Establishment and characterization of immortalized bovine male germline stem cell line

Male germline stem cells （mGSCs） are unique adult germ cells with self-renewal potential and spermatogenesis function in the testis. However, further studies are needed to establish a long-term cultural system of mGSCs in vitro, especially for large animals such as bovine mGSCs. In this study, we first established a stable immortalized bovine male germline stem cell line by transducing Simian virus 40 （SV40） large T antigen. The proliferation of these cells was improved significantly. These cells could express spermatogonial stem cell （SSC）-specific markers, such as PLZF, PGP9.5, VASA, liN28A, and CD49F, both in the mRNA and protein levels. Additionally, these cells could be differentiated into three germ layer cells to enter meiosis, form colonies, and proliferate in the seminiferous tubules of busulfan-induced infertile mice. The immortalized bovine mGSCs maintain the criteria of mGSCs.

Spermatogonial stem cells（SSCs） are unique stem cells in adult body that can transmit genetic information to the next generation. They have self-renewal potential and can continuously support spermatogenesis throughout life of a male animal. However, the SSC population is extremely small, isolation and purification of the SSCs is challenging, especially for livestock animals. It has been confirmed that CDH1（cadherin-1, also known as E-cadherin） can be expressed in undifferentiated SSCs of mouse and rats, but it has not been verified in sheep. Here, CDH1 was found as a novel surface marker for sheep SSCs. In this paper, sheep antiCDH1 polyclonal antibodies were prepared and its activity was checked. Using the obtained antibodies and immunohistochemistry analysis, we confirmed that CDH1 can be expressed by SSCs in sheep testis.

The fatigue resistance of casting polyurethane(CPU)is crucial in various sectors,such as construction,healthcare,and the automotive *** its importance,no studies have reported on the fatigue threshold of *** study employed an advanced Intrinsic Strength Analyzer(ISA)to evaluate the fatigue threshold of CPUs,systematically exploring the effects of three types of isocyanates(PPDI,NDI,TDI)that contribute to hard segment structures based on the cutting *** multiple advanced characterization techniques(XRD,TEM,DSC,AFM),the results indicate that PPDI-based polyurethane exhibits the highest fatigue threshold(182.89 J/m^(2))due to a highest phase separation and a densely packed spherulitic structure,although the hydrogen bonding degree is the lowest(48.3%).Conversely,NDI-based polyurethane,despite having the high hydrogen bonding degree(53.6%),exhibits moderate fatigue performance(122.52 J/m^(2)),likely due to a more scattered ***-based polyurethane,with the highest hydrogen bonding degree(59.1%)but absence of spherulitic structure,shows the lowest fatigue threshold(46.43 J/m^(2)).Compared to common rubbers(NR,NBR,EPDM,BR),the superior fatigue performance of CPU is attributed to its well-organized microstructure,polyurethane possesses a higher fatigue threshold due to its high phase separation degree and orderly and dense spherulitic structure which enhances energy dissipation and reduces crack propagation.

A new passive control approach,utilizing bionic slanting riblets,is employed to mitigate the flow close to the blade endwall in a linear cascade,and its effectiveness and mechanism in controlling corner separation are investigated through numerical *** slanting riblets are positioned at the endwall upstream of the cascade channel,and the influence of riblet height,yaw angle and relative position on the control of corner separation is *** findings indicate that the application of slanting riblets can efficiently counteract corner separation across the stable operational ***,the introduction of riblets with a height of merely 0.1 times the boundary layer thickness results in a significant reduction in total pressure loss by up to 14.53%,while simultaneously enhancing the static pressure coefficient by 21.74%.Flow analysis reveals that minute vortices produced within the riblet channels tend to coalesce,forming a potent large-scale vortex near the boundary layer's base *** phenomenon results in reduced additional losses compared to conventional vortex ***,the induced vortex promotes enhanced mixing between the mainstream flow and boundary layer,inhibiting the lateral displacement of low-energy fluids within the endwall boundary ***,this delays the onset of separation vortex formation and eliminates vortex rings in the corner region,ultimately enhancing the aerodynamic efficiency of the cascade.

The crevice corrosion of the reinforcing steel in the carbonated simulated concrete pore solutions(SCPSs)containing 0.04 mol/L chloride was *** comparison,the steel without crevice covered on its surface was also *** showed that the crevice corrosion in the uncarbonated SCPS was not triggered and the steel remained passivity by suppressing the anodic dissolution in the crevice *** SCPS was carbonated to have pH values of 10.5 and 11.5,the crevice corrosion was easier to be activated than the widely reported pitting corrosion owing to the rapid oxygen depletion within the crevice,but the crevice corrosion damage might be alleviated to a certain extent due to the relative compact surface film formed inside the *** further carbonation led to the activation dissolution of both the crevice interior and exterior,causing the more serious corrosion *** process of the crevice corrosion was systematically analyzed by combining the electrochemical measurement results and Evens polarization diagram.

Understanding the mechanical properties of the lithologies is crucial to accurately determine the horizontal stress *** investigate the correlation between the rock mass properties and maximum horizontal stress,the three-dimensional(3D)stress tensors at 89 measuring points determined using an improved overcoring technique in nine mines in China were adopted,a newly defined characteristic parameter C_(ERP)was proposed as an indicator for evaluating the structural properties of rock masses,and a fuzzy relation matrix was established using the information distribution *** results indicate that both the vertical stress and horizontal stress exhibit a good linear growth relationship with *** is no remarkable correlation between the elastic modulus,Poisson's ratio and depth,and the distribution of data points is scattered and ***,there is no obvious relationship between the rock quality designation(RQD)and *** maximum horizontal stress σ_(H) is a function of rock properties,showing a certain linear relationship with the C_(ERP)at the same *** addition,the overall change trend of σ_(H) determined by the established fuzzy identification method is to increase with the increase of C_(ERP).The fuzzy identification method also demonstrates a relatively detailed local relationship betweenσ_H and C_(ERP),and the predicted curve rises in a fluctuating way,which is in accord well with the measured stress data.

Transition metal-based compounds can serve as pre-catalysts to obtain genuine oxygen evolution reaction（OER） electrocatalysts in the form of oxyhydroxides through electrochemical ***,the role and existence form of leached oxygen anions are still ***,we selected iron selenite-wrapped hydrated nickel molybdate（denoted as NiMoO/FeSeO） as a pre-catalyst to study the oxyanion *** is surprising to find that SeO2-exists in the catalyst in the form of intercalation,which is different from previous studies that suggest that anions are doped with residual elements after electrochemical activation,or adsorbed on the catalyst *** experiment and theoretical calculations show that the existence of SeO42-intercalation effectively adjusts the electronic structure of NiFeOOH,promotes intramolecular electron transfer and O-O release,and thus lowers the reaction energy *** expected,the synthesized NiFeOOH-SeO only needs 202 and 285 mV to attain 100 and 1000 mA cm-2in 1 M ***,the anion exchange membrane water electrolyzer（AEMWE） consisting of NiFeOOHSeO anode and Pt/C cathode can reach 1 A cm-2at 1.70 V and no significant attenuation within 300 *** findings provide insights into the mechanism,by which the intercalated oxyanions enhance the OER performance of NiFeOOH,thereby facilitating large-scale hydrogen production through AEMWE.
