Tr.nsp.sable elements(TEs)constitute the most abundant p.r.ions of p.ant genomes and can dr.matically shap. host genomes dur.ng p.ant *** also p.ay imp.r.ant r.les in cr.p.***,whether.TEs themselves ar. also selected dur.ng cr.p.domestication has r.mained ***,we identify an active long ter.inal r.p.at(LTr.r.tr.tr.nsp.son,HUO,as a p.tential tar.et of selection dur.ng r.ce domestication and *** is a low-cop.-number.LTr.r.tr.tr.nsp.son,and is active under.natur.l gr.wth conditions and tr.nsmitted thr.ugh male gametogenesis,p.efer.ntially inser.ing into genomic r.gions cap.ble of *** exists in all wild r.ce accessions and about half of the ar.haeological r.ce gr.ins(1200–7000 year. ago)and landr.ces sur.eyed,but is absent in almost all moder. var.eties,indicating its gr.dual elimination dur.ng r.ce domestication and *** analyses showed that HUO is subjected to str.ct gene silencing thr.ugh the r.A-dir.cted DNA methylation *** r.sults also suggest that multip.e HUO cop.es may tr.gger.genomic instability thr.ugh alter.ng genome-wide DNA methylation and small r.A biogenesis and changing global gene exp.ession,r.sulting in decr.ased disease r.sistance and yield,coinciding with its elimination dur.ng r.ce ***,our.study suggests that negative selection of an active r.tr.tr.nsp.son might be imp.r.ant for.genome stability dur.ng cr.p.domestication and br.eding.

In or.er.to enhance the glutathione p.r.xidase（Gp.） catalytic activity of the selenium-containing single-chain var.able fr.gments（Se-scFv）, a novel human scFv was designed on the basis of the str.ctur. of human antibody and op.imized via bioinfor.atics methods such as homologous sequence analysis, thr.e-dimensional（3D） model building, binding-site analysis and docking. The DNA sequence of the new human scFv was synthesized and cloned into the exp.ession vector.p.T22b（＋）, then the scFv p.otein was exp.essed in soluble for. in Escher.chia coli BL21（DE3） and p.r.fied by Ni2＋-immobilized metal affinity chr.matogr.p.y（IMAC）. The ser.ne r.sidue of scFv in the active site was conver.ed into selenocysteine（Sec） with the chemical modification method, thus, the human Se-scFv with Gp. activity was obtained. The Gp. activity of the Se-scFv p.otein was char.cter.zed. Comp.r.d with other.Se-scFv, the new human Se-scFv showed similar.efficiency for.catalyzing the r.duction of hydr.gen p.r.xide by glutathione. It exhibited p. and temp.r.tur. dep.ndent catalytic activity and a typ.cal p.ng-p.ng kinetic mechanism.

Autism sp.ctr.m disor.er.(ASD) is a neur.develop.ental disor.er.with consider.ble clinical and genetic *** this study,we identified all classes of genomic var.ants fr.m whole-genome sequencing (WGS) dataset of 32 Chinese tr.os with ASD,including de novo mutations,inher.ted var.ants,cop. number.var.ants (CNVs) and genomic str.ctur.l var.ants.A higher.mutation r.te (p.isson test,p.2.2×10^(-16)) in exonic (1.37×10^(-8)) and 3'-UTr.r.gions (1.42×10^(-8)) was r.vealed in comp.r.son with that of whole genome (1.05×10^(-8)).Using an integr.ted model,we identified 87 p.tentially r.sk genes (p.0.01) fr.m 4832 genes har.or.ng var.ous r.r. deleter.ous var.ants,including CHD8 and Nr.N2,imp.ying that the disor.er. may be in favor.to *** p.r.icular.fr.quent r.r. inher.ted mutations of sever.l micr.cep.aly-associated genes (ASp.,WDr.2,and ZNF335)wer. found in *** chr.mosomal str.ctur. analyses,we found four.de novo CNVs and one de novo chr.mosomal r.ar.angement event,including a de novo dup.0}cation of UBE3A-containing r.gion at 15q11.2-q13.1,which causes Angelman syndr.me and micr.cep.aly,and a disr.p.ed TNr.due to de novo chr.mosomal tr.nslocation t (1;5) (q25.1;q33.2).Taken together.our.r.sults suggest that abnor.alities of centr.somal function and chr.matin r.modeling of the micr.cep.aly-associated genes may be imp.0}cated in p.thogenesis of *** of WGS as a new yet efficient technique to illustr.te the full genetic sp.ctr.m in comp.ex disor.er.,such as ASD,could p.ovide novel insights into p.thogenesis,diagnosis and tr.atment.

基于长三角典型城市大气VOCs排放清单识别的8个VOCs主要排放行业,选择11家代表性企业,实测研究了VOCs治理装置、排放现状、排放组成特征,并计算相关行业排放的臭氧生成潜势.结果表明,不同净化技术对非甲烷总烃(NMHC)的去除效率差异大,存在净化后浓度增加的现象,目前的环保装置对废气的处理有待优化.本次采样的大部分企业存在NMHC、苯、甲苯、二甲苯超标现象,其中甲苯的超标情况最严重.对于筛选的8个主要行业,芳香烃和含氧VOCs,是最主要的排放化合物,芳香烃是对臭氧生成贡献最大的化合物.在不同行业中,VOCs组成存在显著差异,因此在制定VOCs减排控制措施时,应优先减排对臭氧生成贡献大的行业.
