Background:The mobilization and redistribution of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)specific T-cells and neutralizing antibodies(nAbs)during exercise is purported to increase immune surveillance and protect against severe coronavirus disease 2019(COVID-19).We sought to determine if COVID-19 vaccination would elicit exercise-responsive SARS-CoV-2 T-cells and transiently alter nAb titers.Methods:Eighteen healthy participants completed a 20-min bout of graded cycling exercise before and/or after receiving a COVID-19 vaccine.All major leukocyte subtypes were enumerated before,during,and after exercise by flow cytometry,and immune responses to SARS-CoV-2 were determined using whole blood peptide stimulation assays,T-cell receptor(TCR)-βsequencing,and SARS-CoV-2 nAb serology.Results:COVID-19 vaccination had no effect on the mobilization or egress of major leukocyte subsets in response to intensity-controlled graded exercise.However,non-infected participants had a significantly reduced mobilization of CD4+and CD8+naive T-cells,as well as CD4+central memory T-cells,after vaccination(synthetic immunity group);this was not seen after vaccination in those with prior SARS-CoV-2 infection(hybrid immunity group).Acute exercise after vaccination robustly mobilized SARS-CoV-2 specific T-cells to blood in an intensity-dependent manner.Both groups mobilized T-cells that reacted to spike protein;however,only the hybrid immunity group mobilized T-cells that reacted to membrane and nucleocapsid antigens.nAbs increased significantly during exercise only in the hybrid immunity group.Conclusion:These data indicate that acute exercise mobilizes SARS-CoV-2 specific T-cells that recognize spike protein and increases the redistribution of nAbs in individuals with hybrid immunity.

A garnet-bearing schist from the southernmost such exposure along the Himalaya in east central Nepal records prograde metamorphism at 32.4±0.3 Ma.Phase equilibria modelling,combined with Ti-in-biotite and quartz caxis thermometry,outline a tight-to-hairpin pressure-temperature(P-T)path extending from~515℃ and 5.5 kbar to peak conditions at~575℃ and 7 kbar followed by deformation during the retrograde phase at 480-515℃ and 6-7 kbar.The new geochronology data place an upper bound on the evolution of metamorphism and deformation in the frontal-most part of the Himalaya,which lasted until 17.5 Ma,as indicated by previously published ^(40)Ar/^(39)Ar data.The P-T-time data from this part of the Himalaya,as well as that from more hinterlandward portions of the orogen,outline a progressive,stepwise,commonly out-of-sequence evolution.Further data from along the orogen indicates that this evolution is not a local phenomenon,but instead characterizes the tectonics of this system as a whole.

Mutations in the KRAS oncogene represent one of the most prevalent genetic alterations in colorectal cancer(CRC),the third leading cause of cancer-related death in the US.In addition to their well-characterized function in driving tumor progression,KRAS mutations have been recognized as a critical determinant of the therapeutic response of CRC.Recent studies demonstrate that KRAS-mutant tumors are intrinsically insensitive to clinically-used epidermal growth factor receptor(EGFR)targeting antibodies,including cetuximab and panitumumab.Acquired resistance to the anti-EGFR therapy was found to be associated with enrichment of KRAS-mutant tumor cells.However,the underlying molecular mechanism of mutant-KRAS-mediated therapeutic resistance has remained unclear.Despite intensive efforts,directly targeting mutant KRAS has been largely unsuccessful.This review summarizes the recent advances in understanding the biological function of KRAS mutations in determining the therapeutic response of CRC,highlighting several recently developed agents and strategies for targeting mutant KRAS,such as synthetic lethal interactions.

Switch mode capacitive pressure sensors are proposed as a new class of microfabricated devices that transform pressure into a mechanically switching capacitance to form an analog-to-digital signal with zero power,high sensitivity,and a high signal-to-noise ratio.A pressure-sensitive gold membrane suspended over a capacitive cavity makes ohmic contact with patterned gold leads on the substrate,closing circuits to fixed on-chip capacitors outside the cavity and leading to significant step responses.This function is achieved by allocating the switch leads on the part of the counter electrode area,while the remaining area is used for touch mode analog capacitive sensing.The sensor microchip is prototyped through a novel design approach to surface micromachining that integrates micro-Tesla valves for vacuum sealing the sensor cavity,showing an unprecedented response to applied pressure.For a gauge pressure range of 0–120 mmHg,the sensor exhibits an increase of 13.21 pF with resultant switch events,each of which ranges from 2.53–3.96 pF every 12–38 mmHg,in addition to the touch mode linear capacitive increase between switches.The equivalent sensitivity is 80–240 fF/mmHg,which is 11–600×more than commercial and reported touch mode sensors operating in similar pressure ranges.The sensor is further demonstrated for wireless pressure tracking by creating a resonant tank with the sensor,showing a 32.5–101.6 kHz/mmHg sensitivity with frequency jumps led by the switch events.The developed sensor,with its promising performance,offers new application opportunities in a variety of device areas,including health care,robotics,industrial control,and environmental monitoring.

The electrochemical properties of catalyst materials are highly dependent on the materials structure and architecture. Herein, nano-on-micro Cu electrodes are fabricated by growing Cu microcrystals on Ni foam substrate, followed by introducing Cu nanocrystals onto the surface of the Cu microcrystals. The introduction of Cu nanocrystals onto the surface of Cu microcrystals is shown to dramatically increase the electrochemically active surface area and thus significantly enhances the catalytic activity of the catalyst electrode towards electro-oxidation of hydrazine. The onset potential （-1.04 V vs. AglAgCI） of the nano-on-micro Cu electrode is lower than those of the reported Cu-based catalysts under similar testing conditions, and a current density of 16 mA-cm-2, which is 2 times that of the microsized Cu electrode, is achieved at a potential of -0.95 V vs. Ag/AgCh Moreover, the nano-on-micro Cu electrode demonstrates good long-term stability.

Purpose A portable fast-neutron imaging system is being developed to provide complementary information to field X-ray imaging.Applications include inspection of vehicles and infrastructure for corrosion,measurement of material levels in containers,and inspection of munitions and suspicious packages.While fast-neuron imaging generally provides lower imaging resolution compared to X-rays,fast-neutron interaction cross-sections have a weak dependence on material z.This enables imaging of low-z materials inside high-z materials.Here,we discuss the limitations and current improvements in fast-neuron imaging.Methods Limitations in portable fast-neutron imaging systems include low D-T neutron generator output,low light pro-duction in znS(Cu)imaging scintillators,low resolution due to scintillator thickness and D-T spot size,and digital-panel darknoise that varies in time and position and that can be 100×larger than the neutron signal.We have made improvements in these areas through development of a segmented high light yield scintillator,panel noise mitigation techniques,and testing of new high-output,small spot size D-T neutron generators.Results The segmented high light yield fast-neutron scintillator demonstrated 5×increase in light compared to znS(Cu).An additional 2×improvement in signal-to-noise was demonstrated with panel-noise mitigation techniques.Our MCNP calculations also show good agreement with neutron imaging results Conclusions We have demonstrated improvements in fast-neutron imaging through development of a segmented high light yield neutron scintillator,mitigation of digital panel noise,and preliminary testing with new high-output,small spot size D-T neutron generators.We have also demonstrated good results modeling fast-neutron images and scatter effects using MCNP.

Did maize domestication and early spread mediate the population genetics of corn leafhopper?

Investigating how crop domestication and early farming mediated crop attributes, distributions, and interactions with antagonists may shed light on today's agricultural pest problems. Crop domestication generally involved artificial selection for traits desirable to early farmers, for example, in creased productivity or yield, and enhanced qualities, though invariably it altered the interactions between crops and insects, and expanded the geographical ranges of crops. Thus, some studies suggest that with crop domestication and spread, insect populations on wild crop ancestors gave rise to pestiferous insect populations on crops. Here, we addressed whether the emergence of corn leafhopper (Dalbulus ma id is) as an agricultural pest may be associated with domestication and early spread of maize (zea mays mays). We used AFLP markers and mitochondrial COI sequences to assess population genetic structuring and haplotype relationships among corn leafhopper samples from maize and its wild relative zea diploperennis from multiple locations in Mexico and Argentina. We uncovered seven corn leafhopper haplotypes contained within two haplogroups, one haplogroup containing haplotypes associated with maize and the other containing haplotypes associated with z. diploperennis in a mountainous habitat. Within the first haplogroup, one haplotype was predominant across Mexican locations, and another across Argentinean locations;both were considered pestiferous. We suggested that the divergence times of the maize-associated haplogroup and of the "pestiferous" haplotypes are correlated with the chronology of maize spread following its domestication. Overall, our results support a hypothesis positing that maize domestication favored corn leafhopper genotypes preadapted for exploiting maize so that they became pestiferous, and that with the geographical expansi on of maize farming, corn leafhopper colonized z. diploperennis, a host exclusive to secluded habitats that serves as a refuge for archaic corn leafhopper genotypic diversity.
