little is known about how chronic inflammation contributes to the progression of hepatoceUular carcinoma （HCC）, especially the initiation of cancer. To uncover the critical transition from chronic inflammation to HCC and the molecular mechanisms at a network level, we analyzed the time-series proteomic data of woodchuck hepatitis virus/*** mice and age-matched wt-C57BL/6 mice using our dynamical network biomarker （DNB） model. DNB analysis indicated that the 5th month after birth of transgenic mice was the critical period of cancer initiation, just before the critical transition, which is consistent with clinical symptoms. Meanwhile, the DNB-associated network showed a drastic inversion of protein expression and coexpression levels before and after the critical transition. Two members of DNB, PLA2G6 and CYP2C44, along with their associated differentially expressed proteins, were found to induce dysfunction of arachidonic acid metabolism, further activate inflammatory responses through inflammatory mediator regulation of transient receptor potential channels, and finally lead to impairments of liver detoxification and malignant transition to cancer. As a c-Myc target, PLA2G6 positively correlated with c-Myc in expression, showing a trend from decreasing to increasing during carcinogenesis, with the minimal point at the critical transition or tipping point. Such trend of homologous PLA2G6 and c-Myc was also observed during human hepatocarcinogenesis, with the minimal point at high-grade dysplastic nodules （a stage just before the carcinogenesis）. Our study implies that PLA2G6 might function as an oncogene like famous c-Myc during hepatocar- cinogenesis, while downregulation of PLA2G6 and c-Myc could be a warning signal indicating imminent carcinogenesis.

Dear Editor,Enterohemorrhagic Escherichia coli(EHEC)0157:H7,a major diarrheagenic pathogen,can cause bloody diarrhea,hemorrhagic colitis,and>90%of hemolytic uremic syndrome in humans(Mead and Griffin,1998).Many previous studies have demonstrated that 0157:H7 could disrupt host ubiquitin(Ub)system by delivering virulence effectors into host cells with the type III secretion system(T3SS).NleL(also named EspX7)emerged as one of such effectors,whose E3-like activity was first identified in vitro in 2011(lin et al.,2011).

HDL quality features revealed by proteome–lipidome connectivity are associated with atherosclerotic disease

Proteins are the main functional molecules of cells,and the tumor proteome offers valuable insights into the signaling transduction between the tumor microenvironment and cancer *** tumor proteome research has two directions:non-targeted protein mass spectrometry and antibody-based ***,the complexity and heterogeneity of tumor tissue pose significant challenges to analyzing tumor-driving related biomarkers at a global level and to accurately determining the immune cell status of tumors using limited target protein *** work innovatively choose an organic solution system for the lysis of a small number of cells,which,combined with a rapid enzymatic digestion process,allowed the identification of 2300 and 6700 proteins from 50 and 3000 cells within a mass spectrometry analysis time of 60 *** system's advantages can be used for in-depth proteomic analysis of immune *** characterized immune cell subpopulations from fresh tumor tissue,adjacent cancer tissue,and peripheral blood mononuclear cells(PBMC) using flow cytometry ***-proteome mass spectrometry analyzed immune cell protein expression variations across individuals and tissue *** aim to continue searching for tumor immune-related biomarkers,exploring the relevance of immune-related proteins in tumor tissue,adjacent tissue,and blood,further linking histological tissue tumor subtyping and clinical information,and developing personalized detection and treatment plans that are faster,more accurate,sensitive,and efficient.

lipoprotein,especially high-density lipoprotein(HDL),particles are composed of multiple heterogeneous subgroups containing various proteins and *** molecular distribution among these subgroups is closely related to cardiovascular disease(CVD).Here,we established high-resolution proteomics and lipidomics(HiPL)methods to depict the molecular profiles across lipoprotein(lipo-HiPL)and HDL(HDL-HiPL)subgroups by optimizing the resolution of anion-exchange chromatography and comprehensive quantification of proteins and lipids on the omics ***,based on the Pearson correlation coefficient analysis of molecular profiles across high-resolution subgroups,we achieved the relationship of proteome–lipidome connectivity(PLC)for lipoprotein and HDL *** application of these methods to high-fat,high-cholesterol diet-fed rabbits and acute coronary syndrome(ACS)patients,we uncovered the delicate dynamics of the molecular profile and reconstruction of lipoprotein and HDL *** note,the PLC features revealed by the HDL-HiPL method discriminated ACS from healthy individuals better than direct proteome and lipidome quantification or PLC features revealed by the lipo-HiPL method,suggesting their potential in ACS ***,we established HiPL methods to trace the dynamics of the molecular profile and PLC of lipoprotein and even HDL during the development of CVD.
