Dysfunction of synaptic endocytic trafficking in Parkinson's disease

Parkinson's disease is characterized by the selective degeneration of dopamine neurons in the nigrostriatal pathway and dopamine deficiency in the striatum.The precise reasons behind the specific degeneration of these dopamine neurons remain largely elusive.Genetic investigations have identified over 20 causative PARK genes and 90 genomic risk loci associated with both familial and sporadic Parkinson's disease.Notably,several of these genes are linked to the synaptic vesicle recycling process,particularly the clathrinmediated endocytosis pathway.This suggests that impaired synaptic vesicle recycling might represent an early feature of Parkinson's disease,followed by axonal degeneration and the eventual loss of dopamine cell bodies in the midbrain via a"dying back"mechanism.Recently,several new animal and cellular models with Parkinson's disease-linked mutations affecting the endocytic pathway have been created and extensively characterized.These models faithfully recapitulate certain Parkinson's disease-like features at the animal,circuit,and cellular levels,and exhibit defects in synaptic membrane trafficking,further supporting the findings from human genetics and clinical studies.In this review,we will first summarize the cellular and molecular findings from the models of two Parkinson's disease-linked clathrin uncoating proteins:auxilin(DNAJC6/PARK19)and synaptojanin 1(SYNJ1/PARK20).The mouse models carrying these two PARK gene mutations phenocopy each other with specific dopamine terminal pathology and display a potent synergistic effect.Subsequently,we will delve into the involvement of several clathrin-mediated endocytosis-related proteins(GAK,endophilin A1,SAC2/INPP5 F,synaptotagmin-11),identified as Parkinson's disease risk factors through genome-wide association studies,in Parkinson's disease pathogenesis.we will also explore the direct or indirect roles of some common Parkinson's disease-linked proteins(alpha-synuclein(PARK1/4),Parkin(PARK2),and lRRK2(PARK8))in synaptic endocytic trafficking.Additionall

The problem of art forgery and infringement is becoming increasingly prominent,since diverse self-media contents with all kinds of art pieces are released on the Internet every day.For art paintings,object detection and localization provide an efficient and ef-fective means of art authentication and copyright protection.However,the acquisition of a precise detector requires large amounts of ex-pensive pixel-level annotations.To alleviate this,we propose a novel weakly supervised object localization(wSOl)with background su-perposition erasing(BSE),which recognizes objects with inexpensive image-level labels.First,integrated adversarial erasing(IAE)for vanilla convolutional neural network(CNN)dropouts the most discriminative region by leveraging high-level semantic information.Second,a background suppression module(BSM)limits the activation area of the IAE to the object region through a self-guidance mechanism.Finally,in the inference phase,we utilize the refined importance map(RIM)of middle features to obtain class-agnostic loc-alization results.Extensive experiments are conducted on paintings,CUB-200-2011 and IlSVRC to validate the effectiveness of our BSE.

On the functions of astrocyte-mediated neuronal slow inward currents

Slow inward currents are known as neuronal excitatory currents mediated by glutamate release and activation of neuronal extra synaptic N-met hyl-D-aspartate receptors with the contribution of astrocytes.These events are significantly slower than the excitatory postsynaptic currents.Parameters of slow inward currents are determined by seve ral factors including the mechanisms of astrocytic activation and glutamate release,as well as the diffusion pathways from the release site towards the extra synaptic recepto rs.Astrocytes are stimulated by neuronal network activity,which in turn excite neurons,forming an astrocyte-neuron feedback loop.Mostly as a consequence of brain edema,astrocytic swelling can also induce slow inward currents under pathological conditions.There is a growing body of evidence on the roles of slow inward currents on a single neuron or local network level.These events often occur in synchro ny on neurons located in the same astrocytic domain.Besides synchronization of neuronal excitability,slow inward currents also set synaptic strength via eliciting timing-dependent synaptic plasticity.In addition,slow inward currents are also subject to non-synaptic plasticity triggered by long-la sting stimulation of the excitatory inputs.Of note,there might be important regionspecific differences in the roles and actions triggering slow inward currents.In greater networks,the pathophysiological roles of slow inward currents can be better understood than physiological ones.Slow inward currents are identified in the pathophysiological background of autism,as slow inward currents drive early hypersynchrony of the neural networks.Slow inward currents are significant contributors to paroxysmal depolarizational shifts/interictal spikes.These events are related to epilepsy,but also found in Alzheimer's disease,Parkinson's disease,and stroke,leading to the decline of cognitive functions.Events with features overlapping with slow inward currents(excitatory,N-methyl-Daspartate-receptor mediated currents with astrocyti

low-rank matrix decomposition with first-order total variation(TV)regularization exhibits excellent performance in exploration of image structure.Taking advantage of its excellent performance in image denoising,we apply it to improve the robustness of deep neural networks.However,although TV regularization can improve the robustness of the model,it reduces the accuracy of normal samples due to its over-smoothing.In our work,we develop a new low-rank matrix recovery model,called lRTGV,which incorporates total generalized variation(TGV)regularization into the reweighted low-rank matrix recovery model.In the proposed model,TGV is used to better reconstruct texture information without over-smoothing.The reweighted nuclear norm and li-norm can enhance the global structure information.Thus,the proposed lRTGV can destroy the structure of adversarial noise while re-enhancing the global structure and local texture of the image.To solve the challenging optimal model issue,we propose an algorithm based on the alternating direction method of multipliers.Experimental results show that the proposed algorithm has a certain defense capability against black-box attacks,and outperforms state-of-the-art low-rank matrix recovery methods in image restoration.

Recent experiments have demonstrated that synthesized complex-frequency waves can impart a virtual gain to molecule sensing systems,which can effectively restore information lost due to intrinsic molecular damping.The enhancement notably amplifies the signal of trace molecular vibrational fingerprints,thereby substantially improving the upper limit of sensitivity.

Stability and Generalization of Hypergraph Collaborative Networks

Graph neural networks have been shown to be very effective in utilizing pairwise relationships across samples.Recently,there have been several successful proposals to generalize graph neural networks to hypergraph neural networks to exploit more com-plex relationships.In particular,the hypergraph collaborative networks yield superior results compared to other hypergraph neural net-works for various semi-supervised learning tasks.The collaborative network can provide high quality vertex embeddings and hyperedge embeddings together by formulating them as a joint optimization problem and by using their consistency in reconstructing the given hy-pergraph.In this paper,we aim to establish the algorithmic stability of the core layer of the collaborative network and provide generaliz--ation guarantees.The analysis sheds light on the design of hypergraph filters in collaborative networks,for instance,how the data and hypergraph filters should be scaled to achieve uniform stability of the learning process.Some experimental results on real-world datasets are presented to illustrate the theory.

Effect of radiative loss mechanisms on FIR thermometric parameters of Nd3+-doped lithium tellurite glasses

Nd3+-doped tellurite glasses are promising materials for thermometers based on the fluorescence intensity ratio（FIR） technique.Nevertheless,at high Nd3+concentrations,energy transfer（ET） processes such as optical reabsorption and cross-relaxation can affect the Nd3+emission,which has been little explored in the literature.Therefore,the present work investigated the use of Nd3+-doped tellurite glass(samples doped with Nd3+at 0.2 mol%,0.5 mol%,2.0 mol%,and 4.0 mol%) in fluorescence thermometers,in the temperature range from 299 to 371 K.The results indicate a strong dependence of the FIR parameters on the Nd3+concentration,due to changes in the emission band profiles caused by optical reabsorption of the Nd3+emissions and cross-relaxation processes.A decrease of the relative sensitivity of the ratio4F5/2→4I9/2/4F3/2→4I9/2is observed for samples doped with higher amounts of Nd3+.The maximum relative sensitivity at 299 K is 3.00%/K,which is the highest value among the reported Nd3+ions.

Exploring the aptitude of the human brain to compensate functional consequences of a lesion damaging its structural architecture is a key challenge to improve patient care in various neurological diseases,to optimize neuroscientifically-informed strategies of postlesional rehabilitation,and ultimately to develop innovative neuro-regenerative therapies.The term‘plasticity’,initially referring to the intrinsic propensity of neurons to modulate their synaptic transmission in a learning situation,was progressively transposed to brain injury research and clinical neurosciences.Indeed,in the event of brain damage,adaptive mechanisms of compensation allow a partial reshaping of the structure and activities of the central nervous system,thus permitting to some extent the maintenance of brain functions.

Time-inconsistent stochastic linear-quadratic control problem with indefinite control weight costs

A time-inconsistent linear-quadratic optimal control problem for stochastic differential equations is studied.we introduce conditions where the control cost weighting matrix is possibly singular.Under such conditions,we obtain a family of closed-loop equilibrium strategies via multi-person differential games.This result extends Yong’s work(2017) in the case of stochastic differential equations,where a unique closed-loop equilibrium strategy can be derived under standard conditions(namely,the control cost weighting matrix is uniformly positive definite,and the other weighting coefficients are positive semidefinite).
