Effect of Scutellariae Radix extract on experimental dextran-sulfate sodium-induced colitis in rats

AIM： To investigate the effect of Scutellariae Radix extract （SRE） on ulcerative colitis （UC） in rats induced by dextran-sulfate sodium （DSS）. METhoDS： Colitis was induced in male Sprague-Dawley （SD） rats （170-180 g） by 4% dextran sulfate sodium （DSS, wt/v, MW 54000） in drinking water for 8 d. The treated rats received 4% DSS and SRE orally （100 mg/kg per day）. Control rats received either tap water or SRE only. Macroscopic assessment which included body weight changes, fecal occult blood and stool consistency were determined daily. At the appointed time, the rats were sacrificed and the entire colons were removed. The colon length and the myeloperoxidase （MPO） activity were measured. The severity of colitis was graded by morphological and histological assessments. The ion transport activity of the colonic mucosa was assessed by electrophysiological technique. RESULTS： Rats treated with oral administration of 4% DSS regularly developed clinical and macroscopic signs of colitis. Treatment with SRE relieved the symptoms, including the reduction in body weight, shortening 2nd ulceration of the colon. Administration of SRE also significantly reduced the histological damage induced by DSS. Moreover, the Isc responses of the colonic mucosa to forskolin, were suppressed after the induction of colitis. The stimulated ion transport activity of DSS-rats treated with SRE displayed significant improvement in the secretory responsiveness. CONCLUSION： SRE was effective in treating acute DSS- induced ulcerative colitis, as gauged by reduced clinical disease, improved macroscopic and histological damage scores, and enhanced recovery of normal colonic secretory function.

Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. however,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy *** together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. This study also indicates that a novel NK-targeted immune evasion strategy is used by influenza virusto shrink both innate and adaptive immune responses.

Making Sense of Interdisciplinary General Education Curriculum Design: Case Study of Common Core Curriculum at the University of hong Kong

A cyanostilbene-based molecule with aggregation-induced emission properties: amplified spontaneous emission, protonation effect and electroluminescence

A terpyridine-substituted cyanostilbene derivative(Z)-2-(4′-([2,2′:6′,2″-terpyridin]-4′-yl)-[1,1′-biphenyl]-4-yl)-3-phenylacrylonitrile(CNSTPy) was synthesized and characterized. The compound exhibits remarkable aggregation-induced emission phenomenon and its single crystal shows a blue emission with fluorescent efficiency as high as 45%. Based on its aggregation state behavior, multiple applications towards exploring its lasing, fluorescence switching and electroluminescence properties were realized. Amplified spontaneous emission(ASE) was observed from the crystal and verified by the variable pump strip method, with a threshold value of ～1.5 m J cm^(-2). The protonation/deprotonation processes accompanied by the formation of different molecular aggregation structure result in the distinct blue and yellow emissions. Additionally, the molecule also shows a moderate electroluminescence performance.

NOD2 and TLR2 ligands trigger the activation of basophils and eosinophils by interacting with dermal fibroblasts in atopic dermatitis-like skin inflammation

The skin of patients with atopic dermatitis （AD） has a unique predisposition for colonization by Staphylococcus aureus （S. aureus）, which contributes to the inflammation and grim prognosis of AD. Although the mechanism underlying the S. aureus-induced exacerbation of AD remains unclear, recent studies have found a pivotal role for pattern recognition receptors in regulating the inflammatory responses in S. aureus infection. In the present study, we used a typical mouse model of AD-like skin inflammation and found that S. aureus-associated nucleotide-binding oligomerization domain-containing protein 2 （NOD2） and toll-like receptor 2 （TLR2） ligands exacerbated AD-like symptoms, which were further deteriorated by the in vivo expansion of basophils and eosinophils. Subsequent histological analyses revealed that dermal fibroblasts were pervasive in the AD-like skin lesions, Co-culture of human dermal fibroblasts with basophils and eosinophils resulted in a vigorous cytokine/chemokine response to the NOD2/TLR2 ligands and the enhanced expression of intercellular adhesion molecule-1 on the dermal fibroblasts. Basophils and eosinophils were primarily responsible for the AD-related cytokine/chemokine expression in the co-cultures. Direct intercellular contact was necessary for the crosstalk between basophils and dermal fibroblasts, while soluble mediators were sufficient to mediate the eosinophil-fibroblast interactions. Moreover, the intracellular p38 mitogen-activated protein kinase, extracellular signal-regulated kinase, and nuclear factor-kappa B signaling pathways were essential for NOD2/TLR2 ligand-mediated activation of basophils, eosinophils, and dermal fibroblasts in AD-related inflammation. This study provides the evidence of NOD2/TLR2-mediated exacerbation of AD through activation of innate immune cells and therefore sheds light on a novel mechanistic pathway bv which S. aureus contributes to the DathoDhvsiology of AD.

Interactions Between Arbuscular Mycorrhizae and Plants in Phytoremediation of Metal-Contaminated Soils: A Review

Metal contamination in the environment is a global concern due to its high toxicity to living organisms and its worldwide distribution. The principal goal of this review is to examine the current strategies and technologies for the remediation of metal- contaminated soils by metal-accumulating plants and assess the roles of arbnscular mycorrhizal （AM） fungi in remediation of soils under hyperaccumulator or non-accumulator plants. The use of plants to remove metals from the environment or reduce the toxicity, known as phytoremediation, is an environmentally sustainable and low cost remediation technology. The mechanisms of the use of hyperaccumulator plants for phytoremediation included solubilization of the metal in the soil matrix, the plant uptake of the metal, detoxification/chelation and sequestration, and volatilization. Recently, some ecologists have found that phytoremediation with the aids of mycorrhizae can enhance efficiency in the removal of toxic metals. AM fungi can facilitate the survival of their host plants growing on metal-contaminated land by enhancing their nutrient acquisition, protecting them from the metal toxicity, absorbing metals, and also enhancing phytostabilization and phytoextraction. Such information may be useful for developing phytoremediation program at metal-contaminated sites.

Background: The welding trade includes many industrial activities in both the formal and informal sectors. Workers in this professional environment do not have access to individual and collective means of protection adapted to the activity, which exposes them to the dust emitted during their activity and therefore contributes to increasing the risk of developing respiratory disorders in this population. Objective: Our study aimed to evaluate the ventilatory function of metal welders in the city of Dakar. Method: This was a comparative study based on the analysis of the spirometric parameters of metal welders working in the city of Dakar (Senegal) with those of a control group of the same kind, selected from the general population. Measurements of forced expiratory volume in the first second (FEV1), forced vital capacity (FVC), peak expiratory flow (PEF) and maximum mid expiratory flow (MMEF) were measured. Results: A total of 75 men between the ages of 19 and 61 were recruited and divided into two groups (43 controls and 32 welders). The welding population had more frequent distal and central obstructions and pulmonary restriction compared to the control group. The metals most commonly used by welders were aluminum for welders who had an obstruction, while for those who had a restriction, iron was the most used metal. Signs of metal fever were frequently found in welders, especially those with ventilatory disorders. Conclusion: The prevalence of respiratory disorders observed is close to that observed in industrial environments, where exposure levels are the highest. The lack of personal protective equipment could also be a significant risk factor in the development of these disorders.

Techno-economic Analysis of Distributed Hydrogen Production from Natural Gas

It is well established that hydrogen has the potential to make a significant contribution to the world energy *** U.S.,majority of hydrogen production plants implement steam methane reforming(SMR) for centralized hydrogen ***,there is a wide lack of agreement on the nascent stage of using hydrogen as fuel in vehicles industry because of the difficulty in delivery and *** performing technological and economic analysis,this work aims to establish the most feasible hydrogen production pathway for automotives in near *** the evaluation,processes such as thermal cracking of ammonia and centralized hydrogen production followed by bulk delivery are eliminated while on-site steam reforming of methanol and natural gas are the most technologically feasible *** two processes are further evaluated by comprehensive economic *** results showed that the steam reforming(SR) of natural gas has a shorter payback time and a higher return on investment(ROI) and internal rate of return(IRR).Sensitivity analysis has also been constructed to evaluate the impact of variables like NG feedstock price,capital of investment and operating capacity factor on the overall production cost of *** on this study,natural gas is prompted to be the most economically and technologically available raw material for short-term hydrogen production before the transition to renewable energy source such as solar energy,biomass and wind power.
