Simple lC Isocratic Methods Development, Validation, and Application in the Analysis of Poor Quality Antimalarial Medicines

liquid chromatographic methods in isocratic mode for the analysis of poor quality medicines are privileged due to their simplicity and facility in methods development. They are generally fast;do not need to be re-equilibrated between sample injections;have larger flexibility with acceptable changes on different column dimensions;and are applicable to lC systems equipped with simple or high developed pumps. In this study, we focused on developing simple isocratic methods using classical mobile phase composed by methanol and ammonium formate buffer for the analysis of most common antimalarial medicines marketed in malaria endemic countries and susceptible of being counterfeit/falsified, substandard and degraded. The selected medicines were quinine and related cinchona alkaloids in tablets and injectable forms;artemether/lumefantrine tablets;and artemisinin compounds (arteether, artemether, and artesunate) in injectable forms. The current methods were developed thanks to simple methodological approach consisting in sequential isocratic runs through adjustment or adaptation of existing methods to obtain optimal analytical conditions without complex design of experiments that might be long and costly. Then, the new methods presented shorter analysis time;allowed increase of sample analysis throughput;and obviously consumed little mobile phase solvents on classical analytical columns: 50 - 250 mm of length (l), 4.6 mm of internal diameter (I.D.), and 3.5 - 5.0 μm of particle size (dp).

Soil Phosphorus Fractions Change in Winter in a Corn-Soybean Rotation with Tillage and Phosphorus Fertilization

Determining how agricultural management practices affect soil phosphorus(P) over the winter may further our understanding of the soil P cycle under specific environmental conditions in eastern Canada. This study assessed changes over winter for soil P fractions and other selected chemical properties as affected by tillage and P fertilization. In 1992, a long-term corn(Zea mays l.) and soybean(Glycine max l.) rotational experiment was established in the province of Quebec, Canada. Soil samples(0–15 cm) were collected in fall 2001 and 2007 after a soybean harvest, and in the following spring 2002 and 2008 before corn seeding, in main plots under moldboard plow and no-till managements and selected subplots fertilized with 0, 17.5, or 35 kg P ha-1and 160 kg N ha-1. Soil samples were analyzed for P fractions and other chemical properties to assess changes over winter for 2001–2002 and 2007–2008. Changes over winter of all soil P fractions were significant for the two periods, indicating the occurrence of soil P transformation and movement over winter. The Mehlich-3-extractable Fe, Al, Ca, and Mg decreased during the two studied periods. Tillage had no significant effect on all soil P fractions. The resin-extractable P in 2001–2002 and Na HCO3-extractable inorganic P and Na OH-extractable organic P during the two winters were significantly increased under P fertilization. This study demonstrated that P in cultivated soils changed during winter as a result of changes in labile P fractions possibly due to the solubilization of residual fertilizer P combined with environmental factors.

Productivity and nutritive quality of dallisgrass (<i>Paspalum dilatatum</i>) as influenced by cutting height and rate of fertilization with poultry litter or commercial fertilizer

Dallisgrass (Paspalum dilatatum) is well adapted to the Black belt region of the southeastern US, and information on its productivity and nutritive quality as influenced by fertility is needed. In each yr of a 2-yr study, an existing dallisgrass pasture that had been subdivided into 48 plots of 9.3 m2 each was fertilized with the equivalent of 34 (34N), 67 (67N), 101 (101N) or 134 (134N) kg N/ha from poultry litter (Pl) or commercial fertilizer (CF;NH4NO3). In both years, primary-growth and vegetative regrowth forage was harvested in mid-August and late September, respectively, and forage from each harvest was clipped to either a 5- or 10-cm stubble height. Forage cut to a 5-cm height yielded 71% more (P lds were not different between CF and Pl treatments across years and fertilization rates. Concentration of crude protein (CP) was greater (P = 0.002) for CF than Pl forage and increased for both fertilizer sources with increasing rates of N application. Forage concentrations of cell-wall constituents were not different between CF and Pl treatments. Forage amended with CF had a higher concentration of Ca, Mg and Mn than Pl-amended forage;however, forage amended with Pl had a higher concentration of P and K than CF-amended forage. There was no effect of fertilizer source on forage concentration of Al, Cu or Zn. Results indicate that Pl and CF were comparable for supporting productivity and nutritive quality of dallisgrass on Black belt soils.

目的探讨p53核酶对肝癌细胞突变型抑癌基因p53的抑制作用。方法应用计算机设计并合成针对突变型p53(249位密码子AGG→AGT)的锤头状核酶RZ,构建其体外转录和真核表达载体,检测核酶对突变型p53(mtp53)的体外切割作用,并在lipofect AMINE^(TM)2000的介导下转染肝癌细胞MHCC97,应用逆转录聚合酶联反应(RT—PCR)检测核酶对肝癌细胞突变型p53的抑制作用。结果测序证实核酶基因被正确克隆入体外转录载体pBSKU6和真核载体pEGFPC1中。体外切割效率为42％,而野生型p53(wtp53)没有被切割。在lipofectAMINETM2000的介导下成功转染肝癌细胞MHCC97,RT—PCR检测证实突变型p53的mRNA水平明显下降,细胞内的切割效率为69％。结论 p53核酶可成功抑制肝癌细胞中突变型p53的表达,为肝癌的基因治疗提供了一个新的选择。

Competitive metabolism of l-arginine:arginase as a therapeutic target in asthma

Exhaled breath nitric oxide （NO） is an accepted asthma biomarker. lung concentrations of NO and its amino acid precursor, l-arginine, are regulated by the relative expressions of the NO synthase （NOS） and arginase isoforms. Increased expression of arginase I and NOS2 occurs in murine models of allergic asthma and in biopsies of asthmatic airways. Although clinical trials involving the inhibition of NO-producing enzymes have shown mixed results, small molecule arginase inhibitors have shown potential as a therapeutic intervention in animal and cell culture models. Their transition to clinical trials is hampered by concerns regarding their safety and potential tox- icity. In this review, we discuss the paradigm of arginase and NOS competition for their substrate l-arginine in the asthmatic airway. We address the functional role of l-arginine in inflammation and the potential role of arginase inhibitors as therapeutics.

Electronic sensors based on biomaterials can lead to novel green technologies that are low cost,renewable,and eco-friendly.Here we demonstrate bioelectronic ammonia sensors made from protein nanowires harvested from the microorganism Geobacter sulfurreducens.The nanowire sensor responds to a broad range of ammonia concentrations(10 to 10^6 ppb),which covers the range relevant for industrial,environmental,and biomedical applications.The sensor also demonstrates high selectivity to ammonia compared to moisture and other common gases found in human breath.These results provide a proof-of-concept demonstration for developing protein nanowire based gas sensors for applications in industry,agriculture,environmental monitoring,and healthcare.

The Gurupi belt,in north-northeastern Brazil,is a mobile belt developed in the south-southwestern margin of the Sao luis cratonic fragment and crops out as a tectonic and erosional window within the Phanerozoic cover.Field,petrographic,geochemical,geochronological,and Nd isotopic information(new and published)constrain the timing and types of magmatic associations present in the belt and the tectonic settings in which they formed.The Rhyacian was the main period of magmatic activity,which can be grouped into two main stages.(1)~2185-2130 Ma:pre-collisional,juvenile,calc-alkaline magnesian and calcic ferroan granitoid suites,and minor calc-alkaline and tholeiitic mafic plutonism(now amphibolites),formed in intra-oceanic to transitional/continental arcs;and intra-or back-arc volcano-sedimentary basin.(2)~2125-2070 Ma:syn-(two-mica granites)to late-collisional(potassic to shoshonitic granites and quartz-syenite)plutonic suites produced after crustal thickening and melting,with localized migmatization,that intruded during the compressive D1 deformational phase and concomitantly with greenschist to amphibolite metamorphism.There is a zonation of the Rhyacian episodes,with intra-oceanic stages occurring to the northeast,and the continental arc and collisional phases occurring to the southwest,indicating the presence of an active continental margin to the southwest,and subduction from NE to SW(present-day configuration).This magmatic framework is a continuation to the south of what is described for the Sao luis cratonic fragment to the north,and the orogenic scenario is identical to what is observed for the same period in the West African Craton(Eburnean/Birrimian orogen),which additionally supports previous geological correlations.In the Neoproterozoic,a few magmatic occurrences are recognized.An extensional event allowed the intrusion of an anorogenic,nepheline syenite at ca.730 Ma,which was followed by the intrusion of a crustal,calc-alkaline microtonalite,of uncertain tectonic setting,at 624 Ma.Both intrusions under

Identification of pests and assessment of their damage on Carapa procera and lophira lanceolata in Burkina Faso,West Africa

Nontimber forest products are a source of income for women in rural African communities. However, these products are frequently damaged by insect pests. The present study investigates the diversity and damage rates of insect pests that attack Carapa procera seeds and lophira lanceolata fruits. The experiment was set up in western Burkina Faso and, for C. carapa, consisted of pests collected from seeds that had fallen to the ground and from stockpiled seeds. For l. lanceolata, pests were collected from fruits on the trees, and on the ground. The collected samples were sent to the laboratory to estimate the proportion of damaged seeds/fruits and rear the insects. The results showed that Ephestia spp., Tribolium castaneum, Oryzeaphilus spp., and Tenebroides mauritanicus were the pests of Carapa procera seeds and lophira lanceolata fruits. Ephestia spp. was recorded as the main pest of both C. proeera and l. lanceolata, whereas T. castaneum was only detected from seeds of l. lanceolata. For C. procera, the stocks were the most infested （29 %） by Ephestia spp. The infestation rate of fruits of l. lanceolata by Ephestia spp, on trees （31.42 ± 3.75 %） was less than the rate of fruits by T. castaneum on the ground （44.00 ± 3.5 %）. The different body sizes of Ephestia spp. may indicate the occurrence of two putative species, one from C. procera and another one from l. lanceolata. This work provides important information that could contribute to the setting up of a local-scale sustainable management framework for oil tree pests in Burkina Faso and surrounding countries.
