Background:The convergence of smartphone technology and artificial intelligence(AI)has revolutionized the landscape of ophthalmic care,offering unprecedented opportunities for diagnosis,monitoring,and management of ocular ***,there is a lack of systematic studies on discussing the integration of smart-phone and AI in this *** text:This review includes 52 studies,and explores the integration of smartphones and AI in ophthalmology,delineating its collective impact on screening methodologies,disease detection,telemedicine initiatives,and patient *** collective findings from the curated studies indicate promising performance of the smartphone-based AI screening for various ocular diseases which encompass major retinal diseases,glaucoma,cataract,visual impairment in children and ocular surface ***,the utilization of smartphone-based imaging modalities,coupled with AI algorithms,is able to provide timely,efficient and cost-effective screening for ocular *** modality can also facilitate patient self-monitoring,remote patient monitoring and enhancing accessibility to eye care services,particularly in underserved *** involving data pri-vacy,algorithm validation,regulatory frameworks and issues of trust are still need to be ***,evaluation on real-world implementation is imperative as well,and real-world prospective studies are currently ***:Smartphone ocular imaging merged with AI enables earlier,precise diagnoses,personalized treat-ments,and enhanced service accessibility in eye *** is crucial to navigate ethical and data security challenges while responsibly leveraging these innovations,promising a potential revolution in care access and global eye health equity.

Aim:To investigate the effect of the novel immunosuppressant mycophenolicacid（MPA）on cytokine production and apoptosis of the peripheral blood mono-nuclear cells（PBMC）of patients with systemic lupus erythematosus（SLE）.Methods:The levels of IL-10,IL-12,IFN-γ,sFas and sFasL in the supernatants ofcultured PBMC from 41 SLE patients was determined by the ABC-EliSA *** percentage of IFN-γIL- 10-,IFN-γ-IL-10+,and IFN-γ+IL-10+subsets in CD4+ceils were detected by three-color flow *** percentage of apoptoticTh cells was detected by AV-FITC/PI flow *** from 22 sex-andage-comparable healthy people were used as normal ***:The lev-els of IL-10,IL-12,and IFN-γwere all significantly elevated in the supernatants ofcultured PBMC from SLE samples,compared with normal *** enhancedproductions of IL-10,IL-12,and IFN-γ by PBMC from SLE both spontaneouslyand stimulated by phytohaemagglutinin（PHA）were significantly reduced by *** percentages of CD4+IFN-γIL-10+and CD4+IFN-γ+IL-10+cell subsets in cul-tured PBMC from SLE were significantly increased,but decreased when MPA wasadded into the *** being cultured in vitro for 48h,the PBMC of SLEpatients showed a higher secretion of sFasL as well as a higher percentage *** significantly increased the apoptotic percentage of SLE PBMC,but reduced the secretion of sFasL and ***:MPA reduces theabnormal production of SLE-associated cytokines,such as IL-10,IL-12,and IFN-γ,inhibits the increase of CD4+IFN-γ+IL-10,CD4+IFN-γIL-10+and CD4+IFN-γ+IL-10+subset;and promotes the apoptosis of PBMC in SLE patients,which may beassociated with the therapeutic mechanism of MPA for SLE.

The Chinese tree shrew(Tupaia belangeri chinensis)has emerged as a promising model for investigating adrenal steroid synthesis,but it is unclear whether the same cells produce steroid hormones and whether their production is regulated in the same way as in ***,we comprehensively mapped the cell types and pathways of steroid metabolism in the adrenal gland of Chinese tree shrews using single-cell RNA sequencing,spatial transcriptome analysis,mass spectrometry,and *** compared the transcriptomes of various adrenal cell types across tree shrews,humans,macaques,and *** showed that tree shrew adrenal glands expressed many of the same key enzymes for steroid synthesis as humans,including CYP11B2,CYP11B1,CYB5A,and *** analysis confirmed the production of aldosterone,cortisol,and dehydroepiandrosterone but not dehydroepiandrosterone sulfate in the tree shrew adrenal ***,genes in adrenal cell types in tree shrews were correlated with genetic risk factors for polycystic ovary syndrome,primary aldosteronism,hypertension,and related disorders in humans based on genome-wide association ***,this study suggests that the adrenal glands of Chinese tree shrews may consist of closely related cell populations with functional similarity to those of the human adrenal *** comprehensive results(publicly available at http://***:16245/scAGMap/)should facilitate the advancement of this animal model for the investigation of adrenal gland disorders.

To enhance the clarity and transparency of reporting in trials involving Chineseherbal medicine(CHM)formulas,the CONSORT extension specifically tailored for CHM formulas(CONSORT-CHM Formulas 2017)was introduced in 2017[1].This statement,along with its corresponding checklist,outlines key elements essential for reporting in randomized controlled trials(RCTs)related tothe rationale,design,analysis,and interpretation of interventions using CHM *** CONSORT-CHM Formulas 2017 elaborated on 7 of 25 CONSORT checklist items,namely title,abstract and keywords,backgrounds and objectives,participants,interventions,outcomes,generalizability,and interpretation.

Background Diarrhea is a common complication of hematopoietic stem cell transplantation(HSCT)and is associated with substantial morbidity,but its etiology is often *** of diarrhea in this population include infectious causes,chemotherapy-or medication-induced mucosal injury and graft-versus-host disease(GVHD).Distinguishing these potential causes of diarrhea is challenging since diarrheal symptoms are often multifactorial,and the etiologies often overlap in transplant *** objectives of this study were to evaluate whether the FilmArray gastrointestinal(GI)panel would increase diagnostic yield and the degree to which pre-transplantation colonization predicts post-transplantation *** From November 2019 to February 2021,a total of 158 patients undergoing HSCT were prospectively included in the *** specimens were obtained from all HSCT recipients prior to conditioning therapy,28±7 days after transplantation and at any new episode of *** stool samples were tested by the FilmArray GI panel and other clinical microbiological *** The primary cause of post-transplantation diarrhea was infection(57/84,67.86%),followed by medication(38/84,45.24%)and GVHD(21/84,25.00%).Ninety-five of 158 patients were colonized with at least one gastrointestinal pathogen before conditioning therapy,and the incidence of infectious diarrhea was significantly higher in colonized patients(47/95,49.47%)than in non-colonized patients(10/63,15.87%)(P1).Fourteen of 19(73.68%)patients who were initially colonized with norovirus pre-transplantation developed a post-transplantation norovirus ***-four of 62(38.71%)patients colonized with Clostridium difficile developed a diarrheal *** addition,FilmArray GI panel testing improved the diagnostic yield by almost twofold in our study(55/92,59.78%vs.30/92,32.61%).Conclusions Our data show that more than half of pediatric patients who were admitted for HSCT were colonized

Partial AZFc duplications not deletions are associated with male infertility in the Yi population of Yunnan Province,china

Objective:There are many reports on associations between spermatogenesis and partial azoospermia factor c(AZFc) deletions as well as duplications;however,results are conflicting,possibly due to differences in methodology and ethnic *** purpose of this study is to investigate the association of AZFc polymorphisms and male infertility in the Yi ethnic population,residents within Yunnan Province,chinaMethods:A total of 224 infertile patients and 153 fertile subjects were selected in the Yi ethnic *** study was performed by sequence-tagged site plus/minus(STS+/) analysis followed by gene dosage and gene copy definition analysis.Y haplotypes of 215 cases and 115 controls were defined by 12 binary markers using single nucleotide polymorphism on Y chromosome(Y-SNP) multiplex assays based on single base primer extension ***:The distribution of Y haplotypes was not significantly different between the case and control *** frequencies of both gr/gr(7.6% vs.8.5%) and b2/b3(6.3% vs.8.5%) deletions do not show significant ***,single nucleotide variant(SNV) analysis shows no significant difference of gene copy definition between the cases and ***,the frequency of partial duplications in the infertile group(4.0%) is significantly higher than that in the control group(0.7%).Further,we found a case with sY1206 deletion which had two CDY1 copies but removed half of DAZ ***:Our results show that male infertility is associated with partial AZFc duplications,but neither gr/gr nor b2/b3 deletions,suggesting that partial AZFc duplications rather than deletions are risk factors for male infertility in Chinese-Yi population.

Aporphine alkaloids have diverse pharmacological activities;however,our understanding of their biosynthesis is relatively *** studies have classified aporphine alkaloids into two categories based on the configuration and number of substituents of the D-ring and have proposed preliminary biosynthetic pathways for each *** this study,we identified two specific cytochrome P450 enzymes(CYP80G6 and CYP80Q5)with distinct activities toward(S)-configured and(R)-configured substrates from the herbaceous perennial vine Stephania tetrandra,shedding light on the biosynthetic mechanisms and stereochemical features of these two aporphine alkaloid ***,we characterized two CYP719C enzymes(CYP719C3 and CYP719C4)that catalyzed the formation of the methylenedioxy bridge,an essential pharmacophoric group,on the A-and D-rings,respectively,of aporphine *** the functional characterization of these crucial cytochrome P450 enzymes,we reconstructed the biosynthetic pathways for the two types of aporphine alkaloids in budding yeast(Saccharomyces cerevisiae)for the de novo production of compounds such as(R)-glaziovine,(S)-glaziovine,and *** study provides key insight into the biosynthesis of aporphine alkaloids and lays a foundation for producing these valuable compounds through synthetic biology.

The human immunodeficiency virus （HIV） lentiviral vector is an ideal vector for gene therapy. In the present study, the wild-type HIV-1 genome was segregated into four plasmids, and an optimized novel HIV-1 lentiviral vector containing green fluorescent protein and vesicular stomatitis virus G pseudo-capsule was constructed. The plasmids were pHR-CMV-EGFP, pCMVΔ8.9, pRSV-Rev, pCMV-VSV-G. The four plasmid system was co-transfected into 293T cells, and green fluorescent protein expression was observed. The present study obtained lentiviral particles by high-speed centrifugation, and the lentiviral particle titer was 4 × 108 TU/mL after centrifugation. Thus, an optimized novel HIV-1 lentiviral vector was successfully constructed.

A genome-wide association study(GWAS)identifies trait-associated loci,but identifying the causal genes can be a bottleneck,due in part to slow decay of linkage disequilibrium(LD).A transcriptome-wide association study(TWAS)addresses this issue by identifying gene expression-phenotype associations or integrating gene expression quantitative trait loci with GWAS ***,we used self-pollinated soybean(Glycine max[L.]Merr.)as a model to evaluate the application of TWAS to the genetic dissection of traits in plant species with slow LD *** generated RNA sequencing data for a soybean diversity panel and identified the genetic expression regulation of 29286 soybean *** TWAS solutions were less affected by LD and were robust to the source of expression,identifing known genes related to traits from different tissues and developmental *** novel pod-color gene L2 was identified via TWAS and functionally validated by genome *** introducing a new exon proportion feature,we significantly improved the detection of expression variations that resulted from structural variations and alternative *** a result,the genes identified through our TWAS approach exhibited a diverse range of causal variations,including SNPs,insertions or deletions,gene fusion,copy number variations,and alternative *** this approach,we identified genes associated with flowering time,including both previously known genes and novel genes that had not previously been linked to this trait,providing insights complementary to those from *** summary,this study supports the application of TWAS for candidate gene identification in species with low rates of LD decay.
