Neuronal protein-tyrosine phosphatase 1B hinders sensory-motor functional recovery and causes affective disorders in two different focal ischemic stroke models

Ischemic brain injury causes neuronal death and *** activates protein-tyrosine phosphatase 1B(PTP1B).Here,we tested the significance of PTP1B activation in glutamatergic projection neurons on functional recovery in two models of stroke:by photothrombosis,focal ischemic lesions were induced in the sensorimotor cortex(Sm stroke)or in the peri-prefrontal cortex(peri-PFC stroke).Elevated PTP1B expression was detected at 4 days and up to 6 weeks after *** ablation of PTP1B in neurons of neuronal knockout(NKO)mice had no effect on the volume or resorption of ischemic lesions,markedly different effects on functional recovery were *** stroke caused severe sensory and motor deficits(adhesive removal test)in wild type and NKO mice at 4 days,but NKO mice showed drastically improved sensory and motor functional recovery at 8 *** addition,peri-PFC stroke caused anxiety-like behaviors(elevated plus maze and open field tests),and depression-like behaviors(forced swimming and tail suspension tests)in wild type mice 9 and 28 days after stroke,respectively,with minimal effect on sensory and motor ***-PFC stroke-induced affective disorders were associated with fewer active(FosB+)neurons in the PFC and nucleus accumbens but more FosB+neurons in the basolateral amygdala,compared to sham-operated *** contrast,mice with neuronal ablation of PTP1B were protected from anxiety-like and depression-like behaviors and showed no change in FosB+neurons after peri-PFC *** together,our study identifies neuronal PTP1B as a key component that hinders sensory and motor functional recovery and also contributes to the development of anxiety-like and depression-like behaviors after ***,PTP1B may represent a novel therapeutic target to improve stroke *** procedures for animal use were approved by the Animal Care and Use Committee of the University of Ottawa Animal Care and Veterinary Service(protocol 1806)on July 27,2018.

The extent of male mate choosiness is driven by a trade-off between various environmental factors associated with the costs of mate acquisition,quality assessment and opportunity *** knowledge about natural variation in male mate choosiness across different populations of the same species,however,remains *** this study,we compared male mate choosiness across 10 natural populations of the freshwater amphipod Gammarus roeselii(Gervais 1835),a species with overall high male mating investments,and evaluated the relative influence of population density and sex ratio(both affecting mate availability)on male mate *** investigated amplexus establishment after separating mating pairs and presenting focal males with a novel,size-matched female from the same *** analysis revealed considerable effects of sex ratio and(to a lesser extent)population density on time until amplexus establishment(choosiness).male amphipods are able to perceive variable social conditions(e.g.,sex ratio)and modify their mating strategy accordingly:We found choosiness to be reduced in increasingly male-biased populations,whereas selectivity increases when sex ratio becomes female *** this,our study expands our limited knowledge on natural variations in male mate choosiness and illustrates the importance of sex ratio(i.e.,level of competition)for male mating decisions in natural *** for variation in sex ratios,therefore,allows envisioning a distinctive variation of choosiness in natural populations and highlights the importance of considering social background information in future behavioral studies.

Cathodic activated stainless steel mesh as a highly active electrocatalyst for the oxygen evolution reaction with self-healing possibility

The oxygen evolution reaction(OER)represents one of the major bottlenecks for broad-based applications of many clean energy storage/conversion *** key to solving this problem lies in developing high-performing,cost effective and stable catalysts for the ***,we demonstrate that ubiquitous stainless steel mesh(SSm)materials activated by a facile cathodization treatment can be employed as a high performing OER catalyst,as showcased by the impressively low overpotentials of 275 and 319 mV to reach the benchmark current densities of 10 and 100 mA cm^−2(1.0 m KOH),*** SSm also exhibits excellent performance in a two-electrode water electrolyzer,which requires a low cell voltage of 1.58 at 10 mA cm^−2 and outperforms many of water electrolyzers using earth-abundant OER ***,cathodized SSm with minor performance degradation after the stability test can also be readily healed by subjecting it to an additional cathodization *** is disclosed that the superior performance of cathodized SSms stems from the surface enrichment of OER active Ni(oxy)hydroxide,facile gas-bubble removal and transportation over the unique mesh-structured surfaces,while the abundant reservoir of nickel in the bulk allows healing of the catalyst by a facile cathodization.

Anti-Saccharomyces cerevisiae antibody titers are stable over time in Crohn's patients and are not inducible in murine models of colitis

AIm： To investigate ASCA production over time in CD and murine colitis in order to further our understanding of their etiology. mATERIALS AND mETHODS： Sixty-six CD patients were compared to ulcerative colitis （UC） and irritable bowel syndrome patients with respect to ASCA production as measured by ELISA. ASCA IgG or IgA positivity as well as change in titers over a period of up to 3 years （△tgG/A） was correlated with clinical parameters such as CD activity index （CDm） and C-reactive protein levels （CRP）. moreover, two murine models of colitis （DSS and IL-10 knock out） were compared to control animals with respect to ASCA titers after oral yeast exposure. RESULTS： ASCA IgG and IgA titers are stable over time in CD and non-CD patients. Fistular disease was associated with a higher rate of ASCA IgA positivity （P = 0.014）. Ileal disease was found to have a significant influence on the △tgG of ASCA （P = 0.032）. There was no correlation found between ASCA positivity or △tgG/A and clinical parameters of CD： CDAI and CRP. In mice, neither healthy animals nor animals with DSS-induced or spontaneous colitis exhibited a marked increase in ASCA titers after high-dose yeast exposure. On the other hand, mice immunized intraperitoneally with mannan plus adjuvant showed a marked and significant increase in ASCA titers compared to adjuvant-only immunized controls （P = 0.014）. CONCLUSION： The propensity to produce ASCA in a subgroup of CD patients is largely genetically predetermined as evidenced by their stability and lack of correlation with clinical disease activity parameters. Furthermore, in animal models of colitis, mere oral exposure of mice to yeast does not lead to the induction of marked ASCA titers irrespective of concomitant colonic inflammation. Hence, environment may play only a minor role in inducing ASCA.

Light metal compound casting

‘Compound casting’simplifies joining processes by directly casting a metallic melt onto a solid metal substrate. A continuously metallurgic transition is very important for industrial applications, such as joint structures of spaceframe constructions in transport industry. In this project, ‘compound casting’ of light metals is investigated, aiming at weight-saving. The substrate used is a wrought aluminium alloy of type AA5xxx, containing magnesium as main alloying element. The melts are aluminium alloys, containing various alloying elements (Cu, Si, Zn), and magnesium. By replacing the natural oxygen layer with a zinc layer, the inherent wetting difficulties were avoided, and compounds with flawless interfaces were successfully produced (no contraction defects, cracks or oxides). Electron microscopy and EDX investigations as well as optical micrographs of the interfacial areas revealed their continu- ously metallic constitution. Diffusion of alloying elements leads to heat-treatable microstructures in the vicinity of the joining interfaces in Al-Al couples. This permits significant variability of mechanical properties. Without significantly cutting down on wettability, the formation of low-melting intermetallic phases (Al3mg2 and Al12mg17 ImPs) at the interface of Al-mg couples was avoided by applying a protective coating to the substrate.

Biology and control of the fruit borer, Conopomorpha sinensis Bradley on litchi （Litchi chinensis Sonn.） in northern Thailand

Fruit and leaf shoot of litchi （Litchi chinensis） infestation by naturally occurring populations of the fruit bprer, Conopomorpha sinensis larvae and C. sinensis parasitization rates were determined in litchi orchards in northern Thailand at high （ca. 1 400 m ASL） and low （ca. 800 m ASL） elevations with different, non-overlapping fruiting seasons. In addition, spray applications of Bacillus thuringiensis var. aizawai, the natural compound spinosad and the chemical pesticide imidacloprid were conducted in the field to test the effects of these agents on C. sinensis fruit infestation rate. Fruit infestation rate and fruit growth, studied in a low elevation orchard, were sigmoidal and showed a highly significant positive correlation （P ≤ 0.01）. Leaf shoot infestation rate in the low elevation orchard decreased in the course of the fruiting season （march until may）, but increased in the high elevation orchard, where no fruits were present within the same period of time. Together, these results indicate that females of C. sinensis clearly prefer fruits over shoots for oviposition. If no fruits are available, which was the case in the high elevation orchard during that time, they are constrained to lay their eggs on shoots. The parasitization rate of C. sinensis larvae in untreated fruits was 54.3%. The parasitoid species were Chelonus chailini （90.8%） and Phanerotoma sp. （9.2%）. In the leaf shoots, C. sinensis was found to be parasitized by Phanerotoma sp. and Apanteles briareus. The applications of the different active agents had no significant effect on C. sinensis fruit infestation compared to the untreated control. Besides the cryptic life of the larvae, this is most probably explained by the fact that C. sinensis has several generations during the fruiting season. Because of the high parasitization rates recorded, an alternative and more effective control measure than the application of pesticides might be the augmentative release of parasitoids.

Faecal pyruvate kinase isoenzyme type m2 for colorectal cancer screening:A meta-analysis

AIm:To present a critical discussion of the efficacy of the faecal pyruvate kinase isoenzyme type m2(faecal m2-PK) test for colorectal cancer(CRC) screening based on the currently available ***:A literature search in Pubmed and Embase was conducted using the following search terms:fecal Tumor m2-PK,faecal Tumour m2-PK,fecal m2-PK,faecal m2-PK,fecal pyruvate kinase,faecal pyruvate kinase,pyruvate kinase stool and m2-PK ***:Stool samples from 704 patients with CRC and from 11 412 healthy subjects have been investigated for faecal m2-PK concentrations in seventeen independent *** mean faecal m2-PK sensitivity was 80.3%;the specificity was 95.2%.Four studies compared faecal m2-PK head-to-head with guaiacbased faecal occult blood test(gFOBT).Faecal m2PK demonstrated a sensitivity of 81.1%,whereas the gFOBT detected only 36.9% of the *** independent studies investigated the sensitivity of faecal m2-PK for adenoma(n = 554),with the following sensitivities:adenoma m in diameter:25%;adenoma > 1 cm:44%;adenoma of unspecified diameter:51%.In a direct comparison with gFOBT of adenoma > 1 cm in diameter,47% tested positive with the faecal m2-PK test,whereas the gFOBT detected only 27%.CONCLUSION:We recommend faecal m2-PK as a routine test for CRC *** m2-PK closes a gap in clinical practice because it detects bleeding and nonbleeding tumors and adenoma with high sensitivity and specificity.

High-performance oxygen permeation membranes:Cobalt-free Ba_(0.975)La_(0.025)Fe_(1-x)Cu_(x)O_(3)-δceramics

A new group of cobalt-free perovskite oxides,Ba_(0.975)La_(0.025)Fe_(1-x)Cu_(x)O_(3-δ)(BLFC,x=0.05-0.15),was designed,characterized and applied as oxygen permeation *** was found that BLFC oxides with Cu doping range of 0.075-0.15 maintain cubic perovskite phase in a wide range of *** Cu introduced at the B-site results in a gradual increase of the electrical conductivity,which is attributed to the denser overlapping of electron clouds of CueO *** increasing Cu content,the oxygen vacancy concentration increases and the oxygen ion migration energy decreases,leading to the highest oxygen permeation flux of 1.59 mL cm^(-2)min^(-1)recorded for Ba_(0.975)La_(0.025)Fe_(0.9)Cu_(0.1)O_(3-δ)1mm thick membrane at 950℃.However,the oxygen permeability decreases with further Cu doping,which may be correspond to a presence of defect ***_(0.975)La_(0.025)Fe_(0.9)Cu_(0.10)O_(3-δ)membrane with 0.7mm thickness delivers stable oxygen permeation flux of 1.57 mL cm^(-2)min^(-1)for 200 h at 900℃.All of the obtained results indicate that the developed BLFC with optimized Cu content(i.e.x=0.1)is a very promising material for usage in oxygen separation applications.

Accuracy and Consistency of Respiratory Inductive Plethysmography for Overnight Tidal Volume measurement

To validate the accuracy and consistency of respiratory inductive plethysmography (RIP) in measuring tidal volume after an overnight sleep, tidal volumes of 18 patients with suspected sleep disordered breathing and 8 normal volunteers were measured simultaneously with RIP (V TRIP ) and with an ultrasonic airflow meter (V TUFm ) before and after an unstrained overnight sleep on supine and lateral decubitus. The bias of the V TRIP was expressed as (V TRIP V TUFm )/ V TUFm ·100 %, limits of agreement between V TRIP and V TUFm was measured by averaged bias ±2 s . Results showed that in normal subjects, the bias of RIP before and after overnight sleep was precise and consistent in both supine (0.7 % and -1.6 %) and lateral decubitus (3.7 % and -0.56 %). In these patients, the bias of RIP before and after sleep in supine also remained small (1.9 % and 1.7 %), but it became larger in lateral decubitus (24.5 % and 20.4 %) and 11.5 % exceeded the limits of agreement observed in the evening. The patients′ body mass indices (BmI) were higher than those of normal subjects (median 34.2 vs. 27.8 kg/m 2). Pooled data showed that the bias of V TRIP in the morning on lateral decubitus but not on supine was correlated to BmI (Spearman R=0.32, n =52, P =0.02). Thus, we were led to conclude that the accuracy of V TRIP overnight was precise and consistent in normal subjects, but the deviation of V TRIP measured on lateral decubitus in patients especially in those with excessive obesity was greater, thus, the method should not be used for quantitative determination.
