Effect of moxibustion on expression profile of miRNAs in Tripterygium glycoside-induced decreased ovarian reserve

OBJECTIVE: To explore the possible regulatory mechanism of microRNA(miRNA) in moxibustion treatment for decreased ovarian reserve(DOR). METHODS: The DOR model was constructed by intragastrical Tripterygium glycoside suspension administration, and moxibustion therapy was simultaneously given. The morphological ovarian changes were observed by hematoxylin and eosin staining. The miRNA expression profile was detected by RNA sequencing, and bioinformatics analysis was performed. Cytoscape software 3.6.1 was used to establish a regulatory network and differentially expressed miRNAs were verified by reverse transcription quantitative polymerase chain reaction(RT-qPCR). RESULTS: Decreased number of mature follicles, increased atresia follicles, and abnormal granulosa cell morphology were observed in the model group compared with the control group. The moxibustion group demonstrated increased mature follicles, decreased atretic follicles, and significantly decreased abnormal morphology of granulosa cells compared with the model group. Additionally, RNA sequencing results manifested significantly up-regulated miRNA expressions(miR-92b-3p, miR-26-5p＿R + 1＿1ss10TC, miR-206-3p, miR-9993b-3p＿1ss6GA, miR-7857-3p＿R-1, miR-219a-2-3p＿1ss10GC, miR-3968-p5＿1ss10AT, and PC-5p-6478＿1795) and down-regulated miR-664-2-5p＿R + 1 in the model group, compared with the control group, and the moxibustion group reversed abnormal disorder levels of these miRNAs. Moreover, these differentially expressed miRNAs were mainly involved in the phosphatidylinositol-3-kinase/protein kinase B signaling pathway and nuclear factor erythropoietin-2-related factor 2/heme oxygenase 1 signaling pathway. Finally, network and RT-qPCR verification revealed miR-9993b-3p＿1ss6GA as the most critical miRNA. CONCLUSION: This experiment proved the effectiveness of moxibustion in improving the ovarian reserve of rats by regulating miRNA expression, especially miR-9993b-3p＿1ss6GA.

目的在分析变形量为25%的小变形拉拔加工Cu-0.08Ag合金组织与性能的基础上,研究不同退火温度对小变形拉拔Cu-0.08Ag合金微观组织、力学性能和导电性能的影响规律,为Cu-Ag合金电磁线漆包工艺的制定提供参考。方法利用电子背散射衍射(EBSD)技术对Cu-0.08Ag合金小变形拉拔及200~500℃不同温度退火1 h后的样品进行组织表征,结合显微硬度计、拉伸试验机和电阻计等手段对性能进行综合分析。结果经变形量为25%的小变形拉拔后,Cu-0.08Ag合金等轴晶组织部分晶粒沿变形方向被拉长,和取向占比均增大,应力集中分布在晶粒内,屈服强度提高了270%,而电导率仅下降了4.1%IACS。小变形拉拔Cu-0.08Ag合金的软化温度为350℃。与小变形拉拔态相比,在300℃及以下退火时显微组织和力学性能未发生明显变化,而电导率提升了2.9%IACS。在400℃及以上退火时,无畸变的等轴晶粒取代变形晶粒,并产生大量的退火孪晶,晶粒取向随机分布,力学性能和电导率与小变形拉拔前的相当。结论在300℃以下退火后,小变形拉拔Cu-0.08Ag合金在保持较高屈服强度的同时,其电导率也获得了提升,可获得强度和电导率良好的匹配关系。

Discrimination of periodontal pathogens using Raman spectroscopy combined with machine learning algorithms

在工程机械和矿山机械的重载工作条件下,斜齿锥齿轮在啮合时易引发振动与冲击,对其实施动力学分析显得尤为关键。运用有限元方法分析斜齿锥齿轮副的性能特征,通过设置齿轮副接触条件和施加载荷,识别出系统阻尼、启动加速时间及负载对动态啮合特性及振动响应,并采用正交实验法和极差法得出三个影响因素对斜齿锥齿轮振动的响应程度,揭示了齿面接触力和角速度随时间变化的规律。经过仿真结果表明,阻尼系数和加速时间的变化会显著影响齿轮啮合的稳定时间,其中较大阻尼系数有助于缩短稳定时间,而较短的加速时间导致更大的初始冲击,负载条件主要决定了啮合后的振动幅度,负载过大会导致啮合质量变差,从而影响齿轮的正常运行。

重金属离子具有环境持久性、生物累积性和生物毒性高等特点,对人类的生命安全造成巨大威胁。因此,开发灵敏度高、选择性强、快速便捷的重金属离子检测方法具有重要意义。发光金属纳米簇(Metal nanoclusters,MNCs)因其生物相容性好、尺寸小易于标记、光稳定性强和毒性低等优点,在重金属离子检测方面受到广泛关注。金属纳米簇传感的灵敏度与其荧光性能密切相关。深刻理解金属纳米簇荧光的增强机制,是提升基于金属纳米簇对重金属离子传感性能的基石。基于此,本文总结归纳了金属纳米簇荧光性能的增强机制并介绍了金属纳米簇在重金属离子检测方面的应用及其发展前景,为高效金属纳米簇的开发和应用提供参考。

Assessment of Co-Cr-Ni ternary system by CALPHAD technique

The Co-Cr-Ni ternary system was critically assessed using the CALPHAD technique. The solution phases including the liquid, γ(Co,Ni), ɛCo and αCr phases were described by a substitutional solution model. The σ phase was characterized by a three-sublattice model of (Co,Ni)8Cr4(Co,Cr,Ni)18 in order to reproduce its homogeneity range determined by experiments. A self-consistent set of thermodynamic parameters for each phase was derived.

Phase equilibria in the Mg-rich corner of the Mg-Zn-La system at 350°C

The phase equilibria in the Mg-rich corner of the Mg-Zn-La system at 350°C have been investigated by scanning electron microscopy, X-ray diffraction, and electron probe microanalysis. It has been shown that the linear compound (Mg,Zn)17La2 existed in the Mg-Zn-La system at 350°C. The linear compound (so-called T phase) was with the C-centred orthorhombic crystal structure induced by the solution of significant quantities of the third element. The three-phase region α(Mg) + MgZn(La) + T and the two-phase region composed of the α(Mg) and the linear-compound T phase existed in the Mg-rich corner of the Mg-Zn-La system at 350°C.

Thermodynamic prediction of the composition of Fe-based amorphous alloys

The liquidus univariant lines of the Fe-Nb-B ternary system have been thermodynamically calculated by means of CALPHAD method and Fe-based thermodynamic data. It is found that there are two eutectic reactions in the Fe-rich corner,that is,（1） L（Fe-3Nb-15B） →α＋γ＋ M2B （1430 K）,and （2） L（Fe-10Nb-27B） → FeB ＋ Lc14 ＋ M2B （1575 K）. Moreover,the eutectic points are very close to the compositions with high glass forming ability determined experimentally. This means that it is feasible to design the compositions of multicomponent bulk metallic glasses by looking for the eutectic points in the Fe-Nb-B system by means of thermodynamic calculation.

Effect of Cu on the Spinodal Decomposition of AlZn Alloy with Symmetrical Composition

The effect of Cu addition on the spinodal decomposition of the Al-Zn alloy with symmetrical compositions has been investigated by X-ray diffraction analysis. It is found that the single fcc phase can be obtained in the AlZn alloy with the addition of 2 at. pct Cu after solution treatment at 400℃ and water quenching to room temperature. The modulation structure occurs in two types of alloys aged at room temperature for 30 min. The spinodal process remarkably slows down above or at the three phase equilibrium temperature, particularly at room temperature due to the addition of 2 at. pct Cu. The spinodal time increases by 20 times when aging at 300℃ and by more than 30 times when aging at room temperature.
