The occurrence of pharmaceuticals in water bodies and drinking water poses risks for the envi-ronment and human health,thus it is necessary to study methodologies that allow the efficient removal of these *** this work,corn cob-derived biochar was obtained by ZnCl_(2)-activation,and subsequent carbonization at 700℃.The effect of contact time,temperature,pH,and initial concentration on the adsorption capacity of acetaminophen(ACE)and amoxicillin(AMX)was determined through batch *** addition,the kinetics,isotherms,and ther-modynamics parameters were *** activated biochar exhibited a maximum adsorp-tion capacity of 332.08 mg/g for ACE and 175.86 mg/g for *** adsorption kinetics and adsorption isotherm of ACE corresponded to the pseudo-second order and langmuir model,***,pseudo-first-order kinetics and the Freundlich isotherm model were well-fitted to AMX *** ACE and AMX co-adsorption had a synergistic effect on AMX but an antagonistic effect on ACE removal,achieving a maximum adsorption capacity of 193.51 and 184.58 mg/g,*** the other hand,fixed-bed column experiments showed that the adsorption capacity depends on the influent concentration,and the breakthrough curve fits the Thomas and Yoon-nelson *** mechanism adsorption studies showed that surface inter-actions(hydrogen bonding formation and n-𝜋interactions)are the main driving forces for the adsorption process,and pore filling is the rate-limiting *** this way,the prepared biochar exhibits a high potential for the adsorption of pharmaceutical compounds from water.

Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCl), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCl/Gelatin membranes mainly in PCl/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCl/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. Connexin 43 (CX43) is one of the major components of gap junctions, the structures responsible for the intercellular communication and transmission of the electrical impulse in the left ventricle. There is limited information on the histological changes of CX43 with age and their effect on electrophysiology, especially in humans. Here, we analyzed left ventricular biopsies from living donors starting at midlife to characterize age-related CX43 remodeling. We assessed its quantity, degree of lateralization, and spatial heterogeneity together with fibrotic deposition. We observed no significant age-related remodeling of CX43. Only spatial heterogeneity increased slightly with age, and this increase was better explained by biological age than by chronological age. Importantly, we found that CX43 features varied considerably among individuals in our population with no relevant relationship to age or fibrosis content, in contrast to animal species. We used our experimental results to feed computational models of human ventricular electrophysiology and to assess the effects of interindividual differences in specific features of CX43 and fibrosis on conduction velocity, action potential duration, and arrhythmogenicity. We found that larger amounts of fibrosis were associated with the highest arrhythmic risk, with this risk being increased when fibrosis deposition was combined with a reduction in CX43 amount and/or with an increase in CX43 spatial heterogeneity. These mechanisms underlying high arrhythmic risk in some individuals were not associated with age in our study population. In conclusion, our data rule out CX43 remodeling as an age-related arrhythmic substrate in the population beyond midlife, but highlight its potential as a proarrhythmic factor at the individual level, especially when combined with increased fibrosis.

Metabolic collaboration between cells in the tumor microenvironment has a negligible effect on tumor growth

Background:This study aimed to determine the effect of different carbohydrate(CHO)doses on exercise capacity in patients with McArdle disease—the paradigm of“exercise intolerance”,characterized by complete muscle glycogen unavailability—and to determine whether higher exogenous glucose levels affect metabolic responses at the McArdle muscle cell(in vitro)***:Patients with McArdle disease(n=8)and healthy controls(n=9)underwent a 12-min submaximal cycling constant-load bout followed by a maximal ramp test 15 min after ingesting a non-caloric *** a randomized,double-blinded,cross-over design,patients repeated the tests after consuming either 75 g or 150 g of CHO(glucose:fructose=2:1).Cardiorespiratory,biochemical,perceptual,and electromyographic(EMG)variables were ***,glucose uptake and lactate appearance were studied in vitro in wild-type and McArdle mouse myotubes cultured with increasing glucose concentrations(0.35,1.00,4.50,and 10.00 g/l).Results:Compared with controls,patients showed the“classical”second-wind phenomenon(after prior disproportionate tachycardia,myalgia,and excess electromyographic activity during submaximal exercise,all pnd an impaired endurance exercise capacity(-51%ventilatory threshold and55%peak power output,both pless of the CHO dose(placebo),CHO intake increased blood glucose and lactate levels,decreased fat oxidation rates,and attenuated the second wind in the ***,only the higher dose increased ventilatory threshold(+27%,p=0.010)and peak power output(+18%,p=0.007).In vitro analyses revealed no differences in lactate levels across glucose concentrations in wild-type myotubes,whereas a doseresponse effect was observed in McArdle ***:CHO intake exerts beneficial effects on exercise capacity in McArdle disease,a condition associated with total muscle glycogen *** of these benefits are dose dependent.

Microbial electricity-driven anaerobic phenol degradation in bioelectrochemical systems

Microbial electrochemical technologies have been extensively employed for phenol ***,previous research has yielded inconsistent results,leaving uncertainties regarding the feasibility of phenol degradation under strictly anaerobic conditions using anodes as sole terminal electron *** this study,we employed high-performance liquid chromatography and gas chromatography-mass spectrometry to investigate the anaerobic phenol degradation *** findings provide robust evidence for the purely anaerobic degradation of phenol,as we identified benzoic acid,4-hydroxybenzoic acid,glutaric acid,and other metabolites of this ***,no typical intermediates of the aerobic phenol degradation pathway were ***-chamber reactors(t0.4 V ***)exhibited a phenol removal rate of 3.5±0.2 mg l^(-1) d^(-1),while two-chamber reactors showed 3.6±0.1 and 2.6±0.9 mg l^(-1) d^(-1) at anode potentials of t0.4 and t 0.2 V,*** results also suggest that the reactor configuration certainly influenced the microbial community,presumably leading to different ratios of phenol consumers and microorganisms feeding on degradation products.

认识区域大气花粉组成及其形成条件有利于明确不同类型花粉组合的气候与环境意义。利用布卡大气花粉采样器在珠穆朗玛峰北坡开展了连续2年(2012—2013年)的大气花粉观测研究。基于后向轨迹和潜在来源区域模型,探讨了秋季主要组分桤木属花粉的传输路径与潜在来源区域,分析了桤木属花粉与其植物分布和大气环流的关系及气候指示意义。结果显示:(1)桤木属花粉季气团传输路径主要来自于采样点西南方向;(2)桤木属花粉潜在来源区域与其气团传输路径基本一致,主要是喜马拉雅山脉中段,包括尼泊尔中部和东部以及西藏南部等地区;(3)桤木属花粉数量、传输路径和来源区域的年际变化与大气环流有关,受高空西风影响的西南气团对桤木属花粉影响更大。研究结果可以为认识珠穆朗玛峰北坡外来花粉的气候意义提供科学依据。

采用热力学计算和实验技术研究铝角的Al-Ca-Fe体系相图,包括液相面投影图和凝固反应。结果表明,与铝固溶体(Al)平衡的不是Al_(3)Fe相,而是一种组成符合化学式Al_(10)CaFe_(2)的三元化合物。通过包晶转变l+Al_(3)Fe→(Al)+Al_(10)CaFe_(2)(638℃,3.3%Ca和0.5%Fe,摩尔分数)发生从二元化合物到三元化合物的转变。与针状Al_(3)Fe相夹杂物不同,三元化合物的初晶和共晶具有致密的形貌。使用第一性原理计算和X射线衍射分析确定三元化合物Al_(10)CaFe_(2)的晶格结构。此外,近共晶合金Al-6%Ca-1%Fe(质量分数)在500~600℃退火后具有细晶组织,过量相的总体积分数约为25%。因此,Al-Ca-Fe体系可用于制造新的铝基复合合金。
