BACKGROUND 3,6-dichlorobenzo[b]thiophene-2-carboxylic acid(BT2)is a benzothiophene carboxylate derivative that can suppress the catabolism of branched-chain amino acid(BCAA)-associated mammalian target of rapamycin complex 1(mTORC1)activation.Previous studies have demonstrated the therapeutic effects of BT2 on arthritis,liver cancer,and kidney injury.However,the effects of BT2 on ulcerative colitis(UC)are unknown.AIM To investigate the anti-UC effects of BT2 and the underlying mechanism.METHODS Mouse UC models were created through the administration of 3.5%dextran sodium sulfate(DSS)for 7 d.The mice in the treated groups were administered salazosulfapyridine(300 mg/kg)or BT2(20 mg/kg)orally from day 1 to day 7.At the end of the study,all of the mice were sacrificed,and colon tissues were removed for hematoxylin and eosin staining,immunoblot analyses,and immunohistochemical assays.Cytokine levels were measured by flow cytometry.The contents of BCAAs including valine,leucine,and isoleucine,in mouse serum were detected by liquid chromatography-tandem mass spectrometry,and the abundance of intestinal flora was analyzed by 16S ribosomal DNA sequencing.RESULTS Our results revealed that BT2 significantly ameliorated the inflammatory symptoms and pathological damage induced by DSS in mice.BT2 also reduced the production of the proinflammatory cytokines interleukin 6(IL-6),IL-9,and IL-2 and increased the anti-inflammatory cytokine IL-10 level.In addition,BT2 notably improved BCAA catabolism and suppressed mTORC1 activation and cyclooxygenase-2 expression in the colon tissues of UC mice.Furthermore,highthroughput sequencing revealed that BT2 restored the gut microbial abundance and diversity in mice with colitis.Compared with the DSS group,BT2 treatment increased the ratio of Firmicutes to Bacteroidetes and decreased the abundance of Enterobacteriaceae and Escherichia-shigella.CONCLUSION Our results indicated that BT2 significantly ameliorated DSS-induced UC and that the latent mechanism involved the suppression of BCAA-as

Microstructural characterization and mechanical properties of(TiC+TiB)/TA15 composites prepared by an in-situ synthesis method

Titanium matrix composites reinforced with ceramic particles are considered a promising engineering material due to their combination of high specific strength,low density,and high modulus.In this study,the TA15-based composites reinforced with a volume fraction of 10% to 25%(TiB+TiC)were prepared using powder metallurgy and casting technique.Microstructural characterization and phase constitution were examined using optical microscopy(OM),scanning electron microscopy(SEM),and X-ray diffraction(XRD).In addition,the microhardness,room temperature(RT)and high temperature(HT)tensile properties of the composites were evaluated.Results revealed that the reinforcements are distributed uniformly even in the composites with a high volume of TiB and TiC.However,as the volume fraction exceeds 15%,TiB and TiC particles become coarsening and exhibit rod-like and dendritic-like morphology.Microhardness increases gradually from 321.2 HV for the base alloy to a maximum of 473.3 HV as the reinforcement increases to 25vol.%.Tensile test results indicate that a reinforcement volume fraction above 20% is beneficial for enhancing tensile strength and yield strength at high temperatures,but it has an adverse effect on room temperature elongation.Conversely,if the reinforcement volume fraction is below 20%,it can improve high-temperature elongation when the temperature exceeds 600℃.

Six million Q factor micro fused silica shell resonator with teeth-like tines released by femtosecond laser-assisted chemical etching

The quality factor(Q factor) is a crucial performance parameter for resonators. In this paper, a novel release method for highquality micro fused silica shell resonators with teeth-like tines with good surface morphology is proposed. This method is based on femtosecond laser-assisted chemical etching. First, the optimal energy range of femtosecond laser modification is obtained through mechanism analysis. Second, the optimal parameters for a straight line and arc pattern are determined by optimizing the average output power, processing speed, and processing spacing. The results demonstrate why edge breakage in rounded corners is easy under different parameters. Finally, according to these conclusions, the processing is performed on a micro fused silica shell resonator with a Q factor exceeding 6 million. In addition, subsurface damage is rare throughout the fabrication process, and the surface roughness of the released cross section reaches the nanometer level. The improved Q factor helps suppress mechanical thermal noise and reduce zero bias and zero bias drift, constituting the primary method for enhancing the performance of the resonant gyroscope.

Distribution and geochemical significance of alkylbenzenes for crude oil with different depositional environments and thermal maturities

A total of 45 alkylbenzenes were detected and identified in crude oils with different depositional environments and thermal maturities from the Tarim Basin,Beibuwan Basin,and Songliao Basin using comprehensive two-dimensional gas chromatography time-of-flight mass spectrometry(GC×GCTOFMS).By analyzing the distribution characteristics of C0-C5alkylbenzenes,it is found that the content of some alkylbenzenes varies greatly in crude oils.Based on the distribution characteristics of 1,2,4,5-tetramethylbenzene(Te MB)and 1,2,3,4-Te MB,the ratio of 1,2,4,5-Te MB to 1,2,3,4-Te MB is proposed to indicate the organic matter origin and depositional environment of ancient sediments.Oil samples originated mainly from lower hydrobiont,algae,bacteria and source rocks deposited under reducing/anoxic conditions have low 1,2,4,5-/1,2,3,4-Te MB values(less than 0.6),while oil samples originated mainly from terrestrial higher plants and source rocks deposited under oxic/sub-oxic conditions have higher 1,2,4,5-/1,2,3,4-Te MB values(greater than 1.0).The significant difference of 1,2,4,5-/1,2,3,4-Te MB values is controlled by 1,2,4,5-Te MB content.1,2,4,5-Te MB content in oils derived from source rocks deposited in oxidized sedimentary environment(greater than 1.0 mg/g whole oil)is higher than that in oils from source rocks deposited in reduced sedimentary environment(less than 1.0 mg/g whole oil).1,2,4,5-/1,2,3,4-Te MB ratio might not or slightly be affected by evaporative fractionation,biodegradation and thermal maturity.1,2,4,5-/1,2,3,4-Te MB ratio and 1,2,4,5-Te MB content can be used as supplementary parameter for the identification of sedimentary environment and organic matter input.It should be noted that compared to the identification of organic matter sources,the 1,2,4,5-/1,2,3,4-Te MB parameter is more effective in identifying sedimentary environments.

Here,we characterize the temporal and spatial dynamics of forest community structure and species diversity in a subtropical evergreen broad-leaved forest in China.We found that community structure in this forest changed over a 15-year period.Specifically,renewal and death of common species was large,with the renewal of individuals mainly concentrated within a few populations,especially those of Aidia canthioides and Cryptocarya concinna.The numbers of individual deaths for common species were concentrated in the small and mid-diameter level.The spatial distribution of community species diversity fluctuated in each monitoring period,showing a more dispersed diversity after the 15-year study period,and the coefficient of variation on quadrats increased.In 2010,the death and renewal of the community and the spatial variation of species diversity were different compared to other survey years.Extreme weather may have affected species regeneration and community stability in our subtropical monsoon evergreen broad-leaved forests.Our findings suggest that strengthening the monitoring and management of the forest community will help better understand the long-and short-term causes of dynamic fluctuations of community structure and species diversity,and reveal the factors that drive changes in community structure.

Atomic-scale mechanism of effect of co-separation of elements at interface of ferrite and austenite on Cr-depleted zone of duplex stainless steel

Three-dimensional atom-probe tomography and first-principles calculation combined with density functional theory were used to study the effect of the co-segregation of different elements formed during the solidification process of S32205 duplex stainless steel on the Cr-depleted zone at the interface between ferrite and austenite.It was found that the co-segregation of different elements formed during the solidification process of duplex stainless steel can also form Cr-depleted zone at the interface between ferrite and austenite.Moreover,Mo,Si,B,C and P atoms promote co-segregation with Cr atoms,which promotes the formation of Cr-depleted zone at the interface between ferrite and austenite in duplex stainless steel.Mo and Si strongly promote the segregation of Cr at the interface between ferrite and austenite,thereby promoting the formation of Cr-depleted zone.B,C and P elements also promote the segregation of Cr element at the interface between ferrite and austenite and the formation of Cr-depleted zone,but their effect is weaker than that of Mo and Si elements.These conclusions provide a new theoretical basis for improving the intergranular corrosion performance of duplex stainless steel.

The influence of yttrium and manganese additions on the degradation and biocompatibility of magnesium-zinc-based alloys:In vitro and in vivo studies

The repair and regeneration of bone defects are highly challenging orthopedic problems.Recently,Mg-based implants have gained popularity due to their unique biodegradation and elastic modulus similar to that of human bone.The aim of our study is to develop a magnesium alloy with a controllable degradation that can closely match bone tissue to help injuries heal in vivo and avoid cytotoxicity caused by a sudden increase in ion concentration.In this study,we prepared and modified Mg-3Zn,Mg-3Zn-1Y,and Mg-2Zn-1Mn by hot extrusion,and used Mg-2.5Y-2.5Nd was as a control.We then investigated the effect of additions of Y and Mn on alloys'properties.Our results show that Mn and Y can improve not only compression strength but also corrosion resistance.The alloy Mg-2Zn-1Mn demonstrated good cytocompatibility in vitro,and for this reason we selected it for implantation in vivo.The degraded Mg-2Zn-1Mn implanted a bone defect area did not cause obvious rejection and inflammatory reaction,and the degradation products left no signs of damage to the heart,liver,kidney,or brain.Furthermore,we find that Mg-2Zn-1Mn can promote an osteoinductive response in vivo and the formation of bone regeneration.

Perioperative remedial antiviral therapy in hepatitis B virus-related hepatocellular carcinoma resection:How to achieve a better outcome

BACKGROUND Although the benefits of antiviral therapy for hepatitis B virus(HBV)-related hepatocellular carcinoma(HCC)have been proven,researchers have not con-firmed the differences in patient outcomes between patients who received preoperative antiviral therapy for a period of time(at least 24 wk)and patients who received remedial antiviral therapy just before radical resection for HBV-related HCC.AIM To investigate the efficacy of perioperative remedial antiviral therapy in patients with HBV-related HCC.METHODS A retrospective study of patients who underwent radical resection for HBV-related HCC at the First Affiliated Hospital of Xi’an Jiaotong University from January 2016 to June 2019 was conducted.Considering the history of antiviral therapy,patients were assigned to remedial antiviral therapy and preoperative antiviral therapy groups.RESULTS Kaplan–Meier analysis revealed significant differences in overall survival(P<0.0001)and disease-free survival(P=0.035)between the two groups.Multivariate analysis demonstrated that a history of preoperative antiviral treatment was independently related to improved survival(hazard ratio=0.27;95%confidence interval:0.08-0.88;P=0.030).CONCLUSION In patients with HBV-related HCC,it is ideal to receive preoperative long-term antiviral therapy,which helps patients tolerate more extensive hepatectomy;however,remedial antiviral therapy,which reduces preoperative HBV-DNA levels to less than 4 Log10 copies DNA/mL,can also result in improved outcomes.

Identification of reservoir types in deep carbonates based on mixedkernel machine learning using geophysical logging data

Identification of reservoir types in deep carbonates has always been a great challenge due to complex logging responses caused by the heterogeneous scale and distribution of storage spaces. Traditional cross-plot analysis and empirical formula methods for identifying reservoir types using geophysical logging data have high uncertainty and low efficiency, which cannot accurately reflect the nonlinear relationship between reservoir types and logging data. Recently, the kernel Fisher discriminant analysis(KFD), a kernel-based machine learning technique, attracts attention in many fields because of its strong nonlinear processing ability. However, the overall performance of KFD model may be limited as a single kernel function cannot simultaneously extrapolate and interpolate well, especially for highly complex data cases. To address this issue, in this study, a mixed kernel Fisher discriminant analysis(MKFD) model was established and applied to identify reservoir types of the deep Sinian carbonates in central Sichuan Basin, China. The MKFD model was trained and tested with 453 datasets from 7 coring wells, utilizing GR,CAL, DEN, AC, CNL and RT logs as input variables. The particle swarm optimization(PSO) was adopted for hyper-parameter optimization of MKFD model. To evaluate the model performance, prediction results of MKFD were compared with those of basic-kernel based KFD, RF and SVM models. Subsequently, the built MKFD model was applied in a blind well test, and a variable importance analysis was conducted. The comparison and blind test results demonstrated that MKFD outperformed traditional KFD, RF and SVM in the identification of reservoir types, which provided higher accuracy and stronger generalization. The MKFD can therefore be a reliable method for identifying reservoir types of deep carbonates.

Establishing and clinically validating a machine learning model for predicting unplanned reoperation risk in colorectal cancer

BACKGROUND Colorectal cancer significantly impacts global health,with unplanned reoperations post-surgery being key determinants of patient outcomes.Existing predictive models for these reoperations lack precision in integrating complex clinical data.AIM To develop and validate a machine learning model for predicting unplanned reoperation risk in colorectal cancer patients.METHODS Data of patients treated for colorectal cancer(n=2044)at the First Affiliated Hospital of Wenzhou Medical University and Wenzhou Central Hospital from March 2020 to March 2022 were retrospectively collected.Patients were divided into an experimental group(n=60)and a control group(n=1984)according to unplanned reoperation occurrence.Patients were also divided into a training group and a validation group(7:3 ratio).We used three different machine learning methods to screen characteristic variables.A nomogram was created based on multifactor logistic regression,and the model performance was assessed using receiver operating characteristic curve,calibration curve,Hosmer-Lemeshow test,and decision curve analysis.The risk scores of the two groups were calculated and compared to validate the model.RESULTS More patients in the experimental group were≥60 years old,male,and had a history of hypertension,laparotomy,and hypoproteinemia,compared to the control group.Multiple logistic regression analysis confirmed the following as independent risk factors for unplanned reoperation(P<0.05):Prognostic Nutritional Index value,history of laparotomy,hypertension,or stroke,hypoproteinemia,age,tumor-node-metastasis staging,surgical time,gender,and American Society of Anesthesiologists classification.Receiver operating characteristic curve analysis showed that the model had good discrimination and clinical utility.CONCLUSION This study used a machine learning approach to build a model that accurately predicts the risk of postoperative unplanned reoperation in patients with colorectal cancer,which can improve treatment decisions and prognosis.
