Survivin expression induced by doxorubicin in cholangiocarcinoma

AIM:To study the role of survivin expression induced by chemotherapy agent (doxorubicin) in the development and anti-chemotherapy of ***:Expression of survivin was detected by SP immunohistochemical technique in 33 cases of cholangiocarcinorna,28 cases of adjacent noncancerous bile duct, and 5 cases of benign bile duct lesions. Low concentration of doxorubicin (0.05mg/l) was added in cultured cholangiocarcinoma cell line (QBC939). The expression of survivin was detected by RT-PCR and Western blot at 24h and 48h after adding ***:Survivin was expressed in 24 of 33 cholangiocarcinoma cases (72.7%).In contrast,no expression of survivin in adjacent noncancerous and benign bile duct lesions was observed (P<0.01). No correlation was found between survivin expression and clinical *** could markedly (P<0.001) up-regulate survivin mRNA and protein expression of QBC939 ***:Overexpression of survivin in cholangiocarcinomas may play an important role in the development of cholangiocarcinoma, its relationship with prognosis of cholangiocarcinoma deserves further investigation. Higher expression of survivin is induced by doxorubicin in *** expression may resist apoptosis induced by chemotherapy agents.

Crowd behaviors analysis is the‘state of art’research topic in the field of computer vision which provides applications in video surveillance to crowd safety,event detection,security,*** presents some of the works related to crowd behavior detection and *** crowd behavior detection,varying density of crowds and motion patterns appears to be complex occlusions for the *** work presents a novel crowd behavior detection system to improve these *** proposed crowd behavior detection system is developed using hybrid tracking model and integrated features enabled neural *** object movement and activity in the proposed crowded behavior detection system is assessed using proposed GSLM-based neural *** based neural network is developed by integrating the gravitational search algorithm with LM algorithm of the neural network to increase the learning process of the *** performance of the proposed crowd behavior detection system is validated over five different videos and analyzed using *** experimentation results in the crowd behavior detection with a maximum accuracy of 93%which proves the efficacy of the proposed system in video surveillance with security concerns.

AIM: To investigate the therapeutic effect of somatostatin receptor type 2 (SSTR2) gene transfection on pancreatic carcinoma xenografts in vivo in experimental cancers. METHODS: Human pancreatic cancer cell line Panc-1 was inoculated subcutaneously into the back of nude mice. When tumor nodules were grown as large as about 5 mmx5 mm days after inoculation, the mice were randomly divided into 3 groups (6 mice in each group). Group Ⅰ served as untreated control group. Group Ⅱ received an intratumoral injection of a combination of human cytomegalovirus promoter-6C (pCMV-6C) and lipofectamine 2000. Group Ⅲ received an intratumoral injection of a combination of pCMV-6C-SSTR2 and lipofectamine 2000. The rate of tumor growth was compared among these three groups. The expression of SSTR2 in these tumors was detected by immunohistochemistry and Western-blot. Apoptosis index (AI) in these tumors was examined by using TUNEL in situ. RESULTS: Intratumoral injection of a combination of pCMV-6C-SSTR2 and lipofectamine 2000 resulted in the expression of SSTR2 protein. The tumor size and weight in group Ⅲ (0.318±0.098 cm3, and 0.523±0.090 g, respectively) were significantly lower than those in group I (2.058±0.176 cms, and 1.412±0.146 g, respectively) and group Ⅱ (2.025±0.163 cm3, and 1.365±0.116 g, respectively) (P<0.05) The AI in group Ⅲ (1.47±0.13%) was significantly higher than that in groupⅠ(0.56±0.09%) and group Ⅱ (0.57±0.11%) (P<0.05). But there were no significant differences between groups Ⅰ and Ⅱ. CONCLUSION: Our data demonstrate that re-expression of SSTR2 gene has antitumor effects on experimental pancreatic cancer. Restoration of SSTR2 gene expression through gene transfer in vivo might be a potential gene therapy strategy for human pancreatic cancer.

Tumor angiogenesis is the proliferation of a network of blood vessels that penetrates into cancerous growths, supplying nutrients and oxygen and removing waste products. The process of angiogenesis plays an important role in many physiological and pathological conditions. Solid tumors depend on angiogenesis for growth and metastasis in a hostile environment. In the prevascular phase, the tumor is rarely larger than 2 to 3 mm3 and may contain a million or more cells. Up to this size, tumor cells can obtain the necessary oxygen and nutrient supplies required for growth and survival by simple passive diffusion. The properties of tumors to release and induce several angiogenic and antiangiogenic factors which play crucial roles in regulating endothelial cell (EC) proliferation, migration, apoptosis or survival, cell-cell and cell-matrix adhesion through different intracellular signaling are thought to be the essential mechanisms during tumor-induced angiogenesis. Tumor angiogenesis actually starts with tumor cells releasing molecules that send signals to surrounding normal host tissue. This signaling activates certain genes in the host tissue that, in turn, make proteins to encourage growth of new blood vessels. In this review, we focus the mechanisms of tumor-induced angiogenesis, with an emphasis on the regulatory role of several angiogenic and anti-angiogenic agents during the angiogenic process in tumors. Advances in understanding the mechanisms of tumor angiogenesis have led to the development of several most effective antiangiogenic and anti-metastatic therapeutic agents and also have provided several techniques for the regulation of cancer's angiogenic switch. The suggestion is made that standard cytotoxic chemotherapy and angiogenesis inhibitors used in combination may produce complementary therapeutic benefits in the treatment of cancer.

Cloud computing has gained widespread popularity over the last *** problem in cloud computing is prejudiced due to enormous demands of cloud ***-heuristic techniques in cloud computing have exhibited high performance in comparison to traditional scheduling *** paper presents a novel hybrid Nesterov Accelerated Gradient-based Cuckoo Search Algorithm(NAGCSA)to address the scheduling issue in cloud *** Accelerated Gradient can address trapping at local minima in CSA by updating the position using future *** local search in the proposed algorithm is performed by using Nesterov Accelerated Gradient,while the global search is performed by using levy *** amalgamation of NAG and CSA helps in cost reduction and time-saving for *** simulation has been carried out on the CloudSim tool on three different real datasets;NASA,HPC2N,and *** results of the proposed hybrid algorithm have been compared with state-of-art scheduling algorithms(GA,PSO,and CSA),and statistical significance is carried on mean,standard deviation,and best for each *** has been established that the proposed algorithm minimizes the execution cost and makespan,hence enhancing the quality of service for users.

A numerical algorithm is developed for the approximation of the solution to certain boundary value problems involving the third-order ordinary differential equation associated with draining and coating flows. The authors show that the approximate so- lutions obtained by the numerical algorithm developed by using uonpolynomial quintic spline functions ave better than those produced by other spline and domain decomposition methods. The algorithm is tested on two problems associated with draining and coating flows to demonstrate the practical usefulness of the approach.

Amyotrophic lateral sclerosis（ALS） and motor neuron diseases（MNDs） are progressive neurodegenerative diseases that affect nerve cells in the brain affecting upper and lower motor neurons（UMNs/LMNs）, brain stem and spinal cord. The clinical phenotype is characterized by loss of motor neurons（MNs）, muscular weakness and atrophy eventually leading to paralysis and death due to respiratory failure within 3–5 years after disease onset. No effective treatment or cure is currently available that halts or reverses ALS and MND except FDA approved drug riluzole that only modestly slows the progression of ALS in some patients. Recent advances in human derived induced pluripotent stem cells have made it possible for the first time to obtain substantial amounts of human cells to recapitulate in vitro “disease in dish” and test some of the underlying pathogenetic mechanisms involved in ALS and MNDs. In this review, I discussed the opportunities and challenges of induced pluropotent stem cells-derived motor neurons for treatment of ALS and MND patients with special emphasis on their implications in finding a cure for ALS and MNDs.

AIM: To explore the difference of somatostatin receptor subtype 2 (SST2R) gene expression in pancreatic cancerous tissue and its adjacent tissue, and the relationship between the change of SST2R gene expression and pancreatic tumor angiogenesis related genes. METHODS: The expressions of SST2R, DPC4, p53 and ras genes in cancer tissues of 40 patients with primary pancreatic cancer, and the expression of SST2R gene in its adjacent tissue were determined by immunohistochemiscal LSAB method and EnVisionTM method. Chi-square test was used to analyze the difference in expression of SST2R in pancreatic cancer tissue and its adjacent tissue, and the correlation of SST2R gene expression with the expression of p53, ras and DPC4 ***: Of the tissue specimens from 40 patients with primary pancreatic cancer, 35 (87.5%) cancer tissues showeda negative expression of SST2R gene, whereas 34 (85%) a positive expression of SST2R gene in its adjacent tissues. Five (12.5%) cancer tissues and its adjacent tissues simultaneously expressed SST2R. The expression of SST2R gene was markedly higher in pancreatic tissues adjacent to cancer than in pancreatic cancer tissues (P0.05). CONCLUSION: There was a significant difference of SST2R gene expression in pancreatic cancer tissues and its adjacent tissues, which might be one cause for the different therapeutic effects of somatostatin and its analogs on pancreatic cancer patients. There were abnormal expressions of SST2R, DPC4, p53 and ras genes in pancreatic carcinogenesis, and moreover, the loss or decrease of SST2R gene expression was significantly negatively correlated with the overexpression of tumor angiogenesis correlated p53 and ras genes, suggesting that SST2R gene togethe

Expression of TREM-1 mRNA in acute pancreatitis

AIM: To explore the expression of triggering receptor expressed on myeloid cells (TREM-I) mRNA in acute pancreatitis (AP).METHODS: Using the reverse transcription polymerase chain reaction (RT-PCR), we examined the expression of TREM-1 mRNA in 10 cases of mild acute pancreatitis (MAP), 8 cases of severe acute pancreatitis (SAP), and 10 cases of healthy control subjects. And we also examined the expression of TREM-1 mRNA in 14 cases of AP (including 10 MAP and 4 SAP) before treatment, after successful therapy and clinically ***: The expression of TREM-1 mRNA in the groups of MAP, SAP patients and healthy control subjects was 0.771±0.274, 1.092±0.331 and 0.459±0.175, respectively;there was a significant difference among the three groups (P<0.05). And there was also a significant difference between the AP patients (0.914±0.341) and healthy control subjects (0.459+0.175) (P<0.05). Moreover, in the 14 cases of AP, before treatment, after successful therapy and clinically cured, the expression of TREM-1 mRNA was 0.905±0.226,0.739±0.169 and 0.633±0.140, respectively, and there was a significant difference among the three stages (P<0.05).CONCLUSION: The expression of TREM-1 mRNA in the patients with AP increases obviously, and correlates with the degree of AP. Furthermore, the expression of TREM-1 mRNA is distinctly different at the different stages of AP. It indicates TREM-1 may play an important role in the occurrence and development of AP.
