In order to investigate the biodiversity of free-living nematodes along the coast of the Yellow Sea,sediment samples were collected in a number of intertidal sites in recent years.In this area,the mean abundance of free-living nematodes is 1663±502 ind 10 cm^(-2),and more than 200 species have been found(Zhai et al.,2020;Hao et al.,2021,2022;Chu et al.,2023).Among them,species from the genera,Daptonema,Thalassomonhystera,Parodontophora and Bathylaimus,are the most common,while the species,Daptonema parabreviseta Huang&Sun,Parodontophora deltensis Zhang,Bathylaimus huanghaiensis Huang&Zhang and Thalassomonhystera siamensis Kito&Aryuthaka are in dominant.Here,a species belongs to the genus Ethmolaimus is identified as new to science,namely E.multispiralis sp.nov.It is also the first time for the genus reported in the Chinese sea area.

Premature ovarian insufficiency(POI)is a heterogeneous female disorder characterized by the loss of ovarian function before the age of 40.It represents a significant detriment to female fertility.However,the known POI-causative genes currently account for only a fraction of cases.To elucidate the genetic factors underlying POI,we conducted whole-exome sequencing on a family with three fertile POI patients and identified a deleterious missense variant in RNF111.In a subsequent replication study involving 1,030 POI patients,this variant was not only confirmed but also accompanied by the discovery of three additional predicted deleterious RNF111 variants.These variants collectively account for eight cases,representing 0.78%of the study cohort.A further study involving 500 patients with diminished ovarian reserve also identified two additional RNF111 variants.Notably,RNF111 encodes an E3 ubiquitin ligase with a regulatory role in the TGF-β/BMP signaling pathway.Our analysis revealed that RNF111/RNF111 is predominantly expressed in the oocytes of mice,monkeys,and humans.To further investigate the functional implications of RNF111 variants,we generated two mouse models:one with a heterozygous missense mutation(Rnf111+/M)and another with a heterozygous null mutation(Rnf111^(+/-)).Both mouse models exhibited impaired female fertility,characterized by reduced litter sizes and small ovarian reserve.Additionally,RNA-seq and quantitative proteomics analysis unveiled that Rnf111 haploinsufficiency led to dysregulation in female gonad development and negative regulation of the BMP signaling pathway within mouse ovaries.In conclusion,our findings strongly suggest that monoallelic deleterious variants in RNF111 can impair female fertility and induce POI in both humans and mice.

The interaction among type Ⅱ collagen(CⅡ),human DR4 major histocompatibility complex type Ⅱ molecule(MHC Ⅱ)and T-cell receptor(TCR)is associated with the development of rheumatoid arthritis(RA).The activation of T cells can be reduced through exposure to modified CⅡ(263-272)glycopeptide fragment via competitive inhibition with self-antigen.In this work,30 peptides based on the sequence of CⅡ(263-272)were prepared and evaluated for their binding to DR4 protein by surface plasmon resonance(SPR)assay.The effect on the secretion of pro-inflammatory factors by the spleen cells in collagen induced rheumatoid arthritis(CIA)mouse was also investigated.Two N-glycosylated CⅡ peptides were identified to have strong binding to the human recombinant DR4 protein and weak proinflammatory effect.These glycopeptides could be developed as therapeutic saccharide vaccines for the treatment of rheumatoid arthritis(RA).

Predictive cruise control for heavy trucks based on slope information under cloud control system

With the advantage of fast calculation and map resources on cloud control system(CCS), cloud-based predictive cruise control(CPCC) for heavy trucks has great potential to improve energy efficiency, which is significant to achieve the goal of national carbon neutrality. However, most investigations focus on the on-board predictive cruise control(PCC) system,lack of research on CPCC architecture under CCS. Besides, the current PCC algorithms have the problems of a single control target and high computational complexity, which hinders the improvement of the control effect. In this paper, a layered architecture based on CCS is proposed to effectively address the realtime computing of CPCC system and the deployment of its algorithm on vehicle-cloud. In addition, based on the dynamic programming principle and the proposed road point segmentation method(RPSM), a PCC algorithm is designed to optimize the speed and gear of heavy trucks with slope information. Simulation results show that the CPCC system can adaptively control vehicle driving through the slope prediction, with fuel-saving rate of 6.17% in comparison with the constant cruise control. Also,compared with other similar algorithms, the PCC algorithm can make the engine operate more in the efficient zone by cooperatively optimizing the gear and speed. Moreover, the RPSM algorithm can reconfigure the road in advance, with a 91% roadpoint reduction rate, significantly reducing algorithm complexity.Therefore, this study has essential research significance for the economic driving of heavy trucks and the promotion of the CPCC system.

Zeolite-based catalyst hydrocracking of plastics is a potential strategy for mitigating the environmental impacts of plastic wastes and recycling valuable resources,but difficult mass transfer,low concentration of acid sites,and high cost are still barriers to their practical applications.In this paper,we report an excellent hydrocracking catalyst of ZSM-5 nanosheets(Ce/b-ZSM-5)modified by Ce species with high conversion up to 96.3%,C_(3)−C_(5)selectivity up to 80.9%,and good stability during the hydrogenation of low-density polyethylene.Through comprehensive studies,b-ZSM-5 shows higher molecular diffusion efficiency and acid site concentrations compared with normal ZSM-5(n-ZSM-5)and hollow ZSM-5(h-ZSM-5).The introduction of Ce species into b-ZSM-5 further increases the density of Brønsted(B)and Lewis(L)acid sites as active sites,which enhances the adsorption of substrates and facilitates the formation of intermediates and desorption of products.As a result,the hydrocracking activity of Ce/b-ZSM-5 is significantly improved.

During spermiogenesis,haploid spermatids undergo dramatic morphological changes to form slender sperm flagella and cap-like acrosomes,which are required for successful fertilization.Severe deformities in flagella cause a male infertility syndrome,multiple morphological abnormalities of the flagella(MMAF),while acrosomal hypoplasia in some cases leads to sub-optimal embryonic developmental potential.However,evidence regarding the occurrence of acrosomal hypoplasia in MMAF is limited.Here,we report the generation of base-edited mice knocked out for coiled-coil domain-containing 38(Ccdc38)via inducing a nonsense mutation and find that the males are infertile.The Ccdc38-KO sperm display acrosomal hypoplasia and typical MMAF phenotypes.We find that the acrosomal membrane is loosely anchored to the nucleus and fibrous sheaths are disorganized in Ccdc38-KO sperm.Further analyses reveal that Ccdc38 knockout causes a decreased level of TEKT3,a protein associated with acrosome biogenesis,in testes and an aberrant distribution of TEKT3 in sperm.We finally show that intracytoplasmic sperm injection overcomes Ccdc38-related infertility.Our study thus reveals a previously unknown role for CCDC38 in acrosome biogenesis and provides additional evidence for the occurrence of acrosomal hypoplasia in MMAF.

Root Endophyte Shift and Key Genera Discovery in Rice under Barnyardgrass Stress

Despite increasing knowledge of barnyardgrass(Echinochloa crus-galli) interference with rice, relatively little is known how endophytes improve the ability of rice against barnyardgrass stress. Here, we provided a detailed temporal characterization of rice root-associated microbiomes during co-cultivation with barnyardgrass and a comparison with the microbiomes of weed-free rice plants. Alpha diversity analysis indicated that barnyardgrass had the opposite effects on endophytic bacteria and fungi in rice roots, in terms of the community diversity, richness and coverage at the rice seedling stage. Principal coordinate analysis showed that barnyardgrass had only a minor effect on the community composition of endophytes in rice roots at the rice seedling stage, but showed a significant and maximum interference at the heading stage. Rice recruited many endophytes to resist biotic stress from barnyardgrass, especially for fungi. PICRUSt(phylogenetic investigation of communities by reconstruction of unobserved states) predictive analysis indicated that 23 metabolic pathways of bacteria were overrepresented in rice. In addition, the main trophic mode of fungi was pathotroph according to FUNGuild analysis. A positive correlation between bacteria and fungi in rice roots was found via network analysis. Anaeromyxobacter, Azospira and Pseudolabrys were the vital bacteria, Phaeosphaeria and Funneliformis were the dominant fungi in maintaining the stability of the ecological network. These results provided data and a theoretical basis for the in-depth understanding of what role endophytes play in rice resistance to barnyardgrass stress and will have implications on improving the resistance of rice against biotic stress using root microbiota.

创新生态系统是加快形成区域高质量发展经济格局的重要支撑,加速释放的数字生产力为区域创新带来了巨大红利。基于2013—2019年的省级面板数据,使用多重中介模型、调节模型、门槛回归模型及空间模型,结合生态位理论和协同理论等,在创新生态系统视角下探究数字技术与区域创新绩效的关系,得出以下结论:(1)创新生态系统视角下数字技术能够显著提高区域创新绩效,且对技术子系统绩效的影响最大,其次是商业子系统和科学子系统。(2)数字技术影响区域创新绩效的路径中,各子系统的生态位适宜度和子系统间协同不仅存在并行中介作用,也存在链式中介作用。(3)技术子系统和商业子系统的生态位宽度能够正向调节数字技术和区域创新绩效的关系。(4)数字技术发展强化了创新极化的格局。本文拓展了数字技术驱动区域创新相关研究的理论视角,也为各区域通过数字技术塑造多维生态优势提供了参考与启示。

Background:The resurgence of seasonal influenza virus circulation has been seen in 2021-2022 after the tempo-rary suppression in 2020-2021.Neuraminidase inhibitors(NAIs)are widely applied in the clinical treatment of influenza A despite several limitations.Objective:To access the efficacy of Xu’s influenza decoction(XID)in combination therapy with oseltamivir for the treatment of influenza A.Methods:In this retrospective cohort study,the eligible participants were diagnosed with influenza A between June 1,2018,and May 30,2022,in the First Affiliated Hospital of Zhejiang Chinese Medical University.According to whether Xu’s influenza decoction was applied,patients were divided into two groups:treated with or without XID.Propensity score matching(PSM)was used to further adjust the covariates between groups.The primary outcome was to compare time to defervescence via K-M curves,Breslow tests,and Cox regression analysis.In Cox proportional hazards model,a univariate analysis was performed to obtain preliminary results,while a further multivariate analysis was conducted to study the independent factors that influence defervescence.Subgroup analysis was conducted according to body temperature and time from onset to admission.The secondary outcome consisted of routine blood and C-reactive protein(CRP),length of stay,and medical costs.Results:A total of 336 patients with influenza A were enrolled in this study(i.e.,163 patients in the XID+oseltamivir group;173 patients in the oseltamivir group).After 1:1 matching via PSM,230 patients meeting the criteria were included in the analysis,with 115 in each arm.The XID+oseltamivir group had shorter time to defervescence(36 h vs 44 h,P=0.011),shorter length of stay(3 days vs 4 days,P=0.018),and higher defervescence possibility(HR=1.384,95%CI:1.054-1.818).Subgroup analysis indicated that for patients during non-window period(≥48 h)with medium-grade fever(38.1℃-39℃),the XID+oseltamivir combination therapy reduced time to defervescence(P=0.04995/0.004)with a higher defervescence pos
