Assessing gray matter volume in patients with idiopathic rapid eye movement sleep behavior disorder

Idiopathic rapid eye movement sleep behavior disorder(iRBD) is often a precursor to neurodegenerative disease. However, voxel-based morphological studies evaluating structural abnormalities in the brains of iRBD patients are relatively rare. This study aimed to explore cerebral structural alterations using magnetic resonance imaging and to determine their association with clinical parameters in iRBD patients. Brain structural T1-weighted MRI scans were acquired from 19 polysomnogram-confirmed iRBD patients(male:female 16:3; mean age 66.6 ± 7.0 years) and 20 age-matched healthy controls(male:female 5:15; mean age 63.7 ± 5.9 years). Gray matter volume(GMV) data were analyzed based on Statistical Parametric Mapping 8, using a voxel-based morphometry method and two-sample t-test and multiple regression analysis. Compared with controls, iRBD patients had increased GMV in the middle temporal gyrus and cerebellar posterior lobe, but decreased GMV in the Rolandic operculum, postcentral gyrus, insular lobe, cingulate gyrus, precuneus, rectus gyrus, and superior frontal gyrus. iRBD duration was positively correlated with GMV in the precuneus, cuneus, superior parietal gyrus, postcentral gyrus, posterior cingulate gyrus, hippocampus, lingual gyrus, middle occipital gyrus, middle temporal gyrus, and cerebellum posterior lobe. Furthermore, phasic chin electromyographic activity was positively correlated with GMV in the hippocampus, precuneus, fusiform gyrus, precentral gyrus, superior frontal gyrus, cuneus, inferior parietal lobule, angular gyrus, superior parietal gyrus, paracentral lobule, and cerebellar posterior lobe. There were no significant negative correlations of brain GMV with disease duration or electromyographic activity in iRBD patients. These findings expand the spectrum of known gray matter modifications in iRBD patients and provide evidence of a correlation between brain dysfunction and clinical manifestations in such patients. Discovery of a potent FKBP38 agonist that ameliorates HFD-induced hyperlipidemia via mTOR/P70S6K/SREBPs pathway

The mammalian target of rapamycin(mTOR)-sterol regulatory element-binding proteins(SREBPs)signaling promotes lipogenesis.However,mTOR inhibitors also displayed a significant side effect of hyperlipidemia.Thus,it is essential to develop mTOR-specific inhibitors to inhibit lipogenesis.Here,we screened the endogenous inhibitors of mTOR,and identified that FKBP38 as a vital regulator of lipid metabolism.FKBP38 decreased the lipid content in vitro and in vivo via suppression of the mTOR/P70 S6 K/SREBPs pathway.3,5,6,7,8,3’,4’-Heptamethoxyflavone(HMF),a citrus flavonoid,was found to target FKBP38 to suppress the mTOR/P70 S6 K/SREBPs pathway,reduce lipid level,and potently ameliorate hyperlipidemia and insulin resistance in high fat diet(HFD)-fed mice.Our findings suggest that pharmacological intervention by targeting FKBP38 to suppress mTOR/P70 S6 K/SREBPs pathway is a potential therapeutic strategy for hyperlipidemia,and HMF could be a leading compound for development of anti-hyperlipidemia drugs.

Unusual presentation of congenital radioulnar synostosis with osteoporosis,fragility fracture and nonunion:A case report and review of literature

BACKGROUND Congenital radioulnar synostosis(CRUS)is a rare deformity of the upper extremity.It is characterized by loss of rotation of the involved forearm and functional limitations in daily activities.No studies on CRUS with osteoporosis have been reported to date,and osteoporosis is usually recognized as an important dimension of genetic disorder in children.We discuss the possible relationship among this disorder,osteoporosis and fracture nonunion,investigate the strict surgical indications and recommended treatments.CASE SUMMARY A 14-year-old male patient with bilateral CRUS with osteoporosis,fragility fracture and nonunion of fractures in ulna and radius presented our institution for further treatment,complaining of limitation in rotation.The bone mineral density of the hip and lumbar spine was 0.687 g/cm2 and 0.705 g/cm2,respectively,and the Z-score for both was-2.1,which revealed osteoporosis and a high risk of fracture.Tow serum bone turnover markers indicated an imbalance of bone metabolism.Reoperation for ulna fracture with autogenous bone grafting and a postoperative physiotherapy program were adopted rather than the separation of pathological synostosis.Radiological examination,observational posture assessment and limb function scale were evaluated before and 1 year after surgery.At 1 year,the fracture nonunion had almost recovered,forearm movement function on the fracture side was restored,and function on the healthy side was significantly improved compared with that before rehabilitation.CONCLUSION Surgical indications for CRUS vary from person to person.Surgery should not be the first choice of treatment,and physiotherapy is not inferior to surgical treatment.

Ground-state cooling of the mechanical degree of freedom is a fundamental requirement for quantum state transfer between relevant optical and mechanical systems.Here,we fabricate optomechanical crystal cavities with co-localized mechanical and optical modes on a monolithic chip.The typical linewidth of the optical modes at telecom wavelength is as low as 0.95 GHz,and the mechanical resonant frequency is around 5 GHz,which means the optomechanical system can be operated in the resolved sideband regime.With the statistics of the asymmetry in the scattering rates of red and blue detuning driving processes,we initialize and characterize the mechanical system in its quantum ground-state of motion,with a mean thermal phonon occupancy nth=0.018±0.0034,corresponding to a mode temperature of 57.3 m K.It is a general platform for quantum state transfer between relevant optical and mechanical systems,as well as the quantum entanglement between these systems.

Optimal design of hot rolling process for C-Mn steel by combining industrial data-driven model and multi-objective optimization algorithm

A successful mechanical property data-driven prediction model is the core of the optimal design of hot rolling process for hot-rolled strips. However, the original industrial data, usually unbalanced, are inevitably mixed with fluctuant and abnormal values. Models established on the basis of the data without data processing can cause misleading results, which cannot be used for the optimal design of hot rolling process. Thus, a method of industrial data processing of C-Mn steel was proposed based on the data analysis. The Bayesian neural network was employed to establish the reliable mechanical property prediction models for the optimal design of hot rolling process. By using the multi-objective optimization algorithm and considering the individual requirements of costumers and the constraints of the equipment, the optimal design of hot rolling process was successfully applied to the rolling process design for Q345B steel with 0.017% Nb and 0.046% Ti content removed. The optimal process design results were in good agreement with the industrial trials results, which verify the effectiveness of the optimal design of hot rolling process.

We study the dynamics evolution of a two-qubit Heisenberg XXX spin chain under a time-dependentrotating magnetic field.Based on the algebraic structure of the non-autonomous system,the exact solution of theSchrdinger equation is obtained by using the method of algebraic dynamics.Based on the time-dependent analyticalsolution,we further study the entanglement evolution between the two coupled spins for different initial states,and findthat the entanglement is determined by the coefficients of the initial state and the coupling constant J of the system.

Blood typing and transfusion therapy in a patient with A2 subtype acute myeloid leukemia M2:A case report

BACKGROUND Acute myeloid leukemia(AML)is one of the most common types of leukemia in adults.However,AML is relatively rare in the population overall,accounting for only about 1 percent of all cancers.Treatment for AML can be very effective for some patients,yet it leaves others with serious and even life-threatening side effects.Chemotherapy is still the primary treatment for most AML,but over time,leukemia cells become resistant to chemotherapy drugs.In addition,stem cell transplantation,targeted therapy,and immunotherapy are currently available.At the same time,with the progression of the disease,the patient may have corresponding complications,such as coagulation dysfunction,anemia,granulocytopenia,and repeated infection,so transfusion supportive therapy will be involved in the overall treatment regime.To date,few articles have reported on blood transfusion treatment options for patients with ABO subtypes AML-M2.Blood transfusion therapy is an important supportive treatment for AML-M2,and accurate determination of patients'blood type is one of the most important steps in the treatment process.In this study,we explored blood typing and supportive treatment strategies for a patient with A2 subtype AML-M2 to provide the basis for treatment for all patients.CASE SUMMARY In order to determine the blood type of the patient,serological and molecular biological methods were used for reference tests,and the genetic background was studied to determine the patient's final blood type and select the appropriate blood products for infusion treatment.According to the results obtained by serological and molecular biological methods,the blood type of the patient was A2 subtype;the genotype was A02/001;the irregular antibody screening was negative,and anti-A1 was found in the plasma.According to the overall treatment plan,active anti-infection,elevated cells,component blood transfusion support,and other rescue and supportive treatments were given,and the patient successfully passed the stage of myelosuppression after chemothera

Pangolins are among the most critically endangered animals due to heavy poaching and worldwide trafficking. However, their demographic histories and the genomic consequences of their recent population declines remain unknown. We generated high-quality de novo reference genomes for critically endangered Malayan(Manis javanica, MJ) and Chinese(M. pentadactyla, MP) pangolins and re-sequencing population genomic data from 74 MJs and 23 MPs. We recovered the population identities of illegally traded pangolins and previously unrecognized genetic populations that should be protected as evolutionarily distinct conservation units. Demographic reconstruction suggested environmental changes have resulted in a population size fluctuation of pangolins. Additionally, recent population size declines due to human activities have resulted in an increase in inbreeding and genetic load. Deleterious mutations were enriched in genes related to cancer/diseases and cholesterol homeostasis, which may have increased their susceptibility to diseases and decreased their survival potential to adapt to environmental changes and high-cholesterol diets. This comprehensive study provides not only high-quality pangolin reference genomes, but also valuable information concerning the driving factors of long-term population size fluctuations and the genomic impact of recent population size declines due to human activities, which is essential for pangolin conservation management and global action planning.

近年来,宽带隙半导体材料氧化镓在日盲紫外探测领域的应用引起了广泛关注.本文基于溶液法制备了非晶氧化镓薄膜,采用紫外光退火的方式降低了薄膜的制备温度,并且通过铝颗粒修饰氧化镓薄膜表面,提升了氧化镓紫外探测器的性能.紫外退火的方式可将氧化镓薄膜的制备温度降至300℃,有望实现柔性器件的制备.当沉积铝膜厚度在3—5 nm时,可获得分布均匀、直径为2—3 nm的铝颗粒,经修饰的氧化镓薄膜表现出优秀的光电响应性能和日盲探测特性.在254nm光照下,最大光暗电流比可达2.55×10^(4),在紫外波段的抑制比I_(254nm)/I_(365nm)为2.2×10^(4).最佳的探测器响应度和探测率分别为0.771 A/W和1.13×10^(11)Jones,相比于未做修饰的氧化镓紫外探测器提升了约34倍和36倍.然而,铝纳米颗粒的修饰也会引入部分缺陷态,导致氧化镓光电探测器响应下降时间的增大.

为了解斜拉桥施工阶段π型混凝土梁翼缘板斜向裂缝成因并制定合理的防裂措施,以某主跨120 m的π型预应力混凝土梁斜拉桥为背景,针对其悬臂施工阶段主梁翼缘板下表面出现斜向裂缝的现象进行分析。采用ANSYS软件建立该桥施工阶段杆系模型与混凝土梁实体模型,分析梁体自重、斜拉索张拉、预应力张拉、混凝土收缩、混凝土水化热作用下主梁翼缘板下表面应力情况。结果表明:梁体自重及斜拉索张拉作用下,翼缘板下表面未出现拉应力;横向及纵向预应力张拉作用下,翼缘板下表面产生一定的拉应力,但未超过混凝土抗拉强度标准值;混凝土收缩作用下,翼缘板下表面产生极小的拉应力,不会引起裂缝;混凝土养护过程中水化热作用下,翼缘板下表面混凝土与周边混凝土存在一定温差,部分测点第一主应力超过混凝土抗拉强度标准值,是开裂的主要原因。针对混凝土水化热作用影响,提出主肋布置冷却水管与翼缘板下方火炉加热并用的防裂措施,建立热流耦合模型分析防裂效果。结果表明:所采用的措施能够改善主梁养护过程中水化热温度场分布,各测点第一主应力均未超过混凝土抗拉强度标准值。
