Volatile sex pheromones are vital for sexual communication between males and females.Females of the American cockroach,Periplaneta americana,produce and emit two sex pheromone components,periplanone-A(PA)and periplanone-B(PB).Although PB is the major sex attractant and can attract males,how it interacts with PA in regulating sexual behaviors is still unknown.In this study,we found that in male cockroaches,PA counteracted PB attraction.We identified two odorant receptors(ORs),OR53 and OR100,as PB/PA and PA receptors,respectively.OR53 and OR100 were predominantly expressed in the antennae of sexually mature males,and their expression levels were regulated by the sex differentiation pathway and nutrition-responsive signals.Cellular localization of OR53 and OR100 in male antennae further revealed that two types of sensilla coordinate a complex two-pheromone-two-receptor pathway in regulating cockroach sexual behaviors.These findings indicate distinct functions of the two sex pheromone components,identify their receptors and possible regulatory mechanisms underlying the male-specific and age-dependent sexual behaviors,and can guide novel strategies for pest management.

ObjectiveTo explore the regulatory effect of Pien Tze Huang (PZH) on targeting partner of NOB1 (PNO1) and it’s down-stream mediators in colorectal cancer (CRC) cells.MethodsQuantitative polymerase chain reaction was performed to determine mRNA levels of PNO1, TP53, and CDKN1A. Western blotting was performed to determine protein levels of PNO1, p53, and p21. HCT-8 cells were transduced with a lentivirus over-expressing PNO1. Colony formation assay was used to detect cell survival in PNO1 overexpression of HCT-8 cells after PZH treatment. Cell-cycle distribution, cell viability and cell apoptosis were performed to identify the effect of PNO1 overexpression on cell proliferation and apoptosis of HCT-8 cells after PZH treatment. Xenograft BALB/c nude mice bearing HCT116 cells transduced with sh-PNO1 or sh-Ctrl lentivirus were evaluated. Western blot assay was performed to detect PNO1, p53, p21 and PCNA expression in tumor sections. Terminal deoxynucleotidyl transferase dUTP nick end labling (TUNEL) assay was used to determine the apoptotic cells in tissues.ResultsPZH treatment decreased cell viability, down-regulated PNO1 expression, and up-regulated p53 and p21 expressions in HCT-8 cells (Plity, cell cycle arrest and cell apoptosis in vitro (Pliminated the effects of PZH treatment on tumor growth, inhibiting cell proliferation inhibition and apoptosis induction in vivo (Pliferative effect is at least in part by regulating the expression of PNO1 and its downstream targets p53 and p21.

Two-dimensional transition metal dichalcogenides(TMDs)exhibit promising application prospects in the domains of electronic devices,optoelectronic devices and spintronic devices due to their distinctive energy band structures and spin−orbit coupling properties.Cr-based chalcogenides with narrow or even zero bandgap,covering from semiconductors to metallic materials,have considerable potential for wide-band photodetection and two-dimensional magnetism.Currently,the preparation of 2D CrX_(n)(X=S,Se,Te)nanosheets primarily relies on chemical vapor deposition(CVD)and molecule beam epitaxy(MBE),which enable the production of high-quality large-area materials.This review article focuses on recent progress of 2D Cr-based chalcogenides,including unique crystal structure of the CrX_(n)system,phase-controlled synthesis,and heterojunction construction.Furthermore,a detailed introduction of room-temperature ferromagnetism and electrical/optoelectronic properties of 2D CrXn is presented.Ultimately,this paper summarizes the challenges associated with utilizing 2D Cr-based chalcogenides in preparation strategies,optoelectronics devices,and spintronic devices while providing further insights.

Numerical Simulation of Asphaltene Precipitation and Deposition during Natural Gas and CO_(2) Injection

Asphaltene deposition is a signiﬁcant problem during gas injection processes,as it can block the porous medium,the wellbore,and the involved facilities,signiﬁcantly impacting reservoir productivity and ultimate oil recovery.Only a few studies have investigated the numerical modeling of this potential effect in porous media.This study focuses on asphaltene deposition due to natural gas and CO_(2) injection.Predictions of the effect of gas injection on asphaltene deposition behavior have been made using a 3D numerical simulation model.The results indicate that the injection of natural gas exacerbates asphaltene deposition,leading to a signiﬁcant reduction in permeability near the injection well and throughout the reservoir.This reduction in permeability strongly affects the ability of gas toﬂow through the reservoir,resulting in an improvement of the displacement front.The displacement efﬁ-ciency of the injection gas process increases by up to 1.40%when gas is injected at 5500 psi,compared to the scenario where the asphaltene model is not considered.CO_(2) injection leads to a miscible process with crude oil,extracting light and intermediate components,which intensiﬁes asphaltene precipitation and increases the viscosity of the remaining crude oil,ultimately reducing the recovery rate.

Driving safety and accident prevention are attracting increasing global interest.Current safety monitoring systems often face challenges such as limited spatiotemporal coverage and accuracy,leading to delays in alerting drivers about potential hazards.This study explores the use of edge computing for monitoring vehicle motion and issuing accident warnings,such as lane departures and vehicle collisions.Unlike traditional systems that depend on data from single vehicles,the cooperative vehicle-infrastructure system collects data directly from connected and automated vehicles(CAVs)via vehicle-to-everything communication.This approach facilitates a comprehensive assessment of each vehicle’s risk.We propose algorithms and specific data structures for evaluating accident risks associated with different CAVs.Furthermore,we examine the prerequisites for data accuracy and transmission delay to enhance the safety of CAV driving.The efficacy of this framework is validated through both simulated and real-world road tests,proving its utility in diverse driving conditions.

Insight into the capacity degradation and structural evolution of single-crystal Ni-rich cathodes

Single-crystal Ni-rich cathodes are a promising candidate for high-energy lithium-ion batteries due to their higher structural and cycling stability than polycrystalline materials. However, the phase evolution and capacity degradation of these single-crystal cathodes during continuous lithation/delithation cycling remains unclear. Understanding the mapping relationship between the macroscopic electrochemical properties and the material physicochemical properties is crucial. Here, we investigate the correlation between the physical-chemical characteristics, phase transition, and capacity decay using capacity differential curve feature identification and in-situ X-ray spectroscopic imaging. We systematically clarify the dominant mechanism of phase evolution in aging cycling. Appropriately high cut-off voltages can mitigate the slow kinetic and electrochemical properties of single-crystal cathodes. We also find that second-order differential capacity discharge characteristic curves can be used to identify the crystal structure disorder of Ni-rich cathodes. These findings constitute a step forward in elucidating the correlation between the electrochemical extrinsic properties and the physicochemical intrinsic properties and provide new perspectives for failure analysis of layered electrode materials.

HepG2.2.15-derived exosomes facilitate the activation and fibrosis of hepatic stellate cells

BACKGROUND The role of exosomes derived from HepG2.2.15 cells,which express hepatitis B virus(HBV)-related proteins,in triggering the activation of LX2 liver stellate cells and promoting liver fibrosis and cell proliferation remains elusive.The focus was on comprehending the relationship and influence of differentially expressed microRNAs(DE-miRNAs)within these exosomes.AIM To elucidate the effect of exosomes derived from HepG2.2.15 cells on the activation of hepatic stellate cell(HSC)LX2 and the progression of liver fibrosis.METHODS Exosomes from HepG2.2.15 cells,which express HBV-related proteins,were isolated from parental HepG2 and WRL68 cells.Western blotting was used to confirm the presence of the exosomal marker protein CD9.The activation of HSCs was assessed using oil red staining,whereas DiI staining facilitated the observation of exosomal uptake by LX2 cells.Additionally,we evaluated LX2 cell proliferation and fibrosis marker expression using 5-ethynyl-2′-deoxyuracil staining and western blotting,respectively.DE-miRNAs were analyzed using DESeq2.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways were used to annotate the target genes of DE-miRNAs.RESULTS Exosomes from HepG2.2.15 cells were found to induced activation and enhanced proliferation and fibrosis in LX2 cells.A total of 27 miRNAs were differentially expressed in exosomes from HepG2.2.15 cells.GO analysis indicated that these DE-miRNA target genes were associated with cell differentiation,intracellular signal transduction,negative regulation of apoptosis,extracellular exosomes,and RNA binding.KEGG pathway analysis highlighted ubiquitin-mediated proteolysis,the MAPK signaling pathway,viral carcinogenesis,and the toll-like receptor signaling pathway,among others,as enriched in these targets.CONCLUSION These findings suggest that exosomes from HepG2.2.15 cells play a substantial role in the activation,proliferation,and fibrosis of LX2 cells and that DE-miRNAs within these exosomes contribute to the underlying mechanisms.

Mesenchymal-epithelial transition factor amplification correlates with adverse pathological features and poor clinical outcome in colorectal cancer

BACKGROUND Colorectal cancer(CRC)is the third most common cancer and the second most common cause of cancer-related mortality worldwide.Mesenchymal-epithelial transition factor(MET)gene participates in multiple tumor biology and shows clinical potential for pharmacological manipulation in tumor treatment.MET amplification has been reported in CRC,but data are very limited.Investigating pathological values of MET in CRC may provide new therapeutic and genetic screening options in future clinical practice.AIM To determine the pathological significance of MET amplification in CRC and to propose a feasible screening strategy.METHODS A number of 205 newly diagnosed CRC patients undergoing surgical resection without any preoperative therapy at Shenzhen Cancer Hospital of Chinese Academy of Medical Sciences were recruited.All patients were without RAS/RAF mutation or microsatellite instability-high.MET amplification and c-MET protein expression were analyzed using fluorescence in situ hybridization(FISH)and immunohistochemistry(IHC),respectively.Correlations between MET aberration and pathological features were detected using the chi-squared test.Progression free survival(PFS)during the two-year follow-up was detected using the Kaplan-Meier method and log rank test.The results of MET FISH and IHC were com pared using one-way ANOVA.RESULTS Polysomy-induced MET amplification was observed in 14.4%of cases,and focal MET amplification was not detected.Polysomy-induced MET amplification was associated with a higher frequency of lymph node metastasis(LNM)(Plified-and non-amplified MET in a two-year follow-up after the first diagnosis(P=0.001).C-MET scores of 0,1+,2+,and 3+were observed in 1.4%,24.9%,54.7%,and 19.0%of tumors,respectively.C-MET overexpression correlated with higher frequency of LNM(P=0.002),but no significant difference of PFS was detected between patients with different protein lev

Specialized households serve as the primary units within specialized villages in China,and their capacity to withstand risks and external influences significantly shapes the future trajectory of specialized villages and the overall vitality of the rural economy.In this study,we established a measurement indicator system based on the definition of specialized households’resilience,elucidating the logical connection between specialized households’resilience and rural industrial development in China.The musical instrument industry in Lankao County,Henan Province of China,was employed as a case;survey data,the entropy method,and an obstacle diagnosis model were used to examine how instrument production specialized households responded to the challenges posed by Corona Virus Disease 2019(COVID-19)and the tightening of national environmental protection policies,yielding the following key findings:1)there exists substantial variation in the comprehensive resilience levels among different specialized households;2)the ability to learn and adapt is the most significant contributor to the overall resilience level of specialized households;3)technological proficiency and access to skilled talent emerge as pivotal factors influencing specialized households’resilience;4)the positioning of specialized households within the industrial supply chain and the stability of their income have a direct bearing on their resilience level.The influence of specialized households’resilience on industrial development primarily manifests in the following ways:stronger resilience correlates with increased stability in production and sales,fostering a more proactive approach to future actions.However,heightened exposure to the external macroeconomic environment can lead to a higher rate of export reduction.To enhance the development resilience of entities like specialized households and family farms,and to invigorate rural economic development,escalating investments in rural science and technology and prioritizing the training of technical talen

A greater number of compact and reliable electrostatic capacitors are in demand due to the Internet of Things boom and rapidly growing complex and integrated electronic systems,continuously promoting the development of high-energy-density ceramic-based capacitors.Although significant successes have been achieved in obtaining high energy densities in lead-based ferroelectric ceramics,the utilization of lead-containing ceramies has been restricted due to environmental and health hazards of lead.Lead-free ferroelectric ceramics have garnered tremendous attention and are expected to replace lead-based ceramics in the near future.However,the energy density of lead-free ceramics is still lagging behind that of lead-containing cou.nterparts,severely limiting their applications.Significant efforts have been made to enhance the energy storage performance of lead-free ceramics using multi-scale design strategies,and exciting progress has been achieved in the past decade.This review briefly discusses the energy storage mechanism and fundamental characteristics of a dielectric capacitor,summarizes and compares the state-of-the-art design strategies for high-energy-density lead-free ceramics,and highlights several critical issues and requirements for industrial production.The prospects and challenges of lead-free ceramics for energy storage applications are also discussed.
