Characterization of Episomal Replication of Bovine Papillomavirus Type 1 DNA in Long-Term Virion-Infected Saccharomyces Cerevisiae Culture

We have previously reported that bovine papillomavirus type 1(BPV-1) DNA can replicate its genome and produce infectious virus-like particles in short term virion-infected S. cerevisiae(budding yeast) cultures(zhao and Frazer 2002,Journal of Virology, 76:3359–64 and 76:12265–73). Here, we report the episomal replications of BPV-1 DNA in long term virion-infected S. cerevisiae culture up to 108 days. Episomal replications of the BPV-1 DNA could be divided into three patterns at three stages, early active replication(day 3–16), middle weak replication(day 23–34/45) and late stable replication(day 45–82). Two-dimensional gel electrophoresis analysis and Southern blot hybridization have revealed further that multiple replication intermediates of BPV-1 DNA including linear form, stranded DNA, monomers and higher oligomers were detected in the virion-infected yeast cells over the time course. Higher oligomers shown as covalently closed circular DNAs(cccDNAs) are the most important replication intermediates that serve as the main nuclear transcription template for producing all viral RNAs in the viral life cycle. In this study, the cccDNAs were generated at the early active replication stage with the highest frequencies and then at late stable replication, but they appeared to be suppressed at the middle weak replication. Our data provided a novel insight that BPV-1 genomic DNA could replicate episomally for the long period and produce the key replication intermediates cccDNAs in S. cerevisiae system.

Integrated sensing and communication (ISAC) is a promising technique to increase spectral efficiency and support various emerging applications by sharing the spectrum and hardware between these functionalities. However, the traditional ISAC schemes are highly dependent on the accurate mathematical model and suffer from the challenges of high complexity and poor performance in practical scenarios. Recently, artificial intelligence (AI) has emerged as a viable technique to address these issues due to its powerful learning capabilities, satisfactory generalization capability, fast inference speed, and high adaptability for dynamic environments, facilitating a system design shift from model-driven to data-driven. Intelligent ISAC, which integrates AI into ISAC, has been a hot topic that has attracted many researchers to investigate. In this paper, we provide a comprehensive overview of intelligent ISAC, including its motivation, typical applications, recent trends, and challenges. In particular, we first introduce the basic principle of ISAC, followed by its key techniques. Then, an overview of AI and a comparison between model-based and AI-based methods for ISAC are provided. Furthermore, the typical applications of AI in ISAC and the recent trends for AI-enabled ISAC are reviewed. Finally, the future research issues and challenges of intelligent ISAC are discussed.

Aided by device-to-device(D2D) connections, unmanned aerial vehicle(UAV) can significantly enhance the coverage of wireless communications. In this paper, we consider a data collection system with the assistance of D2D, where two fixed-wing UAVs as aerial base stations cooperatively serve the ground devices. To accommodate more devices, we propose two effective algorithms to establish the multi-hop D2D connections. Then, the user scheduling, UAV trajectory, and transmit power are jointly optimized to maximize the energy efficiency, which is a non-convex problem. Accordingly, we decompose it into three subproblems. The scheduling optimization is first converted into a linear programming. Then, the trajectory design and the transmit power optimization are reformulated as two convex problems by the Dinkelbach method. Finally, an iterative algorithm is proposed to effectively solve the original problem. Simulation results are presented to verify the effectiveness of the proposed scheme.

In this report,we investigated expression of HPV16 wild type E7(Wt-E7) and two codon modified E7 genes(HB1-E7 and HB2-E7) in three cell *** blot and qRT-PCR analysis showed that both wt and HB E7 genes had similar RNA transcription ***,immunoblotting showed that HB1-E7 and HB2-E7 genes produced E7 proteins at least ten-fold higher than Wt-E7 ***16 E7 significantly inhibited expression of pRb that was associated with cell differentiation in primary human keratinocytes and prevented pRb expression in granular keratinocytes and the formation of a cornified envelope in mice injected with EL4-E7 *** also showed that HPV 16 E7 expression substantially upregulated Cyclin D2 and down-regulated cyclin B1 in primary human keratinocyte in vitro and skin epidermis of mice in vivo,leading to development of skin ***,HPV16 E7 protein dramatically inhibited expression of involucrin(keratinocyte differentiation marker),α-spectrin and β-adducin and disrupted the organization of involucrin filaments,spectrin cytoskeleton and the cytoskeletal structure of adducin in keratinocytes in vitro and in epidermis in vivo associated with *** provided evidence that HPV 16 E7 markedly induced expression of phosphorylated β-adducin(Ser662) and translocated the phosphorylated β-adducin(Ser662) to nucleus in keratinocytes,which may be an important event in the pathogenesis of HPV-induced cervical cancer.

Leveraging the interplay between the metal component and the supporting material represents a cornerstone strategy for augmenting electrocatalytic efficiency,e.g.,electrocatalytic CO_(2)reduction reaction(CO_(2)RR).Herein,we employ freestanding porous carbon fibers(PCNF)as an efficacious and stable support for the uniformly distributed SnO_(2)nanoparticles(SnO_(2)PCNF),thereby capitalizing on the synergistic support effect that arises from their strong *** one hand,the interaction between the SnO_(2)nanoparticles and the carbon support optimizes the electronic configuration of the active *** interaction leads to a noteworthy shift of the d-band center toward stronger intermediate adsorption energy,consequently lowering the energy barrier associated with CO_(2)*** a result,the Sn O_(2)PCNF realizes a remarkable CO_(2)RR performance with excellent selectivity towards formate(98.1%).On the other hand,the porous carbon fibers enable the uniform and stable dispersion of SnO_(2)nanoparticles,and this superior porous structure of carbon supports can also facilitate the exposure of the SnO_(2)nanoparticles on the reaction interface to a great ***,adequate contact between active sites,reactants,and electrolytes can significantly increase the metal utilization,eventually bringing forth a remarkable7.09 A/mg mass *** work might provide a useful idea for improving the utilization rate of metals in numerous electrocatalytic reactions.

The effect of rolling schedules on the ridging resistance of ultra-thin ferritic stainless steel(FSS)430 foil was evaluated by studying the microstructure and *** results show that specimens processed with three-pass cold rolling under the reductions of 40%,40%and 31%,respectively,exhibit improved ridging resistance owing to the microstructural refinement and the texture structure optimization.A nearly 40%reduction of ridging height can be achieved using the proposed rolling schedule compared to the other two rolling *** addition,the effect of annealing temperature after cold rolling on the ridging resistance of FSS 430 foil is also found to be crucial,and an optimal annealing temperature of 900℃ is obtained for FSS 430 foil with high ridging ***,the improvement in the ridging resistance of FSS 430 foil can be attributed to the reduction in the fraction of{001}and{114}components by optimization of the rolling and annealing processes.

Cytoplasmic male sterile(CMS)lines play a crucial role in utilization of heterosis in crop ***,the mechanism underlying the manipulation of male sterility in cotton by long non-coding RNA(lncRNA)and brassinosteroids(BRs)remains ***,using an integrative approach combining lncRNA transcriptomic profiles with virus-induced gene silencing experiments,we identify a flower budspecific lncRNA in the maintainer line 2074B,lncRNA67,negatively modulating with male sterility in upland cotton(Gossypium hirsutum).lncRNA67positively regulates cytochrome P274B(GhCYP724B),which acted as an eTM(endogenous target mimic)for *** suppression of GhCYP724B induced symptoms of BR deficiency and male semisterility in upland cotton as well as in tobacco,which resulted from a reduction in the endogenous BR ***724B regulates BRs synthesis by interacting with GhDIM and GhCYP90B,two BRs biosynthesis ***,GhCYP724B suppressed a unique chimeric open reading frame(Aorf27)in 2074A mitochondrial *** expression of Aorf27 in yeast inhibited cellular growth,and over expression of Aorf27 in tobacco showed male ***,the results proved that the miR3367–lncRNA67–GhCYP724B module positively regulates male sterility by modulating BRs *** findings uncovered the function of lncRNA67–GhCYP724B in male sterility,providing a new mechanism for understanding male sterility in upland cotton.
