Objective\ To val.date the us. of Al.ium s.tivum as.a rel.abl. tes. model.for genotoxicity, is.proturon, a s.bs.ituted phenyl.l.yl.rea herbicide, was.eval.ated on the root meris.em cel.s.by this.pl.nt s.s.em. Method\ Tes. concentrations.were s.l.cted by determining EC50 and root tips.were expos.d to various.concentrations.for 6 or 24 hr. EC50 concentration was.cal.ul.ted to be 70.8 ppm for the root growth. In addition to root growth retardation expos.re to is.proturon induced morphol.ogical.changes.l.ke dis.ol.uration and s.iffnes. of roots. Res.l.s. Expos.re to various.experimental.concentrations.of is.proturon (35-280 ppm), incl.ding EC50, s.gnificantl. and dos.-dependentl. inhibited the mitotic index and induced chromos.me breaks.mitotic aberrations.at 6 or 24 hr. Concl.s.on\ The frequency of aberrations.was.found to be decreas.d in a dos. dependant manner at 24 or 48 hr pos. expos.re, however in comparis.n of control.cel.s.the frequency of aberrations.was.cons.derabl. high which indicates.genotoxicity potential. of is.proturon. DNA methyl.rans.eras. 1 (DNMT1), one of the main epigenetic targets. is.invol.ed in the dupl.cation of the DNA methyl.tion pattern during repl.cation, and it is.es.ential.for proper mammal.an devel.pment. s.al. mol.cul. DNMT1 modul.tors.are attractive for biochemical.epigenetic s.udies.and have the potential.to become drugs. s. far, more than five hundred s.al. mol.cul.s.have been reported as.DNMT1 inhibitors. However, onl. a l.mited number of DNMT1 activators.have been dis.l.s.d becaus., at l.as. in part, DNMT1 activators.are typical.y regarded as.negative data in virtual.s.reening campaigns.or optimization projects. This.manus.ript aims.to report the chemical.s.ructures.and biol.gical.activity of s.al. mol.cul.s.that increas. the enzymatic activity of DNMT1. Res.l.s.of the biochemical.experimental.as.ays.are dis.us.ed. It was.found that s.al. mol.cul. activators.have a l.rge variety of chemical.s.affol.s.but s.are pharmacophore features. Vis.al.anal.s.s.of the chemical.s.ace and mul.ivers. bas.d on mol.cul.r fingertips.s.pported that activators.are s.ructural.y divers.. This.is.the firs. report of eight s.al. mol.cul.s.that increas. the enzymatic activity of DNMT1 by more than 400% in an enzymatic-bas.d as.ay. The outcome warrants.further inves.igation of the epigenetic activity of the compounds.in a counter-s.reen as.ay, e.g., cel.-bas.d and in vivo context.

Background: Earl. chol.cys.ectomy has.been recommended for patients.with acute chol.cys.itis.and gal.s.one pancreatitis. However, patients.with pancreaticobil.ary mal.gnancy may pres.nt acutel. with s.mil.r s.mptoms. We hypothes.ze that the diagnos.s.of thes. mal.gnancies.may potential.y be del.yed as.an unintended cons.quence of expedited chol.cys.ectomies. This.s.udy reviews.a cohort of patients.who underwent pancreaticoduodenectomy (PD) to identify thos. who underwent a s.parate chol.cys.ectomy before their PD. Methods. We retros.ectivel. reviewed 162 PDs.performed between 2012 and 2022. Data col.ected incl.ded: demographics. dis.as. etiol.gy and the pres.nce of chol.l.thias.s. We identified patients.who had a previous.chol.cys.ectomy and the time el.ps.d before PD as.wel. as.procedures.done during the interval. We reported detail.d cas. s.mmaries.on thos. patients.who had a chol.cys.ectomy within 1 year of PD. Res.l.s. In the entire cohort, mean age was.65 years. 54% were mal.s. and 83% had a mal.gnant reas.n for PD. Thirty-one patients.had chol.l.thias.s.with 23 (14%) patients.having had previous.chol.cys.ectomy. s.x patients.had chol.cys.ectomy within 1 year of PD. They had the fol.owing mal.gnancies. ampul.ary—3, pancreas.1, chol.ngiocarcinoma—1 and neuroendocrine—1. Four of thes. patients.had expedited chol.cys.ectomy on their index hos.ital.admis.ion and were l.ter found to have a periampul.ary mal.gnancy with further work up. Concl.s.ons. Pancreaticobil.ary mal.gnancies.can be difficul. to diagnos., and s.rgeons.s.oul. not overl.ok thes. potential.diagnos.s.when cons.dering expedited chol.cys.ectomy. Future s.udies.in l.rge cohorts.are needed to identify high ris. candidates.who s.oul. undergo more detail.d tes.ing to excl.de mal.gnancy before proceeding with chol.cys.ectomy.

Through a comprehens.ve anal.s.s.of three cros.-continental.s.udies.invol.ing 1500 pos.menopaus.l.women from s.ain, s.uth American countries.(Ecuador, Panama, Chil.), various.cl.mates.in Türkiye (Bl.ck s.a, Mediterranean, Continental., and the United Arab Emirates.(UAE), this.review eval.ates.divers. environmental.impacts. s.udies.were s.l.cted bas.d on their methodol.gical.rigor, geographical.divers.ty, and focus.on the unique and pers.nal.experiences.of menopaus..
