IL-22/IL-22R1 pathway enhances cholangiocarcinoma progression via ERK1/2 activation

BACKGROUND IL-22 plays a pivotal role in the processes of inflammation and tissue healing., but its role in cholangiocarcinoma (CCA) remains unclear. our study explored the IL-22/IL-22R1 pathway and its impact on CCA progression through the ERK1/2 signaling cascade. AIM To determine the mechanism of the IL-22/IL-22R1 pathway in CCA and provide new directions for its clinical treatment. METHODS IL-22R1 expression was assessed in human and rat CCA tissues utilizing immunohistochemical techniques, Western blot analysis, and quantitative reverse transcription PCR. The impact of IL-22 on CCA cells was assessed in vitro via tests for proliferation, migration, invasion, and apoptosis assays. The rat models of thioacetamide-induced CCA and subcutaneous xenografts in nude mice were used to assess the in vivo effects. ERK1/2 inhibitors were applied to elucidate the mechanistic role of the pathway. RESULTS IL-22R1 was overexpressed in CCA cell lines and tissues. IL-22 treatment increased the phosphorylation of ERK1/2, promoting tumor cell proliferation, migration, invasion, and resistance to apoptosis. ERK1/2 inhibition considerably reversed these effects both in vitro and in vivo. CONCLUSION The IL-22/IL-22R1 axis promotes CCA progression by activating ERK1/2 signaling. Targeting this pathway with ERK1/2 inhibitors offers potential therapeutic strategies for CCA. © The Author(s) 2025. Published by Baishideng Publishing Group Inc. All rights reserved.

Gemcitabine-based combination therapy compared with gemcitabine alone for advanced pancreatic cancer： a meta-analysis of nine randomized controlled trials

BACKGROUND： Pancreatic cancer is one of the most aggressive malignancies and chemotherapy is an effective strategy for advanced pancreatic cancer. Gemcitabine （GEM） is one of first-line agents. However, GEM-based combination therapy has shown promising efficacy in patients with advanced pancreatic cancer. This meta-analysis aimed to compare the efficacy and safety of GEM-based combination therapy versus GEM alone in the treatment of advanced pancreatic *** SOURCES： A comprehensive search of literature was performed using PubMed, EMBASE, Web of Science and Cochrane Central Register of Controlled Trials. A quantitative meta-analysis was performed based on the inclusion criteria from all eligible randomized controlled trials. The outcome indicators included overall survival （OS）, 6-month survival, 1-year survival, progression-free survival/time-to-progression （PFS/TTP）, and toxicities. RESULTS： A total of nine randomized controlled trials involving 1661 patients were included in this meta-analysis. There was significant improvement in the GEM-based combination therapy with regard to the OS （HR=0.85, 95% CI： 0.76-0.95, P=0.003）, PFS （HR=0.76, 95% CI： 0.65-0.90, P-0.002）, 6-month survival （RR=1.09, 95% CI： 1.01-1.17, P=0.03）, and the overall toxicity （RR=l.68, 95% CI： 1.52-1.86, P〈0.01）. However, there was no significant difference in the 1-year ***： GEM-based combination chemotherapy might improve the OS, 6-month survival, and PFS in advanced pancreatic cancer. However, combined therapy also added toxicity.

The electronic density decay lengths of freestanding Pb films are investigated by first-principles *** results show that,like surface energy and work function,the electronic density decay lengthλexhibits pronounced oscillatory behavior as a function of film thickness and this is expected to have an impact on surface chemical *** freestanding Pb(111)films,λoscillates following a bilayer pattern interrupted by crossovers,and the separation between two neighbor crossovers is 9 *** the films on Si(111)substrates,the oscillations of the decay lengths are similar to those of freestanding films except for an extra phase shift.

Inorganic Nanotube/Organic Nanoparticle Hybrids for Enhanced Photoelectrochemical Properties

Inorganic/organic nanohybrids composed of arrayed TiO_2 nanotubes（Ti NTs）/porphyrin nanoparticles（NPs） have been fabricated via a wet chemical approach. The inorganic component, particularly the arrayed one-dimensional（1D） nanostructures, provides high charge-carrier mobility and rapid charge transport. The organic component exhibits extensive visible light absorption and good solution processability. Additionally, the geometric restraint by supramolecular assembly renders an improved photostability. A combination of these two components could thus allow for an efficient solar energy conversion. In this work, a colloid of porphyrin NPs prepared by a solvent exchange method is coated on anodic Ti NTs by means of a dip-coating treatment to form inorganic/organic hybrids. The hybrids exhibit an improvement on solar absorption and a significant enhancement on photocurrent generation at a small bias compared with individual component. Herein, the inorganic/organic nanohybrids are proved to be excellent photoanodes highly responsive to visible light and thus pave a way to discover new inorganic/organic assemblies for high-performance optoelectronic applications, as well as for device integration.

Is short-term therapy really sufficient to eradicate Helicobacter pylori infection?

Objective:The aim of our study was to perform a systematic review and meta-analysis of the efficacy of short-term protocols for Helicobacter pylori eradication and to review the safety and adverse profiles of these eradication ***:literatures were located through electronic searches by PubMed,Medline,ISI Web of Knowledge,and Cochrane library using the relevant *** of important meetings were searched manually in some journal *** bibliographies were identified from the reference lists of identified *** independent reviewers systemically identified randomized controlled trials(RCTs) comparing short-duration protocols vs.7-d proton pump inhibitor(PPI)-based triple protocols,as well as studies reporting eradication rates of short-duration protocols for *** effect size was calculated as relative risk(RR) and 95% confidence intervals(CI) using Review Manager 4.2,and Plightly fewer adverse-effects were found in short-term ***:Although more economical,short-duration protocols are inferior to 7-d PPI-based triple protocols with regarding to eradication rate of *** of more than 3 d,however,may be equivalent to 7-d protocols.

BACKGROUND: liver transplantation is so far the most effective therapeutic modality for end-stage liver diseases, but ischemia/reperfusion (I/R) injury represents a critical barrier to liver transplantation. Primary graft dysfunction and small-for-size syndrome are closely associated with I/R injury. Ischemic preconditioning (IPC) is defined as a brief period of liver ischemia followed by reperfusion, and has demonstrated protections against a prolonged I/R injury and improved the capacity of regeneration. The article aimed to review IPC literatures for the understanding of the effects of IPC on I/R injury involving in the procurement of donor liver and protective mechanisms. DATA SOURCES: A literature search of MEDliNE and Web of Science databases using 'liver transplantation', 'liver regeneration', 'hepatectomy', 'ischemia/reperfusion' and 'ischemic preconditioning' was performed, and then a large amount of related data was collected. RESULTS: The literature search provided a huge amount of evidence for the protective effects of IPC on I/R injury in liver transplantation, including reduction of blood loss in hepatectomy, intraoperative hemodynamic stability and its significant role in liver regeneration. The mechanism involves in balancing inflammatory cytokines, enhancing energy status and mitigating microcirculatory disturbance. CONCLUSION: IPC plays an essential role in hepatectomy before and after harvest of living donor liver and implantation of liver graft.

An empirical model for high energy density lithium-(ion)batteries with ultra-thick electrodes

Increasing the electrode thickness is a significant method to decrease the weight and volume ratio of the inactive components for high energy density of the *** this contribution,we extracted a repeating unit in the configurations and establish the empirical energy density model based on some *** this model,the effects of the electrode thickness on the energy density for lithium-ion batteries(liBs),lithium metal batteries(LMBs),and anode-free lithium batteries(ALBs)are evaluated quantitively with the current parameters of the *** results demonstrate that the structure evolutions from liBs,LMBs to ALBs with the reduction of the anode weight contribution,the energy density can be well improved *** the increase of the thickness of the electrode provide another route to furthe r enhance the energydensity by decreasing the weight contribution of inactive materials;meanwhile the effects for ALBs are higher than LMBs and liBs due to the higher weight ratio of inactive *** empirical energy density model is also applied into the practical system and provide intuitional results to guide the battery design for higher energy density.
