Effects of the COVID-19 pandemic on semen quality in male partners of infertile couples: a hospitalbased observational study

The effects of the coronavirus disease 2019(COVID-19)pandemic on male fertility have received considerable attention because human testes contain high levels of angiotensin-converting enzyme-2 receptors,through which severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)can *** studies showed decreases in semen quality during and after recovery from ***,no semen quality studies have examined the effects of widespread subclinical and mild disease,as well as changes in lifestyle,psychosocial behavior,intake of dietary supplements,and *** cross-sectional study compared semen quality parameters in male partners of infertile couples between men who underwent semen analysis before the COVID-19 pandemic(prepandemic group)and men who underwent semen analysis during the pandemic period(pandemic group);the analysis sought to clarify the overall effects of the *** participants in the pandemic group had experienced clinically overt *** the 239 participants,mean body weight(P=0.001),mean body mass index(P<0.001),median sperm concentration(P=0.014),total sperm count(P=0.006),and total percentages of motile(P=0.013)and abnormal cells(P<0.001)were significantly greater in the pandemic group(n=137)than those in the prepandemic group(n=102).Among abnormal cells,the percentages of cells with excess residual cytoplasm(P<0.001),head defects(P<0.001),and tail defects(P=0.015)were significantly greater in the pandemic group than those in the prepandemic *** the exception of morphology,the overall semenogram results were better in the pandemic group than those in the prepandemic group.

Variation in seed morphometric characters, oil content and azadirachtin content of seeds, in vitro shoot cultures and callus cultures among different populations of Azadirachta indica

We studied seed morphometric characters, oil content and azadirachtin content in a medicinally important and insect repellent tree species Azadirachta indica ***. Ten populations were selected from two central Indian states of Madhya Pradesh and Chhattisgarh and twenty candidate plus trees were selected from each population based on tree attributes. Azadirachtin content was estimated in seeds, in vitro shoots and callus cultures through HPLC. Significant variation（p ／ 0.05） was observed in seed morphometric characters, oil content and azadirachtin content among the ten populations. Maximum value for 100 seed weight was obtained in seeds of Raigarh（30.51 g） which was on par with seeds of Bargi（28.37 g）and Chhatarpur（26.36 g）. The ratio of highest kernel to seed was also obtained in seeds of Raigarh（60.11%）.Kernels from Chhatarpur population recorded the highest oil content（24.20%）, which was equal to that in Sihore（23.75%） and Katni（21.75%）. We recorded highly significant variation in azadirachtin content in seed samples,in vitro shoot samples and callus cultures. Maximum azadirachtin was present in the seeds of Gwalior（0.882%）which was on par with azadirachtin content in seeds of Chhatarpur, Katni, Sihore, Shahdol and *** azadirachtin in the in vitro shoots was present in the cultures of Gwalior（0.218%） which was statistically on par with Sihore（0.176%）. Similarly, in callus cultures of nodal segments maximum azadirachtin was recorded in the Gwalior population（0.033%） which was statistically on par with Sihore（0.028%）. The trees of Gwalior and Sihore populations can be produced in large numbers through vegetative propagation to obtain seeds with higher azadirachtin content.

Utilization of biodiesel waste in the development of botanical-based floating tablet formulation against early stages of mosquitoes

The current paper has elaborated the efficient utilization of liquid biodiesel waste in combination with dillapiole and citronella essential oil as active ***,cellulose and hydrophobic silica were used as inert ingredients,which make the tablet to float over the water ***-FTIR analysis of tablet confirmed the compatibility with citronella oil,dillapiole,liquid biodiesel waste in tablet composition after ***-chemical analysis studies show that tablet parameters are in standard *** analysis shows some porous structures in tablet composition which confirms the floating nature of the *** specific ratio(2:2:1)of citronella oil,dillapiole and liquid biodiesel waste showed maximum mortality,i.e.95%after 24 *** application,the tablet is nontoxic towards the aquatic organisms and water quality remains *** better performance of the tablets has been evaluated in terms of characterization studies,***-FTIR and SEM studies and bioefficacy trials which confirmed the presence of active ingredients responsible for insecticidal activity.

Bioengineered miR-124-3p prodrug selectively alters the proteome of human carcinoma cells to control multiple cellular components and lung metastasis in vivo

Bioengineered miR-328-3p modulates GLUT1-mediated glucose uptake and metabolism to exert synergistic antiproliferative effects with chemotherapeutics

MicroRNAs(miRNAs or miRs)are small noncoding RNAs derived from genome to control target gene *** we have developed a novel platform permitting high-yield production of bioengineered miRNA agents(BERA).This study is to produce and utilize novel fully-humanized BERA/miR-3’28-3p molecule(hBERA/miR-3’28)to delineate the role of miR-328-3p in controlling nutrient uptake essential for cell *** first demonstrated successful high-level expression of hBERA/miR-328 in bacteria and purification to high degree of homogeneity(>98%).Biologic miR-328-3p prodrug was selectively processed to miR-328-3p to suppress the growth of highly-proliferative human osteosarcoma(OS)*** glucose transporter protein type 1,gene symbol solute carrier family 2 member 1(GUJTHSLC2A1),we identified and verified large neutral amino acid transporter 1,gene symbol solute carrier family 7 member 5(LPAT1/SLC7A5)as a direct target for miR-3’*** reduction of LAT1 protein levels by miR-3’28-3p did not alter homeostasis of amino acids within OS cells,suppression of GLUT1 led to a significantly lower glucose uptake and decline in intracellular levels of glucose and glycolytic metabolite ***,combination treatment with hBERA/miR-3’28 and cisplatin or doxorubicin exerted a strong synergism in the inhibition of OS cell *** findings support the utility of novel bioengineered RNA molecules and establish an important role of miR-328-3p in the control of nutrient transport and homeostasis behind cancer metabolism.

Bioengineered miR-27b-3p and miR-328-3p modulate drug metabolism and disposition via the regulation of target ADME gene expression

Drug-metabolizing enzymes, transporters, and nuclear receptors are essential for the absorption, distribution, metabolism, and excretion(ADME) of drugs and xenobiotics. MicroRNAs participate in the regulation of ADME gene expression via imperfect complementary Watson–Crick base pairings with target transcripts. We have previously reported that Cytochrome P450 3A4(CYP3A4) and ATP-binding cassette sub-family G member 2(ABCG2) are regulated by miR-27b-3p and miR-328-3p,respectively. Here we employed our newly established RNA bioengineering technology to produce bioengineered RNA agents(BERA), namely BERA/miR-27b-3p and BERA/miR-328-3p, via fermentation. When introduced into human cells, BERA/miR-27b-3p and BERA/miR-328-3p were selectively processed to target miRNAs and thus knock down CYP3A4 and ABCG2 mRNA and their protein levels,respectively, as compared to cells treated with vehicle or control RNA. Consequently, BERA/miR-27b-3p led to a lower midazolam 10-hydroxylase activity, indicating the reduction of CYP3A4 activity. Likewise,BERA/miR-328-3p treatment elevated the intracellular accumulation of anticancer drug mitoxantrone, a classic substrate of ABCG2, hence sensitized the cells to chemotherapy. The results indicate that biologic miRNA agents made by RNA biotechnology may be applied to research on miRNA functions in the regulation of drug metabolism and disposition that could provide insights into the development of more effective therapies.

Use of recombinant microRNAs as antimetabolites to inhibit human non-small cell lung cancer

During the development of therapeutic microRNAs(miRNAs or miRs),it is essential to define their pharmacological ***,miRNA research and therapy mainly use miRNA mimics synthesized in *** experimental screening of unique recombinant miRNAs produced in vivo,three lead antiproliferative miRNAs against human NSCLC cells,miR-22-3p,miR-9-5p,and miR-218-5p,were revealed to target folate metabolism by bioinformatic *** miR-22-3p,miR-9-5p,and miR-218-5p were shown to regulate key folate metabolic enzymes to inhibit folate metabolism and subsequently alter amino acid metabolome in NSCLC A549 and H1975 *** tracing studies further confirmed the disruption of one-carbon transfer from serine to folate metabolites by all three miRNAs,inhibition of glucose uptake by miR-22-3p,and reduction of serine biosynthesis from glucose by miR-9-5p and-218-5p in NSCLC *** greater activities to interrupt NSCLC cell respiration,glycolysis,and colony formation than miR-9-5p and-218-5p,recombinant miR-22-3p was effective to reduce tumor growth in two NSCLC patient-derived xenograft mouse models without causing any *** results establish a common antifolate mechanism and differential actions on glucose uptake and metabolism for three lead anticancer miRNAs as well as antitumor efficacy for miR-22-3p nanomedicine,which shall provide insight into developing antimetabolite RNA therapies.
