Fine particulate matter(PM_(2.5))exposure is associated with cardiovascular disease(CVD)morbidity and mortality.Mitochondria are sensitive targets of PM_(2.5),and mitochondrial dysfunction is closely related to the occurrence of CVD.The epigenetic mechanism of PM_(2.5)-triggered mitochondrial injury of cardiomyocytes is unclear.This study focused on the mi R-421/SIRT3 signaling pathway to investigate the regulatory mechanism in cardiac mitochondrial dynamics imbalance in rat H9c2 cells induced by PM_(2.5).Results illustrated that PM_(2.5)impaired mitochondrial function and caused dynamics homeostasis imbalance.Besides,PM_(2.5)up-regulated mi R-421 and down-regulated SIRT3 gene expression,along with decreasing p-FOXO3a(SIRT3 downstream target gene)and p-Parkin expression and triggering abnormal expression of fusion gene OPA1 and fission gene Drp1.Further,mi R-421 inhibitor(mi R-421i)and resveratrol significantly elevated the SIRT3 levels in H9c2 cells after PM_(2.5)exposure and mediated the expression of SOD2,OPA1 and Drp1,restoring the mitochondrial morphology and function.It suggests that mi R-421/SIRT3 pathway plays an epigenetic regulatory role in mitochondrial damage induced by PM_(2.5)and that mi R-421i and resveratrol exert protective effects against PM_(2.5)-incurred cardiotoxicity.

Methanol(CH_(3)OH)is a liquid hydrogen(H_(2))source that effectively releases H_(2) and is convenient for transportation.Traditional thermocatalytic CH_(3)OH reforming reaction is used to produce H_(2),but this process needs to undergo high reaction temperature(e.g.,200℃)along with a catalyst and a large amount of carbon dioxide(CO_(2))emission.Although photocatalysis and photothermal catalysis under mild conditions are proposed to replace the traditional thermal catalysis to produce H_(2) from CH_(3)OH,they still inevitably produce CO_(2) emissions that are detrimental to carbon neutrality.Here,we,for the first time,report an ultrafast and highly selective production of H_(2) without any catalysts and no CO_(2) emission from CH_(3)OH by laser bubbling in liquid(LBL)at room temperature and atmospheric pressure.

Participation in agricultural cooperative organizations is an important form of industrial chain organization expansion, but also an important way to effectively reduce industrial chain transaction costs and promote the quality and efficiency improvement of new agricultural business entities.Based on the micro-survey data of 553 family farms in Xinjiang Corps in 2021, this paper uses stochastic frontier approach to measure the business performance of family farms in Xinjiang Corps, and discusses the action mechanism and difference in the influence of participation in agricultural cooperative organizations on the business performance of family farms. The research shows that participation in agricultural cooperative organizations can significantly improve the business performance of family farms through such mechanisms as easing the constraints of farm credit financing, reducing the transaction costs of agricultural products, and promoting the adoption of agricultural technologies. Compared with food farms, cotton and fruit and vegetable family farms have more obvious performance improvement in participating in agricultural cooperative organizations. Moreover, participation in agricultural cooperation organizations has a stronger effect on the performance improvement of farms in northern Xinjiang. It is found that the vertical extension of the industrial chain between”family farm+agricultural cooperative organization” and network marketers, chain supermarkets, industrial leading enterprises and agricultural production and marketing enterprises is conducive to the improvement of the business performance of family farms, among which the performance improvement effect of”family farm+agricultural cooperative organization+network marketers” mode is more obvious. The research provides a scientific reference for deepening the expansion of the industrial chain organization and promoting the quality-oriented growth of family farms in the new era.

Time-dependent characteristics(TDCs)have been neglected in most previous studies investigating the deviation mechanisms of bridge pile foundations and evaluating the effectiveness of preventive measures.In this study,the stress-strain-time characteristics of soft soils were illustrated by consolidation-creep tests based on a typical engineering case.An extended Koppejan model was developed and then embedded in a finite element(FE)model via a user-material subroutine(UMAT).Based on the validated FE model,the time-dependent deformation mechanism of the pile foundation was revealed,and the preventive effect of applying micropiles and stress-release holes to control the deviation was investigated.The results show that the calculated maximum lateral displacement of the cap differs from the measured one by 6.5%,indicating that the derived extended Koppejan model reproduced the deviation process of the bridge cap-pile foundation with time.The additional load acting on the pile side caused by soil lateral deformation was mainly concentrated within the soft soil layer and increased with the increase in load duration.Compared with t=3 d(where t is surcharge time),the maximum lateral additional pressure acting on Pile 2#increased by approximately 47.0%at t=224 d.For bridge pile foundation deviation in deep soft soils,stress-release holes can provide better prevention compared to micropiles and are therefore recommended.

Colorectal cancer(CRC)is one of the most common cancers around the world,and it is one of the leading causes of cancer-related death.1 Although anti-EGFR therapy and immune checkpoint inhibitor(ICl)therapy are becoming more and more important for colorectal therapy,however,clinical outcomes of current treatments for metastatic CRC,especially with ICls,have been shown to be affected by the status of KRAS.Therefore,KRAS mutation status detection has become a very important diagnostic factor for managing metastatic CRC patients.

Anthropogenic environmental changes may affect community assembly through mediating both deterministic(e.g.,competitive exclusion and environmental filtering)and stochastic processes(e.g.,birth/death and dispersal/colonization).It is traditionally thought that environmental changes have a larger mediation effect on stochastic processes in structuring soil microbial community than aboveground plant community;however,this hypothesis remains largely untested.Here we report an unexpected pattern that nitrogen(N)deposition has a larger mediation effect on stochastic processes in structuring plant community than soil microbial community(thoselia,manipulating nine rates(0–50 g N m^(-2)per year)at two frequencies(nitrogen added twice or 12 times per year)under two grassland management strategies(fencing or mowing).We separated the compositional variation of plant and soil microbial communities caused by each treatment into the deterministic and stochastic components with a recently-developed method.As nitrogen addition rate increased,the relative importance of stochastic component of plant community first increased and then decreased,while that of soil microbial community first decreased and then increased.On the whole,the relative importance of stochastic component was significantly larger in plant community(0.552±0.035;mean±standard error)than in microbial community(0.427±0.035).Consistently,the proportion of compositional variation explained by the deterministic soil and community indices was smaller for plant community(0.172–0.186)than microbial community(0.240–0.767).Meanwhile,as nitrogen addition rate increased,the linkage between plant and microbial community composition first became weaker and then became stronger.The larger stochasticity in plant community relative to microbial community assembly suggested that more stochasti

Microstructure and Properties of AlCoCrFeNiTi High-Entropy Alloy Coatings Prepared by Laser Cladding

21-4N(5Cr21Mn9Ni4N)is extensively employed in the production of engine valves,operating under severe conditions.Apart from withstanding high-temperature gas corrosion,it must also endure the impact of cylinder explosion pressure.The predominant failure mode of 21-4N valves is abrasive wear.Surface coatings serve as an effective approach to prevent such failures.In this investigation,Laser cladding technology was utilized to fabricate AlCoCrFeNiTi high entropy alloy coatings onto the surfaces of 21-4N valves.According to the findings,the cladding zone has a normal dendritic microstructure,a good substrate-to-cladding layer interaction,and no obvious flaws.In terms of hardness,the cladding demonstrates an average hardness of 620 HV.The hardness has increased by 140%compared to the substrate.The average hardness of the cladding remains at approximately 520 HV even at elevated temperatures.Regarding frictional wear performance,between 400℃and 800℃,the cladding layer exhibits an average friction coefficient of 0.4,with the primary wear mechanisms being abrasive wear,adhesive wear,and a minor degree of plastic deformation.

Dual-targeting AAV9P1-mediated neuronal reprogramming in a mouse model of traumatic brain injury

Traumatic brain injury results in neuronal loss and glial scar formation.Replenishing neurons and eliminating the consequences of glial scar formation are essential for treating traumatic brain injury.Neuronal reprogramming is a promising strategy to convert glial scars to neural tissue.However,previous studies have reported inconsistent results.In this study,an AAV9P1 vector incorporating an astrocyte-targeting P1 peptide and glial fibrillary acidic protein promoter was used to achieve dual-targeting of astrocytes and the glial scar while minimizing off-target effects.The results demonstrate that AAV9P1 provides high selectivity of astrocytes and reactive astrocytes.Moreover,neuronal reprogramming was induced by downregulating the polypyrimidine tract-binding protein 1 gene via systemic administration of AAV9P1 in a mouse model of traumatic brain injury.In summary,this approach provides an improved gene delivery vehicle to study neuronal programming and evidence of its applications for traumatic brain injury.
