The influence of exendin and GLP-1 on VCAM-1 and ICAM-1 production in endothelium stimulated by TNF-α and glycated albumin

A growing body of evidence indicates that incretins may have pleiotropic beneficial effects beyond lowering glucose blood concentration. The effect of GLP-1 and exendin-4 on coronary arteries endothelium in diabetic and obese individuals has been studied widely. TNF-a is one of adipocytokines. The aim of our study was to evaluate the influence of glycated albumin (GlyAlb;100;500 and 1000 mg/L) and proinflammatory cytokine, TNF-α (2.5 and 10 ng/mL), on expression of ICAM-1 and VCAM-1 in cultured human endothelial cells derived from coronary arteries. The next goal of the study was to evaluate the influence of GLP-1 (10 nM and 100 nM) and its analogue, exendin-4 (1 nM and 10 nM), on the expression of ICAM-1 and VCAM-1 in these cell line. TNF-a statistically significantly increased VCAM-1 production by endothelial cells, whereas GlyAlb statistically significantly augmented the expression of both tested adhesion mole- cules. Exendin-4 and GLP-1 statistically significantly reduced the expression of VCAM-1 in endothelial cells stimulated by GlyAlb in dose-de- pendent manner. When TNF-a was used as the stimulant only exendin-4 in the concentration of 10 nM statistically significantly reduced the expression of VCAM-1. Studied incretins in their both concentrations statistically significantly reduced the expression of ICAM-1 in endothelial cells stimulated by GlyAlb. The influence of TNF- a on the expression of ICAM-1 was statistically significantly reduced by both concentrations of exendin-4 but only by the higher concentration of GLP-1. The results of our present study indicate that incretins may present a group of agents developing pleiotropic effects beyond the reduction of blood glucose concentration. Their vaso-protective and cardioprotective action may be of importance in diabetic and obese individuals.

Insect CAPA neuropeptidesare considered to affect water and ion balance by mediating the physiological metabolism activities of the Malpighian *** previous studies,the CAPA-PK analogue 1895(2Abf-Suc-FGPRLamide)was reported to decrease aphid fitness when administered through microinjection or via topical ***,a further statistically significant decrease in the fitness of aphids and an increased mortality could not be established with pairwise combinations of 1895 with other CAPA *** this study,we assessed the topical application of new combinations of 1895 with five CAPA-PVK analogues on the fitness of *** found that 1895 and CAPA-PVK analogue 2315(ASG-[β3L]-VAFPRVamide)was statistically the most effective combination to control the peach potato aphid Myzus persicae nymphs via topical application,leading to 72%***,the combination(1895+2315)was evaluated against a selection of beneficial insects,that is,a pollinator(Bombus terrestris)and three natural enemies(Chrysoperla carnea,Nasonia vitripennis,and Adalia bipunctata).We found no significant influence on food intake,weight increase,and survival for the pollinator and the three representative natural *** results could facilitate to further establish and generate CAPA analogues as alternatives to broad spectrum and less friendly insecticides.

Chromium Content in the Human Hip Joint Tissues

Objective Chromium has many important functions in the human body. For the osseous tissue, its role has not been clearly defined. This study was aimed at determining chromium content in hip joint tissues. Methods A total of 91 hip joint samples were taken in this study, including 66 from females and 25 from males. The sample tissues were separated according to their anatomical parts. The chromium content was determined by the AAS method. The statistical analysis was performed with U Mann-Whitney＇s non-parametric test, P〈0.05. Results The overall chromium content in tissues of the hip joint in the study subjects was as follows： 5.73 μg/g in the articular cartilage, 5.33 μ/g in the cortical bone, 27.86 μ/g in the cancellous bone, 5.95 μg/8 in the fragment of the cancellous bone from the intertrochanteric region, and 2.28 μ/g in the joint capsule. The chromium contents were observed in 2 group patients, it was 7.04 μ/g in people with osteoarthritis and 22.59 μ/g in people with fractures. Conclusion The observed chromium content was highest in the cancellous bone and the lowest in the joint capsule. Chromium content was significantly different between the people with hip joint osteoarthritis and the people with femoral neck fractures.

Mycelium Growth and Yielding of Black Poplar Mushroom-Agrocybe aegerita （Brig.） Sing. on Different Substrates

Mycelial growth and yield of two strains ofA. aegerita on different substrates was investigated. Mycelial growth on agar media （PDA, standard, wheat, MEA, CYM, PCA, sawdust extracts： alder and mixture of beech and birch （1：1）） and sawdust substrates （birch, beech, oak, maple, alder） was investigated. Petri dishes （Ф 9 cm） for agar media and biological tubes （18 cm long and Ф 2.5 cm） for sawdust substrates were used. Yield and morphological features were studied on birch, beech and mixture of beech and alder （1：1） sawdust substrates. The temperature of inoculation for agar media and sawdust substrates was 25 ℃. For yielding, when mycelium has completely overgrown the substrate the temperature was decreased to 15-17 ℃ to initiate primordia formation. The cultivation was enlightened 10 h/d with daylight lamps （500 Ix）. One crop was harvested after five weeks. The carpophores of black poplar mushrooms were picked up in clusters. There was no statistically important difference between the mycelial growths of the investigated strains. The best growing agar media were PDA （7.3 cm）, MEA （7.2 cm） and wheat （7.1 cm）, both strains showed the slowest mycelium growth on CYM （5.2 cm）. The mycelial growth on sawdust was the best on the beech （7.1 cm） and birch （6.8 cm） sawdust. The best substrate for cultivation ofA. aegerita was mixture of beech and alder sawdust （39.5 g/100 substrate DM）. The dry yield of carpophores were the highest on beech and alder substrate （3.2 g/100 substrate DM）, but dry matter content was the highest on beech sawdust （15.7%）. The heaviest carpophores were harvested from birch sawdust （3.7 g）; the biggest caps from beech and alder sawdust （3.3 cm）. There were no statistically important differences between the mycelium growth and yielding between both investigated strains.

Phytoremediation Potential of Selected Energetic Plants （Miscanthus giganteus L. and Phalaris arundinacea L.） in Dependence on Fertilization

This paper present the influence of fertilization in cultivation Miscanthus giganteus L. and Phalaris arundinacea L. in the process of phytoremediation of Cd, Ni and Zn. Research show that after first year of vegetation, Cd （0.344 mg·kg^-1 dry mass （d.m.）） and Zn （29.47 mg·kg^-1 d.m.） were the most absorbed metals by the biomass ofMiscanthus giganteus L. grown on soil fertilized with municipalsludge （OW）. However, in the case of the reed canary grass, the correlation can be attributed to the Ni （3.76 mg·kg^-1 d.m.） and Zn （122.6 mg·kg^-1 d.m.）. In turn, after the second year of vegetation, all of the tested metals are characterized by a lower absorption as compared to the first year. In the case of reed canary grass, Cd and Zn content in the biomass after the second year of vegetation was similar to the control, while the Ni concentration was lower in relation to the control. In the case of Miscanthus giganteus L., it was observed that the concentration of Ni and Zn in the biomass after the second year of vegetation was similar to the control, while the Cd content varied in dependence on the applied fertilizer.

Pseudoelastic effect in autoparametric non-ideal vibrating system with SMA spring

In this paper a three degrees of freedom autoparametric system with limited power supply is investigated numerically. The system consists of the body, which is hung on a spring and a damper, and two pendulums connected by shape memory alloy （SMA） spring. Shape memory alloys have ability to change their material properties with temperature. A polynomial constitutive model is assumed to describe the behavior of the SMA spring. The non-ideal source of power adds one degree of freedom, so the system has four degrees of freedom. The equations of motion have been solved numerically and pseudoelastic effects associated with the martensitic phase transformation are studied. It is shown that in this type system one mode of vibrations might excite or damp another mode, and that except different kinds of periodic vibrations there may also appear chaotic vibrations. For the identification of the responses of the system＇s various techniques, including chaos techniques such as bifurcation diagrams and time histories, power spectral densities, Poincare maps and exponents of Lyapunov may be used.

Maturation, proliferation and apoptosis of seminal tubule cells at puberty after administration of estradiol, follicle stimulating hormone or both

Aim： To assess proliferative and apoptotic potential of the seminiferous epithelium cells in relation to Sertoli cell maturation in newborn rats under the influence of estradiol, follicle stimulating hormone （FSH） or both agents given together. Methods： From postnatal day （PND） 5 to 15 male rats were daily injected with 12.5 μg of 1713-estradiol benzoate （EB） or 7.5 IU of human purified FSH （hFSH） or EB ＋ hFSH or solvents （control）. On postnatal day 16, autopsy was performed. Sertoli cell maturation/function was assessed by morphometry. Proliferation of the seminiferous epithelium cells was quantitatively evaluated using immunohistochemical labeling against proliferating cell nuclear antigen and apoptosis using the TUN-EL method. Results： Although EB inhibited Sertoli cell maturation and hFSH was not effective, a pronounced acceleration of Sertoli cell maturation occurred after EB ＋ hFSH. Whereas hFSH stimulated Sertoli cell proliferation, EB or EB ＋ hFSH inhibited Sertoli cell proliferation. All treatments significantly stimulated germ cell proliferation. Apoptosis of Sertoli cells increased 9-fold and germ cells 2-fold after EB, and was not affected by hFSH but was inhibited after EB ＋ hFSH. Conclusion： At puberty, estradiol inhibits Sertoli cell maturation, increases Sertoli and germ cell apoptosis but stimulates germ cell proliferation. Estradiol in synergism with FSH, but neither of the hormones alone, accelerates Sertoli cell maturation associated with an increase in germ cell survival. Estradiol and FSH cooperate to induce seminal tubule maturation and trigger first spermatogenesis.

Influence of Dietary Administration of β-hydroxy-β-methylbutyrate （HMB） on the Innate Immunity and Resistance against Bacterial Infections in Pikeperch （Sander lucioperca）

The present study examined the influence of leucine metabolite β-hydroxy-β-methylbutyrate （HMB） on the nonspecific cellular and humoral defence mechanisms and protection against Yersinia ruckeri and Aeromonas salmonicida in juvenile pikeperch （Sander lucioperca）. The fish was fed with a commercial trout pellets, containing either 0 （control group） or 50 mg HMB kg^-1 body weight day^-1 （HMB fed group）. After four weeks feeding, 20 pikeperch from each group were anaesthetised and blood and pronephros samples were taken. The levels of the following immunological parameters were measured： respiratory burst activity of phagocytes, potential killing activity of phagocytes, lymphocytes proliferation stimulated by concanavaline A or lipopolisaccharide, lysozyme activity in serum, ceruloplasmin activity in serum, total protein in serum and total immunoglobulin （Ig） level in serum. A disease challenge test using Yersinia ruckeri or Aeromonas salmonicida was conducted after 4 weeks of feeding. The levels of all immunological parameters excluding ceruloplasmin activity and total protein in serum were statistically significantly higher （P 〈 0.05） in the HMB treated group compared to the control group. Feeding with HMB has led to a significant decrease in the mortality after the challenge trial with Y. ruckeri （30% lower mortality than in the control group） and with theA. salmonicida （40% lower mortality than in the control group）. The current study strongly suggests that feeding pikeperch with HMB in dose 50 mg kg^-1 body weight day^-1 may improve the innate immunity and decrease the mortality rates after experimental infection with pathogenic bacteria Y. ruckeri and A. salmonicida.

Condensing Heat Exchanger

The paper shows a method of designing a heat exchanger recovering heat from the condensation of water vapour contained in flue gases. A heat exchanger condenses water vapour and SO2 （sulphur dioxide） in the presence of inert gases （CO2, CO, N2, O2） contained in flue gases. A mathematical model and a sample design of a heat exchanger were presented. The heat exchange is capable of recovering from a dozen or so to several dozen percent of heat from flue gases escaping into the atmosphere. A second advantage of the heat exchanger is the possibility to reduce the emissions of SO2 considerably. Depending on the parameters, it can be even a sevenfold reduction in the emissions. The main mathematical tool used for designing the condensing heat exchanger is the Colburn-Hougen method. The authors omitted that part of the method which requires iterative calculations. The Mollier diagram was used instead.
