Genetic characterization of Northeast Asian cattle based on sequence polymorphisms in the complete mitochondrial genome

In this study, we analyzed complete mtDNA sequences variation and genetic relationship among taurine, indicine and Bison groups. In total, 107 sequences from different breeds, 45 European (45 Italian), 16 Middle East Asian (seven Iranian and nine Iraqi), 41 Northeast Asian (34 Korean and seven Japanese), two Nellore (Bos indicus) and two American Bison bison (Ame. bison) were obtained from Gen-Bank database. One Korean Hanwoo (Bos taurus) sequence was generated using the SOLiDTM System. In total, 1370 polymorphic sites, representing 8.39% of the complete 107 mtDNA sequences (16,338 bp) were detected and of these, 1186 parsimony informative polymorphic sites were identified. Neighbor-joining tree indicated that Korean, Japanese, Iranian, Iraqi, and Italian cattle were closely related to one another, but are separated from B. Bison. The B. taurus mtDNA polymorphism was greater in the D-loop than in the other regions. The ATP8, ND3, ND5, and ND6 regions were also quite parsimony informative, similar to Cyt b. In addition, this study revealed a distinct genetic difference between Korean cattle and B. indicus.

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

We present innovative nonlinear acoustics for characterizing fatigue-induced micro-damage of austenitic stainless steel 316 subjected to high-cycle *** fatigue-driven deformations are accumulated at several positions near the middle of hourglass-shaped specimens.A bell-shaped curve of acoustic nonlinearity as a function of position is observed,and the variation in acoustic nonlinearity is attributed to the evolution of a lattice defect (dislocation) and stress-induced martensite based on transmission electron microscopy (TEM) *** oblique incidence technique using a longitudinal waveform is a potentially viable method for characterizing the high-cycle fatigue deformation of austenitic stainless steel 316 alloys.

The acoustic nonlinearity of surface waves is studied to evaluate plastic deformation in aluminum alloys.A narrow-band surface wave is successfully generated by a pulsed Nd-YAG laser system consisting of a beam expander and a slit *** degrees of tensile deformation are induced by interrupting the tensile tests so as to obtain aluminum specimens with different degrees of plastic *** normalized acoustic nonlinearity increases as a function of tensile *** experimental results show that the acoustic nonlinearity of a laser-generated surface wave has a good correlation with the level of tensile deformation and has a potential to evaluate microdamage induced by dislocation microplasticity.

Several image-based biomedical diagnoses require high-resolution imaging capabilities at large spatial ***,conventional microscopes exhibit an inherent trade-off between depth-of-field(DoF)and spatial resolution,and thus require objects to be refocused at each lateral location,which is time ***,we present a computational imaging platform,termed E2E-BPF microscope,which enables large-area,high-resolution imaging of large-scale objects without serial *** method involves a physics-incorporated,deep-learned design of binary phase filter(BPF)and jointly optimized deconvolution neural network,which altogether produces high-resolution,high-contrast images over extended depth *** demonstrate the method through numerical simulations and experiments with fluorescently labeled beads,cells and tissue section,and present high-resolution imaging capability over a 15.5-fold larger DoF than the conventional *** method provides highly effective and scalable strategy for DoF-extended optical imaging system,and is expected to find numerous applications in rapid image-based diagnosis,optical vision,and metrology.

Phosphorylated p70S6K in urothelial carcinoma of the tumor recurrence noninvasive low-grade bladder： correlation with

We investigated whether inhibiting phosphorylated p70S6K （p-p70S6K） suppresses the proliferation and growth of noninvasive low-grade urothelial carcinoma （LG-URCa） in vitro and whether p-p70S6K can serve as a predictive biomarker for the recurrence of noninvasive LG-URCa of the bladder in patients. We constructed a tissue microarray （TMA） for 95 LG-URCa and 35 benign urothelium samples and performed immunohistochemical staining for p-p70S6K and p-4E-BP1. A Cox regression model was used to investigate the predictive factors for recurrence of LG-URCa. We investigated the dose-dependent antiproliferative effect of rapamycin, its antiproliferative effect and the growth-inhibition effect of p70S6K siRNA transfection in RT4 and 253J cell lines. The pT1 staged group （P 〈 0.05; hazard ratio （HR）, 2.415） and the high p-p70S6K staining group （P 〈 0.05; HR, 2.249） were independent factors for predicting recurrence. Rapamycin inhibited RT4 and 253J cell proliferation in a dose-dependent manner （r = -0.850, P 〈 0.001 in RT4 cells; r = -0.835, P 〈 0.001 in 253J cells）. RT4 and 253J cell proliferation and growth were inhibited by the transfection of p70S6K siRNA and rapamycin, respectively （P 〈 0.05）. Transfection of p70S6K siRNA resulted in inhibitory effects on cell proliferation and growth that were similar to those of rapamycin. Our results suggest that inhibiting p70S6K phosphorylation is important to prevent recurrence and that p70S6K phosphorylation can be used as a molecular biomarker to predict recurrence of certain LG-URCa of the bladder.

Atypical onset of bicalutamide-induced liver injury

Anti-androgen therapy is the leading treatment for advanced prostate cancer and is commonly used for neoadjuvant or adjuvant treatment. Bicalutamide is a non-steroidal anti-androgen, used during the initiation of androgen deprivation therapy along with a luteinizing hormone-releasing hormone agonist to reduce the symptoms of tumor-related flares in patients with advanced prostate cancer. As side effects, bicalutamide can cause fatigue, gynecomastia, and decreased libido through competitive androgen receptor blockade. Additionally, although not as common, drug-induced liver injury has also been reported. Herein, we report a case of hepatotoxicity secondary to bicalutamide use. Typically, bicalutamideinduced hepatotoxicity develops after a few days; however, in this case, hepatic injury occurred 5 mo after treatment initiation. Based on this rare case of delayed liver injury, we recommend careful monitoring of liver function throughout bicalutamide treatment for prostate cancer.

The temporal and spatial variability of the Yellow Sea Cold Water Mass in the southeastern Yellow Sea, 2009–2011

The Yellow Sea Cold Water Mass （YSCWM） is one of the important water mass in the Yellow Sea （YS）. It is distributed in the lower layer in the Yellow Sea central trough with the temperature less than 10℃ and the salinity lower than 33.0. To understand the variability of the YSCWM, the hydrographic data obtained in April and August during 2009-2011 are analyzed in the southeastern Yellow Sea. In August 2011, relatively warm and saline water compared with that in 2009 and 2010 was detected in the lower layer in the Yellow Sea central area. Although the typhoon passed before the cruise, the salinity in the Yellow Sea central trough is much higher than the previous season. It means that the saline event cannot be explained by the typhoon but only by the intrusion of saline water during the previous winter. In April 2011, actually, warm and saline water （T 〉 10~C, S 〉34） was observed in the deepest water depth of the southeastern area of the Yellow Sea. The wind data show that the northerly wind in 2011 winter is stronger than in 2009 and 2010 winter season. The strong northerly wind can trigger the intrusion of warm and saline Yellow Sea Warm Current. Therefore, it is proposed that the strong northerly wind in winter season leads to the intrusion of the Yellow Sea Warm Current into the Yellow Sea central trough and influenced a variability of the YSCWM in summer.

Mapping the Dementia Research Area at the Micro-Level Using Co-terms Analysis and Positioning for Traditional Herbal Medicine

Objective： To identify the position of traditional herbal medicine in dementia research field using mapping technology. Methods： Keywords for dementia and traditional herbal medicine for treating dementia were used to extract scientific articles from the Web of Science database from January 2000 to July 2010. A co- occurrence matrix was created based on the concurrent set of author＇s keywords occurring in each scientific article, and technology network maps were created from similarity index matrices. Results： Twenty specialized research areas were identified in the dementia field, and the relationship strength was 0.2-0.6. Many research fields were associated with diagnosis and risk factors for dementia. Additionally, the mechanism or cause of dementia is an actively studied field. Traditional herbal medicine for treating dementia was located on a map near the cortical dementia diagnosis and therapy, and frontotemporal dementia research field with a relationship strength of 0.53 and 0.31-0.33 respectively, which demonstrates that traditional herbal medicine for dementia occupies an independent research area with a relationship to existing scientific research fields. Conclusion： Traditional herbal medicine can provide an alternative and complementary approach for treating dementia as evidenced by a scientific mapping analysis.
