Synthesis of O-(2.[^(18)F]fluoroethyl)-L-tyrosine and its biological evaluation in B16 melanoma-bearing mice as PET tracer for tumor imaging

O-(2.[18F]fluoroethyl) -L-tyrosine([18F]FET) ,a fluorine-18 labeled analogue of tyrosine,has been syn-thesized and biologically evaluated in tumor-bearing mice. The whole synthesis procedure is com-pleted within 50 min. The radiochemical yield is about 40%(no decay corrected) and radiochemical purity more than 97% after simplified solid phase extraction. [18F]FET shows rapid,high uptake and long retention in the tumor as well as low uptake in the brain. The ratios of tumor-to-muscle(T/M) and tumor-to-blood(T/B) of [18F]FET are similar to those of [18F]FDG,but the ratios of tumor-to-brain(T/Br) are 2.3 times higher than that of [18F]FDG. Autoradiography of [18F]FET demonstrates a remarkable accumulation in melanoma with high contrast. It appears to be a probable competitive candidate for melanoma imaging with PET.

Aim: To investigate the mitochondria-initiated apoptosis pathway involved in Carbon tetrachloride （CCl4） hepatotoxicity in vitro. Methods: Several cytotoxic-ity endpoints, including WST-8 metabolism, lactate dehydrogenase leakage and morphological changes, were examined. The 5,5'-dithio-bis（2.nitrobenzoic acid） reaction was used to measure reduced glutathione level, and the malondialdehyde level was determined using the thiobarbituric acid assay. The release of cyto-chrome c and Bcl-XLwas detected by Western blot. Caspase-3 activity was mea sured using the fluorogenic substrate Ac-DEVD-AMC. DNA fragmentation wasused to evaluate cell apoptosis. Results: A time-and dose-dependent decrease in cellular glutathione content was observed, along with a concomitant increase in malondialdehyde levels following the application of CCl4. Caspase 3 activity was stimulated at all doses of CCl4, with the most significant activation at 3 mmol/L. Cytochrome c was released obviously after CCl4treatment. A time-dependent decrease in BcI-XLexpression was observed. DNA fragmentation results revealed apoptosis and necrosis following CCl4treatment. Conclusion: Oxidative damage is one of the essential mechanisms of CCl4hepatotoxicity, which triggers apoptosis via the mitochondria-initiated pathway.

Temporal lobe epilepsy is a multifactorial neurological dysfunction syndrome that is refractory,resistant to antiepileptic drugs,and has a high recurrence *** pathogenesis of temporal lobe epilepsy is complex and is not fully *** calcium dynamics have been implicated in temporal lobe ***,the effect of fluctuating calcium activity in CA1 pyramidal neurons on temporal lobe epilepsy is unknown,and no longitudinal studies have investigated calcium activity in pyramidal neurons in the hippocampal CA1 and primary motor cortex M1 of freely moving *** this study,we used a multichannel fiber photometry system to continuously record calcium signals in CA1 and M1 during the temporal lobe epilepsy *** found that calcium signals varied according to the grade of temporal lobe epilepsy *** particular,cortical spreading depression,which has recently been frequently used to represent the continuously and substantially increased calcium signals,was found to correspond to complex and severe behavioral characteristics of temporal lobe epilepsy ranging from gradeⅡto gradeⅤ.However,vigorous calcium oscillations and highly synchronized calcium signals in CA1 and M1 were strongly related to convulsive motor *** inhibition of pyramidal neurons in CA1 significantly attenuated the amplitudes of the calcium signals corresponding to gradeⅠ*** addition,the latency of cortical spreading depression was prolonged,and the above-mentioned abnormal calcium signals in CA1 and M1 were also significantly ***,it was possible to rescue the altered intracellular calcium *** simultaneous analysis of calcium signals and epileptic behaviors,we found that the progression of temporal lobe epilepsy was alleviated when specific calcium signals were reduced,and that the end-point behaviors of temporal lobe epilepsy were *** results indicate that the calcium dynamic between CA1 and M1 may reflect sp

This paper presents a graphics processing unit (GPU) based stylization approach that preserves the fine structure between the original and the stylized images using gradient *** abstraction and painterly stylization methods focused on contrast manipulation only,and thus the detailed salient structures of the input images are always destroyed when performing the current stylization techniques because of limitations like unavoidable salience information loss caused by contrast *** propose an image structure map to naturally model the fine structure existing in the original ***-based structure tangent generation and tangent-guided image morphology are used to construct the structure *** image structure map,unlike an edge map,not only systematically models the boundary information within the imagery but also accentuates the underlying inner structure detail for further *** facilitate the final stylization via parallel bilateral grid and structure-aware stylizing optimization on a GPU-CUDA platform in real *** multiple experiments,the proposed method consistently demonstrates efficient and high quality image stylization performance.

Microstructure and Electromechanical Properties in PMnN-PZT Ceramics Sintered at Different Temperatures

The microstructure and piezoelectric properties of Pb[(Zr0.52.i0.48)0.95(Mn1/3Nb2.3)0.05]03 (PMnN-PZT) ceramics have been investigated at different sintering temperatures from 1070℃ to 12.0℃. The experimental results suggest that grain sizes and electromechanical properties of the ceramics show strong sintering temperature dependences. Double polarization to electric field (P-E) loops of the PMnN-PZT ceramics are assumed to be the result of the pinning effect of the defect dipoles. The results obtained through X-ray photoelectron spectrum suggest that sintering temperature affects valence of the Mn ion and quantity of oxygen vacancies. Hence, a proper explanation is proposed to illustrate the fluctuation of mechanical quality factor (Qm) due to the sintering temperature.

Using atomic force microscopy (AFM), the dynamic process of the in vitro nucleosome reconstitution followed by slow dilution from high salt to low salt was visualized. Data showed that the histone octamers were dissociated from DNA at 1M NaCl. When the salt concentration was slowly reduced to 650 mM and 300 mM, the core histones bound to the naked DNA gradually. Once the salt concentration was reduced to 50 mM the classic 'beads-on-astring' structure was clearly visualized. Furthermore, using the technique of the in vitro reconstitution of nucleosome, the mono- and di- nucleosomes were assembled in vitro with both HS2.ore (-10681 to -10970 bp) and NCR2.(-372.to -194 bp) DNA sequences in the 5’flanking sequence of human b-globin gene. Data revealed that HMG 1/2.and HMG 14/17 proteins binding to both DNA sequences are changeable following the assembly and disassembly of nucleosomes. Optical observations of Sco X-1 with exposure times around 0.5 s were performed with the 1.56 m telescope of the shanghai Astronomical observatory in *** these observations,we studied the fluctuation of the optical flux on three short timescales(5,10,100 s).We found that the standard deviations of fluctuations on these three timescales differ from those of Gaussian distributions by 7 Sigma,5 Sigma and 3 Sigma,*** result suggests that the variations in the intensity on these short timescales differ from pure statistical fluctuations,which,we suggest,is due to the intrinsic variabi{2.ty in the optical emission.

We generalize the algebraic method presented by Fan [***.A:***.36 (2.03) 7009)] to uniformlyconstruct a series of soliton-like solutions and double-llke periodic solutions for nonlinear partial differential equations(NPDE),As an application of the method,we choose a (2.1)-dimensional asymmetric Nizhnik-Novikov-Vesselov equa-tion and successfully construct new and more general solutions including a series of nontraveling wave and coefficientfunctions’ soliton-like solutions,double-like periodic and trigonometric-like function solutions.
