Diosgenin, a steroidal sapogenin, obtained from Trigonella foenum-graecum, Dioscorea, and Rhizoma polgonati, has shown high potential and interest in the treatment of various cancers, such as oral squamous cell carcinoma, laryngeal cancer, esophageal cancer, liver cancer, gastric cancer, lung cancer, cervical cancer, prostate cancer, glioma, and leukemia. This article aims to provide an overview of the in vivo, in vitro,and clinical studies reporting the diosgenin’s anticancer effects. Preclinical studies have shown promising effects of diosgenin on inhibiting tumor cell proliferation and growth, promoting apoptosis, inducing differentiation and autophagy, inhibiting tumor cell metastasis and invasion, blocking cell cycle, regulating immunity and improving gut microbiome. Clinical investigations have revealed clinical dosage and safety property of diosgenin. Furthermore, in order to improve the biological activity and bioavailability of diosgenin, this review focuses on the development of diosgenin nano drug carriers, combined drugs and the diosgenin derivatives. However, further designed trials are needed to unravel the diosgenin’s deficiencies in clinical application.

The Characteristics of Turbulence Structure and Transfer over the Middle Area of the Tibetan Plateau

In this paper, turbulent data obtained from the Damxung site during the Secondary Tibetan Plateau Science Experiment (TIPEX) in 1998 are used to study the characteristics of the turbulent spectra, turbulence transport, and the dissipation rates of turbulent kinetic energy, temperature variance, and humidity variance in the middle area of the Tibetan Plateau. The turbulent spectra of wind velocity, potential temperature, and humidity satisfy the-2/3 power law in the high frequency range. Horizontal transportation of heat and water vapor is negligible compared with vertical transportation under strong unstable conditions, and as the stability parameter z/L increases (where z is the observational height, and L is the Monin Obukhov length), horizontal transportation becomes dominant under near-neutral, neutral, and stable conditions. The non-dimensional temperature and humidity variances are 20% less than the temperature and humidity gradient variances. These deficits appear to increase as the absolute stability parameter increases. Moreover, the effects of turbulence transportation and pressure variance exist throughout the entire stability region.

Biotic and abiotic stress-responsive genes are stimulated to resist drought stress in purple wheat

Triticum aestivum L. cv. Guizi 1(GZ1) is a drought-tolerant local purple wheat cultivar. It is not clear how purple wheat resists drought stress, but it could be related to anthocyanin biosynthesis. In this study, transcriptome data from droughttreated samples and controls were compared. Drought slightly reduced the anthocyanin, protein and starch contents of GZ1 grains and significantly reduced the grain weight. Under drought stress, 16 682 transcripts were reduced, 27 766 differentially expressed genes(DEGs) were identified, and 379 DEGs, including DREBs, were related to defense response. The defense-response genes included response to water deprivation, reactive oxygen, bacteria, fungi, etc. Most of the structural and regulatory genes in anthocyanin biosynthesis were downregulated, with only Ta DFR, Ta OMT, Ta5,3GT, and Ta MYB-4 B1 being upregulated. Ta CHS, Ta F3H, TaCHI, Ta4CL, and TaF3’H are involved in responses to UV, hormones, and stimulus. Ta CHS-2D1, Ta DFR-2D2, Ta DFR-7D, TaOMT-5A, Ta5,3 GT-1B1, Ta5,3GT-3A, and Ta5,3GT-7B1 connect anthocyanin biosynthesis with other pathways, and their interacting proteins are involved in primary metabolism, genetic regulation, growth and development, and defense responses. There is further speculation about the defense-responsive network in purple wheat. The results indicated that biotic and abiotic stress-responsive genes were stimulated to resist drought stress in purple wheat GZ1, and anthocyanin biosynthesis also participated in the drought defense response through several structural genes.

盐酸利托君、间苯三酚、硫酸镁用于孕20周后先兆流产患者的临床研究

目的观察盐酸利托君、间苯三酚、硫酸镁对孕20周后先兆流产患者血清性激素水平和保胎效果的影响。方法将保胎治疗的先兆流产(孕20周后)患者按队列法分为A组(予以盐酸利托君注射液100 mg+5%葡萄糖注射液500 mL,静脉滴注,宫缩抑制后持续滴注12~18 h,之后改为口服盐酸利托君片)、B组(予以间苯三酚注射液40 mg+5%葡萄糖葡萄糖注射液500 mL,静脉滴注,宫缩抑制后停药)和C组(予以硫酸镁注射液20 mL+5%葡萄糖注射液100 mL,快速静脉滴注后再次给予硫酸镁注射液40 mL+5%葡萄糖注射液500 mL,静脉滴注,宫缩抑制后持续滴注12 h)。观察3组患者起效时间和宫缩消失时间、血清性激素水平[孕酮(P)、雌二醇(E2)、人绒毛膜促性腺激素β-亚基(β-hCG)]、药物不良反应、保胎有效率。结果A组40例,B组38例,C组42例。A组、B组和C组的起效时间分别为(1.71±0.34)、(2.29±0.23)和(4.51±1.12)h,组间两两比较在统计学上差异均有统计学意义(均P<0.05)。A组、B组和C组的宫缩消失时间分别为(1.34±0.32)、(2.24±0.26)和(2.36±0.28)d,B组、C组与A组比较在统计学上差异均有统计学意义(P<0.05)。治疗3 d后,A组、B组和C组的血清P水平分别为(78.64±10.34)、(69.35±10.52)和(68.76±11.13)ng·mL^(-1),E2水平分别为(672.25±85.63)、(623.25±92.31)和(624.12±93.65)pg·mL^(-1),β-hCG水平分别为(6.95×10^(4)±1258.65)、(6.75×10^(4)±1274.43)和(6.70×10^(4)±1327.59)mU·mL^(-1),B组、C组与A组比较在统计学上差异均有统计学意义(均P<0.05)。A组、B组、C组的心悸发生率分别为25.00%、0、9.52%,A组与B组比较在统计学上差异均有统计学意义(均P<0.05);头痛发生率分别为2.50%、2.63%、26.19%,C组与A组和B组比较在统计学上差异均有统计学意义(均P<0.05);乏力的发生率分别为5.00%、0、19.05%,C组与B组比较在统计学上差异均有统计学意义(均P<0.05);胃肠道不适的发生率分别为5.00%、0、11.90%,C组与B组比较在统计学上差异有统计学意义(P<0.05)。A组、B组和C组的保胎有效率分别为92.50%、94.74%和73.81%,A组、B组与C组比较在统计学上差异均有统计学意义(均P<0.05)。结论盐酸利托君的起效时间短,可作为控制病情的首选,间苯三酚在保胎效果上与盐酸利托君相当,且在药物不良反应中具有优势,临床对于盐酸利托君使用耐受不佳的患者可优先考虑间苯三酚。

Effects of salinity and desalination on seed germination of six annual weed species

The effects of various salinities and desalination on seed germination of six annual glycophytes （Artemisia sieversiana, A. scoparia, Chloris virgata, Eragrostis pilosa, Chenopodium acuminatum and Chenopodium glaucum） were studied in Horqin Sandy land, Inner Mongolia, China. NaCl solutions of five concentrations （0 mM, as the control, and 50, 100, 200 and 300 mM） were used for saline stress and desalination treatments. Increasing salinity significantly reduced germination percentages of A. sieversiana, A. scoparia, Ch.virgata and Ch. acuminatum, but had no effect on the germination percentages of E. pilosa. Lower salinity levels （50 mM） significantly increased germination percentage of Ch. glaucum. High salinity might be a precondition for germi- nation after desalination for five of the six species, excepting E. pilosa at NaCl concentration of 300 mM in comparison with non-primed seeds. Higher salinity （200 mM） led to some specific ion toxicity and reduced seed viability of A. sieversiana. No specific ion toxicity but an osmotic effect limited the germination of other five species was observed The final germination percentages （salinity stress and desalination） of the six species showed three variations in comparison with the controls, namely, indiscrimination, stimulation, and reduction. Germination responses to salinity and desalination suggested that the six species were separated into three categories. Three species （A. sieversiana, Ch. virgata and Ch. acuminatum） showed similar germination responses to salinity with those of halophyte, but also showed a lower tolerance limit than most halophytes, although this was not always the case. A. scoparia and Ch. glaucum exhibited some ‘salt stimulation’ in seed germination percentages after desalination, whereas E. pilosa did not show any obvious responseto salinity. Therefore, salinity usually induces dormancy of seeds with strong germination capacity in fresh water, but has few, or even positive, effects on seeds with strong innate dormancy.

为了验证GB/T 40904—2021《家具产品及其材料中禁限用物质测定方法偶氮染料》中检测分析24种禁限用芳香胺方法的适用性,本文采用叔丁基甲醚为提取溶剂,绘制标准曲线,确定方法的定量限和检出限,用气相色谱-质谱联用仪对24种禁限用芳香胺进行分析,最后用样品进行重复性和精密度测试验证。分析结果得出:24种禁限用芳香胺在3~36 mg/L线性关系良好,线性相关系数R2>0.995;检出限为5 mg/kg,定量限为15 mg/kg;2,4-二氨基苯甲醚回收率为51.4%,邻甲苯胺、2,4-二氨基甲苯、2,4-二甲基苯胺、2,6-二甲基苯胺回收率范围为65.8%~70.8%,其余的芳香胺回收率范围在71.6%~90.5%,重复性测试的RSD为0.4%~5.6%。结果表明该标准方法精密度和准确度符合要求,方法操作简单,结果复现性好。

Three-stage niobium mineralization at Bayan Obo, China

The Chinese Bayan Obo deposit is a world-class rare earth element(REE) deposit with considerable niobium(Nb) and iron(Fe) resources. A complete genetic understanding on all metals is fundamental for establishing genetic models at Bayan Obo. With extensive research being focused on REE enrichment, the timing and controls of Nb enrichment remain unresolved at Bayan Obo, which is mainly due to the challenges in dating, i.e. multistage thermal events, fine-grained minerals with complex textures and the rare occurrence of uranium-enriched minerals with mature dating methods. Based on robust geological and petrographic frameworks, here we conducted ion probe uranium-lead(U-Pb) dating of ferrocolumbite to unravel the timing, hence the genesis of Nb mineralization. Three types of hydrothermal ferrocolumbites—key Nb-bearing minerals—are identified based on their textures and mineral assemblages. They yield U-Pb ages of 1312 ± 47 Ma(n = 99), 438 ± 7 Ma(n = 93), and 268 ± 5 Ma(n = 19), respectively. In line with deposit geology, we tentatively link the first, second and third stage Nb mineralization to Mesoproterozoic carbonatite magmatism, ubiquitous early Paleozoic hydrothermal activity, and Permian granitic magmatism, respectively. While quantifying the contribution of metal endowment from each stage requires further investigation, our new dates highlight that multi-stage mineralization is critical for Nb enrichment at Bayan Obo, which may also have implications for the enrichment mechanism of Nb in REE deposits in general.

WNT7A promotes tumorigenesis of head and neck squamous cell carcinoma via activating FZD7/JAK1/STAT3 signaling

Wnt signaling are critical pathway involved in organ development, tumorigenesis, and cancer progression. WNT7A, a member of the Wnt family, remains poorly understood in terms of its role and the underlying molecular mechanisms it entails in head and neck squamous cell carcinoma (HNSCC). According to the Cancer Genome Atlas (TCGA), transcriptome sequencing data of HNSCC, the expression level of WNT7A in tumors was found to be higher than in adjacent normal tissues, which was validated using Real-time RT-PCR and immunohistochemistry. Unexpectedly, overexpression of WNT7A did not activate the canonical Wnt-β-catenin pathway in HNSCC. Instead, our findings suggested that WNT7A potentially activated the FZD7/JAK1/STAT3 signaling pathway, leading to enhanced cell proliferation, self-renewal, and resistance to apoptosis. Furthermore, in a patient-derived xenograft (PDX) tumor model, high expression of WNT7A and phosphorylated STAT3 was observed, which positively correlated with tumor progression.These findings underscore the significance of WNT7A in HNSCC progression and propose the targeting of key molecules within the FZD7/JAK1/STAT3 pathway as a promising strategy for precise treatment of HNSCC.

目的探究基于磁共振三维快速液体衰减反转恢复序列(3D-FLAIR MRI)检查评估突发性聋(ISNHL)患者耳聋分级、分型及预后的关系研究。方法选择2020年4月至2023年4月来我院就诊的突发性耳聋(ISNHL)患者328例,患者均行3D-FLAIR MRI检测。收集受试者的一般资料及影像学结果,根据患者的高信号区变化情况分为A组196例(内耳正常、无异常信号显示)、B组73例(表现为高信号吸收)及C组59例(表现高信号未吸收),记录三组受试者的耳聋分级、分型,并进行针对性治疗分析三组预后情况。结果三组的性别、年龄、病程比较,差异无统计学意义(P>0.05),三组的伴随头晕/眩晕、前庭功能异常比例及治疗天数比较,差异有统计学意义(P0.05),A组的痊愈高于B组及C组(P<0.05)。结论3D-FLAIR MRI检查能够有助于临床判断ISNHL患者耳聋分级、分型,并与预后关系密切。
