Curcumin-loaded polymeric nanoparticles for neuro-protection in neonatal rats with hypoxic-ischemic encephalopathy

Hypoxic-ischemic encephalopathy is the leading cause of permanent brain injury in term newborns and currently has no cure. Inflammatory processes play a key role in the progression of this disease and may be amenable to a targeted pharmaceutical intervention. Curcumin is a dietary compound with potent anti-inflammatory, antioxidant, and antiapoptotic properties but is limited in therapeutic applications due to its low aqueous solubility, low bioavailability, and rapid first-pass hepatic metabolism. To address these limitations, loading curcumin into poly（lactic-co-glycolic acid）-poly（ethylene glycol）（PLGA-PeG） nanoparticles may increase relevant pharmacokinetic parameters and allow for effective drug delivery to the brain. Using the Vannucci model of unilateral hypoxic-ischemic brain injury in neonatal rats, we studied the in vivo effect of curcumin-loaded PLGA-PeG nanoparticles on brain uptake and diffusion of curcumin and on neuroprotection. The curcumin-loaded nanoparticles were able to overcome the impaired blood-brain barrier, diffuse effectively through the brain parenchyma, localize in regions of injury, and deliver a protective effect in the injured neonatal brain. The application of curcumin and PLGA-PeG nanoparticle-mediated delivery to a clinically relevant model of neonatal brain injury provides greater opportunities for clinical translation of targeted therapies for hypoxic-ischemic encephalopathy.

Solar wind transport into magnetosphere caused by magnetic reconnection at high latitude magnetopause during northward ImF: Cluster-DSP conjunction observations

An event of Cluster-Double Star conjunction observations of magnetic reconnec-tion at high latitude magnetopause nightside of both cusps and solar wind trans-port into magnetosphere caused by such reconnection process has been investi-gated. During northward ImF, Cluster/SC1 observed accelerated flows and ion heating associated with magnetic reconnection at high latitude magnetopause nightside of southern cusp. And Double Star observed cold dense solar wind plasma transported into dayside magnetosphere. The analysis on such conjunction observations shows that: (1) during northward ImF, magnetic reconnection occurs at high latitude nightside of southern cusp, accompanied by accelerated flows that are observed by Cluster/SC1; (2) the direction of the accelerated flows, with its sunward component Vx, dawnward component Vy, northward component Vz, is quite consistent with the theoretical anticipation under the condition of northward ImF with dawnward component By; (3) reconnection can heat plasma more in par-allel direction than in perpendicular direction, to a level of about 4 keV; (4) with reconnection taking place at high latitude magnetopause nightside of the southern cusp, TC-1 observed cold and dense plasma transported into magnetosphere; (5) by reconnection at high latitude magnetopause nightside of both cusps, solar wind flux tube can be captured by magnetosphere and pulled into dayside magneto-sphere. This event presents further observational evidence for magnetic reconnec-tion at high latitude magnetopause nightside of both cusps as an important mech-anism of sol-ar w-ind transport into magnetosphere.

Preliminary evaluation of a decision support tool for biochar amendment

meta-analyses have shown that only about half of biochar addition studies report improved plant *** improve yield response,here we describe a decision support tool(DST)for selecting a biochar type and amendment rate to meet soil and crop management goals,based on soil and biochar tests values and crop requirements as reported by regional extension *** data from a greenhouse wheat trial,we assessed whether this approach could predict changes in soil chemistry and whether it could identify soils in which amendment would stimulate crop *** data indicated that the DST could provide semi-quantitative predictions of biochar amendment rates needed to meet target soil pH levels,with recommended rates averaging 25%higher than were *** DST was better at predicting biochar application rates to provision P and *** six soil types,post-harvest measurements of extractable-K showed an average 104%recovery of the quantities estimated to be provisioned by conifer wood or wheat straw ***-P recovery averaged 101%in two sandy soils with low P-retention but was considerably lower in four soils with higher P-retention ***-house data also showed that wheat growth improved only when biochar alleviated pH that was substantially below critical thresholds for plant growth,and supported the principle of using crop-specific pH requirements as part of an approach for biochar *** field trials that evaluate yield responses in several nutrient-deficient or acidified soils will be needed to provide a robust evaluation of this DST.

We applied an ex-situ casting procedure to prepare a nanocomposite(NCP)from makrofol polycarbonate(PC)and CdSe *** CdSe nanoparticles were prepared by a thermolysis procedure in the presence of N_(2) gas *** refinement of x-ray data illustrated that the CdSe adopts a cubic zinc blend structure of 6.057 A lattice parameter and 2 nm typical grain *** from the prepared NCP were exposed to γ dosages(20 kGy-250 kGy).The modifications induced in the NCP films owing to γ dosages have been ***γirradiation(50 kGy-250 kGy)causes crosslinks that reduce the optical bandgap from 4.15 eV to 3.81 eV,associated with an increase in dielectric parameters and refractive *** is attributed to an increase in the mass fraction of the disordered regions as specified by x-ray *** PC-CdSe NCP was found to have a reaction to color modification which makes it suitable for saleable reproduction on a printing press.

Nogo receptor expression in microglia/macrophages during experimental autoimmune encephalomyelitis progression

myelin-associated inhibitory factors within the central nervous system（CNS） are considered to be one of the main obstacles for axonal regeneration following disease or injury. The nogo receptor 1（NgR1） has been well documented to play a key role in limiting axonal regrowth in the injured and diseased mammalian CNS. However, the role of nogo receptor in immune cell activation during CNS inflammation is yet to be mechanistically elucidated. microglia/macrophages are immune cells that are regarded as pathogenic contributors to inflammatory demyelinating lesions in multiple sclerosis（mS）. In this study, the animal model of mS, experimental autoimmune encephalomyelitis（eAe） was induced in ngr1^＋/＋ and ngr1^–/– female mice following injection with the myelin oligodendrocyte glycoprotein（mOG_（35–55）） peptide. A fatemap analysis of microglia/macrophages was performed throughout spinal cord sections of eAe-induced mice at clinical scores of 0, 1, 2 and 3, respectively（increasing locomotor disability） from both genotypes, using the CD11 b and Iba1 cell markers. Western immunoblotting using lysates from isolated spinal cord microglia/macrophages, along with immunohistochemistry and flow cytometric analysis, was performed to demonstrate the expression of nogo receptor and its two homologs during eAe progression. myelin protein engulfment during eAe progression in ngr1^＋/＋ and ngr1^–/– mice was demonstrated by western immunblotting of lysates from isolated spinal cord microglia/macrophages, detecting levels of Nogo-A and mOG. The numbers of m1 and m2 microglia/macrophage phenotypes present in the spinal cords of eAe-induced ngr1^＋/＋ and ngr1^–/– mice, were assessed by flow cytometric analysis using CD38 and erg-2 markers. A significant difference in microglia/macrophage numbers between ngr1^＋/＋ and ngr1^–/– mice was identified during the progression of the clinical symptoms of eAe, in the white versus gray matter regions of the spinal cord. Shrinkage of magnetosphere observed by TC-1 satellite during the high-speed solar wind stream

During the interval 06:14―07:30 UT on August 24, 2005, since the earth’s magneto- pause was suddenly compressed by the persistent high-speed solar wind stream with the southward component of the interplanetary magnetic field (ImF), the magnetopause moved inward for about 3.1 Re. meanwhile, TC-1 satellite shifted from northern plasma sheet to the northern lobe/mantle region, although it kept inward flying during the interval 06:00―07:30UT. The shift of TC-1 from the plasma sheet to the lobe/mantle is caused by the simultaneous inward displacements of the plasma sheet and near-earth lobe/mantle region, and their inward movement velocity is larger than the inward motion velocity of TC-1. The joint inward dis-placements of the magnetopause, the lobe/mantle region and the plasma sheet indicate that the whole magnetosphere shrinks inward due to the magnetospheric compression by the high-speed solar wind stream, and the magnetospheric ions are attached to the magnetic field lines (i.e. ‘frozen’ in magnetic field) and move inward in the shrinking process of magnetosphere. The large shrinkage of magne-tosphere indicates that the near-earth magnetotail compression caused by the strong solar wind dynamic pressure is much larger than its thickening caused by the southward component of the ImF, and the locations of magnetospheric regions with different plasmas vary remarkably with the variation of the solar wind dynamic pressure.

Gastroesophageal reflux disease after diagnostic endoscopy in the clinical setting

AIm: To investigate the outcome of patients with symptoms of gastroesophageal reflux disease (GeRD) referred for endoscopy at 2 and 6 mo post endoscopy. meTHODS: Consecutive patients referred for upper endoscopy for assessment of GeRD symptoms at two large metropolitan hospitals were invited to participate in a 6-mo non-interventional (observational) *** two institutions are situated in geographically and socially disparate areas. Data collection was by selfcompletion of questionnaires including the patient assessment of upper gastrointestinal disorders symptoms severity and from hospital records. endoscopic finding using the Los-Angeles classification, symptom severity and it's clinically relevant improvement as change of at least 25%, therapy and socio-demographic factors were assessed. ReSULTS: Baseline data were available for 266 patients and 2-mo and 6-mo follow-up data for 128 and 108 patients respectively. At baseline, 128 patients had erosive and 138 non-erosive reflux disease. Allmost all patient had proton pump inhibitor (PPI) therapy in the past. Overall, patients with non-erosive GeRD at the index endoscopy had significantly more severe symptoms as compared to patients with erosive or even complicated GeRD while there was no difference with regard to medication. After 2 and 6 mo there was a small, but statistically significant improvement in symptom severity (7.02 ± 5.5 vs 5.9 ± 5.4 and 5.5 ± 5.4 respectively); however, the majority of patients continued to have symptoms (i.e. , after 6 mo 81% with GeRD symptoms). Advantaged socioeconomic status as well as being unemployed was associated with greater improvement. CONCLUSION: The majority of GORD patients receive PPI therapy before being referred for endoscopy even though many have symptoms that do not sufficiently respond to PPI therapy.

In response to increasing concerns over climate change,soil health and wine quality,grape growers are seeking new practices(e.g.,biochar application)to minimize their environmental footprint while increasing productivity and the quality of their *** explore the potential of biochar-based amendments to achieve these goals in wine grape production,vineyard trials were established in the fall of *** Oregon sites were chosen with distinct soil types and climates(Willamette Valley and Rogue Valley)but planted with the same grapevine scion/rootstock Pinot noir *** treatments were applied under vines at each location:no biochar-no tillage(NT);no biochar+tillage(B0);18 tons ha^(−1)biochar+tillage(B18);35 tons ha^(−1)biochar+tillage(B35).In 2019,a suite of soil health,plant,and crop variables were measured,and wines were produced after *** incorporation of biochar modified the chemical and physical composition of soils at the two studied locations,increasing the bioavailability of carbon and nitrogen,their gravimetric water content and the concentration of plant available micro and macro *** responses of plant physiology parameters or productivity at either site were found after biochar incorporation when compared with ***,a significant and gradual decrease in the amount of wine tannins was found as a result of biochar application in wines produced from grapes from the Woodhall ***-term field experiments are required to assess the effects of biochar on soil properties,vine physiol-ogy,productivity,and grape and wine quality several years after incorporation.
