BACKGr.UND: Hep.tic ischemia/r.p.r.usion injur. may induce intestinal micr.flor. imbalance. Salvia miltior.hiza is effective in p.omoting blood cir.ulation and counter.cting p.r.xidation in tissues. The aim of the p.esent study was to deter.ine the effects of Salvia miltior.hiza on intestinal mi- cr.flor., endotoxemia, and bacter.al tr.nslocation in r.ts with hep.tic I/r.injur.. METHODS: Sp.ague-Dawley r.ts in sp.cific p.thogen fr.e gr.de wer. divided into 3 gr.up.: gr.up.I(n =6) for.sham op.r.tion: gr.up. ( n = 7) for.liver.ische- mia for.20 minutes and r.p.r.usion for.22 hour.. Gr.up.was also p.etr.ated with 4 ml/day of Salvia miltior.hiza solu- tion (250 mg/kg) by daily gavage for.7 days. The levels of ser.m alanine aminotr.nsfer.se (ALT), asp.r.ate amino- tr.nsfer.se (AST), malondialdehyde ( MDA) and sup.r.- xide dismutase ( SOD ) in liver.tissues, ser.m endotoxin, intestinal bacter.al counts, intestinal mucosal histology and bacter.al tr.nslocation wer. studied. r.SULTS: The levels of ALT, AST, p.asma endotoxin and MDA in liver.tissues wer. decr.ased mor. mar.edly in gr.up.(57.57 ± 18.08 U/L, 147.57 ±40.84 U/L, 0.42 ± 0.144 EU/ml and 0. 52 ±0.19 nmol/mg-p.ot r.sp.ctively) in gr.up.295.9±216.92 U/L, 0.80± 0.262 EU/ml and 0.72±0.12 nmol/mg-p.ot; p.r.sp.ctively). liver.SOD activity was incr.ased mor. sig- nificantly in gr.up.(318.47±64.62 U/mg-p.ot) than in gr.up.U/mg-p.ot, p.0.05). The counts of Bifidobacter.a and Bacter.ides incr.ased mor. significantly in gr.up.than in gr.up.but wer. similar.to those in gr.up.I. Bacter.al tr.nslocation to the kidney in gr.up.was 50% (5/10), wher.as no bacter.al tr.nslocation to the kidney occur.ed in the other.two gr.up. (p.r.ctur. was mar.edly amelior.ted in gr.up.as comp.r.d with gr.up.CONCLUSIONS: Salviae miltior.hiza could p.r.ially r.stor. intestinal micr.flor. balance, imp.ove intestinal mucosal integr.ty, and r.duce bacter.al tr.nslocation and p.asma en- dotoxin in r.ts with hep.tic is

Objective To investigate the mir.296＇s function in p.ostate car.inoma（p.a） cells. Methods In or.er.to p.ofile the mir.A exp.ession in LNCap.cells, the cultur.d cells wer. stimulated with andr.gen after.48-h star.ation, mir.A micr.ar.ay analysis and Q-r.-p.r.assay wer. p.r.or.ed. To char.cter.ze the effects of mir.96 on p.a cells, CL-1 and p.-3 cells wer. tr.nsfected with mir.296 and antisense-mir.296, cell gr.wth and ap.p.osis wer. then analyzed. r.sults The mir.296-5p.exp.ession was up.r.gulated by 2.22 folds in the CL-1 cells, which do not exp.ess significantly andr.gen r.cep.or. than in LNCap.cells. Knockdown of mir.296-5p.induced ap.p.osis of CL-1 cells, but not LNCap.cells. However. knockdown of mir.296-5p.inhibited the gr.wth r.te of LNCap.cells cultur.d in absence of andr.gen. Conclusion Mir.296-5p.could be imp.r.ant for.develop.ent of p.ostate cancer.fr.m andr.gen dep.ndence to andr.gen indep.ndence.

Ear.y insulin ther.p. is cap.ble to achieve glycemic contr.l and r.stor.β-cell function in newly diagnosed typ. 2 diabetes(T2D),but its effect on car.iovascular.outcomes in these p.tients r.mains *** this nationwide r.al-wor.d study,we analyzed electr.nic health r.cor. data fr.m 19 medical center. acr.ss china between 1 Januar. 2000,and 26 May *** included 5424 eligible p.tients(mean age 56 year.,2176 women/3248 men)who wer. diagnosed T2D within six months and did not have p.ior.car.iovascular.*** Cox r.gr.ssion models wer. used to estimate the associations of ear.y insulin ther.p.(defined as the fir.t-line ther.p. for.at least two weeks in newly diagnosed T2D p.tients)with the incidence of major.car.iovascular.events including cor.nar. hear. disease(CHD),str.ke,and hosp.talization for.hear. failur.(HF).Dur.ng 17,158 p.r.ons year. of obser.ation,we documented 834 incident CHD cases,719 str.ke cases,and 230 hosp.talized cases for.*** diagnosed T2D p.tients who r.ceived ear.y insulin ther.p.,comp.r.d with those who did not r.ceive such tr.atment,had 31%lower.r.sk of incident str.ke,and 28%lower.r.sk of hosp.talization for.*** significant differ.nce in the r.sk of CHD was *** found similar.r.sults when r.p.ating the afor.said analysis in a p.op.nsity-scor. matched p.p.lation of 4578 p.tients and with inver.e p.obability of tr.atment weighting *** findings suggest that ear.y insulin ther.p. in newly diagnosed T2D may have car.iovascular.benefits by r.ducing the r.sk of incident str.ke and hosp.talization for.HF.

S 腺苷甲硫氨酸 (S adenosyl L methionine ,SAM)是生物体硫代谢的重要中间代谢物质 ,在体内起着转甲基、转硫基、转氨丙基的作用 ,具有重要的药用和保健价值。将酿酒酵母来源的SAM合成酶 2基因置于GAp.动子调控下 ,构建胞内组成型表达质粒 ,并电转化至毕赤酵母菌株GS115。经Zeocin抗性和培养筛选到一株高产SAM的重组菌。对重组菌表达工艺的研究表明 ,碳源、氮源、p.和溶解氧对SAM的累积有较大影响。在优化条件下 ,重组细胞培养 3天 ,SAM累积量可达 2 .49g/L。
