet improved（P 〈 0.01） the average daily gain and feed： gain ratio compared with those fed the LP diet, and the MP diet tended to increase（P = 0.09） the weight of LDM. Moreover, the ATP content and energy charge value were varied among muscle samples from different locations of pigs fed the reduced protein diets. We also observed that pigs fed the MP diet up-regulated（P 〈 0.05） muscular m RNA expression of all the selected key genes, except that myosin heavy chain（My HC） IIb,My HC IIx, while m RNA expression of ubiquitin ligases genes was not affected by dietary CP level. Additionally, the activation of mammalian target of rapamycin complex 1（m TORC1） pathway was stimulated（P 〈 0.05） in skeletal muscle of the pigs fed the MP or AP diet compared with those fed the LP ***： The results suggest that the pigs fed the MP diet could catch up to the growth performance and the LDM weight of the pigs fed the AP diet, and the underlying mechanism may be partly due to the alteration in energy status, modulation of muscle fiber characteristics and m TORC1 activation as well as its downstream effectors in skeletal muscle of different locations in growing-finishing pigs.

Myokine interleukin-15 expression profile is different in suckling and weaning piglets

Interleukin-15(IL-15) is a cytokine highly expressed in skeletal muscle. The objective of the present study was to investigate the development of muscle IL-15 expression in suckling piglets and in early weaning piglets(day 14) at each level, that is, mRNA, protein, and secretion. Eight litters(eight piglets per litter) of newborn healthy piglets(Large × White × Landrace) with a similar initial weight(1618.0 ± 140.1 g) were chosen and divided into two groups, Group one used suckling piglets that were killed, respectively, at days 1,7,14,21, and group two used early(day 14) weaning piglets that were killed respectively, at days 15, 17, 19, 21. In group one, IL-15 gene expression levels increased significantly(P 0.05)among piglets at other ages. These findings indicated that increased IL-15 mRNA expression did not result in a corresponding increase of its protein expression. In group two, which used early weaning piglets from days 15-19, IL-15 mRNA and protein expression levels increased constantly(P ling piglets. Moreover, there was no gain of body weight(P > 0.05) compared with suckling piglets at day 14 of age. However, IL-15 protein expression levels in early weaning piglets at day 21 of age dropped significantly(P ling piglets at day 21 of age, while body weight increased(P ling piglets at day 21 of age. In both groups, the serum IL-15 levels of piglets decreased significantly(P ling piglets and in weaning piglets. It is speculated that IL-15 may play an important role in counteracting the effects of early weaning stress.

Shen Qi Wan attenuates renal interstitial fibrosis through upregulating AQP1

Renal interstitial fibrosis(RIF)is the crucial pathway in chronic kidney disease(CKD)leading to the end-stage renal ***,the underlying mechanism of Shen Qi Wan(SQW)on RIF is not fully *** the current study,we investigated the role of Aquaporin 1(AQP1)in SQW on tubular epithelial-to-mesenchymal transition(EMT).A RIF mouse model induced by adenine and a TGF-β1-stimulated HK-2 cell model were etablished to explore the involvement of AQP 1 in the protective effect of SQW on EMT in vitro and in ***,the molecular mechanism of SQW on EMT was explored in HK-2 cells with AQP1 *** results indicated that SQW alleviated kidney injury and renal collagen deposition in the kidneys of mice induced by adenine,increased the protein expression of E-cadherin and AQP1 expression,and decreased the expression of vimentin andα-smooth muscle actin(α-SMA).Similarly,treatmement with SQW-containing serum significantly halted EMT process in TGF-β1 stimulated HK-2 *** expression of snail and slug was significantly upregulated in HK-2 cells after knockdown of ***1 knockdown also increased the mRNA expression of vimentin andα-SMA,and decreased the expression of *** protein expression of vimentin increased,while the expression of E-cadherin and CK-18 significantly decreased after AQP1 knockdown in HK-2 *** results revealed that AQP1 knockdown promoted ***,AQP1 knockdown abolished the protective effect of SQW-containing serum on EMT in HK-2 *** sum,SQW attentuates EMT process in RIF through upregulation of the expression of AQP1.

Longitudinal patterns of spring-intermonsoon phytoplankton biomass,species compositions and size structure in the Bay of Bengal

Vertical distributions of phytoplankton biomass, compositions and size structure were investigated during the spring-intermonsoon （April 22 to 30） of 2010 along transact 10°N of the Bay of Bengal, northern Indian Ocean. Surface phytoplankton biomass （Chl a） was （0.065±0.009） μg/L, being greater than 80% of which was contributed by pico-phytoplankton （〈3 μm）. The Chl a concen- tration vertically increased to the maximal values at deep chlorophyll maximum （DCM） layer that shoaled eastwards from 75 to 40 m. The Chl a biomass at DCM layer generally varied between 0.2 and 0.4 μg/L, reaching the maximum of 0.56 μg/L with micro-phytoplankton cells （~20 #m） accounting for 58% and nano- （3-20μm） or pico-cells for 15% and 27%, respectively. In particular, the cells concentration coupling well with phosphate level was observed at middle layer （75-100 m） of 87° to 89°E, dominated by micro-cells diatoms （e.g., Chaetoceros atlanticus v. neapolitana, Chaetoceros femur and Pseudonitzsehia sp.） and cyanobacteria （i.e., Trichodesmium hildebrandtii）, With the ceils concentration reached as high as 4.0 × 10^4 and 4.3 × 10^4 cells/L. At the rest of the trans- act however, dinoflagellates （e.g., Amphisdinium carterae and Prorocentrum sp.） were the dominant species, with the cells concentration varying from 0.3×10^3 to 6.8×10^3 cells/L. Our results also indicate that the regulation of large cells （micro-, nano-） on phytoplankton biomass merely occurred at DCM layer of the Bay.
