Isolation and characterization of human polyreactive pneumococcal polysaccharide antibodies

Natural antibodies serve as the body’s first line of defense against pneumococcal challenge. Polyreactive human pneumococcal polysaccharide IgG antibodies have not been extensively studied. We analyzed human polyreactive antibodies that bind multiple pneumococcal polysaccharides, including PPS14 and PPS23F. These antibodies were isolated from single pneumococcal polysaccharide specific B cells allowing for the analysis of human immunoglobulins with natively paired variable regions. Although isolated individually, these antibodies demonstrated similar characteristics. most antibodies possessed a variable light chain with a CDR3 length made up of nine amino acids and relatively high number of flexible amino acids in combined VH/VL. While these antibodies were polyreactive and structurally alike, kinetic analysis revealed unique KD values. Variable chains are responsible for antigen recognition whereas antibody fine specificity is affected by isotype structure. To investigate the contribution of the constant region of these isotypes and their effect on antibody avidity to pneumococcal polysaccharide, the polyreactive variable regions were expressed as IgG1 or IgG2 and subjected to kinetic analysis. The IgG1 antibodies uniformly had a stronger avidity to PPS14 and PPS23F compared to IgG2. To further document the importance of the constant region in antibody avidity and fine specificity, analysis of antibody F(ab)’2 fragment binding to PPS14 and PPS23F resulted in similar KD values. These studies suggest that antigen recognition by polyreactive antibodies is determined by a conserved variable light chain CDR3 length and longer, more flexible variable heavy CDR3s when compared to pneumococcal polysaccha-ride-specific sequences while differences in specific avidities are modulated by antibody isotype.

Assessment of UAV Based Vegetation Indices for Nitrogen Concentration Estimation in Spring Wheat

Unmanned Aerial Vehicles (UAVs) have become increasingly popular in recent years for agricultural research. High spatial and temporal resolution images obtained with UAVs are ideal for many applications in agriculture. The objective of this study was to evaluate the performance of vegetation indices (VIs) derived from UAV images for quantification of plant nitrogen (N) concentration of spring wheat, a major cereal crop worldwide. This study was conducted at three locations in Idaho, United States. A quadcopter UAV equipped with a red edge multispectral sensor was used to collect images during the 2016 growing season. Flight missions were successfully carried out at Feekes 5 and Feekes 10 growth stages of spring wheat. Plant samples were collected on the same days as UAV image data acquisition and were transferred to lab for N concentration analysis. Different VIs including Normalized Difference Vegetative Index (NDVI), Red Edge Normalized Difference Vegetation Index (NDVIred edge), Enhanced Vegetation Index 2 (EVI2), Red Edge Simple Ratio (SRred edge), Green Chlorophyll Index (CIgreen), Red Edge Chlorophyll Index (CIred edge), medium Resolution Imaging Spectrometer (mERIS) Terrestrial Chlorophyll Index (mTCI) and Red Edge Triangular Vegetation Index (core only) (RTVIcore) were calculated for each flight event. At Feekes 5 growth stage, red edge and green based VIs showed higher correlation with plant N concentration compare to the red based VIs. At Feekes 10 growth stage, all calculated VIs showed high correlation with plant N concentration. Empirical relationships between VIs and plant N concentration were cross validated using test data sets for each growth stage. At Feekes 5, the plant N concentration estimated based on NDVIred edge showed one to one correlation with measured N concentration. The Usage of Saccharomyces cerevisiae in microbial Fuel Cell System for Electricity Energy Production

A laboratory-scale mFC （microbial fuel cell） was studied in order to determine the capacity and the efficiency of electricity production in microbial fuel cell system by using microorganisms. The medium used is an isolated culture ofSaccharomces cereviciae. A number of media has been evaluated to provide the best growth phase for Saccharomces cereviciae using optical density method with spectrophotometer at a wavelength of 550 nm. Nation 117, Lyntech, USA is used as proton exchange membrane and graphites with surface area 1.46 x 10.3 m2 are used as cathodes. Cathode chamber is fulfilled with a mixture of electrolyte compounds K3Fe（CN）6 and a buffer solution. microbes that have been cultured are applied into anode chamber. A number of factors need to be controlled so that microbes can generate electrical energy efficiently, such as by measuring the degree of acidity and DO value in the anode compartment. In this research, the usage of riboflavin as a mediator is also investigated, while the usage of riboflavin increase production efficiency 53.90% compare to mFC without riboflavin as a mediator.

目的建立IS6110限制性片段多态性分析（IS6110一RFLP）标准方法并评价该方法的分型能力。方法采用核酸提取、PCR、限制性内切酶分析、Southern杂交、琼脂糖凝胶电泳等技术，结合Gel—Proanalyzer3．1和BioNumerics（Version5．0）软件，对78株结核分枝杆菌插入序列IS6110-RFLP进行分析。结果确定标准化的IS6110-RFLP技术，包括核酸提取、PCR、限制性内切酶分析、Southern杂交、琼脂糖凝胶电泳等实验步骤及标化参数的相关数据分析软件的使用；采用该技术，将78株结核分枝杆菌分为75个不同的基因型，分别归属于11个基因簇，其中有52株归属于同一个基因簇，占菌株总数的66．7％（52／78）。结论建立标准化的IS6110-RFLP技术方案，该方法具有很强的基因分型和株水平鉴定能力，可用于结核病的病原学监测。

many gastrointestinal complaints and motility disorders are described in patients suffering from different eating disorders. These have a negative impact on refeeding. This report evaluated–on admission-gastric emptying of a standardized solid meal with 13C octanoic acid breath test in anorexic adolescents. The results were compared with age and gender matched controls. Gastric emptying data were related to a subjective symptom score. 21 Anorexic girls and 3 boys (mean age: 15.6 y ± 1.3 y), body mass index (mean 15.6 kg/m2 ± 1.6 kg/m2), weight loss (mean: 22.2% ± 10.7%) were studied. T? of the gastric emptying time exceeded the P95 in 14/24 (58%) patients, classified as having delayed gastric emptying (DGE). One patient (4%) had a T? exceeding P75 and was classified as slow (SGE), whereas 9/24 (38%) patients had normal gastric emptying (NGE). In comparison to NGE, subjects with DGE had significantly higher symptom scores (p = 0.01) and more weight loss. The DGE and SGE patients lost 25.29% and 21.38% of weight respectively, whereas a weight loss of 17.64% was found in anorexics with NGE. In conclusion, gastric emptying is delayed in patients with anorexia. This is associated with more significant gastrointestinal symptoms and higher weight loss.

Forgiveness education in fibromyalgia: A qualitative inquiry

The purpose of this qualitative study was to assess perspectives on the acceptability and the potential applicability of a forgiveness education in patients with fibromyalgia. The concept and tools of forgiveness were presented to thirteen women (age: 40 - 54 years) with a previous diagnosis of fibromyalgia. Subjects participated in 1 of 2 focus groups following a 90-minute education session in which forgiveness was presented as an emotion-focused coping strategy to deal with interpersonal stressors. Qualitative assessment of focus group discussions reveals 3 themes: 1) forgiveness is healthy and reduces pain, 2) forgiveness is within a patient’s personal control, and 3) forgiveness education is similar to other types of patient education and is well received. Our results suggest that forgiveness education is acceptable and feasible in patients with fibromyalgia. This justifies further exploration of forgiveness as an emotion-focused self- management strategy to decrease psychological distress.

获取骨骼和牙釉质中的锶同位素比值来研究史前人群中的非本地个体,已经在全球范围内逐渐开展。在美国中西部地区开展的考古研究逐渐将人口流动视为影响文化交流变迁的一个重要因素。由于美国中西部地区在地质上的差异较小,因而相应的锶的多样性可能也比较低,这对于利用锶同位素划定这一区域非本地人的灵敏度是一个问题。利用考古遗址的动物遗存,我们探索了这一区域锶同位素比值的多样性。研究结果显示锶同位素比值的多样性是可以被测量的,并且这一结果证明了利用锶同位素分析解决美国中西部地区人口来源以及迁徒问题的可能性。
