Genome reprogramming for synthetic biology

The ability to go from a digitized DNA sequence to a predictable biological function is central to synthetic biology. Genome engineering tools facilitate rewriting and implementation of engineered DNA sequences. Recent development of new programmable tools to reengineer genomes has spurred myriad advances in synthetic biology. Tools such as clustered regularly interspace short palindromic repeats enable RNA-guided rational redesign of organisms and implementation of synthetic gene systems. New directed evolution methods generate organisms with radically restructured genomes. These restructured organisms have useful new phenotypes for biotechnology, such as bacteriophage resistance and increased genetic stability. Advanced DNA synthesis and assembly methods have also enabled the construction of fully synthetic organisms, such as J. Craig Venter Institute （JCVI）-syn 3.0. Here we summarize the recent advances in programmable genome engineering tools.

Alpha-adrenoceptor antagonism by Crassostrea gigas oyster extract inhibits noradrenalineinduced vascular contraction in Wistar rats

OBJECTIVE： Crassostrea gigas oyster extract has been reported to have antioxidant, antihypertensive and lipid-lowering properties that may be useful for treating cardiovascular diseases. This study aimed to evaluate the effect of C. gigas oyster extract on cardiovascular function in tissues from healthy rats. METHODs： single-cell microelectrode and isolated thoracic aortic organ bath studies were performed on tissues from 8-week-old healthy Wistar rats, using varying concentrations of C. gigas oyster extract. To elucidate a mechanism of action for the oyster’s vasoactive properties, concentration response curves were carried out in the presence of a calcium channel inhibitior（verapamil）, a nitric oxide synthase inhibitor（N G-nitro-L-arginine methyl ester）, a potassium channel inhibitor（4-aminopyridine）, in addition to the α-adrenoceptor inhibitor prazosin. REsULTs： Oyster solution at 7 500 mg/m L inhibited noradrenaline-induced contraction in isolated aortic rings. Cardiac electrophysiology results showed that neither concentration of oyster solution was able to significantly reduce action potential duration at all phases of repolarisation in left ventricular papillary muscles from healthy animals. CONCLUsION： When administered to healthy vascular tissue, C. gigas oyster extract inhibits contraction induced by noradrenaline. This effect is likely to be mediated through α-adrenoceptor inhibition, and to a lesser extent, calcium modulating activity.

Deconvolution is a challenging inverse problem,particularly in techniques that employ complex engineered pointspread functions,such as microscopy with propagation-invariant ***,we present a deep-learning method for deconvolution that,in lieu of end-to-end training with ground truths,is trained using known physics of the imaging ***,we train a generative adversarial network with images generated with the known point-spread function of the system,and combine this with unpaired experimental data that preserve perceptual *** method rapidly and robustly deconvolves and super-resolves microscopy images,demonstrating a two-fold improvement in image contrast to conventional deconvolution *** contrast to common end-to-end networks that often require 1000-10,000s paired images,our method is experimentally unsupervised and can be trained solely on a few hundred regions of *** demonstrate its performance on light-sheet microscopy with propagation-invariant Airy beams in oocytes,preimplantation embryos and excised brain tissue,as well as illustrate its utility for Bessel-beam *** method aims to democratise learned methods for deconvolution,as it does not require data acquisition outwith the conventional imaging protocol.

Curcumin-loaded polymeric nanoparticles for neuro-protection in neonatal rats with hypoxic-ischemic encephalopathy

Hypoxic-ischemic encephalopathy is the leading cause of permanent brain injury in term newborns and currently has no cure. Inflammatory processes play a key role in the progression of this disease and may be amenable to a targeted pharmaceutical intervention. Curcumin is a dietary compound with potent anti-inflammatory, antioxidant, and antiapoptotic properties but is limited in therapeutic applications due to its low aqueous solubility, low bioavailability, and rapid first-pass hepatic metabolism. To address these limitations, loading curcumin into poly（lactic-co-glycolic acid）-poly（ethylene glycol）（PLGA-PEG） nanoparticles may increase relevant pharmacokinetic parameters and allow for effective drug delivery to the brain. Using the Vannucci model of unilateral hypoxic-ischemic brain injury in neonatal rats, we studied the in vivo effect of curcumin-loaded PLGA-PEG nanoparticles on brain uptake and diffusion of curcumin and on neuroprotection. The curcumin-loaded nanoparticles were able to overcome the impaired blood-brain barrier, diffuse effectively through the brain parenchyma, localize in regions of injury, and deliver a protective effect in the injured neonatal brain. The application of curcumin and PLGA-PEG nanoparticle-mediated delivery to a clinically relevant model of neonatal brain injury provides greater opportunities for clinical translation of targeted therapies for hypoxic-ischemic encephalopathy.

The notion of communication is in essence that of information transmission in its simplest form—the semiotic encoding and decoding of messages that occur in every type of language,be that written,verbal,non-verbal,or *** form in an Aristotelian syllogism,this paper argues that graffiti constitutes its own art form that must be equally represented,examined,appreciated,and *** exploring how graffiti is art,how art is language,and therefore how graffiti is language,this paper seeks to expand its audience’s understanding of how artworks are underpinned by a similar semiotic structure to that of the written or *** theory and case studies of contemporary graffiti artists and their works,the notion of graffiti as a language that can be read and contextualized comes to life in a way that is both grounded in history and stunningly creative.

We have recently discovered a unique CD34loCD133lo cell population in the human fetal liver （FL） that gives rise to cells in the hepatic lineage. In this study, we further characterized the biological functions of FL CD341~CD133~~ cells. Our findings show that these CD341~CD133I~ cells express markers of both endodermal and mesodermal lineages and have the capability to differentiate into hepatocyte and mesenchymal lineage cells by ex vivo differentiation assays. Furthermore, we show that CD34~~CD 133I~ cel Is express growth factors that are important for human hematopoietic stem cell （HsC） expansion： stem cell factor （sCF）, insulin-like growth factor 2 （IGF2）, C-X-C motif chemokine 12 （CXCL12）, and factors in the angiopoietin-like protein family. Co-culture of autologous FL HsCs and allogenic HsCs derived from cord blood with CD34loCD133lo cells supports and expands both types of *** findings are not only essential for extending our understanding of the HsC niche during the development of embryonic and fetal hematopoiesis but will also potentially benefit adult stem cell transplantations in clinics because expanded HsCs demonstrate the same capacity as primary cells to reconstitute the human immune system and mediate long-term hematopoiesis in vivo. Together,CD34loCD133lo cells not only serve as stem/progenitor cells for liver development but are also an essential component of the HsC niche in the human FL.
