Breast cancer is the predominant form of carcinoma among women worldwide,with 70%of advanced patients developing bone metastases,with a high mortality *** this sense,the bone marrow(BM)mesenchymal stem/stromal cells(MSCs)are critical for BM/bone homeostasis,and failures in their functionality,transform the BM into a premetastatic niche(PMN).We previously found that BM-MSCs from advanced breast cancer patients(BCPs,infiltrative ductal carcinoma,stage III-B)have an abnormal *** work aims to study some of the metabolic and molecular mechanisms underlying MSCs shift from a normal to an abnormal profile in this group of patients.A comparative analysis was undertaken,which included self-renewal capacity,morphology,proliferation capacity,cell cycle,reactive oxygen species(ROS)levels,and senescence-associatedβ‑galactosidase(SA‑β‑gal)staining of BMderived MSCs isolated from 14 BCPs and 9 healthy volunteers(HVs).Additionally,the expression and activity of the telomerase subunit TERT,as well as telomere length,were *** levels of pluripotency,osteogenic,and osteoclastogenic genes(OCT-4,SOX-2,M-CAM,RUNX-2,BMP-2,CCL-2,M-CSF,and IL-6)were also *** results showed that MSCs from BCPs had reduced,self-renewal and proliferation *** cells also exhibited inhibited cell cycle progression and phenotypic changes,such as an enlarged and flattened ***,there was an increase in ROS and senescence levels and a decrease in the functional capacity of TERT to preserve telomere *** also found an increase in pro-inflammatory/pro-osteoclastogenic gene expression and a decrease in pluripotency gene *** conclude that these changes could be responsible for the abnormal functional profile that MSCs show in this group of patients.

Spatial and temporal patterns of forest fires in the Central Monte:relationships with regional climate

Background Natural and anthropogenic wildfires burn large areas of arid and semi-arid forests with significant socio-economic and environmental impacts. Fire regimes are controlled by climate, vegetation type, and anthropogenic factors such as ignition sources and human-induced disturbances. Projections of climate and land-use change suggest that these controlling factors will change, altering fire regimes in the near future. In the southern Central Monte, Mendoza, Argentina, the factors that modulate the fire temporal and spatial variability are poorly understood. We reconstructed the fire history of southeast of Mendoza from 1984 to 2023 and investigated the relationships between fire extent and climate variability at seasonal and interannual scales. Burned areas were determined using Google Earth Engine by processing Landsat 5-TM, Landsat 7-ETM+, and Landsat 8-OLI-TIRS sensor *** The region exhibited high spatial and temporal variability in fire occurrence, being a mosaic of areas with different fire histories and recovery times. Between 1985 and 2023, fire recurrence ranged from sites unburned to sites with up to 14 fires. The occurrence of large fires was strongly favored by a combination of a year with abundant spring–early summer precipitation, which favors fuel accumulation, followed by a year of low spring–early summer precipitation. Precipitation and burnt area showed a very pronounced 6–7 year cycle, suggesting a dominant climatic control on fire *** Fire distribution in southeastern Mendoza forests is not homogeneous, resulting in a mosaic of patches with different fire histories. This heterogeneity may be related to vegetation patterns and land use. The temporal variability of fires is strongly influenced by climate variability, which would promote fuel production and subsequent drying. Intelligent speed adaptation for visibility technology affects drivers’speed selection along curves with sight limitations

Sight obstructions along road curves can lead to a crash if the driver is not able to stop the vehicle in *** is a particular issue along curves with limited available sight,where speed management is necessary to avoid unsafe situations(e.g.,driving off the road or invading the other traffic lane).To solve this issue,we proposed a novel intelligent speed adaptation(ISA)system for visibility,called V-ISA(intelligent speed adaptation for visibility).It estimates the real-time safe speed limits based on the prevailing sight conditions.V-ISA comes with three variants with specific feedback modalities(1)visual and(2)auditory information,and(3)direct intervention to assume control over the vehicle ***,we investigated the efficiency of each of the three V-ISA variants on driving speed choice and lateral behavioural response along road curves with limited and unsafe available sight distances,using a driving *** also considered curve road geometry(curve direction:rightward ***).Sixty active drivers were recruited for the *** half of them(experimental group)tested the three V-ISA variants(and a V-ISA off condition),the other half always drove with the V-ISA off(validation group).We used a linear mixed-effect model to evaluate the influence of V-ISA on driver *** V-ISA variants were efficient at reducing speeds at entrance points,with no discernible negative impact on driver lateral *** rightward curves,the V-ISA intervening variant appeared to be the most effective at adapting to sight *** of the current study implies that V-ISA might assist drivers to adjust their operating speed as per prevailing sight conditions and,consequently,establishes safer driving conditions.

Modelling of the variation of granular base materials resilient modulus with material characteristics and humidity conditions

This study aims to quantify the susceptibility of granular materials used in pavements to changes in moisture content and propose a correlation model to incorporate this susceptibility into seasonal *** fines content and the percentage of fractured coarse aggregates were identified as direct indicators of the resilient modulus susceptibility to changes in water *** results showed that the percentage of fractured coarse aggregates particles(FR)has a more significant impact on the resilient modulus(Er)of crushed granular materials used in pavement construction than the combined indicator of the fines content and sample volumetrics(nf).Crushed granular materials with a higher percentage of fractured coarse aggregates are relatively insensitive to changes in the degree of saturation,but become more sensitive as the fine fraction porosity *** adjusted model was proposed based on the existing formulation,but considers a complex parameter to describe and adjust the sensitivity of base granular materials to variations in moisture content with respect to fabrication charac-teristics,fines content and volumetric *** model shows that the variation of Er values is below10%for fully crushed granular ***,it reaches approximately±12%for materials with 75%of crushed coarse aggregates andþ40%and-25%for materials with FR=50%.This model could help select good ag-gregates characteristics and adjust grain-size distribution for environments where significant moisture content variations can occur in the pavement system,such as in the Province of Quebec(Canada).As it is based on pa-rameters that can be easily determined or estimated,it also represents a valuable tool for detailed design and analysis that can consider material characteristics.

We and others have published extensively on the benefits of physical activity(PA),exercise training,and cardiorespiratory fitness(CRF)to decrease the prevalence of cardiovascular diseases(CVDs)and CVD and all-cause morbidity and mortality.^(1-5)Both PA and CRF are important;however,CRF,which is mostly obtained through PA,although there is a genetic component,seems to be even more important than PA for predicting prognosis.^(1,4-6)Certainly,the wealth of data from across the world has indicated that increasing PA to enhance CRF while mitigating sedentary behavior would go a long way to lessen the world-wide obesity epidemic and reduce CVD and all-cause mortality.

Transcriptomic responses to drought stress in the Patagonian southern beech Nothofagus alpina

Background Deciphering the genetic architecture of drought tolerance could allow the candidate genes identification responding to water stress. In the Andean Patagonian forest, the genus Nothofagus represents an ecologically relevant species to be included in different genomic studies. These studies are scarce in South American ecosystems however represent an important source of genomic data in order to interpret future climate-change environment scenarios of these emblematic forests. Here, we achieved the assemblage of the transcriptome of N. alpina while searching for key genes of activated or suppressed metabolic pathways in response to drought *** De novo transcriptome assembly resulted in 104,030 transcripts. Following confirmation of drought conditions, based on reduction of leaf water potential and stomatal conductance, a differential gene expression analysis resulted in 2720 significantly expressed genes(1601 up-regulated and 1119 down-regulated). Enrichment analysis(over-representation analysis and gene set enrichment analysis) resulted in more than one hundred stress-responsive term ontologies(i.e. biological processes) and pathways. Terms such as response to abscisic acid and pathways such as plant hormone signal transduction or starch and sucrose metabolism were over-represented. Protein–protein interaction assessment resulted in networks with significantly expressed top common hub gene clusters(e.g. planttype cell wall biogenesis among down-regulated or ABA-signalling among up-regulated). These networks evidenced important regulators at gene expression such as transcriptional *** Responses of N. alpina seedlings to drought stress were evidenced by the activation of several genes linked to GO biological processes and KEGG pathways, which were mainly based on over-expression of specific protein kinases, phosphatases, synthases and transcription factors.
