Deep tectonics and seismogenic mechanisms of the seismic source zone of the Jishishan M_(s)6.2 earthquake on December 18,2023,at the northeast margin of the Tibetan Plateau

On December 18,2023,an M_(s)6.2 earthquake occurred in Jishishan,Gansu Province,China.This earthquake happened in the eastern region of the Qilian Orogenic Belt,which is situated at the forefront of the NE margin of the Tibetan Plateau(i.e.,Qinghai-Tibet Plateau),encompassing a rhombic-shaped area that intersects the Qilian-Qaidam Basin,Alxa Block,Ordos Block,and South China Block.In this study,we analyzed the deep tectonic pattern of the Jishishan earthquake by incorporating data on the crustal thickness,velocity structure,global navigation satellite system(GNSS)strain field,and anisotropy.We discovered that the location of the earthquake was related to changes in the crustal structure.The results showed that the Jishishan M_(s)6.2 earthquake occurred in a unique position,with rapid changes in the crustal thickness,Vp/Vs,phase velocity,and S-wave velocity.The epicenter of the earthquake was situated at the transition zone between high and low velocities and was in proximity to a low-velocity region.Additionally,the source area is flanked by two high-velocity anomalies from the east and west.The principal compressive strain orientation near the Lajishan Fault is primarily in the NNE and NE directions,which align with the principal compressive stress direction in this region.In some areas of the Lajishan Fault,the principal compressive strain orientations show the NNW direction,consistent with the direction of the upper crustal fast-wave polarization from local earthquakes and the phase velocity azimuthal anisotropy.These features underscore the relationship between the occurrence of the Jishishan M_(s)6.2 earthquake and the deep inhomogeneous structure and deep tectonic characteristics.The NE margin of the Tibetan Plateau was thickened by crustal extension in the process of northeastward expansion,and the middle and lower crustal materials underwent structural deformation and may have been filled with salt-containing fluids during the extension process.The presence of this weak layer makes it easier for strong ea

Hemostatic mechanism of Jianpi Yiqi Shexue decoction(健脾益气摄血方) in treatment of immune thrombocytopenia

OBJECTIVE: To explore the early hemostatic mechanism of Jianpi Yiqi Shexue decoction(健脾益气摄血方, JYSD) in treating immune thrombocytopathy(ITP), based on the functional homeostasis of brain-intestine axis and blood neurotransmitter METHODS: Non-drug treatment cases: Healthy volunteers were selected as normal control group and compared with patients with dysfunctional uterine bleeding, gastrointestinal tumors with bleeding and ITP, to detect the changes of blood 5-hydroxytryptamine(5-HT), β-endorphin(β-EP), vasoactive intestinal peptide(VIP) and compare the changes of blood neurotransmitters in patients with different disease symptoms. Drug treatment cases: According to the randomized controlled multicenter clinical trial, 272 ITP patients were randomly divided into three groups: treatment group(JYSD) combined group(JYSD + Prednisone) control group(Prednisone). The changes of blood neurotransmitter(5-HT, β-EP, VIP) before and after treatment were detected on the basis of peripheral blood platelet(PLT) and grade score. RESULTS: Non-drug treatment cases: compared with the normal control group, the 5-HT level was higher, and the VIP and β-EP levels were both lower in the ITP group(P < 0.001), and the 5-HT, VIP and β-EP levels in the Gastrointestinal tumors with bleeding group were also lower compared with the normal control group(P < 0.05, 0.001). Drug treatment cases: The PLT grading scores of the combination group and the control group after treatment were lower than that before treatment(P < 0.05, 0.001). The PLT grading score of the 3 groups were compared in pairs after treatment: the combination group was the lowest among the 3 groups, which was better than the treatment group, but no better than the control group(vs the treatment group, P = 0.005, vs the control group, P = 0.709). The statistical results of full analysis set(FAS) and per protocol set(PPS) were consistent. Decreased LDHB expression in breast tumor cells causes NK cell activation and promotes tumor progression

Objective: Abnormal metabolism is the underlying reason for breast cancer progression. Decreased lactate dehydrogenase B(LDHB) has been detected in breast cancer but the function of LDHB remains unknown.Methods: Western blot was used to analyze LDHB expression in breast cancer cells. The impact of LDHB on tumor cell migration and invasion was determined using Transwell assays, wound healing assays, and a mouse lung metastasis model. Subcutaneous tumor formation, a natural killer(NK) cell cytotoxicity assay, and flow cytometry evaluated NK cell activation. Immunofluorescence and quantitative real-time PCR detected NK cell activation markers. Kaplan-Meier analysis evaluated the effect of immune cell infiltration on prognosis. Single-sample gene set enrichment analysis determined NK cell activation scores. A support vector machine predicted the role of LDHB in NK cell activation.Results: In this study we showed that LDHB inhibits the breast cancer cell metastasis and orchestrates metabolic reprogramming within tumor cells. Our results revealed that LDHB-mediated lactic acid clearance in breast cancer cells triggers NK cell activation within the tumor microenvironment. Our findings, which were confirmed in a murine model, demonstrated that LDHB in tumor cells promotes NK cell activation and ultimately results in the eradication of malignant cells. Clinically, our study further validated that LDHB affects immune cell infiltration and function. Specifically, its expression has been linked to enhanced NK cell-mediated cytotoxicity and improved patient survival. Furthermore, we identified LDHB expression in tumors as an important predictor of NK cell activation, with strong predictive ability in some cancers.Conclusions: Our results suggest that LDHB is a promising target for activating the tumor immune microenvironment in breast cancer, where LDHB-associated lactic acid clearance leads to increased NK cell activity. ResNeSt-biGRU: An Intrusion Detection Model Based on Internet of Things

The rapid expansion of Internet of Things (IoT) devices across various sectors is driven by steadily increasingdemands for interconnected and smart technologies. Nevertheless, the surge in the number of IoT device hascaught the attention of cyber hackers, as it provides them with expanded avenues to access valuable data. Thishas resulted in a myriad of security challenges, including information leakage, malware propagation, and financialloss, among others. Consequently, developing an intrusion detection system to identify both active and potentialintrusion traffic in IoT networks is of paramount importance. In this paper, we propose ResNeSt-biGRU, a practicalintrusion detection model that combines the strengths of ResNeSt, a variant of Residual Neural Network, andbidirectionalGated RecurrentUnitNetwork (biGRU).Our ResNeSt-biGRUframework diverges fromconventionalintrusion detection systems (IDS) by employing this dual-layeredmechanism that exploits the temporal continuityand spatial feature within network data streams, a methodological innovation that enhances detection accuracy.In conjunction with this, we introduce the PreIoT dataset, a compilation of prevalent IoT network behaviors, totrain and evaluate IDSmodels with a focus on identifying potential intrusion traffics. The effectiveness of proposedscheme is demonstrated through testing, wherein it achieved an average accuracy of 99.90% on theN-BaIoT datasetas well as on the PreIoT dataset and 94.45% on UNSW-NB15 dataset. The outcomes of this research reveal thepotential of ResNeSt-biGRU to bolster security measures, diminish intrusion-related vulnerabilities, and preservethe overall security of IoT ecosystems.

Deformation monitoring of long-span railway bridges based on SBAS-InSAR technology

The deformation monitoring of long-span railway bridges is significant to ensure the safety of human life and property.The interferometric synthetic aperture radar(In SAR)technology has the advantage of high accuracy in bridge deformation monitoring.This study monitored the deformation of the Ganjiang Super Bridge based on the small baseline subsets(SBAS)In SAR technology and Sentinel-1A data.We analyzed the deformation results combined with bridge structure,temperature,and riverbed sediment scouring.The results are as follows:(1)The Ganjiang Super Bridge area is stable overall,with deformation rates ranging from-15.6 mm/yr to 10.7 mm/yr(2)The settlement of the Ganjiang Super Bridge deck gradually increases from the bridge tower toward the main span,which conforms to the typical deformation pattern of a cable-stayed bridge.(3)The sediment scouring from the riverbed cause the serious settlement on the bridge’s east side compared with that on the west side.(4)The bridge deformation negatively correlates with temperature,with a faster settlement at a higher temperature and a slow rebound trend at a lower temperature.The study findings can provide scientific data support for the health monitoring of long-span railway bridges.

Uncertainty quantification of inverse analysis for geomaterials using probabilistic programming

Uncertainty is an essentially challenging for safe construction and long-term stability of geotechnical engineering.The inverse analysis is commonly utilized to determine the physico-mechanical parameters.However,conventional inverse analysis cannot deal with uncertainty in geotechnical and geological systems.In this study,a framework was developed to evaluate and quantify uncertainty in inverse analysis based on the reduced-order model(ROM)and probabilistic programming.The ROM was utilized to capture the mechanical and deformation properties of surrounding rock mass in geomechanical problems.Probabilistic programming was employed to evaluate uncertainty during construction in geotechnical engineering.A circular tunnel was then used to illustrate the proposed framework using analytical and numerical solution.The results show that the geomechanical parameters and associated uncertainty can be properly obtained and the proposed framework can capture the mechanical behaviors under uncertainty.Then,a slope case was employed to demonstrate the performance of the developed framework.The results prove that the proposed framework provides a scientific,feasible,and effective tool to characterize the properties and physical mechanism of geomaterials under uncertainty in geotechnical engineering problems.

Five-view three-dimensional reconstructionfor ultrafast dynamic imaging of pulsedradiation sources

Multiaxial neutron/x-ray imaging and three-dimensional (3D) reconstruction techniques play a crucial role in gaining valuable insights intothe generation and evolution mechanisms of pulsed radiation sources. Owing to the short emission time (∼200 ns) and drastic changes of thepulsed radiation source, it is necessary to acquire projection data within a few nanoseconds in order to achieve clear computed tomography3D imaging. As a consequence, projection data that can be used for computed tomography image reconstruction at a certain moment are oftenavailable for only a few angles. Traditional algorithms employed in the process of reconstructing 3D images with extremely incomplete datamay introduce significant distortions and artifacts into the final image. In this paper, we propose an iterative image reconstruction methodusing cylindrical harmonic decomposition and a self-supervised denoising network algorithm based on the deep image prior method. Weaugment the prior information with a 2D total variation prior and a 3D deep image prior. Single-wire Z-pinch imaging experiments have beencarried out at Qin-1 facility in five views and four frames, with a time resolution of 3 ns for each frame and a time interval of 40 ns betweenadjacent frames. Both numerical simulations and experiments verify that our proposed algorithm can achieve high-quality reconstructionresults and obtain the 3D intensity distribution and evolution of extreme ultraviolet and soft x-ray emission from plasma.

Highly selective photocatalytic oxidation of methane to methyl hydroperoxide

Partial oxidation of methane into primary oxidation products with high value remains a challenge.In this work,photocatalytic oxidation of methane(CH_(4))with high methyl hydroperoxide(CH_(3)OOH)selectivity is achieved using pure titanium oxide(TiO_(2))without any cocatalyst at room temperature and atmospheric pressure.The CH_(3)OOH production rate can reach up to 2050±88μmol·g^(-1)·h^(-1) at pH≈7.0 with 100%selectivity in the liquid product.The stable reaction cycle can reach more than 30 times.This low-cost system achieves superior CH_(4) conversion activity and selectivity compared with similar work.The energy of hydrogen peroxide(H_(2)O_(2))to adsorbed hydroperoxyl radical(^(*)OOH)has a significantly lower reaction energy than conversion to adsorbed hydroxyl radical(*OH)on the(210)surface of the TiO_(2).The^(*)OOH preferentially combines with methyl radical(·CH_(3))to form the most energetically favorable CH_(3)OOH.The mild oxidative environment of this system prevents the reduction of CH_(3)OOH to CH_(3)OH or over-oxidation of CH_(4),which ensures the final CH_(3)OOH with high selectivity and stability.This work provided a low-cost but highly efficient method to achieve partial oxidation with superior selectivity,i.e.,to convert CH_(4) into high-value chemicals.

Presence of a long nuclear-localization signal sequence in homeodomain transcription factor Nkx 1.2

Homeodomains,a 60-amino acid sequence encoded by 180 nucleotides,are highly conserved DNA-binding motifs that are present in a variety of transcription factors in species ranging from yeast to humans.The NKX proteins belong to the homeodomain(HD)-containing transcription factor family.They play vital roles in the regulation of morphogenesis.NKX1-2 is one member of the NKX subfamily.At present,information about its nuclear localization signal(NLS)sequence is limited.We studied the NLS sequence of zebrafish Nkx1.2 by introducing sequence changes such as deletion,mutation,and truncation,and identified an NLS motif(QNRRTKWKKQ)that is localized at the C-terminus of the homeodomain.Moreover,the deletion of two amino acid residues(RR)in this NLS motif prevents Nkx1.2 from entering the nucleus,indicating that the two amino acids are essential for Nkx1.2 nuclear localization.However,the NLS motif alone is unable to target cytoplasmic protein glutathione S-transferase(GST)to the nucleus.An intact homeodomain is necessary for mediating the complete nuclear transport of cytoplasmic protein.Unlike most nuclear import proteins with short NLS sequences,a long NLS is present in zebrafish Nkx1.2.We also demonstrated that the sequences of homeodomain of NKX1.2 are well conserved among different species.This study is informative to verify the function of the NKX1.2 protein.

The RNA m^(6)A demethylase ALKBH5 drives emergency granulopoiesis and neutrophil mobilization by upregulating G-CSFR expression

Emergency granulopoiesis and neutrophil mobilization that can be triggered by granulocyte colony-stimulating factor(G-CSF)through its receptor G-CSFR are essential for antibacterial innate defense.However,the epigenetic modifiers crucial for intrinsically regulating G-CSFR expression and the antibacterial response of neutrophils remain largely unclear.N6-methyladenosine(m^(6)A)RNA modification and the related demethylase alkB homolog 5(ALKBH5)are key epigenetic regulators of immunity and inflammation,but their roles in neutrophil production and mobilization are still unknown.We used cecal ligation and puncture(CLP)-induced polymicrobial sepsis to model systemic bacterial infection,and we report that ALKBH5 is required for emergency granulopoiesis and neutrophil mobilization.ALKBH5 depletion significantly impaired the production of immature neutrophils in the bone marrow of septic mice.In addition,Alkbh5-deficient septic mice exhibited higher retention of mature neutrophils in the bone marrow and defective neutrophil release into the circulation,which led to fewer neutrophils at the infection site than in their wild-type littermates.During bacterial infection,ALKBH5 imprinted production-and mobilization-promoting transcriptome signatures in both mouse and human neutrophils.Mechanistically,ALKBH5 erased m^(6)A methylation on the CSF3R mRNA to increase the mRNA stability and protein expression of G-CSFR,consequently upregulating cell surface G-CSFR expression and downstream STAT3 signaling in neutrophils.The RIP-qPCR results confirmed the direct binding of ALKBH5 to the CSF3R mRNA,and the binding strength declined upon bacterial infection,accounting for the decrease in G-CSFR expression on bacteria-infected neutrophils.Considering these results collectively,we define a new role of ALKBH5 in intrinsically driving neutrophil production and mobilization through m^(6)A demethylation-dependent posttranscriptional regulation,indicating that m^(6)A RNA modification in neutrophils is a potential target for treating bacteria
