Inhibition of G9a by a small molecule inhibitor, UNC0642,induces apoptosis of human bladder cancer cells

Urinary bladder cancer（UBC）is characterized by frequent recurrence and metastasis despite the standard chemotherapy with gemcitabine and cisplatin *** modifiers are often dysregulated in cancer development,thus they can serve as an excellent drug targets for cancer ***,we investigated whether G9a,one of the histone H3 methyltransferases,was associated with UBC *** first analyzed clinical data from public databases and found that G9a was significantly overexpressed in UBC *** TCGA Provisional dataset showed that the average expression level of G9a in primary UBC samples（n = 408）was 1.6-fold as much as that in normal bladder samples（n = 19;P1）.Then we used small interfering RNA to knockdown G9a in human UBC T24 and J82 cell lines in vitro,and observed that the cell viability was significantly decreased and cell apoptosis ***,we choosed UNC0642,a small molecule inhibitor targeting G9a,with low cytotoxicity,and excellent in vivo pharmacokinetic properties,to test its anticancer effects against UBC cells in vitro and in *** with UNC0642 dose-dependently decreased the viability of T24,J82,and 5637 cells with the IC50 values of 9.85±0.41,13.15±1.72,and 9.57±0.37 μM,***,treatment with UNC0642（1-20 μM）dose-dependently decreased the levels of histone H3 K9 me2,the downstream target of G9a,and increased apoptosis in T24 and J82 *** nude mice bearing J82 engrafts,administration of UNC0642（5 mg/kg,every other day,i.p.,for 6 times）exerted significant suppressive effect on tumor growth without loss of mouse body ***,administration of UNC0642 significantly reduced Ki67 expression and increased the level of cleaved Caspase 3 and BIM protein in J82 xenografts evidenced by immunohistochemistry and western blot analysis,*** together,our data demonstrated that G9a may be a promising therapeutic target for UBC,and an epigeneticsbased therapy by UNC0642 is suggested.

It is well-known that insulin can activate both PDK/Akt pathway,which is responsible for glucose uptake,and MAPK pathway,which is crucial for insulin resistance *** it is unclear exactly how the two pathways coordinate to regulate insulin sensitivity upon hyperinsulinism stress of type 2 diabetes mellitus(T2DM).Here we show that an early response transcription factor Egr-1 could tilt the signaling balance by blocking PI3K/Akt signaling through PTEN and augmenting Erk/MAPK signaling through GGPPS,resulting in insulin resistance in ***-1,PTEN and GGPPS are upregulated in the fat tissue of T2DM patients and db/db ***-1 overexpression in epididymal fat induced systematic insulin resistance in wild-type mice,and loss of Egr-1 function improved whole-body insulin sensitivity in diabetic mice,which is mediated by Egr-1 controlled PDK/Akt and Erk/MAPK signaling balance. Therefore,we have revealed,for the first time,the mechanism by which Egr-1 induces insulin resistance under hyperinsulinism stress,which provides an ideal pharmacological target since inhibiting Egr-1 can simultaneously block MAPK and augment PDK/Akt activation during insulin stimulation.

近60年重庆市日照时数时空变化特征及影响因素分析

为探究重庆市日照时数的时空变化规律,基于重庆34个气象站点1961-2020年逐日日照时、风速、温度、总云量、相对湿度资料,使用M-K突变检验、小波分析、时空冷热点模式探测等方法对地区日照时数的时空变化及其影响因素进行分析。结果表明:①重庆日照时数呈现“东北高、东南低”的分布特点。高值集中在巫山、巫溪等地;低值集中在彭水等地。②年均日照时数呈极显著下降趋势,突变年份1979年,振荡主周期为46年。除春季外其他季节均呈现下降趋势。③时空冷热点表明近重庆近60年的日照时数呈现混合聚类、振荡冷点2种模式。④相关系数的季节性差异表明各因素与日照时数的关系并非定值,其中风速、总云量、相对湿度与日照时数相关性四季均显著,降水与温度与日照仅在夏季关系较为紧密。

Background and Aims:As practice patterns and hepatitis C virus(HCV)genotypes(GT)vary geographically,a global real-world study from both East and west covering all GTs can help inform practice policy toward the 2030 HCV elimination *** study aimed to assess the effectiveness and tolerability of DAA treatment in routine clinical practice in a multinational cohort for patients infected with all HCV GTs,focusing on GT3 and ***:We analyzed the sustained virological response(SVR12)of 15,849 chronic hepatitis C patients from 39 Real-World Evidence from the Asia liver Consortium for HCV clinical sites in Asia Pacific,North America,and Europe between 07/01/2014–07/01/***:The mean age was 62±13 years,with 49.6%*** demographic breakdown was 91.1%Asian(52.9%Japanese,25.7%Chinese/Taiwan residents,5.4%Korean,3.3%Malaysian,and 2.9%Vietnamese),6.4%White,1.3%Hispanic/Latino,and 1%Black/***,34.8%had cirrhosis,8.6%had hepatocellular carcinoma(HCC),and 24.9%were treatment-experienced(20.7%with interferon,4.3%with direct-acting antivirals).The largest group was GT1(10,246[64.6%]),followed by GT2(3,686[23.2%]),GT3(1,151[7.2%]),GT6(457[2.8%]),GT4(47[0.3%]),GT5(1[0.006%]),and untyped GTs(261[1.6%]).The overall SVR12 was 96.9%,with rates over 95%for GT1/2/3/6 but 91.5%for ***12 for GT3 was 95.1%overall,98.2%for GT3a,and 94.0%for ***12 was 98.3%overall for GT6,lower for patients with cirrhosis and treatment-experienced(TE)(93.8%)but≥97.5%for tretment-naive patients regardless of cirrhosis *** multivariable analysis,advanced age,prior treatment failure,cirrhosis,active HCC,and GT3/4 were independent predictors of lower SVR12,while being Asian was a significant predictor of achieving ***:In this diverse multinational realworld cohort of patients with various GTs,the overall cure rate was 96.9%,despite large numbers of patients with cirrhosis,HCC,TE,and GT3/***12 for GT3/6 with cirrhosis and TE was lower but st

土壤中普遍存在微塑料和重金属等复合污染物,二者极难降解且易形成复合污染,改变土壤的理化性质,并影响植物的生长与生理生态特征.为研究土壤微塑料与重金属的复合污染对土壤性质和植物生长的影响,选取粒径3μm的聚苯乙烯微塑料(PS-MPs)和重金属镉(Cd),设置不同PS-MPs暴露量(0、10、50、100、200和400 mg·kg^(-1))并联合Cd污染(0、1.2和6.0 mg·kg^(-1))条件下,研究土壤的理化性质变化及其对生菜(Lactuca sativa)种子萌发、幼苗生长的影响.结果表明,随着PS-MPs与Cd复合污染强度的增加,土壤有机质(SOM)、有效磷(AP)、碱解氮(AHN)和速效钾(AK)等主要理化因子均显著降低,在PS-MPs暴露量为400 mg·kg^(-1)时降到最低值;同时,PS-MPs联合Cd的污染也会显著降低生菜种子的萌发率,但PS-MPs低暴露量(10 mg·kg^(-1)和50mg·kg^(-1))会减缓Cd(6.0 mg·kg^(-1))污染对生菜种子萌发的抑制作用,而PS-MPs高暴露量(200 mg·kg^(-1)和400 mg·kg^(-1))会加剧Cd(6.0 mg·kg^(-1))对生菜种子的抑制作用.受不同含量Cd污染后,生菜幼苗鲜重、干重和株高等生长指标随PS-MPs暴露量增加均呈先升后降趋势;叶绿素含量、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)等指标均呈下降趋势,而丙二醛(MDA)含量总体呈上升趋势.土壤的主要理化指标与生菜幼苗的MDA呈显著负相关,而与幼苗的其它各项指标则呈显著正相关.PS-MPs和重金属Cd复合污染能通过改变土壤理化性质从而影响植物种子萌发及其幼苗生理生态特征,研究结果对于评估土壤-植物系统中微塑料和重金属复合污染的生态效应能提供一定的参考借鉴.
