Biased activity of soluble guanylyl cyclase:the Janus face of thymoquinone

The natural c.m.ound thymoquinone, extracted from Nigella sativa(black c.m.n), is widely used in humans for its anti-oxidative properties. Thymoquinone is known for its acute endotheliumindependent vasodilator effects in isolated rat aortae and pulmonary arteries, depending in part on activation of adenosine triphosphate-sensitive potassium channels and inhibition of voltage-dependent calcium channels. The c.m.ound also improves endothelial dysfunction in mesenteric arteries of ageing rodents and in aortae of rabbits treated with pyrogallol, by inhibiting oxidative stress. Serendipitously,thymoquinone was found to augment contractions in isolated arteries with endothelium of both rats and pigs. The endothelium-dependent augmentation it causes counterintuitively depends on biased activation of soluble guanylyl cyclase(sGc) producing inosine 3',5'-cyclic.m.nophosphate(cyclic ImP) rather than guanosine 3',5'-cyclic.m.nophosphate. This phenomenon shows a striking mechanistic similarity to the hypoxic augmentation previously observed in porcine coronary arteries. The cyclic ImP preferentially produced under thymoquinone exposure causes an increased contractility of arterial smooth muscle by interfering with calcium homeostasis. This brief review summarizes the vascular pharmacology of thymoquinone, focussing in particular on how the c.m.ound causes endothelium-dependent contractions by biasing the activity of sGc.

Revealing the residual stress distribution in laser welded Eurofer97 steel by neutron diffraction and Bragg edge imaging

Eurofer97 steel is a primary structural material for applications in fusion reactors. Laser welding is a promising technique to join Eurofer97 plasma-facing c.m.onents and overc.m. remote handling and maintenance challenges. The interaction of the induced residual stress and the heterogeneous microstructure degrades the mechanical performance of such fusion c.m.onents. The present study investigates the distribution of residual stress in as-welded and post-heat treated Eurofer97 joints. The mechanistic connections between microstructure, material properties, and residual stress are also studied. Neutron diffraction is used to study the through-thickness residual stress distribution in three directions,and neutron Bragg edge imaging(NBEI) is applied to study the residual strain in high spatial *** microstructures and micro-hardness are characterised by electron backscatter diffraction and nanoindentation, respectively. The m-shaped residual stress distribution through the thickness of the as-welded weldment is observed by neutron diffraction line scans over a region of 1.41 × 10 mm^(2). These profiles are cross-validated over a larger area(∼56 × 40 mm^(2)) with the higher spatial resolution by NBEI. The micro-hardness value in the fusion zone of the as-welded sample almost doubles from 2.75 ± 0.09 GPa to 5.06 ± 0.29 GPa due to a c.m.ination of residual stress and cooling-induced martensite. conventional post weld heat treatment(PWHT) is shown to release ∼90% of the residual stress but not fully restore the microstructure. By c.m.aring its hardness with that of stress-free samples, it is found that the microstructure is the primary contribution to the hardening. This study provides insight into the prediction of structural integrity for critical structural c.m.onents of fusion reactors.

Afteruse development of former landfll sites in Hong Kong

Landfll is one of the major methods in disposing of municipal solid waste(mSW) in Hong Kong. There are now 13 closed landfll sites in Hong Kong, and a restoration program has been launched since 1999 to transform the closed landfll sites back into recreational area. The mSW underneath these closed landfll sites will biodegrade slowly and gradually, releasing toxic gases and leachate. As there have only been a few afteruse development ever c.m.leted in Hong Kong, this paper describes the consideration in planning and design of two former landfll sites in Hong Kong–Ngau chi Wan(NcW) and Jordan Valley(JV) landflls, in particular a discussion on the mSW settlement. Various models in predicting the longterm mSW settlement have been available. This paper reviews the applicability of these models to predict mSW settlement. However, as the predicted settlements can be signifcantly in error, they need sitespecifc calibration and validation. This paper therefore presents the settlement monitoring data in these former landfll sites over the last 30 years, and suggests modifying the Bjarngard and Edgers’ method for estimating this long-term settlement as well as the effect of additional surcharge due to afteruse development. Based on the regression of these data, c.m.ression indices cce and c at different stages of the settlement in the modifed method have been calibrated and validated.

The anatomical architecture of the human liver and the diversity of its immune c.m.onents endow the liver with its physiological function of immune c.m.etence. Adaptive immunity is a major arm of the immune system that is organized in a highly specialized and systematic.m.nner, thus providing long-lasting protection with immunological memory. Adaptive immunity consists of humoral immunity and cellular immunity. cellular immunity is known to have a crucial role in controlling infection, cancer and autoimmune disorders in the liver. In this article, we will focus on hepatic virus infections, hepatocellular carcinoma and autoimmune disorders as examples to illustrate the current understanding of the contribution of T cells to cellular immunity in these maladies. cellular immune suppression is primarily responsible for chronic viral infections and cancer. However, an uncontrolled auto-reactive immune response accounts for autoimmunity. consequently, these immune abnormalities are ascribed to the quantitative and functional changes in adaptive immune cells and their subsets, innate immunocytes, chemokines, cytokines and various surface receptors on immune cells. A greater understanding of the c.m.lex orchestration of the hepatic adaptive immune regulators during homeostasis and immune c.m.etence are much needed to identify relevant targets for clinical intervention to treat immunological disorders in the liver.
