The therapeutic outcomes of conventional oral medications against ulcerative colitis(UC)are restricted by inefficient drug delivery to the colitis mucosa and weak capacity to modulate the inflammatory ***,a fluorinated pluronic(FP127)was synthesized and employed to functionalize the surface of mulberry leaf-derived nanoparticles(MlNs)loading with resveratrol nanocrystals(RNs).The obtained FP127@RN-MlNs possessed exosome-like morphologies,desirable particle sizes(around 171.4 nm),and negatively charged surfaces(−14.8 mV).The introduction of FP127 to RN-MlNs greatly improved their stability in the colon and promoted their mucus infiltration and mucosal penetration capacities due to the unique fluorine effect.

For the first time, sulfanilamide(sFD) was determined in otologic solution, human urine and serum by electroanalytical techniques on glassy carbon electrode(GCE). The cyclic voltammetry(CV) experiments showed an irreversible oxidation peak at t 1.06 V in 0.1 mol/l BRBs(p H ? 2.0) at 50 m V/s. Different voltammetric scan rates(from 10 to 250 m V/s) suggested that the oxidation of sFD on the GCE was a diffusioncontrolled process. square-wave voltammetry(sWV) method under optimized conditions showed a linear response to sFD from 5.0 to 74.7 μmol/l(R ? 0.999) with detection and quantification limits of 0.92 and3.10 μmol/l, respectively. The developed sWV method showed better results for detection limit and linear range than the chronoamperometry method. It has been successfully applied to determine sFD concentration in pharmaceutical formulation, human urine and serum samples with recovery close to 100%.

Background: Genomic growth curves are general y defined only in terms of population mean; an alternative approach that has not yet been exploited in genomic analyses of growth curves is the Quantile Regression(QR). This methodology allows for the estimation of marker effects at different levels of the variable of interest. We aimed to propose and evaluate a regularized quantile regression for sNP marker effect estimation of pig growth curves, as well as to identify the chromosome regions of the most relevant markers and to estimate the genetic individual weight trajectory over time(genomic growth curve) under different quantiles(levels).Results: The regularized quantile regression(RQR) enabled the discovery, at different levels of interest(quantiles), of the most relevant markers al owing for the identification of QTl regions. We found the same relevant markers simultaneously affecting different growth curve parameters(mature weight and maturity rate): two(AlGA0096701 and AlGA0029483)for RQR(0.2), one(AlGA0096701) for RQR(0.5), and one(AlGA0003761) for RQR(0.8). Three average genomic growth curves were obtained and the behavior was explained by the curve in quantile 0.2, which differed from the ***: RQR allowed for the construction of genomic growth curves, which is the key to identifying and selecting the most desirable animals for breeding purposes. Furthermore, the proposed model enabled us to find, at different levels of interest(quantiles), the most relevant markers for each trait(growth curve parameter estimates) and their respective chromosomal positions(identification of new QTl regions for growth curves in pigs). These markers can be exploited under the context of marker assisted selection while aiming to change the shape of pig growth curves.

The cytokine granulocyte-macrophage-colony stimulating factor (GM-CsF) possesses the capacity to differentiate monocytes into macrophages (MØs) with opposing functions, namely, proinflammatory M1-like MØs and immunosuppressive M2-like MØs. Despite the importance of these opposing biological outcomes, the intrinsic mechanism that regulates the functional polarization of MØs under GM-CsF signaling remains elusive. Here, we showed that GM-CsF-induced MØ polarization resulted in the expression of cytokine-inducible sH2-containing protein (CIs) and that CIs deficiency skewed the differentiation of monocytes toward immunosuppressive M2-like MØs. CIs deficiency resulted in hyperactivation of the JAK-sTAT5 signaling pathway, consequently promoting downregulation of the transcription factor Interferon Regulatory Factor 8 (IRF8). loss- and gain-of-function approaches highlighted IRF8 as a critical regulator of the M1-like polarization program. In vivo, CIs deficiency induced the differentiation of M2-like macrophages, which promoted strong Th2 immune responses characterized by the development of severe experimental asthma. Collectively, our results reveal a CIs-modulated mechanism that clarifies the opposing actions of GM-CsF in MØ differentiation and uncovers the role of GM-CsF in controlling allergic inflammation.

Papillary fistulotomy vs conventional cannulation for endoscopic biliary access:A prospective randomized trial

AIM To compare the cannulation success, biochemical profile, and complications of the papillary fistulotomy technique vs catheter and guidewire standard *** From July 2010 to May 2017, patients were prospectively randomized into two groups: Cannulation with a catheter and guidewire(Group Ⅰ) and papillary fistulotomy(Group Ⅱ). Amylase, lipase and C-reactive protein at T0, as well as 12 h and 24 h after endoscopic retrograde cholangiopancreatography, and complications(pancreatitis, bleeding, perforation) were *** We included 102 patients(66 females and 36 males, mean age 59.11 ± 18.7 years). Group Ⅰ and Group Ⅱ had 51 patients each. The successful cannulation rates were 76.5% and 100%, respectively(P = 0.0002). Twelve patients(23.5%) in Group Ⅰ had a difficult cannulation and underwent fistulotomy, which led to successful secondary biliary access(Failure Group). The complication rate was 13.7%(2 perforations and 5 mild pancreatitis) vs 2.0%(1 patient with perforation and pancreatitis) in Groups Ⅰ and Ⅱ, respectively(P = 0.0597). CONClUsION Papillary fistulotomy was more effective than guidewire cannulation, and it was associated with a lower profile of amylase and lipase. Complications were similar in both groups.
