Background:There is an unmet need for accurate non-invasive methods to diagnose non-alcoholic steatohepatitis(NASH).Since impedance-based measurements of body composition are simple,repeatable and have a strong association with non-alcoholic fatty liver disease(NAFLD)severity,we aimed to develop a novel and fully automatic machine learning algorithm,consisting of a deep neural network based on impedance-based measurements of body composition to identify NASH[the bioeLectrical impEdance Analysis foR Nash(LEARN)algorithm].Methods:A total of 1.259 consecutive subjects with suspected NAFLD were screened from six medical centers across china,of which 766 patients with biopsy-proven NAFLD were included in final *** patients were randomly subdivided into the training and validation groups,in a ratio of 4:*** LEARN algorithm was developed in the training group to identify NASH,and subsequently,tested in the validation ***:The LEARN algorithm utilizing impedance-based measurements of body composition along with age,sex,pre-existing hypertension and diabetes,was able to predict the likelihood of having *** algorithm showed good discriminatory ability for identifying NASH in both the training and validation groups[area under the receiver operating characteristics(AUROC):0.81.95%CI:0.77-0.84 and AUROC:0.80,95%CI:0.73-0.87,respectively].This algorithm also performed better than serum cytokeratin-1. neoepitope M30(CK-1. M30)level or other non-invasive NASH scores(including HAIR,ION,NICE)for identifying NASH(P value1..Additionally,the LEARN algorithm performed well in identifying NASH in different patient subgroups,as well as in subjects with partial missing body composition ***:The LEARN algorithm,utilizing simple easily obtained measures,provides a fully automated,simple,non-invasive method for identifying NASH.

We performed a combined angle-resolved photoemission spectroscopy and scanning tunneling microscopy study of the electronic structure of electron-doped Ca_(0.83)La_(0.1.)Fe_(2)As_(2).A surface reconstruction associated with the dimerization of As atoms is observed directly in the real space,as well as the consequent band folding in the momentum *** this band folding effect,the Fermi surface topology of this material is similar to that reported previously for BaFe_(1.85)Co_(0.1.)As_(2),with Γ-centered hole pockets quasi-nested to M-centered electron pockets by the antiferromagnetic wave *** no superconducting gap is observed by ARPES possibly due to low superconducting volume fraction,a gap-like density of states depression of 7.7±2.9 meV is determined by scanning tunneling microscopy.

目的:探讨miR-34c-5p与poFUT1.靶向关系及对胎盘血管形成的影响。方法:利用ENCORI数据库预测miR-34c-5p与poFUT1.结合位点,采用双荧光素酶报告实验验证。随机选取2023年4至1.月在郑州大学第三附属医院住院的胎儿生长受限(FGR)孕妇20例(FGR组),选择同期正常孕妇20例为对照。采用实时荧光定量PCR法检测两组胎盘组织中miR-34c-5p和poFUT1.mRNA的表达。取脐静脉内皮细胞,分组转染miR-34c-5p模拟物及其对照(NC)、miR-34c-5p抑制物及其NC、si-poFUT1.NC、si-poFUT1.si-poFUT1.miR-34c-5p抑制物,采用CCK-8法、Transwell实验以及成管实验检测细胞转染24、48、72、96 h后的增殖能力,转染48 h后的侵袭能力和成管能力。结果:FGR组和对照组胎盘组织中miR-34c-5p表达水平分别为(1.57±0.39)、(1.09±0.1.),poFUT1.mRNA表达水平分别为(0.51.0.1.)、(1.06±0.1.),FGR组胎盘组织中miR-34c-5p表达水平高于对照组,poFUT1.mRNA表达水平低于对照组(P1.NC组相比,si-poFUT1.侵袭细胞数和管腔节点数减少,细胞增殖活性降低(P1.miR-34c-5p抑制物组上述变化较si-poFUT1.部分逆转(P1.响血管内皮细胞功能,从而影响胎盘的血管形成,参与FGR的发生。

Discrete Pt(Ⅱ)metallacycles have attracted particular attention for the chemotherapeutic treatment of ***,a single chemotherapy cannot simultaneously balance efficiency and safety because the continuous administration throughout the entire therapy period will lead to inefficient therapy and potentially long-term systemic ***,the development of a novel organoplatinum(Ⅱ)metallacycle with multimodal treatment capabilities is urgently needed to overcome these ***,a discrete Pt(II)metallacycle(SCY)bearing the near-infrared(NIR)photosensitizer heptamethine cyanine was fabricated and further encapsulated by amphiphilic 1.2-distearoyl-sn-glycero-3-phosphoethanolamine-N-methoxy(polyethyleneglycol)(DSPEmPEG)to form P@SCY *** cyanine,which has excellent photoconversion efficiency,can generate reactive oxygen species(ROS)and heat simultaneously,and the cyanine moiety can target mitochondria in cancer cells due to their quaternary ammonium salt cations,which improve the effect of *** to its excellent phototherapy and chemotherapy properties,P@SCY exhibited remarkable trimodal therapeutic effects[chemo-/photodynamic therapy(PDT)/photothermal therapy(PTT)]against cancer cells(HepG2 cells,MCF-7 cells,and 4T1.cells)in ***,in vivo results also confirmed thatP@SCY had superior antitumor properties with minimal side effects in the 4T1.tumor *** presents a practicable approach to develop a multifunctional organoplatinum(Ⅱ)metallacycle for multimodal tumor therapy.

目的:探讨内吗啡肽-1.EM-1.对高糖诱导的人外周血来源树突状细胞(PBDC)成熟表型、细胞因子分泌、T细胞增殖和TLR4表达的影响,探讨EM-1.DC免疫功能的调节机制。方法:本研究外周血样品取自1.-20岁学生志愿者,将外周血单个核细胞(PBMNC)诱导成未成熟的树突状细胞(imDC)。使用高浓度葡萄糖作为刺激因素,EM-1.为干预因素,将实验分为正常葡萄糖(NG)、高糖(HG)、加入EM-1.高葡萄糖(EM)、加入EM-1.纳洛酮的高葡萄糖(Nal)共5组。应用流式细胞术检测PBDC表型变化,EliSA检测PBDC与自体淋巴细胞共培养分泌的细胞因子的变化,CFSE法检测T淋巴细胞的增殖情况,RT-PCR法检测PBDC表面TLR4的表达。结果:与HG组比较,EM组PBDC表面分子CD86、CCR7和CD36表达上调(P1.,而CD83的表达无统计学意义。EM组PBDC分泌的IL-1.,IL-1.和PBDCs刺激的T淋巴细胞增殖指数均下降。与EM组比较,Nal组CD86,CCR7,CD36表达降低(P1.,而CD83表达几乎没有变化(P>0.05)。Nal组T淋巴细胞增殖指数极显著升高(P1.。HG组TLR4灰度比值高于NG组,EM组明显低于HG组(P1.。结果表明,高葡萄糖可以促进PBDC TLR4的表达,而EM-1.制TLR4的表达。结论:EM-1.调高糖诱导的PBDC表面分子CD86,CCR7和CD36的表达,抑制高糖诱导的P BDC成熟。分泌的炎性细胞因子IL-1.和IL-1.抑制来自PBDC的T淋巴细胞的增殖,而纳洛酮抑制EM-1.作用。EM-1.制高糖诱导的PBDC表面TLR4的表达。

Background Children as a population have high antimicrobial prescribing rates which may lead to high resistance of bacteria according to data from some single-center surveys of antibiotic prescribing rates in china. The acquirement of base{1.ne data of antibiotic prescribing is the basis of developing intervention strategies on inappropriate antimicrobial prescriptions. Few studies show clearly the pattern and detailed information on classes of antibiotics and distribution of indications of antibiotic prescriptions in children in china. This study aims to assess the antibiotic prescribing patterns among children and neonates hospita{1.zed in 1. hospitals in china. Methods A 24-hour point prevalence survey on antimicrobial prescribing was conducted in hospita{1.zed neonates and chil-dren in china from December 1.t, 201. to February 28th, 201.. Information on the antibiotic use of patients under 1. years of age who were administered one or more on-going antibiotics in the selected wards over a 24-hour period was collected. These data were submitted to the GARPEC (Global Antimicrobial Resistance, Prescribing and Efficacy in Children and Neonates) web-based app{1.cation ( https ://pidrg -datab ***/redca p/ ). For statistical analysis, Microsoft Excel 2007 and SPSS 22.0 were used. Results The antibiotic data were collected in 35 wards in 1. hospitals from 9 provinces. In total, 67.76% (975/1.39) of the patients (n = 1.39) were given at least one antibiotic, including 58.1. (1.3/298) of neonates (n = 298) and 70.3% (802/1.41. of children (n = 1.41.. In neonates, the three most frequently prescribed antibiotics were third-generation cephalosporins (41.7%), penicil{1.ns plus enzyme inhibitor (23.8%), and carbapenems (1..2%). The expression level of CRM1.and CDK5 was detected by immunohistochemistry. The correlations between CRM1.and CDK5 expression and clinicopathological factors were explored. Univariate and multivariate survival analyses were used to identify prognostic factors for GC. Receiver operating characteristic analysis was used to compare the accuracy of the prediction of clinical outcome by the parameters. RESULTS The expression of CRM1.was significantly related to size of primary tumor(P = 0.005), Borrmann type(P = 0.006), degree of differentiation(P = 0.004), depth of invasion(P = 0.008), lymph node metastasis(P = 0.01.), TNM stage(P = 0.002) and distant metastasis(P = 0.01.). The expression of CDK5 was significantly related to sex(P = 0.048) and Lauren's classification(P = 0.01.). Multivariate Cox regression analysis identified that CRM1.and CDK5 co-expression status was an independent prognostic factor for overall survival(OS) of patients with GC. Integration of CRM1.and CDK5 expression could provide additional prognostic value for OS compared with CRM1.or CDK5 expression alone(P = 0.001..CONCLUSION CRM1.and CDK5 co-expression was an independent prognostic factors for GC. Combined CRM1.and CDK5 expression could provide a prognostic model for OS of GC.
