Nano Research volume 13,pages2268–2279(2020)Cite this article 131 Accesses 1 Altmetric Metrics details Abstract Insufficient angiogenesis in the chronic wound of the diabetic is one of the most important causes that making the wound unable to heal *** this work,a cobalt-based metal–organic framework(ZIF-67)was introduced as a carrier for loading a pro-angiogenic small molecular drug(dimethyloxalylglycine,DMOG).To achieve a long-term angiogenic therapy on the diabetic wound beds,a dual cooperative controllable release system has been designed by incorporating the drug-loaded ZIF-67 nanoparticles into the micro-patterned PLLA/Gelatin nanofibrous *** results showed that DMOG was incorporated into ZIF-67 with a high loading ratio(359.12 mg/g),and the drug-loaded ZIF-67 nanoparticles were well embedded in the circular patterned ***,the DMOG as well as Co ions could continuously release from the scaffold for more than 15 *** in vitro studies showed that the released Co ions and DMOG from the micropatterned nanofibrous scaffolds could synergistically promote the proliferation,migration and tube formation of the human umbilical vein endothelial cells(HUVECs)by inducing a hypoxia response and upregulating the expression of angiogenesis-related genes such as HIF-1α,VEGF and ***,the in vivo results demonstrated that the composite scaffolds could significantly enhance angiogenesis,collagen deposition and eliminate inflammation in the diabetes *** results indicate that the cobalt-based metal–organic framework as a dual cooperative controllable release system provides a new strategy for enhancing angiogenesis and promoting diabetic wound healing.

Effects of fasting and refeeding on expression of MAFbx and MuRF1 in chick skeletal muscle

The present study investigated the effects of fasting and refeeding on the expression of proteasorne-related genes and their downstream targets in the skeletal muscles of chicks. Seven-day-old chicks were fasted for 24 or 48 h and then refed for 4 h. The expression levels of MAFbx and MuRF1, which function as E3 ligases in the ubiquitin-proteasome system, were investigated at the mRNA and protein levels. MAFbx and MuRF1 expression were increased by fasting and these increases were downregulated by refeeding. The expression of the target proteins of these E3 ligases, MyoD and M-CK, was also analyzed. The levels of these proteins were downregulated by fasting, and these decreases were rescued by refeeding. The results of this study indicate that fasting stimulates MAFbx and MuRF1 expression in chicks, possibly leading to increased degradation of their corresponding target proteins.

Aerobic exercise mitigates high-fat diet-induced cognitive decline through suppressing liver-brain-axis-mediated ATF3 and neuroinflammation

It is well known that appropriate aerobic exercise can effectively alleviate fatty liver and enhance brain function. The concept of multi-organ crosstalk coordinating disease progression has become the current research hot topic. However， there remains an urgent need to elucidate its specific mechanisms. This study aimed to explore the impact of a high-fat diet （HFD） on liver health and cognitive function， and to further uncover the regulatory effect of aerobic exercise by liver-specific ATF3 knockout (ATF3cKO) mice in a “liver-brain” axis mode. The 5-week-old C57BL/6 and ATF3cKOmice were fed with HFD for 32 weeks， and sequentially subjected to aerobic exercise intervention at the 20thweek for another 12 consecutive weeks. Meanwhile， C57BL/6 mice were provided with a normal diet as the control group. The functional parameters of liver and brain of all mice were assessed. Cognitive capacity of all mice was assessed by the Morris water maze （MWM）. Inflammatory factors in the serum and brain of mice were quantified using enzyme-linked immunosorbent assay （EliSA）， and the expression of inflammasomes was detected by immunohistochemistry （IHC）. Additionally， the activation of NF-κB and PI3K signal pathways was analyzed by Western blotting. In this study， HFD impaired hepatic and brain functions， while aerobic exercise and liver-specific ATF3 knockout suppressed inflammatory factors in the peripheral circulation through hepatoprotective mechanisms， thereby attenuating cerebral inflammation and preserving neurological integrity， as well as mitigating HFD-induced cognitive decline.

We report the clinical and laboratory findings and successful management of seven patients with critical coronavirus disease 2019(COVID-19)requiring mechanical ventilation(MV).The patients were diagnosed based on epidemiological history,clinical manifestations,and nucleic acid *** diagnosis with COVID-19 of critical severity,the patients were admitted to the intensive care unit,where they received early noninvasive-invasive sequential ventilation,early prone positioning,and bundle pharmacotherapy regimen,which consists of antiviral,anti-inflammation,immune-enhancing,and complication-prophylaxis *** patients presented fever(n=7,100%),dry cough(n=3,42.9%),weakness(n=2,28.6%),chest tightness(n=1,14.3%),and/or muscle pain(n=1,14.3%).All patients had normal or lower than normal white blood cell count/lymphocyte count,and chest computed tomography scans showed bilateral patchy shadows or ground glass opacity in the *** acid testing confirmed COVID-19 in all seven *** median MV duration and intensive care unit stay were 9.9 days(interquartile range,6.5-14.6 days;range,5-17 days)and 12.9 days(interquartile range,9.7-17.6 days;range,7-19 days),*** seven patients were extubated,weaned off MV,transferred to the common ward,and discharged as of the writing of this ***,we concluded that good outcomes for patients with critical COVID-19 can be achieved with early noninvasive-invasive sequential ventilation and bundle pharmacotherapy.

A recent outbreak of pneumonia in Wuhan, China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19). We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen, China. To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V, we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients. Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy *** cytokines were linked to 2019-nCoV loads. Fifteen cytokines, namely M-CSF, IL-10, IFN-α2,IL-17, IL-4, IP-10, IL-7, IL-1 ra, G-CSF, IL-12, IFN-γ, IL-1α, IL-2, HGF and PDGF-BB, were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics. Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines, some of which could be potential biomarkers of disease severity of 2019-n Co V infections. These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease. Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.

The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other *** we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,*** 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 *** viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease ***,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung ***,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease ***,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung *** results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.

敦煌吐鲁番文书中记栽了唐代沙州、西州官府民间大量使用非本地所产白练的现象。本文指出,沙州、西州的白练来自剑南道。从剑南至凉州、沙州,向伊州、西州、庭州,剑南道白练进入丝绸之路。通过与官府运输和客商贸易,剑南丝绸深入西域腹地;通过绢马(驼)互市,与游牧部族贸易,剑南丝绸走向了欧亚草原。本文估算盛唐时期每年有三百多万匹剑南道白练进入唐西北边疆,剑南道的白练,改变了河西地区的货币体系。
