Pathophysiological significance of gallbladder volume changes in gallstone diseases

AIM: To study the pathophysiological significance of gallbladder volume (GBV) and ejection fraction changes in gallstone ***: The fasting GBV of gallstone patients with acute cholecystitis (n = 99), chronic cholecystitis (n = 85) and non-gallstone disease (n = 240) were measured by preoperative computed tomography. Direct saline injection measurements of GBV after cholecystectomy were also performed. The fasting and postprandial GBV of 65 patients with gallstones and chronic cholecystitis and 53 healthy subjects who received health examinations were measured by abdominal ultrasonography. Proper adjustments were made after the correction factors were calculated by comparing the preoperative and postoperative measurements. Pathological correlations between gallbladder changes in patients with acute calculous cholecystitis and the stages defined by the Tokyo International Consensus Meeting in 2007 were made. Unpaired Student's t tests were used. P < 0.05 was deemed statistically significant. RESULTS: The fasting GBV was larger in late stage than in early/second stage acute cholecystitis gallbladders (84.66 ± 26.32 cm 3 , n = 12, vs 53.19 ± 33.80 cm 3 , n = 87, P = 0.002). The fasting volume/ejection fraction of gallbladders in chronic cholecystitis were larger/lower than those of normal subjects (28.77 ± 15.00 cm 3 vs 6.77 ± 15.75 cm 3 , P < 0.0001)/(34.6% ± 10.6%, n = 65, vs 53.3% ± 24.9%, n = 53, P < 0.0001). CONCLUSION: GBV increases as acute cholecystitis progresses to gangrene and/or empyema. Gallstone formation is associated with poorer contractility and larger volume in gallbladders that contain stones.

The cell membrane is a critical barrier for cellular homeostasis,integral to signaling and intercellular communication,and vital for understanding cellular functions and disease *** its microenvironment is crucial for uncovering the molecular basis of physiological and pathological processes associated with the cell membrane,driving the development of bioanalytical toolkits capable of dynamically monitoring the cell surface *** the continuous advancement of functional nucleic acids and dynamic DNA nanotechnology,DNA nanodevices with controllable nanosized geometry,specific molecular recognition,and selective membrane-localization properties offer a versatile platform for probing the cell membrane *** this review,we summarize the current biosensing and membrane-anchoring mechanisms of DNA nanodevices and highlight their use in studying key cell membrane events,including membrane lipid dynamics,transmembrane transport,receptor dimerization,and signal ***,we discuss the challenges and potential future applications of DNA nanodevices in advancing cell membrane biology research and biomedical applications.

A Compact UHF Antenna Based on Hilbert Fractal Elements and a Serpentine Arrangement for Detecting Partial Discharg

Efforts to protect electric power systems from faults have commonly relied on the use of ultra-high frequency(UHF)antennas for detecting partial discharge(PD)as a common precursor to ***,the effectiveness of existing UHF antennas suffers from a number of challenges such as limited bandwidth,relatively large physical size,and low detection *** present study addresses these issues by proposing a compact microstrip patch antenna with fixed dimensions of 100 mm×100 mm×1.6 *** results of computations yield an optimized antenna design consisting of 2nd-order Hilbert fractal units positioned within a four-layer serpentine arrangement with a fractal unit connection distance of 3.0 ***,the optimized antenna design achieves a detection bandwidth for which the voltage standing wave ratio is less than 2 that is approximately 97.3%of the UHF frequency range(0.3–3 GHz).Finally,a prototype antenna is fabricated using standard printed circuit board technology,and the results of experiments demonstrate that the proposed antenna is capable of detecting PD signals at a distance of 8 m from the discharge source.

Low-refractive-index particles play significant roles in physics,drug delivery,biomedical science,and other ***,they have not attained sufficient utilization in active manipulation due to the repulsive effect of *** this work,the establishment of customized dark traps is demonstrated to fulfill the demands of versatile manipulation of low-refractive-index *** customized dark traps are generated by assembling generalized perfect optical vortices based on the free lens modulation method,by which the beams’shape,intensity,and position can be elaborately designed with size independent of topological *** the customized dark traps with high quality and high efficiency,rotation along arbitrary trajectories with controllable speed,parallel manipulation,and sorting of low-refractive-index particles by size can be *** unprecedented flexibility and quality,the customized dark traps provide tremendous potential in optical trapping,lithography,and biomedicine.

Etomidate protects retinal ganglion cells from hydrogen peroxide-induced injury via Nrf2/HO-1 pathway

AIM:To determine whether etomidate(ET)has a protective effect on retinal ganglion cells(RGCs)injured with hydrogen peroxide(H_(2)O_(2))and to explore the potential mechanism underlying the antioxidative stress effect of ***:Cultured RGCs were identified by double immunofluorescent labeling of microtubule-associated protein 2 and *** injury model of H_(2)O_(2)-induced RGCs oxidative stress was established in *** were pretreated with different concentrations of ET(1,5,and 10μmol/L)for 4h,followed by further exposure to H_(2)O_(2)at 1000μmol/*** counting kit 8 and Annexin V/propidium iodide assays were applied to detect the viabilities and apoptosis rates of the RGCs at 12,24,and 48h after H_(2)O_(2)*** levels of nitric oxide,malondialdehyde,and glutathione in culture media were measured at these time *** reverse transcription polymerase chain reaction(qRT-PCR)and Western blot were performed to observe the effects of ET on the messenger RNA and protein expression of inducible nitric oxide synthase(iNOS),nuclear factor erythroid 2-related factor 2(Nrf2),heme oxygenase 1(HO-1),glutathione peroxidase 1 and the level of conjugated acrolein in RGCs at 12,24,and 48h after H_(2)O_(2)stimulation and in the retina at 12h after optic nerve transection(ONT).RESULTS:The applications of 5 and 10μmol/L of ET significantly increased the viability of *** from qRT-PCR indicated a decrease in the expression of iNOS and an increase in the expressions of Nrf2 and HO-1 in ETpretreated RGCs at 12,24 and 48h after H_(2)O_(2)stimulation,as well as in ET-treated retinas at 12h after *** blot analysis revealed a decrease in the expression of iNOS and levels of conjugated acrolein,along with an increase in the expressions of Nrf2 and HO-1 in ET-pretreated RGCs in vitro and ET-treated retinas in ***:ET is a neuroprotective agent in primary cultured RGCs injured by H_(2)O_(2).The effect of ET is dosedependent with the

Melt-cast explosives are the most widely used energetic materials in military composite explosives,researchers have been unremittingly exploring high-energy and insensitive melt-cast *** this work,a series of dinitrophenyl-oxadiazole compounds were designed and *** compounds have an ideal low melting point(80-97℃),good detonation performance(detonation velocity D=6455-6971 m/s,detonation pressure P=18-19 GPa)and extreme insensitive nature(impact sensitivity≥60 J,friction sensitivity>360 N).All these compounds were well characterized by nuclear magnetic resonance,fourier transform infrared spectroscopy,elemental *** 2,3 were unambiguously confirmed by X-ray single crystal diffraction *** a result,their overall properties are superior to traditional melt-cast explosives trinitrotoluene(TNT)and dinitroanisole(DNAN)which may have excellent potential applications in insensitive melt-cast explosives.

Background:Shenzao dripping pill(SZDP)is empirically prescribed for treating cardiac ***,there is a lack of comprehensive knowledge regarding the underlying mechanisms contributing to its therapeutic *** objective of this study is to investigate the underlying mechanism of SZDP against chronic myocardial ischemia(CMI)in a rat ***:In this study,we utilized electrocardiographic and echocardiographic detection along with pathological tissue analysis to evaluate the efficacy of *** integration of network pharmacology and metabolomics was conducted to investigate the *** docking and molecular dynamics simulations were used to validate the binding energy between the compounds of SZDP and the associated ***:The results showed that SZDP was able to improve T wave voltage,reverse CMI abnormalities in ejection fraction and fractional shortening,and restore histopathological heart *** results indicated that disturbances of metabolic profile in CMI rats were partly corrected after SZDP administration,mainly affecting purine metabolism.13-Docosenamide may be the potential metabolic biomarker of the therapeutic application of SZDP for *** network pharmacology and metabolomics,thiopurine S-methyltransferase(TPMT),xanthine dehydrogenase/oxidase(XDH),bifunctional purine biosynthesis protein ATIC(ATIC),and cytochrome p4501A1(CYP1A1)were identified as possible targets of SZDP to exert therapeutic effects by enhancing the metabolic levels of L-Tryptophan,Deoxyribose 1-phosphate and Phosphoribosyl ***:SZDP has a therapeutic effect on CMI by regulating metabolite levels,acting on the targets of TMPT,XDH,ATIC,and CYP1A1,and reducing cardiomyocyte injury and myocardial fibrosis.
